ABSTRACT

The chemokine receptor CCR5 is a major co-receptor for HIV-1 entry into the host cell. Deletion of 32 bp (Δ32) alters the receptor structure and is associated with the protection against infection. The distribution of allelic variant depends on several factors influencing the epidemiology of HIV infections. Thus, the present study sought to estimate the allelic frequency of the CCR5 gene variant / CCR5Δ32 in blood donor candidates with and without positive serology for HIV-1+ at the HEMOAM Foundation. 239 candidates were enrolled and divided into two groups, HIV-1+ (101 individuals) and HIV- controls (138 individuals). After collecting peripheral blood, DNA was extracted and allele-specific PCR for identification of CCR5Δ32 polymorphism, was performed. The results obtained were analyzed using Stata (v.13). The groups were of similar ages, predominantly male and the distribution of genotypes and alleles were in Hardy-Weinberg equilibrium (p=0.725 and p=0.879, respectively). The highest frequency was wild genotype, followed by the heterozygous genotype in both groups (control and the HIV-1+ ). When the frequencies in HIV-1+ subgroups were analyzed, the absence of the allelic variant CCR5Δ32 subgroup ELISA(+) Westen Blot(+) was noted. Therefore, our data indicate that CCR5Δ32 polymorphism has a low frequency in the population studied.

ABSTRACT

We analyzed the kinetics of cytokine production by mononuclear cells from 17 patients who had been treated for paracoccidioidomycosis, using the stimulus of gp43 peptide groups (43kDa glycoprotein of Paracoccidioides brasiliensis) at 0.1 and 1µM, gp43 (1µg/ml) and crude Paracoccidioides brasiliensis antigen (PbAg; 75µg/ml). IFN-gamma production was a maximum at 144 hours in relation to the G2 and G8 peptide groups at 1µM and was greatest at 144 hours when stimulated by gp43 and by PbAg. The maximum TNF-alpha production was at 144 hours for the G2 group (0.1µM) and for gp43. IL-10 production was highest after 48 and 72 hours for G7 and G6 at 1µM, respectively. We also suggest the best time for analysis of IL4 production. These results may contribute towards future studies with gp43 peptides and encourage further investigations with the aim of understanding the influence of these peptides on the production of inflammatory and regulatory cytokines.


Analisamos a cinética da produção de citocinas de células mononucleares de 17 pacientes com paracoccidioidomicose tratada, usando como estímulo: grupos de peptídeos da gp43 (glicoproteina de 43kDa de Paracoccidioides brasiliensis) a 0,1 e 1µM, gp43 (1µg/mL) e antígeno bruto de Paracoccidioides brasiliensis - AgPb (75µg/mL). A produção de IFN-gama foi máxima em 144 horas frente aos grupos de peptídeos G2 e G8 a 1µM e maior em 144 horas quando estimuladas por gp43 e por AgPb. A produção de TNF-alfa foi máxima em 144 horas para G2 (0,1µM) e para gp43. A produção de IL-10 foi maior após 48 e 72 horas para G7 e G6 a 1µM, respectivamente. Sugerimos também o melhor período para a análise da produção de IL4. Tais resultados podem contribuir para estudos com peptídeos da gp43, estimulando investigações posteriores visando entender a influência de tais peptídeos na produção de citocinas inflamatórias e regulatórias.

ABSTRACT

A freqüência dos HLA foi analisada em 25 Judeus Ashkenazitas, não consangüíneos, residentes em São Paulo, Brasil, com dermatofitose crônica causada por T. rubrum e em 25 indivíduos sadios, pertencentes ao mesmo grupo étnico dos pacientes. Observou-se valor estatisticamente significante (p<0,05) para HLA-B14 associado a resistência à dermatofitose crônica enquanto HLA-DQB1*06 (p=0,05) possivelmente relacionado a susceptibilidade. Estes achados indicam que o desenvolvimento da dermatofitose crônica pode ser influenciado por genes localizados no cromossomo 6, na região do complexo principal de histocompatibilidade.

ABSTRACT

The frequency of HLA (Human Leucocyte Antigens) was analyzed in 25 non-consanguineous Brazilian Ashkenazic Jews, resident in the city of São Paulo, Brazil, suffering from chronic dermatophytosis caused by T. rubrum, and in 25 non-infected individuals belonging to the same ethnic group. Statistically significant values (p 0.05) were observed for HLA-B14 associated with resistance to chronic dermatophytosis and HLA-DQB1*06 (p=0.05) possibly related to susceptibility. These findings suggest that genes on the chromosome 6, in the region of the major histocompatibility complex, may influence the development of chronic dermatophytosis.

ABSTRACT

Seeking to improve the laboratory diagnosis of Cutaneous Tuberculosis, a study was carried out on the application of PCR technique in macerated, decontaminated(with 4 percent H2SO4 for elimination of normal microbiot), neutralized (with 4 percent NaOH)biopsies tissues samples stored at -20liC. Of the 37 samples submitted for study, 16.22 percent were positive by microscopy for acid-fast bacilli (concentrated method) and in 43.24 percent the Mycobacterium tuberculosis was isolated in L que wenstein-Jensen medium. Using a M. tuberculosis complex specific primer set (gene sequence for 16S rDNA), the mycobacterial DNA was detected in 24.32 percent of the biopsies. The sensitivity and specificity of PCR were 43.7 percent and 90.4 percent, respectively. Due to low sensitivity and discrepant results between bacteriological techniques and PCR methodology, the samples were repeated in a new PCR with primers for the IS6110 target. The sensitivity and specificity of PCR for the IS6110 target obtained 100 percent in comparison with the culture method. The results confirm the effectiveness of PCR methodology using primers for the IS6110 gene sequence and permit the PCR method to be applied to frozen cutaneous biopsies sent by services that do not identify the M. tuberculosis by the biology molecular method.
