ABSTRACT

Objective:To analyze the broadband ultrasound attenuation (BUA), speed of sound (SOS), standard deviation of bone density (T-Score) and stiffness index (SI) with bone mineral density (BMD) in elderly female patients with knee osteoarthritis and their diagnostic efficacy of osteoporosis.Methods:Fifty elderly female patients with knee osteoarthritis admitted o Tangshan People′s Hospital from January 2021 to January 2022 were selected as the observation group, and 40 healthy female patients during the same period were selected as the control group. The results of BUA, SI, T-score and SOS in observation group and control group were analyzed and compared. The BUA, SI, T-score and SOS in elderly women with knee osteoarthritis at different ages and with different bone densities were compared, and the diagnostic value of BUA, SI, T-score and SOS in osteoporosis was analyzed by receiver operating characteristic (ROC) curve.Results:The BUA, SI and T-score of observation group were lower than that in the control group, while SOS was higher than that in the control group (all P<0.05). Among elderly female patients with knee osteoarthritis of different ages, the older the age, the lower the BUA, SI and T-score (all P<0.05), while there was no statistical significance in SOS of elderly female patients with knee osteoarthritis of different ages ( P>0.05). In elderly women with knee osteoarthritis with different BMD grades, the BUA, SI, and T-score in the osteoporosis group were lower than those in the osteopenia group and the normal bone group, and the BUA, SI, and T-score in the osteopenia group were lower than those in the normal bone group; the SOS in the osteoporosis group was higher than those in the osteopenia group and the normal bone group, and the SOS in the osteopenia group was higher than those in the normal bone group (all P<0.05). BUA, SOS, T-score and SI had high sensitivity and specificity in the diagnosis of osteoporosis in elderly women with knee osteoarthritis (all P<0.05). Conclusions:BMD in elderly women with knee osteoarthritis is associated with BUA, SI, T-score, and SOS, and has high diagnostic value for osteoporosis.

ABSTRACT

Objective To explore the diagnostic value of DWI after secretin stimulation for the diagnosis of mild chronic pancreatitis (CP). Methods This was a prospective study. Ninety-nine consecutive individuals including 23 healthy volunteers, 11 risk volunteers, 15 mild CP patients, 14 moderate CP patients and 36 severe CP patients underwent secretin DWI and faecal elastase 1(FE-1) testing. The subjects were grouped by Cambridge classification about endoscopic retrograde cholangiography (ERCP), CT and ultrasonography. Secretin stimulated diffusion weighted imaging(S-DWI), the ADCs, time to peak ADCs and FE-1 were performed on all subjects. The changes of pancreatic ADC values were observed before and after the injection of secretin. All ADCs and FE-1 were compared between groups with single factor analysis of variance, and the correlation between ADCs and FE-1 was determined with Pearson analysis. ROC curves were performed to identify the diagnostic efficacy of DWI related measures. Results Eight patients with severe CP were excluded because the significant atrophy of the pancreatic parenchyma prohibited the evaluation of ADC measurement. Ninety-one individuals were divided into five groups including 23 healthy volunteers, 11 risk volunteers, 15 mild CP patients, 14 moderate CP patients and 28 severe CP patients. The mean baseline and peak ADCs were higher in the healthy volunteers than in other groups, with significant differences (P<0.05). There was no ADC peak in severe CP patients. There were significant differences between the mean baseline ADCs and the peak ADCs in the other groups (P<0.05). The mild and moderate CP groups showed a delayed peak. The area under curve (AUC) of the mean baseline and peak ADCs, time to peak ADCs for differentiating mild CP was 0.818, 0.912 and 0.965, respectively. Using 4.67 min as the cutoff value, time to peak ADCs were most accurate for differentiating healthy from risk patients and those with evident pancreatitis, yielding a sensitivity of 80.0%and a specificity of 100.0%. Good correlations between baseline and peak ADCs, time to peak ADCs, and FE-1 were shown(r=0.57, 0.72 and-0.84, P<0.01). Conclusions Using the peak and time to peak ADCs may improve the detection of risk and mild CP. Secretin-enhanced DWI is a noninvasive, convenient and accurate method.

