ABSTRACT

Aim: This study was to evaluate the value of diffusion-weighted imaging (DWI) in predicting the efficacy of radiotherapy for esophageal cancer from xenograft model level. Subjects and Methods: Thirty-two tumor-bearing mice from the Eca-109 cell line nude mice models were established. The experimental group (n = 16) received a single dose of 15 Gy (6MV X-ray), whereas the control group (n = 16) did not receive any treatment. The tumor volume and apparent diffusion coefficient (ADC) were obtained. The cell density, tissue necrosis ratio, and CD31 expression were determined at matched time points. Results: The tumor volume was smaller in the experimental group than in the control group (P < 0.05) on the 7th day after radiotherapy (1.580 ± 0.965 cm3 vs. 2.671 ± 0.915 cm3). The ADC values were higher in the experimental group than in the control group on the 3rd day (P < 0.05) (998.15 ± 163.76 ×10− 6 mm2/s vs. 833.32 ± 142.15 ×10− 6 mm2/s). On the 3rd day after radiotherapy, the differences in cell density and necrosis ratio between the two groups were statistically significant; the tumor cell density was lower in the experimental group (25.56 ± 1.40%) than in the control group (33.48 ± 4.18%) (P < 0.05), and the proportion of tissue necrosis was higher in the experimental group (32.19 ± 1.21%) than in the control group (29.16 ± 2.16%) (P < 0.05). The negative and weak positive rate of CD31 expression in the experimental group was higher than the control group, whereas the generally positive and strong positive rate of CD31 expression was significantly lower than the control group in the early stage (P < 0.05). Conclusion: ADC values may change at the early stage before the morphological changes of tumors. Changes in cell density and necrosis ratio of transplanted tumors correspond to the changes in ADC values. DWI can be used for the early prediction of esophageal cancer radiotherapy efficacy

ABSTRACT

BACKGROUND@#Patients with temporal lobe epilepsy (TLE) originating from different seizure onset zones had distinct electrophysiological characteristics and surgical outcomes. In this study, we aimed to investigate the relationship between the origin and prognosis of TLE, and the stereoelectroencephalography (SEEG) features.@*METHODS@#Thirty patients with TLE, who underwent surgical treatment in our functional neurosurgery department from January 2016 to December 2017, were enrolled in this study. All patients underwent anterior temporal lobectomy after an invasive pre-operative evaluation with SEEG. Depending on the epileptic focus location, patients were divided into those with medial temporal lobe seizures (MTLS) and those with lateral temporal lobe seizures (LTLS). The Engel classification was used to evaluate operation effectiveness, and the Kaplan-Meier analysis was used to detect seizure-free duration.@*RESULTS@#The mean follow-up time was 25.7 ± 4.8 months. Effectiveness was 63.3% for Engel I (n = 19), 13.3% for Engel II, 3.3% for Engel III, and 20.0% for Engel IV. According to the SEEG, 60.0% (n = 18) had MTLS, and 40.0% (n = 12) had LTLS. Compared with the MTLS group, the operation age of those with LTLS was significantly greater (26.9 ± 6.9 vs. 29.9 ± 12.5 years, t = -0.840, P = 0.009) with longer epilepsy duration (11.9 ± 6.0 vs. 17.9 ± 12.1 years, t = -1.801, P = 0.038). Patients with MTLS had a longer time interval between ictal onset to seizure (67.3 ± 59.1 s vs. 29.3 ± 24.4 s, t = 2.017, P = 0.008). The most common SEEG ictal pattern was a sharp/spike-wave rhythm in the MTLS group (55.6%) and low-voltage fast activity in the LTLS group (58.3%). Compared with the LTLS group, patients with MTLS had a more favorable prognosis (41.7% vs. 77.8%, P = 0.049). Post-operative recurrence was more likely to occur within three months after the operation for both groups, and there appeared to be a stable long-term outcome.@*CONCLUSION@#Patients with MTLS, who accounted for three-fifths of patients with TLE, showed a more favorable surgical outcome.

