ABSTRACT

Objective:To analyse the evaluation and experience of clinical effect of minimally invasive drainage and craniotomy in the treatment of patient with epidural hematoa.Methods:100 cases of patients who were diagnosed as epidural hematoma from January 2015 to January 2016 were selected and randomly divided into two groups,where the control group were given craniotomy,and the observation group were given minimal invasive drainage.The effect of the surgery and the life qualities before and after treatment of the two groups were compared.Results:The effect of observation group is better than that of control group (P < 0.05);craniocerebral defect and the re-hemorrhage incidence occurrence in the observation group were obviously lower than those in the control group,(0 %,6 % vs 12 %,20 %) (P<0.05).Conclusion:Compared with craniotomy,minimal invasive drainage in clinical treatment of epidural hematoma showed advantage in at smaller trauma,faster recovery,and better effect,thus deserving further promotion of clinic application.

ABSTRACT

Objective To examine the prevalence of chikungunya virus in brain tissue samples from rat?like animals in Xiamen, Shenzhen and Guangzhou, and to explore whether the rat?like animals are potential sources of human chikungunya fever infections and the host of the virus. Methods Rat?like animals were trapped in residential areas, city parks, hospitals, markets and schools in Xiamen, Shenzhen and Guangzhou (Yuexiu and Baiyun districts) between January 2013 and June 2016. Brain tissue samples of the trapped animals were collected under sterile. Chikungunya virus was detected by using reverse transcription polymerase chain reaction (RT?PCR). Results Totally 1092 rat?like animals were trapped, which belonged to 7 species, 3 genera, 2 families, 2 orders. Rattus norvegicus was the dominant species in the indoor environment, Rattus losea was dominant in wild environment, and 1092 brain tissue samples were collected. No detectable chikungunya virus was found in the brain tissue samples by RT?PCR. Conclusion There is a low possibility that rat?like animals act infectious sources of human chikungunya fever infections and the host of the virus.

ABSTRACT

Objective To investigate the correlation between three sperm indexes and the outcome of in vitro fertilization and embryo transfer(IVT-ET) before and after semen treatment.Methods The semen derived from 298 male patients treated with IVT-ET from the Affiliated Hospital of Southwest Medical University was retrospectively analyzed.Routine analysis of semen before treatment was performed according to the WHO standard.Vitrolife density gradient centrifugation combined with upstream method was used to analyze sperm parameters after sperm analysis,and the observation and statistics were performed after fertilization and sequential culture to day 3.Results The total viability of spermatozoa before and after semen treatment,the percentage of normal spermatozoa,sperm density,and other differences were statistically significant (P<0.05);The normal sperm morphology rate was positively correlated with fertilization rate(r=0.487,P<0.01) and cleavage rate(r=0.250,P<0.05);The sperm vitality of post-processed semen was positively related to the fertilization rate (r =0.249,P<0.05,);and significantly cleavage rate (r =0.272,P<0.05);After treatment,sperm density was positively correlated with 2 PN(r=0.609,P<0.01) and multiple PN fertilization rates(r=0.243,P<0.05).Conclusion Our experimental results indicate that the parameters of post-processed sperm treated at the day when the oocytes were collected may be more helpful in predicting the fertilization rate and cleavage rate of IVF-ET.Therefore,the IVF-ET treatment protocols should be chose based on the normal sperm morphology rate and sperm vitality after semen treatment.

