ABSTRACT

To evaluate the risks of hair dye exposure, we investigated cellular and molecular effects of Arianor Ebony dye, which is a mixture of azo and anthraquinone dyes, used in the composition of the black color. Cytotoxicity, genotoxicity, and gene expression of relevant molecules of apoptotic and oxidative stress mechanisms were investigated in HepG2 cells exposed to Arianor Ebony. Results showed that the dye did not induce cytotoxicity to exposed cells at a concentration up to 50 µg/mL compared to the negative control. However, genotoxic assays indicated that the dye was able to damage the genetic material at a concentration of 25 µg/mL, with induction factor values of exposed cells two- to five-fold higher than those recorded for the negative control. Moreover, the lowest observed effect concentration was 12.5 µg/mL. For gene expression, relevant changes were observed in cytochrome c and caspase 9, which decreased in cells incubated with the dye in a dose-dependent manner when compared with the negative control. In parallel, the expression of genes for antioxidant enzymes was increased in exposed cells, suggesting the presence of metabolic routes that protect cells against the toxic effect of the dye, avoiding exacerbated cellular death. Results suggested that the dye disrupted cellular homeostasis through mitochondrial dysfunction, which may be hazardous to human health. Thus, further investigations are necessary to deeply understand the mechanisms of action of the dye, considering its toxic potential found in our ex vivo assays.

ABSTRACT

This study aimed to estimate and compare racial inequality in low birth weight (LBW), preterm birth (PTB), and intrauterine growth restriction (IUGR) in two Brazilian birth cohorts. This was a cross-sectional study nested within two birth cohorts in Ribeirão Preto (RP) and São Luís (SL), whose mothers were interviewed from January to December 2010. In all, 7430 (RP) and 4995 (SL) mothers were interviewed. The maternal skin color was the exposure variable. Associations were adjusted for socioeconomic and biological covariates: maternal education, per capita family income, family economic classification, household head occupation, maternal age, parity, marital status, prenatal care, type of delivery, maternal pre-pregnancy BMI, hypertension, hypertension during pregnancy, and smoking during pregnancy collected from questionnaires applied at birth. Statistical analysis was done with the chi-squared test and logistic regression. In RP, newborns from mothers with black skin color had a higher risk of LBW and IUGR, even after adjusting for socioeconomic and biological variables (P<0.001). In SL, skin color was not a risk factor for LBW (P=0.859), PTB (P=0.220), and IUGR (P=0.062), before or after adjustment for socioeconomic and biological variables. The detection of racial inequality in these perinatal outcomes only in the RP cohort after adjustment for socioeconomic and biological factors may be reflecting the existence of racial discrimination in the RP society. In contrast, the greater miscegenation present in São Luís may be reflecting less racial discrimination of black and brown women in this city.

ABSTRACT

The beneficial effects of drugs that act via nicotinic acetylcholine receptors (nAChRs) on Parkinson's disease (PD) symptomatology may explain the negative correlation between cigarette smoking and risk of this neurological condition. Varenicline, an α4β2 nAChR partial agonist approved for smoking cessation treatments, could be valuable for PD treatment. Here, we investigated varenicline effects in a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) PD mouse model. From postnatal day (PN) 90 to PN119, male C57BL/6 mice were exposed daily to varenicline (2 mg/kg) by gavage. After that, MPTP was injected (30 mg/kg, ip) once a day for five days. At PN125, locomotor and anxiety-like effects were assessed with the open field test. At PN126, immobile behavior was assessed with the forced swimming test. At PN127, the frontal cerebral cortex was collected to evaluate dopamine and DOPAC levels. To verify whether varenicline was protective during the MPTP insult, a separate group of MPTP animals received varenicline from PN90 to PN124. MPTP reduced cortical dopamine content and increased dopamine turnover. Those effects were not reversed by varenicline treatment. Interestingly, varenicline reversed the MPTP-induced hyperactivity in the open field. Both maintenance of varenicline treatment during MPTP exposure or its interruption before MPTP exposure elicited similar results. No alterations were observed in anxiety-like behavior or in immobility time. Altogether, these findings suggested that varenicline treatment reduced the MPTP-induced hyperactivity, but did not protect against dopaminergic damage. Based on this partial protective effect, varenicline could exert neuroprotective effects on circuits that control motor activity in PD.

