ABSTRACT

Objective: To investigate the effect of phosphatidylinositol-4-phosphate 3-kinase, catalytic subunit type 2 beta (PIK3C2B) gene silencing on proliferation and invasion of human prostate cancer PC-3 cells in vitro, and to explore its possible mechanism. Methods: PIK3C2B-siRNAs including PIK3C2B-siRNA1 and PIK3C2B-siRNA2 and the negative control (NC)-siRNA were designed and synthesized, then transfected into PC-3 cells by LipofectAMINE 2000. The expression levels of PIK3C2B mRNA and protein were detected by real-time fluorescent quantitative-PCR and Western blotting, respectively. The proliferation and invasion abilities of PC-3 cells after transfection with PIK3C2B-siRNA were detected by MTT method and Transwell cell invasion assay, respectively. The expression levels of protein kinase B (PKB, Akt) signal pathway-related proteins Akt and phospho-Akt in PC-3 cells after transfection with PIK3C2B-siRNA were detected by Western blotting. Results: The expression levels of PIK3C2B mRNA and protein in PC-3 cells after transfection with PIK3C2B-siRNA were down-regulated. The inhibition effect of PIK3C2B-siRNA2 was stronger (P < 0.01). The proliferation abilities of PC-3 cells at 48 and 72 h after transfection with PIK3C2B-siRNA2 were significantly inhibited as compared with those of the cells transfected with NC-siRNA or not transfected with any siRNAs (all P < 0.05); the invasion ability of PC-3 cells was also decreased after transfection with PIK3C2B-siRNA2 (all P < 0.05). The expression level of Akt protein in PC-3 cells transfected with PIK3C2B-siRNA2 had no significant change, but the expression level of phospho-Akt protein was down-regulated (P < 0.05). Conclusion: PIK3C2B may promote the proliferation and invasion of prostate cancer PC-3 cells in vitro by activating Akt signaling pathway.
