ABSTRACT

The current study explored the hepatotoxicity among closed-ring genipin, open-ring tautomer of genipin and gardenia blue that generated from genipin and amino acid reaction using HepaRG cells to identify the material basis of genipin-induced hepatotoxicity in vitro. The effects of temperature, pH value and different kinds of amino acids on the chemical structure tautomerism between closed-ring and open-ring tautomer of genipin and the production of gardenia blue were investigated firstly, which aimed to explicit the conditions that could distinguish the closed-ring genipin and its open-ring tautomer, and the conditions generating gardenia blue, which were applied to prepare different kinds of gardenia blue; the CCK-8 kit was employed to analyze the hepatotoxicity of closed-ring genipin, open-ring tautomer of genipin and gardenia blue. From the results, it was found that, the structure transformation from close-ring to open-ring of genipin could be inhibited under the condition with acid environment; being essential groups to generate gardenia blue, the primary amino group and the open-ring tautomer of genipin reacting to generate the dihydropyridine ring was probably the key structure of gardenia blue; the structure characteristics existed apparent distinction at the reactive temperature of 37 ℃ and 80 ℃; compared to the culture condition with pH = 7.4, the concentration of genipin with close-ring in culture medium was significantly increased at pH = 5, but the cell viability did not decreased; the cell toxicity of gardenia blue was apparently lower than open-ring tautomer of genipin, and even some kinds of gardenia blue showed growth promoting effect on HepaRG cells. Here, it was suggested potentially that open-ring tautomer of genipin be the important material basis to induce hepatotoxicity, which could provide a cue and lay a foundation for the elucidation of the underlying mechanism of genipin-induced hepatotoxicity.

ABSTRACT

Objective: To investigate the impact of obstructive sleep apnea syndrome (OSAS) on pregnancy outcomes, especially the relationship between OSAS and hypertensive disorders in pregnancy (HDP). Methods: A total of 228 pregnant women with high risk of OSAS who underwent sleep monitoring during pregnancy in Peking University People's Hospital from January 2021 to April 2022 were collected by reviewing their medical records for retrospective analysis. According to the diagnosis of OSAS, the pregnant women were divided into OSAS group (105 cases) and non-OSAS group (123 cases). The non-parametric Mann-Whitney U test, χ2 test or Fisher's exact test were used to compare the general data and maternal and fetal outcomes between the two groups, and the occurrence of each type of HDP was further compared. Results: (1) Compared with the non-OSAS group, the median pre-pregnancy body mass index (23.6 vs 27.6 kg/m2) and the proportion of snoring [28.9% (33/114) vs 59.2% (61/103)] in the OSAS group were higher, and the differences were both statistically significant (both P<0.001). (2) The incidence of HDP [67.6% (71/105) vs 39.0% (48/123)] and gestational diabetes mellitus [GDM; 40.0% (42/105) vs 26.8% (33/123)] of pregnant women in the OSAS group were higher than those in the non-OSAS group, and the median delivery week was shorter than that in the non-OSAS group (38.4 vs 39.0 weeks). The differences were all statistically significant (all P<0.05). Between-group differences for the delivery way, postpartum hemorrhage, the rate of intensive care unit admission, preterm birth, small for gestational age infants, neonatal asphyxia, the rate of neonatal intensive care unit admission, newborn birth weight and the proportion of umbilical artery blood pH<7.00 were not statistically significant (all P>0.05). (3) Compared with the non-OSAS group, the incidence of chronic hypertension [11.4% (14/123) vs 22.9% (24/105)] and chronic hypertension with superimposed pre-eclampsia [11.4% (14/123) vs 30.5% (32/105)] were higher in the OSAS group, and the differences were both statistically significant (both P<0.01). Conclusion: OSAS is related to HDP (especially chronic hypertension and chronic hypertension with superimposed pre-eclampsia) and GDM, which could provide a practical basis for the screening, diagnosis and treatment of OSAS in pregnant women at high risk.