ABSTRACT

Fetuses exposed to alcohol and/or tobacco are at risk for perinatal adversities. However, little is currently known about the association of the separate or concomitant use of alcohol and tobacco with infant motor and cognitive development. Thus, the objective of the present study was to investigate the association between maternal consumption of alcohol and/or tobacco during pregnancy and the motor and cognitive development of children starting from the second year of life. The study included 1006 children of a cohort started during the prenatal period (22-25 weeks of pregnancy), evaluated at birth and reevaluated during the second year of life in 2011/2013. The children were divided into four groups according to the alcohol and/or tobacco consumption reported by their mothers at childbirth: no consumption (NC), separate alcohol consumption (AC), separate tobacco consumption (TC), and concomitant use of both (ACTC). The Bayley Scale of Infant and Toddler Development Third Edition screening tool was used for the assessment of motor and cognitive development. Adjusted Poisson regression models were used to determine the association between groups and delayed development. The results indicated that only the ACTC group showed a higher risk of motor delay, specifically regarding fine motor skills, compared to the NC group (RR=2.81; 95%CI: 1.65; 4.77). Separate alcohol or tobacco consumption was not associated with delayed gross motor or cognitive development. However, the concomitant use of the two substances increased the risk of delayed acquisition of fine motor skills.

ABSTRACT

Feline Infectious Peritonitis (FIP) is a progressive and fatal disease in domestic and wild cats, caused by Feline Infectious Peritonitis Virus (FIPV). The disease is characterized by an immunomediated reaction against the virus in various organs. This work described a case report of fibrinous epicarditis caused by FIPV. A male cat, three years old, died and was received to be necropsied. Grossly, soft, multifocal to coalescing, whitish fibrinous exudate, measuring up the 2 centimeters of thickness, was observed in the epicardium, mostly at the apex of the heart. Microscopically, severe, multifocal to coalescing inflammatory infiltrate was observed in the epicardium, composed mainly by macrophages, plasmocytes and lymphocytes, associated with fibrin deposition. Immunohistochemistry was performed for FIPV and was positive in the areas of inflammation in the epicardium. To the author´s knowledge, this is the second report of epicarditis due to FIPV in a cat. Therefore, epicarditis should be considered a differential diagnosis of cardiac diseases in Feline Medicine.(AU)


A Peritonite Infecciosa Felina (PIF) é uma doença progressiva e fatal de felinos domésticos e selvagens, causada pelo vírus da Peritonite Infecciosa Felina (FIPV). A doença é caracterizada por uma reação imunomediada contra o vírus em vários órgãos. Este trabalho descreveu um relato de caso de epicardite fibrinosa causada pelo FPIV. Um gato macho, com três anos de idade, veio a óbito e foi recebido para necropsia. Macroscopicamente, foi observado exsudato fibrinoso esbranquiçado, friável, multifocal, medindo até 2 centímetros de espessura, no epicárdio, principalmente no ápice cardíaco. Microscopicamente, foi observado no epicárdio um infiltrado inflamatório misto, multifocal, composto por macrófagos, plasmócitos e linfócitos, associado a deposição de fibrina. A imuno-histoquímica foi positiva para o FIPV nas áreas de inflamação no epicárdio. Os autores descrevem o segundo relato de caso na literatura científica de epicardite causada pelo FIPV em um gato. Portanto, a epicardite deve ser considerada como diagnóstico diferencial em doenças cardíacas em Medicina Felina.(AU)

ABSTRACT

Acute toxicity test (LD-50) using toxic shock syndrome toxin (TSST-1) was tested in BALB/c, C57BL/6 and Swiss mice. Animals (n = 10) were intraperitoneally injected with TSST-1 (0.01-10.0µg/mouse) followed 4h later by potentiating dose of lipopolysaccharide (75.0µg of LPS - E. coli O111:B4) and cumulative mortality was recorded over 72h. Control animals received either TSST-1 or LPS alone. The data were submitted to qui-Square test and acute toxicity test was calculated by probit analysis (confidence limits expressed as µg toxin/kg). BALB/c mice was the most sensitive (20.0µg/kg, 95 percent confidence limits: 9.0-92.0) followed by C57BL/6 (38.5µg/kg, 95 percent confidence limits: 9.11- 401.6). Data from Swiss mice was not conclusive, indicating only low sensitivity. Selection of the animal model and standardization of the experiment are fundamental for the development of serum neutralization tests used for final quality control of vaccine production.


