ABSTRACT

Interleukin-1 receptor associated kinase 4 (IRAK-4), acting as a serine threonine kinase, is considered as a key signal node for the transduction of IL-1R family and TLRs signal pathway. Studies have found that IRAK-4 has a hand in many signal pathways, involving the inflammatory response of human joints, intestines, liver and nervous system, as well as other autoimmune diseases. It is also one of the causes of drug resistance of some cancer cells. Therefore, IRAK-4 tends to be an effective therapeutic target for inflammatory diseases and cancer. The prospects for the development of drugs in this pathway is to develop novel IRAK-4 small molecule inhibitors and investigate their safety and effectiveness, enrich the clinical treatment of inflammatory and cancer diseases finally. This paper classified and summarized the latest research progress on small molecule inhibitors of IRAK-4 signaling pathway according to structures of the compounds, in order to provide assistances and references for the research and development of related drugs.

ABSTRACT

Object To investigate the changes of neurovascular coupling (NVC) in the contralateral cerebral motor region under the action of repetitive transcranial magnetic stimulation (rTMS) at different frequencies. Methods Some experiments were designed, the electroencephalography (EEG) signals and the near-infrared spectra (NIRS) of the subjects were obtained under different rTMS stimulation frequencies, and EEG power spectrum of each frequency band and NIRS blood oxygen signals to the contralateral movement area were computed and the change characteristics and correlation were analyzed. Results For different stimulation frequencies, the energy of each frequency band of EEG increased after the stimulation, and then decreased rapidly. The oxyhemoglobin (HbO2) concentration was decreased during stimulation, and then raised. The change rate of the two signals varied with different stimulation frequencies, while the change trend was the same. The EEG power spectrum was negatively correlated with the change of HbO2 concentration after stimulation, and it had delayed effect in varying degrees in the period after stimulation. Conclusion The effect of rTMS on the coupling of nerves and vessels in the contralateral cerebral motor region can be reliably revealed by the study of EEG combined with NIRS. This study will be helpful to the development of effective RTMS treatment parameters, and the results are expected to be applied in clinical treatment.

ABSTRACT

Objective To observe the effect of Yijinjing Supporting Pile on the superficial microcirculation blood perfusion units (MBPU) on Du meridian in subjects with yang-deficiency constitution. Methods After the classification and determination of constitution according to the traditional Chinese medicine,19 volunteers (8 males and 11 females) with Yang-deficiency constitution were selected and provided with standard Yijinjing Supporting Pile training for 3 months. The laser Doppler flowmeter system was used to detect the superficial MBPU on the acupoints of Du meridian including Dazhui,Zhiyang and Mingmen,Results Before the training, the average MBPU value of Zhiyang was higher than Mingmen, but lower than Dazhui (P<0.05,P<0.01).After the training, the average MBPU value of all the acupoints increased significantly (P<0.01).Conclusion Yijinjing Supporting Pilecan improve the superficial MBPU of Du meridian in subjects with Yang-deficiency constitution, and regulate the energy metabolism of the Du meridian.