@#[ [Abstract] ]Objective: To analyze and compare the clinical efficacy and safety of dendritic cell cytokine-induced killer cells (DCCIK) combined with palliative therapy or chemotherapy in the treatment of advanced pancreatic carcinoma. Methods: A retrospective study was carried on 50 patients with advanced pancreatic carcinoma who were hospitalized in department of oncology of Shanxi Dayi Hospital during September 2012 to February 2016. The patients were divided into four groups according to the therapy they received (palliative treatment group, palliative+DC-CIK treatment group, chemotherapy group and chemotherapy+DC-CIK treatment group); the immunological function, quality of life and survival time of patients were analyzed; and the efficacy and safety of DC-CIK cell therapy was also evaluated. Results: The percentages of CD8+ T cells and NKT cells in DC-CIK combined therapy groups were significantly improved compared with that of pre-treatment, and the percentages of CD3+, CD8+, NK, NKT cells were increased compared with control groups (P<0.05). The quality of life of patients was significantly improved (P<0.05), while median PFS and median OS were improved but without statistical significance (P>0.05). Conclusion: Compared with palliative therapy and chemotherapy alone, combined DC-CIK immunotherapy can effectively improve the cellular immunity function and quality of life in patients with advanced pancreatic cancer. However, there was no significant extension in overall survival.

ABSTRACT

<p><b>OBJECTIVE</b>To test the feasibility of correcting conjunctival sac narrowing following orbital implantation using polyester fiber heart patches instead of the skin autograft.</p><p><b>METHODS</b>Twelve patients of conjunctival sac narrowing after orbital implantation (including 3 with orbital implant exposure) admitted in Nanfang Hospital between 2012 and 2016 received surgical correction of the conjunctival sac using polyester fiber heart patches. During the surgery, the central conjunctival sac was opened, the exposed area was covered with suitable polyester fiber heart patches, and the palpebral margin was sutured.</p><p><b>RESULTS</b>Three months after the operation, 10 patients showed improved appearance after implantation of the prosthetic eye. Two patients received a second operation to remove the patches due to graft rejection and infections and skin autograft was implanted for reconstruction of the conjunctival sac.</p><p><b>CONCLUSION</b>Polyester fiber heart patches are ideal materials for repairing Conjunctival sac narrowing and orbital implant exposure, but this approach is not suitable in cases of severe narrowing or occlusion of the conjunctival sac.</p>

ABSTRACT

This study was purposed to investigate the incidence and the model of ABO hemolytic disease in newborn (ABO-HDN) and the results of the three hemolysis test, so as to provide the evidences for clinical diagnosis and therapy. A total of 227 cases of maternal-fetal ABO incompatibility from January 2013 to October 2013 in the First Affiliated Hospital of Xiamen University were enrolled in the study. The ABO blood group of newborn and mother was detemined and three hemolysis tests (direct antiglobulin test, free antibody test, RBC antibody release test) were performed. The results indicated that in 227 cases of ABO incompatible pregnancies,186 cases were ABO-HDN (81.94%). There was no significant difference in the incidence between O-A and O-B incompatible pregnancies (P > 0.05). The positive ratio of direct antiglobulin test, free antibody test and RBC antibody release test were 59.14% (110/186), 84.78% (156/186) and 94.62% (176/186) respectively. It is concluded that the incidence of ABO-HDN is high. The main models of ABO-HDN were O-A and O-B. There was no significant difference in the incidence between O-A and O-B incompatible pregnancies. Three hemolysis tests are high sensitivity and are helpful in early diagnosis and early treatment of HDN.