A toxicidade aguda (DL-50) da toxina da síndrome do choque tóxico (TSST-1) foi testada em linhagens de camundongos BALB/c, C57BL/6 e Suíça. Os animais (n=10) inoculados intraperitoneal com doses crescentes de toxina (0,01 - 10,0µg/animal) receberam 4h após 75µg de LPS (E. coli O111: B4). A toxicidade aguda (DL50) foi observada por um período de 72h e os dados submetidos ao teste de qui- quadrado. Os resultados e os limites de confiança foram expressos em µg de toxina/kg. A linhagem BALB/c apresentou maior sensibilidade (20µg/kg - limite de confiança a 95 por cento entre 9,0- 92,0), seguida da C57BL/6 (38,5µg/kg - limite de confiança a 95 por cento entre 9,11 - 401,6). A amplitude dos limites de confiança deve-se à natureza da toxina, ao mecanismo de ação, a via de inoculação e ao animal utilizado. A seleção do modelo animal e a padronização do experimento são fundamentais para o desenvolvimento de testes de soro neutralização para fins de controle de qualidade do processo de produção de vacinas.

ABSTRACT

Purified epsilon prototoxin of Clostridium perfringens type D was produced, purified, and detoxified by the stoiechiometric method of non-radioactive iodine incorporation. Different degrees of iodination were perfomed and the toxicity of the derivatives were analysed by in vivo studies. Toxicity decreased inversely to the iodine incorporation. Eletrophoretic analysis showed different levels of stability of samples kept under different temperatures 4ºC, - 20ºC, and -80 ºC. The iodinated prototoxins were stocked for a period of four months.


A prototoxina epsílon de Clostridium perfringens tipo D foi produzida, purificada e destoxificada por estoiquiometria. Diferentes quantidades de iodo foram incorporadas à estrutura protéica da prototoxina e a toxicidade dos derivados foi analisada em estudos in vivo. Verificou-se que a toxicidade diminuiu à medida que os átomos de iodo foram incorporados à prototoxina. A análise eletroforética demonstrou a estabilidade das amostras quando armazenadas a 4ºC, -20ºC e -80ºC. A prototoxina iodada foi estocada por um período de quatro meses.

ABSTRACT

Acute rejection of a transplanted organ is characterized by intense inflammation within the graft. Yet, for many years transplant researchers have overlooked the role of classic mediators of inflammation such as prostaglandins and thromboxane (prostanoids) in alloimmune responses. It has been demonstrated that local production of prostanoids within the allograft is increased during an episode of acute rejection and that these molecules are able to interfere with graft function by modulating vascular tone, capillary permeability, and platelet aggregation. Experimental data also suggest that prostanoids may participate in alloimmune responses by directly modulating T lymphocyte and antigen-presenting cell function. In the present paper, we provide a brief overview of the alloimmune response, of prostanoid biology, and discuss the available evidence for the role of prostaglandin E2 and thromboxane A2 in graft rejection.

ABSTRACT

Avaliou-se o estresse pós-operatório, durante sete dias, em 30 cadelas, aleatoriamente distribuídas em dois grupos de 15 animais submetidos à ovário-histerectomia (OVH) pelas abordagens laparoscópica (grupo I) e aberta (grupo II). Os grupos foram comparados mediante dosagens do cortisol plasmático. Não se observou aumento do cortisol plasmático no pré-operatório imediato (tempo 1) e quando o animal estava sob anestesia geral imediatamente antes do procedimento cirúrgico (tempo 2). Nos dois grupos houve aumento do cortisol apenas no período intra-operatório (tempo 3) e uma hora após o retorno anestésico (tempo 4), sendo maior nas cadelas submetidas à OVH por laparoscopia. No pós-operatório, os níveis de cortisol das cadelas de ambos os grupos apresentaram-se normais e equivalentes aos do pré-operatório. Independente da abordagem utilizada, a resposta ao estresse foi semelhante para os dois grupos.