ABSTRACT

BACKGROUND: Early mammalian embryonic development and implantation is a very complex process, which is regulated by a variety of cytokines. Moreover, epidermal growth factor, transforming growth factor, leukemia inhibitory factor, insulin-like growth factor and vascular endothelial growth factor play an important role in this regulation process. OBJECTIVE: To introduce cytokines related to early embryonic development, including types, structural features and molecular mechanisms, and meanwhile to review the effects of epidermal growth factor, transforming growth factor, leukemia inhibitory factor, insulin-like growth factor and vascular endothelial growth factor in the early embryonic development. METHODS:A literature retrieval was performed to search relevant articles published from 1989 to 2015 using the keywords of “embryos development, epidermal growth factor, transforming growth factor, leukemia inhibitory factor, insulin-like growth factor, vascular endothelial growth factor”. Active effects of these five kinds of cytokines in overcoming the mammalian embryonic development block were reviewed and summarized to provide theoretical and experimental basis for further optimization of the embryonic culture system in vitro. RESULTS AND CONCLUSION: Cel growth factors involved in embryonic development is the focus of the study. Insulin-like growth factor is capable of promoting the embryonic growth; epidermal growth factor exhibits multiform functions in the embryonic development; leukemia inhibitory factor directly affects oocyte cel development, and progesterone increases the expression of leukemia inhibitory factor to provide a good cytokine microenvironment for embryonic development; vascular endothelial growth factor promotesin vitro oocyte maturation, and plays a very important role in various stages of embryonic development. To clarify these regulatory patterns of cytokines cannot only contribute toin vitro embryonic culture, but also reduce embryonic apoptosis, which is of great significance for promoting embryonic development.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the prevalence of serum antibodies against Japanese encephalitis virus (JEV) in bats.</p><p><b>METHODS</b>Blood samples from the heart were obtained from bats captured in Guangdong and Hainan Provinces in 2013. The anti-JEV antibodies in bat sera were tested using indirect ELISA and virus neutralization test.</p><p><b>RESULTS</b>A total of 201 bat serum samples were tested, in which the total positivity rate of anti-JEV antibodies was 46.27% (93/201). The positive rate of anti-JEV antibodies in bats from Hainan and Guangdong Provinces was 88.89% (48/54) and 30.61% (45/147), respectively. All the samples from Rousettus leschenaultia, Miniopterus schreibersii, Pipistrellus abramus, and Rhinolophus macrotis were positive for anti-JEV antibodies, and up to 95.56% (43/45) of the samples from Miniopterus schreibersii (from Hainan Province) yielded positive results. Of the 28 samples with positive results by indirect ELISA, 15 showed positive results in virus neutralization test (53.57%) with neutralization antibody titers ranging from 1:10 to 1:28.22.</p><p><b>CONCLUSION</b>Bats from different regions and of different species can be naturally infected with JEV and have a high prevalence of anti-JEV antibodies in their sera. The role of bats in the natural cycle of JEV awaits further study.</p>

ABSTRACT

BACKGROUND:The successful establishment of human embryonic stem cel lines in vitro is of great significance to human embryonic development mechanism and developmental biology, cel and tissue transplantation in the treatment of certain diseases. OBJECTIVE:To summarize the progress of in vitro culture of embryos and establishment of embryonic stem cel lines, to explore the influential factors for in vitro culture of embryos, and the methods of culturing human discarded embryos, isolating inner cel mass and establishing embryonic stem cel lines, as wel as the establishing conditions for embryonic stem cel lines. METHODS:With the key words of“embryo, embryonic stem cel s, coculture, sequential culture”, the first author searched CNKI and SCI databases for literatures concerning in vitro culture and transplantation of embryos and establishment of embryonic stem cel lines published from 2000 to 2014. Systematic evaluation was conducted. Final y, 58 literatures were retained for result analysis. RESULTS AND CONCLUSION:The culturing condition for embryos in vitro is the key factor affecting embryo transfer outcomes, including culture medium component and culture system. In previous studies, the component and application of culture medium have changed greatly, and the culture system has altered from single culture to coculture and sequential culture. Ethical issues and embryonic origin restrictions restrict the establishment of human embryonic stem cel lines. Clinical y discarded low-quality embryos can be used as one of the material sources to establish human embryonic stem cel lines, which can effectively lessen the problem of embryo shortage during the establishment of human embryonic stem cel lines and reduce ethical disputes.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the location and characteristics of microdeletions of Y chromosome azoospermia factor (AZF) genes in infertile males with azoospermia and severe oligozoospermia in southern Sichuan.</p><p><b>METHODS</b>Multiplex PCR was used to detect 18 sequence tagged sites (STS) involved in Y chromosome AZF microdeletions among 224 infertile males (including 134 azoospermia cases and 90 severe oligozoospermia cases) and 70 healthy males.</p><p><b>RESULTS</b>Among the 224 infertile males, the overall frequency of microdeletions was 12.1% (27/224), and were 13.4% (18/134) in those with azoospermia and 10.0% (9/90) in those with severe oligozoospermia. The most frequent microdeletions have occurred in the AZFc region (51.9%). Compared with the 6 STS loci recommended by European Academy of Andrology and European Molecular Genetics Quality Network, 22.7% more deletions were detected based on the 18 STS loci selected from the AZF region.</p><p><b>CONCLUSION</b>Identification of Y chromosome microdeletions has a significant implication on the diagnosis of male infertility. The most frequent microdeletions have occurred in the AZFc region in southern Sichuan. To use more sequence tagged sites for the screening can improve the reliability and detection rate of Y chromosome microdeletions.</p>

ABSTRACT

Objective To investigate the influence of processed total motile sperm (PTMS) count of husband on clinical pregnan-cy rate of intrauterine insemination(IUI) .Methods We retrospectively analyzed a total of 229 cycles of IUI among 131 patients in our hospital during the past three years .The cycles were divided into 5 groups according to the PTMS count :group A(0 .05) among five groups .Conclusion Ideal clinical pregnancy can be achieved when the PTMS count is between 3 × 106 and 5 × 106 .