ABSTRACT

Background Corneal neovascularization ( CNV ) is a complication of many ocular surface diseases. It often worsen the pathological course.Effective therapy for CNV is still researching. Objective This study was to investigate the inhibitory effect of irradiation on CNV. Methods CNV models were established in 70 right eyes of 70 clean Wistar rats by corneal alkali burning. The models were randomized into β ray 10 Gy once irradiation group( 2 eyes),β ray 7 Gy multiple irradiation group( 17 eyes),β ray 10 Gy multiple irradiation group( 17 eyes),1% cyclosporin A ( CsA ) eye drops group ( 17 eyes) and model group ( 17 eyes),and 6 matched normal rats were used as normal controls. All treatments started from the first day of the corneal alkali burning.CNV length and area were measured under the slit lamp every day.Corneal samples and homogenate were prepared 3,5,7 days after corneal alkali burning. The expressions of bcl-2,bax,vascular endothelial growth factor(VEGF) in rat corneas were detected by immunochemistry,VEGF proteins and VEGF mRNA were detected by Western blot and reverse transcription polymerase chain reaction (RT-PCR),respectively. Results Corneal ulceration was found in the βray 10 Gy once irradiation group and β ray 10 Gy multiple irradiation group.CNV length and area were much less in the β ray 7 Gy multiple irradiation group and 1% CsA eye drops group compared with the model group on the seventh day after experiment( length:q=14.40,24.20,P<0.01 ;area:q=17.80,14.00,P<0.01 ).Immunochemistry revealed that compared with the model group, expressions of bcl-2 and VEGF proteins were weaker, but the expression of bax protein was stronger in the β ray 7 Gy multiple irradiation group and 1% CsA eye drops group.RT-PCR showed that the expression of VEGF mRNA in cornea was lower in the β ray 10 Gy multiple irradiation group,β ray 7 Gy multiple irradiation group and 1% CsA eye drops group in comparison with that in model group, and the results from Western blot showed the same pattern as RT-PCR. Conclusions Low dose irradiation of 90Sr-90Y ophthalmic applicator inhibits CNV formation after alkali burn. The study provide a new understanding of the irradiation for the treatment of CNV.

ABSTRACT

<p><b>OBJECTIVE</b>The aim of this study was to measure and analyze the anatomic characteristics of the pulp chamber of Chinese permanent anterior teeth radiographically.</p><p><b>METHODS</b>There were 137 human permanent anterior teeth to be used. The crown length, crown width and crown thickness were measured by vernier caliper. Labial-lingual and mesial-distal radiographs were obtained by parallel technique and Photoshop 8.0 was adopted for measuring the largest labial-lingual length, the largest mesial-distal length, the pulp chamber height and the distance between the lingual surface of the crown and the corresponding wall of the pulp chamber; the ratios of the largest labial-lingual length to the crown thickness, the largest mesial-distal length to the crown width and the pulp chamber height to the crown length were calculated. Finally the coefficients of variance were caculated.</p><p><b>RESULTS</b>(1) The ratios of the largest mesial-distal length to the crown width of maxillary and mandibular canines were statistically different from those of the other anterior teeth. There was not statistically significant difference for those ratios either between maxillary and mandibular canines or among incisors. (2) Indexes of the pulp chamber had higher variance than those of the external morphology. (3) The distance between the lingual surface of the crown and the corresponding wall of the pulp chamber had the smallest coefficient of variance. (4) The largest mesial-distal length of the pulp chamber had the largest coefficient of variance, except for the mandibular canines.</p><p><b>CONCLUSION</b>This study provided anatomical basis for clinical work; the distance between the lingual surface of the crown and the corresponding wall of the pulp chamber could be used as a depth reference for the endodontic access, and the access should not be extended excessively in mesial-distal direction.</p>