ABSTRACT

Avaliou-se a evolução clínica pós-operatória de 30 cadelas sem raça definida, durante sete dias, aleatoriamente distribuídas em dois grupos de 15 animais, submetidas à ovário-histerectomia (OVH) pelas abordagens laparoscópica (grupo I) e aberta (grupo II). Avaliaram-se os parâmetros de comportamento, fisiológicos e de complicações na ferida cirúrgica. Foi utilizada uma escala descrita para avaliação da dor e das complicações pós-operatórias. Não foram encontradas diferenças significativas entre os grupos quanto às variáveis: locomoção, postura, interferência na ferida cirúrgica, tensão abdominal, vocalização, apetite, evacuação, freqüências cárdiaca e respiratória e temperatura corporal. Quando as variáveis de comportamento e fisiológicas foram avaliadas em conjunto (escore 1), observou-se maior dor pós-operatória apenas no segundo dia do pós-operatório nas cadelas submetidas à cirurgia aberta. Quando as complicações das feridas cirúrgicas foram avaliadas em conjunto (escore 2), observou-se maior ocorrência dessas nos animais do grupo 1. O escore total (somatória dos escores 1 e 2) mostrou que a recuperação pós-operatória foi semelhante nas duas abordagens estudadas.

ABSTRACT

Avaliou-se, de forma prospectiva, a ovário-histerectomia (OVH) nas abordagens laparoscópica e aberta, mediante a comparação de parâmetros intra-operatórios como tempo cirúrgico, complicações (hemorragia, lesões viscerais e vasculares), dificuldades técnicas e custos. Foram utilizadas 30 cadelas, sem raça definida, com peso entre 6,5 e 19,0kg, aleatoriamente distribuídas em dois grupos de 15 animais. Os animais do grupo I foram submetidos à OVH laparoscópica enquanto os do grupo II à OVH aberta. O tempo cirúrgico médio na abordagem laparoscópica foi significativamente maior (61,6± 14,15 minutos) quando comparada com a aberta (21,13±4,3 minutos), enquanto que o grau de sangramento foi menor na laparoscopia. Foram observadas lesão no baço com agulha de Veress ou trocarte em três animais. Houve ampliação da incisão para retirada do útero e ovários em dois animais do grupo I e em um animal do grupo II. Não houve indicação para conversão da técnica laparoscópica. Concluiu-se que ambas as abordagens mostraram-se seguras e eficientes para realização do procedimento. O tempo cirúrgico foi maior na laparoscópica enquanto que a ocorrência de hemorragia foi menor. A OVH laparoscópica mostrou-se mais onerosa.

ABSTRACT

Estudou-se a ocorrência de Salmonella spp. em forros de caixa de transporte e em órgãos de pintos de corte de um dia, por meio de cultura bacteriológica convencional. Foram avaliadas 378 amostras de 18 lotes pertencentes a três empresas integradoras de frangos de corte do Estado de Goiás. Verificou-se a ocorrência de Salmonella spp. em 55,6 por cento (10/18) dos lotes sendo 10 lotes positivos oriundos de caixa de transporte e um lote de órgãos de pinto de um dia. A freqüência de isolamento de Salmonella spp. nos forros de caixas foi de 11,1 por cento, e nos órgãos de pintos de corte de um dia, 3 por cento. Os resultados sugerem a transmissão vertical do agente. Foram isolados os sorovares Salmonella Enteritidis e Salmonella Heidelberg, o primeiro mais freqüente (92,3 por cento)