ABSTRACT

Objective:To observe and compare the therapeutic effect of metoprolol by routine increasing dose method and rapid titration method on acute myocardial infarction (AMI).Methods:A total of 60 inpatients,who were di-agnosed with AMI within 24h and without contraindications for metoprolol,were randomly divided into two groups:routine therapy group (received metoprolol using routine methods,the dose was added in seven days)and rapid ti-tration group (metoprolol was added in three days using titration).The dosage maintained with 190 mg/d after both groups reaching the target dose of 190mg/d;then therapeutic effects were observed in both groups.Results: ①There were no re-myocardial infarction,rehospitalization caused by heart failure and sudden death etc.in both groups;② Patients received echocardiography in outpatients after three months.Compared with routine increasing dose group,there was significant reduction in left ventricular end-diastolic diameter [LVEDd,(55.00±7.56)mm vs.(50.00± 5.81)mm]and significant rise in left ventricular ejection fraction [LVEF,(49.13 ± 10.18)% vs. (57.84±10.34)%]in rapid titration group,P <0.01 both.Conclusion:Rapid titration method could make the pa-tients rapidly reach the targeted dose of metoprolol and inhibit renin release earlier,block the renin-angiotensin sys-tem,and improve myocardial remodeling and cardiac function.

ABSTRACT

<p><b>OBJECTIVE</b>The purpose of this study was to observe the behavior of bone formation around the titanium-hollow porous cylinder implant composed of bovine bone morphogentic protein(bBMP).</p><p><b>METHODS</b>Porous-hollow cylinder titanium implant composed of bBMP was implanted into mandibule of dogs. Multiple fluorescent was labeled at different times and then LSCM was used to observe the newly formed bone around the complex implant.</p><p><b>RESULTS</b>The newly formed bone around the complex implant in experimental group was more obvious than that in other groups.</p><p><b>CONCLUSION</b>Earlier, longer and more new-bone formation can be induced by porous-hollow cylinder titanium implant composed of bBMP, and LSCM is an effective method to observe new bone formation around implant.</p>

ABSTRACT

<p><b>OBJECTIVE</b>The purpose of this study was to establish a stable, high-efficient and low-toxic way of transfecting nm23-H1 into Tca8113 line cells, and then to find out whether nm23-H1 could affect the invasion and metastases ability of Tca8113 line cells.</p><p><b>METHODS</b>nm23-H1 was transfected into Tca8113 line cells with Lipofect. The different expressions of nm23-H1 between transfected and non-transfected line cells were detected by the method of immunohistochemistry. The difference of the invasion and metastases ability between transfected and non-transfected line cells was detected by transwell-room and wash techniques. The change of chemo-sensitivity was evaluated by MTT.</p><p><b>RESULTS</b>Using pCMV-NEO-BAM system to keep stable expression of nm23-H1, the significant difference of NDPKA expression between transfected and non-transfected Tca8113 line cells was discovered; The metastases ability of transfected Tca8113 line cells decreased significantly; The chemo-sensitivity of transfected Tca8113 line cells to CDDP increased significantly.</p><p><b>CONCLUSION</b>nm23-H1 can inhibit the metastases of Tca8113 line cells and increase the chemo-sensitivity to CDDP significantly.</p>

ABSTRACT

objective: To observe the distribution of pegylated liposmal doxorubicin(PLD) in animal model of tongue cancer. Methods: Tongue cancer model was established in 40 golden hamster, PLD or free doxorubicin at the dose of 9 mg/kg was injected perifocally into each of 20 mice, the concentration of the drugs in lypmph node and blood was measured with HPLC. Results: After injection of free ADM peak concerntration in blood was acheived in 3 h and in lymph nodes in 1 h, while after injection of PLD, that was in 16 and 6 h respectively. The concertration of PLD was heigher than that of ADM in both blood and lymph node from 16 or 6 h till 216 h after injetion.Conclusion: PLD can be regarded as valuable drug delivery system in the treatment of oral cancer.