ABSTRACT

R-spondin (Rspo)s proteins are a new group of Wnt/beta-catenin signaling agonists. These signaling molecules are known to be involved in the developmental stages of skeletal system. Recent studies in various murine osteoblast models have proposed that Rspo1 may interact with Wnt signaling pathway to induce differentiation in osteoblasts. Though findings in murine osteoblasts implicate a synergestic role of Rspo1 with Wnt signaling, still no study has addressed the similar role in more clinically applicable osteoblast models i.e., human cell lines or primary cells. Therefore, in the present study, we investigated the possible role of Rspo1 during differentiation process of human in vitro osteoblast cell models like primary osteoblasts or human osteoprogenitor cell line hFOB1.19 along with murine preosteoblast cell line MC3T3 E-1. Our results showed increase in Rspo1 at transcript level during differentiating phase of human primary osteoblasts and human FOB1.19 cells. We also found that Rspo1 (100 ng/mL) acts additively with Wnt3a to activate Wnt signaling, as confirmed by luciferase activity after transfection of TOPFLASH construct to hFOB1.19 cells. Similar additive role of Rspo1 and Wnt3a was apparent in alkaline phosphatase (ALP) activity analysis of human primary cells. Moreover, a reduction in ALP activity was observed with knock-down of Rspo1 by transfected shRNA in hFOB1.19 cells. These results suggested the possibility of autocrine regulation by Rspo1 on the osteogenic activities in human in vitro osteoblast models. Furthermore, these results were corroborated in MC3T3-E1, murine osteoblast cell model. Osteoblastic differentiation was induced by transfection of Rspo1 which was confirmed by increased ALP staining and qRT-PCR analysis of osteogenic markers, such as Runx2 and osteocalcin. In conclusion, present study highlights the role of Rspo1 in bone remodeling where it activates Wnt signaling to induce differentiation, as shown in human as well murine in vitro osteoblast cell models.

ABSTRACT

Objective To investigate the relation among the results of thyroid fine needle aspiration cytology(FNAC),thyroid ultrasonography(US)and histopathologic diagnosis about the thyroid nodules detected by physical examination,meanwhile to analyze the etiopathogenesis of the nodtries and to evaluate the risk of thyroid cancer and the clinical diagnostic value of FNAC.Methods The data of thyroid FNAC results of the thyrroid nodules detected by physical examination in 271 cases were analyzed and compared with thyroid US and histopathologie diagnosis.Resuit (1)The FNAC results showed that the incidences of malignant and suspected mali-gnant lesions were 1.48%and 5.90%respectively.The rate of benign lesions was 78.60%and that of goiter was 29.15%,Hashimoto's thyroiditis 26.57%and thyroid adenoma 15.13%.Benign lesions were more common than malignant ones.(2)Comparison of the FNAC and US results of the thyroid nodules showed that of 96 cases with single nodule the rates of malignancy and suspected malignancy were 3.12%and 7.29%,but of 137 cases with multiple nodules the rates of the two lesions were 0.73%and 6.57%.In 108 cases with smaller nodules(≤1.5 cm)the rate of malignancy and suspected malignaney found with FNAC were 0.93%and 7.41%,while in 125 cases with greater nodules (>1.5 cm)the rate of the two lesions were 2.40%and 6.42%.In 99 solid nodules the rates of malignancy and suspected malignancy were 2.02%and 12.12%,while in 85 cystic or mixed nodules the rates of the two lesions were 2.35%and 2.35%.In the above-mentioned three groups,only the suspected malignaney rate in solid nodules was higher than these in cystic or mixed ones with significant difference (P=0.013).(3)As compared with the cytological and histological diagnoses in 24 cases,the diagnostic accuracy of FNAC was 75.00%and the rates of false positive and false negative were 25.00%and 0,respectively.Conclusions The common causes of the thyroid nodules detected in physieal examination are goiter,Hashimoto's thyroiditis and thyroid adenoma.FNAC is a reliable method to define the benign or malignant nature of thyroid nodules with a high diagnostic accuracy.US features of the nodule alone,no matter it is single,solid or of greater size do not sufficiently increase the incidence of thyroid carcinoma.