ABSTRACT

The objective of the present study was to determine the effects hypoxia and temperature on the cardiovascular and respiratory systems tems and plasma glucose levels of the winter bullfrog Rana calesbeiana. Body temperature was maintained at 10, 15, 25 and 35 degrees Celsius for measments of breathing frequency, heart rate, arterial blood pressure metabolic rate, plasma glucose levels, blood gases and acid-base status. Reducing body temperature from 35 to 10 degrees Celsius decreased (P<0.001) heart rate (bpm) from 64.0 ñ 3,1 (N =5) to 12.5 + 2.5 (N = 6) and blood pressure (mmHg) (P<0.05) from 41.9 ñ 2.1 (N = 5) to 33.1 ñ 2.1 (N = 6), whereas no significant changes were observed under hypoxia. Hypoxia-induced changes in breathing frequency and acid-base status were proportional to body ternperature, being pronounced at 25 degrees Celsius less so at l5 degrees Celsius, and absent at 10 degrees Celsius. Hypoxia at 35 degrees Celsius was lethal. Under normoxia, plasma glucose concentration (mg/dl) decreased (P<0.01 from 53.0 ñ 3.4 (N = 6) to 35.9 ñ 1.7 (N = 6) at body temperatures 35 and 10 degrees Celsius, respectively. Hypoxia had no significant effect on plasma glucose concentration at 10 and 15 degrees Celsius, but a 25 degrees Celsius there was a significant increase under conditions of 3 per cent inspired O2. The arterial PO2 and pH values were similar to those reported in previous studies on non-estivating Rana calesbeiana, but PaCO2 (37.5 ñ 1.9 mmHg, N = 5) was 3-fold higher, indicating increased plasma bicarbonate levels. The estivating bullfrog may be exposed not only to low temperatures but also to hypoxia. These animals show temperature-dependent responses that may be beneficial since during low body temperatures but also to hypoxia. ABSTRACT

Objetivo. Medir a permeabilidade epitelial pulmonar em indivíduos sadios nao-tabagistas, estabelecendo padrao de normalidade, e correlacionar os achados com a idade, sexo, peso, altura e superfície corporal. Material e Método. Foram estudados 24 voluntários sadios nao-tabagistas, utilizando a taxa de depuraçao pulmonar do (99m)Tc-DTPA como índice da permeabilidade epitelial pulmonar. Os dados obtidos foram correlacionados com sexo, idade, peso, altura e superfície corporal. Resultados. As médias encontradas para as taxas de depuraçao pulmonar do (99m)Tc-DTPA foram de 0,82 + 0,19 por cento min(-1), para o pulmao esquerdo, e de 0,88 + 0,23 por cento min(-1), para o pulmao direito. Nao se evidenciou relaçao estatísticamente significante com sexo, idade, peso, altura e superfície corporal. Conclusao. Os valores encontrados estao dentro da variaçao interindividual descrita pela literatura e passam a ser adotados como referenciais em nosso meio.

ABSTRACT

E estudado o comportamento da injecao intravenosa de um corante (azul patente V) na determinacao dos pontos otimos de resseccao de alcas intestinais desvascularizadas. Cinco caes foram submetidos a desvascularizacao de dois segmentos de intestino delgado com comprimento em torno de 15cm. Entre estes preservou-se a irrigacao de um outro de ñ30cm de extensao que funcionava com alca controle. Os pontos otimos de resseccao (POR) das alcas desvascularizadas eram determinados apos a injecao endovenosa periferica do corante. A existencia de destruicao tissular nivel dos POR e respectivamente a 1,2 e 3cm dos referidos pontos (tanto para o lado da alca normal, como para o lado da alca desvascularizada) foi determinada atraves de estudo histopatologico de fragmentos da parede intestinal correspondentes aos mesmos...

Foram estudados, pelo método da taxa de sobrevida acumulada, 174 pacientes que permaneceram por mais de três meses em hemodiálise num Centro Satélite da Baixada Fluminense. Apesar das baixas condiçöes sócio'culturais da populaçäo servida, o desempenho do Centro foi semelhante aos dados nacionais e internacionais. Assim, 58,62% eram brancos, 53,45% do sexo masculino e a média de idade 40,46 anos )(+ ou - 13,55%). Ao final do período de observaçäo (1980 a 1986), 47,70% persistiram em tratamento hemodialítico na Segumed, 22,42% foram transplantados, enquanto 29,89% morreram. A curva de sobrevivência para os indivíduos de baixo risco foi de 91,84% e 78% aos 12, 24 e 36 meses, respectivamente, enquanto para o grupo inteiro foi de 85,73 e 65%. O estudo de diferentes subgrupos aponta hipertensäo maligna, doenças sistêmicas, idade e raça negra como fatores determinantes de pior prognóstico