ABSTRACT

This study examined the secretion capacity of the gonadal and adrenal cortex systems and the morphology of the adrenal cortex in male rats treated with Tu-chung (Eucommia ulmoides OLIV) extract, the main component of Tu-chung extract, geniposide, or both agents during exercise load testing.<BR>Twenty-four 4-week-old male Wistar rats were used. The rats were divided into four groups : those treated with Tu-chung extract and geniposide (n=7), those treated with Tu-chung extract (n=6), those treated with geniposide (n=7) and a control group treated with distilled water. The dose of each agent was 0.1 ml/100g body weight. The agents were administered orally for 25 days. For the exercise load test, a treadmill for small animals was used, with a tilting angle set at 0. Exercise load testing was performed for 30 min (2-min warm up and 28-min running) daily for 25 days. The running speed was 20 m/min for the first 5 days, and then increased by 5 m/min every 5 days.<BR>The following results were obtained.<BR>1. The relative weight of the adrenal gland (gland weight/100 g body weight) in the group treated with Tu-chung extract and geniposide was significantly higher than that in the group treated with geniposide or the control group (p<0.01) . The relative weight of the adrenal gland in the group treated with Tu-chung extract was significantly higher than that in the group treated with geniposide or the control group (p<0.01, p<0.001) . Furthermore, the relative gland weight in the group treated with geniposide was significantly higher than that in the control group (p<0.05) .<BR>2. In the group treated with Tu-chung extract and geniposide, the group treated with Tu-chung extract and the group treated with geniposide, the relative gland weight of the testis (testis weight/100g body weight) was significantly higher than that in the control group (p<0.001) . However, there were no significant differences among the three groups.<BR>3. The 24-h urinary excretion of 17-ketosteroid (17-KS) in the group treated with Tu-chung extract and geniposide was significantly higher than that in the group treated with Tu-chung extract, the group treated with geniposide or the control group (p<0.01, p<0.01, p<0.001) . In the group treated with geniposide, the 24-h urinary excretion of 17-KS was significantly higher than that in the control group (p<0.05) .<BR>4. In the three groups treated with Tu-chung extract and/or geniposide, serum testosterone levels were significantly higher than those in the control group (p<0.001, p0.05, p<0.05) .<BR>5. Concerning the morphology of the adrenal cortex, the thickness of the reticular layer of the adrenal cortex was most markedly increased in the group treated with Tu-chung extract and geniposide, followed in order by the group treated with Tu-chung extract, the group treated with geniposide and the control group.<BR>Administration of Tu-chung extract and the main component of Tu-chung extract, geniposide, during exercise load testing significantly increased the weights of the adrenal gland and testis, and promoted testosterone secretion in the adrenal cortex reticular layer and testis.<BR>These findings suggest that geniposide plays an important role in the pharmacological action of Tu-chung.

ABSTRACT

We evaluated the effect of Tu-Chung (<I>Eucommia ulmoides</I> OLIV.) extract on anabolic action in castrated exercise and non-exercise rats in which the effects of male sex hormone from the testis were excluded. Castration was performed on 32 male Wistar rats aged 4 weeks. The rats were then divided into 4 groups: a non-exercise group treated with Tu-Chung extract (non Ex. Tu-chung G, n = 8), on exercise group treated with the extract (Ex. Tu-Chung G, n = 8), a non-exercise control group not treated with the extract (non Ex. Cont. G, n8), = and an untreated exercise control group (Ex. Cont. G, n=8) .<BR>The Tu-Chung extract was administered orally at a dose of 1g/kg body weight once daily for 4 weeks. Distilled water was given by a similar method to the control groups. As the exercise load, the rats exercised on an animal treadmill at a starting speed of 20 m/min with an increase of 10 m/min every week for 30 min without rest daily for 4 weeks.<BR>The following results were obtained:<BR>1. The relative weight of the adrenal gland (gland weight/100 g body weight ) after 4 weeks was significantly higher in the non Ex. Tu-Chung G than in the non Ex. Cont. G (p<0.001) and in the Ex. Tu-Chung G than in the Ex. Cont. G or the non Ex. Cont. G (p<0.001 each) .<BR>2. The relative weight of the kidneys (kidney weight/100 g body weight) after 4 weeks was significantly higher in the non Ex. Tu-Chung G than in the non Ex. Cont. G (p<0.001) and was slightly higher in the Ex. Tu-Chung G than in the Ex. Cont. G.<BR>3. The relative weight of the musculus levator ani (muscle weight/100g body weight) after 4 weeks was significantly higher in the non Ex. Tu-Chung G than in the non Ex. Cont. G (p<0.001) and also in the Ex. Tu-Chung G than in the Ex. Cont. G (p<0.001) .<BR>4. The 17-KS level in a 24h urine sample after 4 weeks was significantly higher in the non Ex. Tu-Chung G than in the non Ex. Cont. G or the Ex. Tu-Chung G (p<0.001 each) and also in the Ex. Tu-Chung G than in the Ex. Cont. G (p<0.001) .<BR>5. The total urinary nitrogen level after 4 weeks was significantly lower in the non Ex. Tu-Chung G than in the non Ex. Cont. G (p<0.001) or the exercise group treated with the extract (p<0.05) and also in the Ex. Tu-Chung G than in the Ex. Cont. G (p<0.05) .<BR>These results suggest that administration of Tu-Chung extract significantly increases the relative weight of the adrenal gland, enhances androgen secretion from the reticular layer of the adrenal cortex, and promotes protein anabolic action in castrated rats. In addition, this extract appears to increase the adaptation ability of the adrenal cortex to the stress caused by exercise.