Objective To understand the incidence of subclinical thyroid dysfunction in adult population of Beijing.Methods Serum levels of thyroid-stimulating hormone(TSH),T3,T4 and thyroid peroxidase antibody(TPOAb)were measured for 1966 persons living in Beijing by physical examinations in 2002,and 1646 of them(83.72%)were followed-up in 2004.Results Overall incidence of subclinical hypothyroidism and subclinical hyperthyroidism were 1.72 percent and 0.25 percent,respectively.Incidence of subclinical hypothyroidism in females aged >40 years was significantly higher than that in those ≤40 years(χ2=10.869,P<0.01),but no significant difference between those >40 and ≤40 years in males was found.Incidence of subclinical hypothyroidism in females was significantly higher than that in males >40 years of age(χ2=24.122,P<0.01),but no significant difference between females and males ≤40 years was found.No significant difference in incidence of subclinical hypothyroidism was noticed between varied age groups and between males and females.Incidence of subclinical hypothyroidism was significantly higher in those with history of subclinical hypothyroidism than that in those without it(χ2= 6.898,P<0.01).No significant difference in incidence of subclinical hypothyroidism between those with family history of thyroid disease and those without it was found.Incidence of subclinical hypothyroidism was significantly higher in those with positive TPOAb than that in those with negative one(χ2=14.084,P<0.01),but no significant difference in incidence of subclinical hyperthyroidism between those with positive and negative TPOAb was found.Conclusion Incidence of subclinical hypothyroidism was a little bit higher in population of Beijing area by physical examinations,particularly in females,elder age and positive TPOAb.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate and select excellent germplasm resources of Artemisia annua L., providing basic data for data base and breeding of A. annua.</p><p><b>METHOD</b>Seventy-two germplasms of A. annua, which were collected from main production areas, were planted in germplasm resources garden in Jingxi and Nanning under the same conditions. The samples were gathered in the squaring stage. Artemisinin was extracted by supersonic wave and determined by ultraviolet spectrophotometry.</p><p><b>RESULT</b>The leaf's yield and the content of artemisinin in the samples of different germplasms were varied significantly. Effect of environment on artemisinin content was more than that of hereditary factors. The content of artemisinin was different when the same material of A. annua grew in different place. The content of artemisinin in next year's plant was decreasing.</p><p><b>CONCLUSIONS</b>Seven germplasms from south of China have been selected, their content of artemisinin was above 0.9% and calculated yield was more than 2250 kg x hm(-2). The content of artemisinin has been affected by hereditary factors and variation of the growth environment.</p>

ABSTRACT

Objective: We tired to develop a method of detecting the expression level of RRM1 mRNA in tumor tissues and peripheral blood by real-time fluorescent quantitative PCR and compare the detection methods and results between the two different samples. We aimed to provide the basis for predicting clinical outcome from the expression of RRMI gene. Methods: The plasmid standard of RRM1 gene was constructed. Real-time quantitative analysis was performed using ABI7000 PCR kit. The PCR condition was optimized and the standard curve was established to detect the clinical samples. Results: The RRMI gene expression was detected in lung cancer tissues and normal lung tissues from 18 patients and in peripheral blood samples from 17 patients. The relative expression of RRM1 normalized by β-actin was 4.46 × 10-3 in lung cancer tissues, 3.43 × 10-3 in normal tissues, 2.54 × 10-3 in peripheral blood. The linear correlation between Ct value and the logarithm of original concentration was favorable. Conclusion: The expression of RRM1 mRNA in non-small cell lung cancer tissues and peripheral blood was successfully detected by using SYBR Green I fluorescence real-time quantitative PCR assay. There is no significant difference between the two sources of samples. The detection method has relatively higher sensitivity and specificity and can be used in clinical analysis.

