ABSTRACT

Objective: The Streptomyces phage phiC31 integrase offers a sequence-specific method of transgenesis with a robust long-term gene expression. PhiC31 has been successfully developed in a variety of tissues and organs for purpose of in vivo gene therapy. The objective of the present experiment was to evaluate PhiC31-based site-specific transgenesis system for production of transgenic bovine embryos by somatic cell nuclear transfer and intracytoplasmic sperm injection


Materials and Methods: In this experimental study, the application of phiC31 integrase system was evaluated for generating transgenic bovine embryos by somatic cell nuclear transfer [SCNT] and sperm mediated gene transfer [SMGT] approaches


Results: PhiC31 integrase mRNA and protein was produced in vitro and their functionality was confirmed. Seven phiC31 recognizable bovine pseudo attachment sites of phage [attP] sites were considered for evaluation of site specific recombination. The accuracy of these sites was validated in phic31 targeted bovine fibroblasts using polymerase chain reaction [PCR] and sequencing. The efficiency and site-specificity of phiC31 integrase system was also confirmed in generated transgenic bovine embryo which successfully obtained using SCNT and SMGT technique


Conclusion: The results showed that both SMGT and SCNT-derived embryos were enhanced green fluorescent protein [EGFP] positive and phiC31 integrase could recombine the reporter gene in a site specific manner. These results demonstrate that attP site can be used as a proper location to conduct site directed transgenesis in both mammalian cells and embryos in phiC31 integrase system when even combinaed to SCNT and intracytoplasmic sperm injection [ICSI] method

ABSTRACT

Background: The cyclin E2 [CYCE2] is an important regulator in the progression and development of NSCLC, and its ectopic expression promoted the proliferation, invasion, and migration in several tumors, including Non-Small Cell Lung Cancer [NSCLC]. However, the upregulation of CYCE2 in NSCLC cells suggested that it has a key role in tumorigenicity. In addition, the RAS family proteins as oncoproteins were activated in many major tumor types and its suitability as the therapeutic target in NSCLC was proposed. Considering the crucial role of microRNAs, it was hypothesized that altered expression of hsa-miR-30d-5p and hsa-let-7b might provide a reliable diagnostic tumor marker for diagnosis of NSCLC


Method: Real-time RT-PCR approach could evaluate the expression alteration of hsa-miR-30d-5p and hsa-let-7b and it was related to the surgically resected tissue of 24 lung cancer patients and 10 non-cancerous patients. The miRNAs expression was associated with clinicopathological features of the patients


Results: Hsa-miR-30d showed a significant downregulation [p=0.0382] in resected tissue of NSCLC patients compared with control group. Its expression level could differentiate different stages of malignancies from each other. The ROC curve analysis gave it an AUC=0.73 [p=0.037] which was a good score as a reliable biomarker. In contrast, hsa-let-7b was significantly overexpressed in tumor samples [p=0.03]. Interestingly, our findings revealed a significant association of hsa-let-7b in adenocarcinoma tumors, compared to Squamous Cell Carcinomas [SCC] [p<0.05]. Also, analysis of ROC curve of hsa-let-7b [AUC=0.74, p-value=0.042] suggests that it could be as a suitable biomarker for NSCLC


Conclusion: Together, these results suggest a possible tumor suppressor role for hsa-miR-30d in lung tumor progression and initiation. Moreover, upregulation of hsa-let-7b was associated with the tumor type

ABSTRACT

Background: Morbid obesity is rising around the world. It can cause unpleasant appearance and body image. Most of the studies have aimed to evaluate the psychopathology of overweight and obesity and paying attention to mental well-being in morbid obese individuals is rare. Therefore, this study aimed to assess the relationship between body image and psychological well-being in morbid obese patients


Methods: This cross-sectional study, using simple random sampling method, was done on 124 morbid obese patients who referred to obesity clinic in Shiraz from 2016 to 2017. The data were collected by body image index and psychological well-being questionnaire. Results were analyzed using descriptive statistics, Pearson correlation coefficient test, ANOVA, and Regression analysis


Results: The results showed a significant relationship between body image and psychological wellbeing [r=0.43] [P<0.001], and between the total score of the body image and all the subscales of psychological well-being except autonomy and purpose in life [P<0.05]. There was also a significant relationship between the total score of psychological well-being and all the subscales of body image [P<0.05]. However, there was no significant difference between the mean scores of the body image and those of psychological well-being in different categories of body mass index [BMI] [P>0.05]


Conclusion: Final results indicated that body image defects caused by obesity could lie in negative psychological well-being in all aspects. This study can promote health clinicians' knowledge in supporting of mental status of obese individuals. It is suggested that preventing and supporting intervention should be performed as effective methods for encountering and coping with psychological effects of obesity

ABSTRACT

Background:Colorectal cancer [CRC] is one of the most common cancers among men and women worldwide. Cancer metastasis is the main cause of death in patients with cancer. NEBL [nebulette, Gene ID: 10529] protein interacts with thin filaments in the cell and may functionally destabilize focal adhesion composition. There are some studies on NEBL gene expression alteration in cancer. In the presented study we aimed to analyze NEBL gene expression in patients with colorectal cancer to explore possible association of this gene with clinicopathological features in CRC


Methods:Sixty-seven fresh samples of colorectal tumors and adjacent normal tissues were collected from Iranian patients with CRC. Real time polymerase chain reaction was performed to measure the level of NEBL gene expression and its association with clinico-pathological features


Results:A significant overexpression with 3 fold increse was seen in NEBL mRNA level in tumoral tissues compared with the adjacent normal tissues. In addition there was a significant association between NEBL gene expression with lymph node metastasis in patients with CRC


Conclusion:The overexpression of NEBL has the capacity to be considred as a prognostic biomarker in patients with CRC

ABSTRACT

Depression is common in chronic medical illnesses including HF. It has been established that depression has different rates in different races and the reported depression rates among hospitalized patients range from 13% to 77%; nevertheless, there is remarkably little information on the prevalence of depression and the impact of demographic and health status in the north of IRAN. The goal of this study was to determine the prevalence of depression in hospitalized heart failure [HF] patients, as well as the impact of age, gender, and functional status on this group of patients. In this study, carried out in one heart center, a total of 196 hospitalized HF patients with New York Heart Association [NYHA] functional classes 2- 3 and 4 and an ejection fraction [EF] <40% were given questionnaires to assess depression. Depression was evaluated with the Beck Depression Inventory [BDI] questionnaire. Depression was defined as a score on the BDI of > 17. A total of 23.5% of the patients scored as depressed. Men were more likely [33%] to be depressed than women and had a higher mean BDI score [p=0.004]. Patients classified as NYHA functional class 3 were more likely to score as depressed than class 2 patients [p= 0.001], and patients with a higher BDI score had a lower EF [r=0.25, p=0.001]. Depression is common in patients with HF. Men and patients with lower health status are more likely to be depressed. Pharmacologic or non- pharmacologic treatment of depression in HF patients should be considered