ABSTRACT

Objective To investigate the application of MRI targeting contrast agent ( P-Gd-Probe) for P selectin in evaluating the severity grades of acute pancreatitis (AP).Methods L-arginine solution (100mg/100kg) were intraperitoneally injected for 3 times once an hour to establish AP model in SD rats. Control group was injected with normal saline in a equal volume .All rats were divided into acute edematous pancreatitis (AEP) group (6 h and 12 h after model establishment)and acute necrotizing pancreatitis (ANP) group (24 h and 48 h after model establishment ) based on the pathology .The rats were examined by T 1 WI plain and enhanced scanning at 6 h, 12 h, 24 h and 48 h after injection , and P-Gd-probe was as the enhanced contrast agent.The signal noise ratio ( SNR) of the pancreas were measured before and after enhancement .Then the pancreas tissues were harvested for pathological examination and P selectin expression in pancreatic tissue was detected using immunohistochemical analysis .Results Inflammation was observed in pancreatic tissue at 6h after establishment , and became more serious as the modeling time extended .P selectin expression was increased as pancreatitis inflammation became more serious .The SNR of control group was stable before and after enhancement.The SNR before and after enhancement was 17.22 ±1.35 and 37.38 ±1.66 in AEP group, and 16.29 ±1.39 and 58.18 ±1.03 in ANP group.The SNR after enhancement was higher than that before enhancement, and the differences were significantly different ( t=-49.59 and -86.09, P<0.001 ). Conclusions Monoclonal antibody MRI contrast agent targeting P selectin is helpful in evaluating the severity of AP.

ABSTRACT

Moyamoya disease (MMD) is a chronic progressive cerebrovascular disease with its reason remaining unclear; the disease has different morbidities among different races, with East Asia countries including China having higher incidence. The pathogenesis of MMD is still unclear and the clinical characteristics lack specificity, making it difficult for early diagnosis. Researches have shown that revascularization surgery can slow down the ischemic process, but no sufficient evidence indicates that revascularization surgery can be used for hemorrhagic MMD. Imaging approachs, including morphological and functional imaging, are the main diagnosis, evaluation and follow-up method for cerebrovascular diseases. This review aimed to summarize the application and research on multiple imaging methods for MMD.

ABSTRACT

This study was aimed to investigate the effect of down-regulating the CXCR4 expression on cell cycle and cell apoptosis of human T-ALL Jurkat cells. The CXCR4 specific siRNA plasmid vector was constructed and then transfected into the cultured Jurkat cell line by DMRIE-C. The expression of CXCR4 mRNA was detected by RT-PCR, the cell distribution in cell cycle and cell apoptosis were determined by flow cytometry. The experiments were divided into 3 groups: group A (blank control), group B (non-silencing dsRNA as negative control) and group C (CXCR4 siRNA). The results showed that the expression level of CXCR4 mRNA in Jurkat cells transfected with CXCR4 siRNA (group C) decreased and cell proportion in G(0)/G(1) phase increased as compared with group A (56.9% +/- 1.4% vs 68.3% +/- 2.4% and 35.8% +/- 1.9% vs 18.1% +/- 1.2% respectively) (p < 0.01), cell proportion in G(2)/M and S phase decreased as compared with group A (19.8% +/- 1.7%, 44.4% +/- 2.1% vs 27.2% +/- 1.5%, 54.7% +/- 2.8% respectively) (p < 0.01). The apoptosis rate of Jurkat cells in group C increased as compared with group A (20.9% +/- 2.0% vs 3.13% +/- 0.9% respectively) (p < 0.01), and the comparison between group A and B showed no statistical difference. It is concluded that the CXCR4 specific siRNA can effectively down-regulate the CXCR4 mRNA expression, which induces the cell apoptosis and cell cycle arrest, thereby inhibits the Jurkat cell proliferation.