ABSTRACT

<p><b>OBJECTIVE</b>To analyse retrospectively the diagnosis and treatment of severe pneumonia in kidney transplantation recipients.</p><p><b>METHODS</b>Between January 1999 and December 2003, 172 adult patients underwent kidney transplantation at our department. In all severe pneumonia cases, empirical therapy was initiated with aztreonam, erythromycin and ganciclovir. And the therapy was switched to proper antibiotics according to the results of sensitivity testing. Responsible pathogen was detected by study of BAL (bronco-alveolar-lavage), sputum and blood specimen. Analyses included cell differential count, cytopathologic examination and cultures for bacteria, fungi and viruses. The immunosuppressive therapy was drastically reduced. Hypoxia was relieved by BiPAP (Bi-level Positive Airway Pressure) or mechanical ventilation if necessary.</p><p><b>RESULTS</b>Seventeen cases (9.9%) of pneumonia were observed in the 172 recipients, only 11 (65%) patients experienced severe pneumonia, 1 (9%) of them died. Fever was the most common symptom on presentation (82%). On presentation 46% of the patients presented with classical clinical syndrome of fever accompanied by cough and dyspnea. Positive rate of BAL and blood culture were 100% and 46% respectively. BiPAP and mechanical ventilation were required in 6 and 2 cases respectively.</p><p><b>CONCLUSION</b>BAL is preferred for early detection of responsible pathogen. A combination of drastic reduction of the immunosuppressive regimen, implementation of appropriate empirical antibiotics, proper BiPAP or mechanical ventilation are important.</p>

ABSTRACT

To construct a novel baculovirus expression system of Spodoptera litura multicapsid nucleopolyhedrovirus, the 5' end and 3' end-flanking fragments of ph gene were amplified from the genome DNA of SpltMNPV, Japan-C3 strain using two pairs of primers synthesized according to SpltMNPV China-G2 strain genome DNA sequence published in GenBank. To obtain the transfer vector pSplt-gfp, the fragment of gfp gene was inserted into this vector between two fragments tandem linked into pUC18. The spli cells were cotransfected with pSplt-gfp and the wild SpltMNPV genome DNA. The recombinant virus containing gfp was selected with the limited dilution method. The fluorescence can be observed in the spli cells and the 3rd instar larvae after 24 and 48 hours by infection of the recombinant virus, respectively. The result showed that the recombinant virus was obtained successfully. It will be helpful to establish Spodoptera litura multicapsid nucleopolyhedrovirus expression system and more effective pesticide for Spodoptera litura.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate a new technique for treatment of the symblepharon caused by physical and chemical burn.</p><p><b>METHODS</b>Thirty-eight patients with 40 eyes were undergoing the treatment. It was carried out to reconstruct the conjunctival fornix by pulling the remained conjunctive backward from the corneal limbus in serious stages.</p><p><b>RESULTS</b>Thirty-eight patients (40 eyes) were treated with this technique, with the successful results of 13 patients (13 eyes) in 2 times, 19 patients (21 eyes) in 3 and 6 patients (6 eyes) in 1. Only one case was failure.</p><p><b>CONCLUSION</b>The above mentioned technique could be a safe and effective way for treatment of symblepharen.</p>

ABSTRACT

Gemcitabine is important pyrimidine antimetabolites drug. Ribonucleotide reductase is oneof its targets whose overexpression has been observed in several gemcitabine- resistant cell lines. Meanwhile,the NSCLC patients with high RRM1 expression levels have a relatively poor survival when treated with gem-citabine. To this day, the relationship between RRM1 expression levels in peripheral blood mononuclear cellsand gemcitabine is still uncertain and it deserves further research.

ABSTRACT

Repetitive(repetitive extragenic palindromic,REP,and enterobaterial repetitive intergenic consensus,ERIC) sequences in conjunction with polymerase chain reaction technique(REP and ERIC PCR) were used to fingerprint the genomes of 27 isolates of indigenous soybean rhizobia from Xinjiang.The indigenous soybean rhizobia in Xinjiang can be clustered into relative genetic similarities of approximately 0.5,of which one group mainly includes all slow-growing rhizobia,another mainly includes all fast-growing stains. REP and ERIC PCR analysis demonstrate a substantial genetic variability within members of Xinjiang indigenous soybean rhizobial populations, which reveals that genetic similarities have certain geographical correlation, and isolates from the same site have relative higher similarities.The results show that REP and ERIC PCR analysis give effective means in genetic diversity and population structure analysis of soybean rhizobia.
