ABSTRACT

Objective Young and middle-aged people have become the fastest growing group of diabetes, but the compliance of regular exercise is poor. It is urgent to formulate effective intervention strategies or measures to promote the change of their behavior. The article is Objective ly based on the attitude-social influence-self efficacy model, to investigate the stages distribution and influencing factors of regular exercise behavior in young and middle-aged patients with type 2 diabetes.Methods A convenience sampling was used to select 430 young and middle-aged type 2 diabetic patients from the department of endocrinology in two Nanjing 3A grade hospitals. A cross-sectional survey was conducted with a general questionnaire, a regular exercise behavioral stage questionnaire, a diabetes behavioral attitude and social impact scale, and a diabetes self-efficacy scale.Results The proportion of regular exercise behavior in young and middle-aged patients with type 2 diabetes is as follows. There were 14.9% patients at the precontemplation, 25.8% at the contemplation,15.8% at the preparation, 12.3% at the action, 31.2% at the maintenance. Logistic regression analysis showed that on-the-job (OR=1.77), accepting health education (OR=0.49), HbA1c level (OR=0.78), attitude (OR=5.32), subjective norm (OR=2.43), social support (OR=1.87) and self-efficacy (OR=1.28) are main factors to affect the stages of physical activity (P<0.05) .Conclusion Most of the young and middle-aged patients with type 2 diabetes are in the pre-action stages of regular exercise behavior, which needs to be improved, and many factors can affect the stages of regular exercise behavior. Clinical medical staff should provide individualized interventions for these patients with reference to the characteristics of different behavioral stages of patients.

ABSTRACT

AIM: To establish an experimental model of high intraocular pressure in mice by laser photocoagulation and to prepare for future research. METHODS: Experimental model of high intraocular pressure was induced unilaterally in 44 C57BL/6 mice. The fellow eye served as a control. TONO-PEN AVIA Tonomter was used to measure intraocular pressure (IOP) to guarantee IOP value at 1, 2, 4, 8wk. Slit-lamp biomicroscopy was performed throughout the period and the structural changes were assessed histologically. And then, their eyes were enucleated, postfixed, cryoprotected, and embedded in optimal cutting temperature medium. After hematoxylin and eosin stain ( HE stain ) , cryosections of the retina were observed under light microscope. TdT-mediated biotin-dUTP nick end labeling ( TUNEL ) was performed on the retinal sections to determine apoptosis rate. RESULTS: IOP of laser-treated eyes was significantly higher than that of control eyes from 1-8wk (P CONCLUSION: The laser photocoagulation of limbus causes chronic elevation of IOP and this method may be a promising experimental model for the investigation of biological mechanisms of glaucomatous retinal ganglion cell damage.

ABSTRACT

Objective To investigate the status of Ehrlichia (E.)chaffeensis and A naplasma (A.) phagocytophilum infection among farming populations and domestic animals in the rural area of Beijing,China.Methods Blood samples from 562 farmers and 163 blood samples including 90 goats,71 ox and 2 dogs,were collected.Specificity of IgG antibodies against E.chaffeensis and A.phagocytophilum were tested by micro-indirect immunofluorescent assay (mIFA).16S rRNA genes of A.phagocytophilum were amplified from the domestic animal blood samples and products were sequenced and analyzed by nested PCR.Results The positive rates of E.chaffeensis and A.phagocytophilum antibody were 16.5% and 14.0% respectively for farmers.The total positive rates of A.phagocytophilum were 2.3% and 0 for both goats and oxen respectively.No antibody was found for the 2 tested dogs.The PCR positive rates were 48.9% and 23.9% for goats and oxen respectively.Three dominant varieties of A.phagocytophilum were demonstrated in goats and oxen.Conclusion The prevalence rates of E.chaffeensis and A.phagocytophilum were identified in the rural areas of Beijing.

ABSTRACT

<p><b>OBJECTIVES</b>To investigate the function and possible mechanisms of PIAS3 expression on the invasion of TJ905 cells.</p><p><b>METHODS</b>PIAS3 overexpression vectors were constructed and PIAS3 siRNA were chemically synthesized, which were separately transfected into TJ905 cells for upregulation or downregulation of PIAS3 expression levels in TJ905 cells. After that, the invasive effects of TJ905 cells were measured by Transwell assay, and the expression of PIAS3, tissue inhibitor of metalloproteinases (TIMP)3, matrix metalloprotease (MMP)-2, and MMP-9 were identified by Western blot.</p><p><b>RESULTS</b>In vitro transfection efficiency of plasmids and oligonucleotides were separately 85.3% ± 3.1% and 95.1% ± 2.9%. PIAS3 overexpression plasmid transfection in vitro could effectively improve the expression of PIAS3 protein in TJ905 cells and inhibit the invasion of TJ905 cells (P < 0.05), and cell penetration ratio reduced from 87.9% ± 9.3% to 37.3% ± 7.9% compared with control group, while it upregulated TIMP3 and downregulated MMP-2, MMP-9 protein expression (P < 0.05); PIAS3 siRNA transfection could inhibit the PIAS3 protein expression of TJ905 cells and promote the invasion of TJ905 cells (P < 0.05), and cell penetration ratio increased from 83.9% ± 7.1% to 93.2% ± 3.1% compared with control group, while it downregulated TIMP3 and upregulated MMP-2, MMP-9 protein expression (P < 0.05).</p><p><b>CONCLUSION</b>PIAS3 expression is closely related to the invasion properties of glioma TJ905 cells.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the mRNA expression of transferrin receptor 1 (TfR1) and iron regulatory protein 1 (IRP1) in the full-term placenta from different maternal iron status, and explore the mechanism of placental iron transport and regulation.</p><p><b>METHODS</b>The mRNA level of TfR1 and IRP1 in full-term placentae was detected by reverse transcription polymerase chain reaction (RT-PCR) in normal group (N), iron deficiency group (ID) and iron deficiency anemia group (IDA).</p><p><b>RESULTS</b>(1) The expression of TfR1 mRNA in N group was 0.4813 +/- 0.1891, in ID group was 0. 6647 +/- 0.2788, and in IDA group was 0.9767 +/- 0.2858. There was significant difference between IDA group and N group or ID group (t = 0.002, P < 0.01 or t = 0.028, P < 0.05), and was no difference between ID group and N group (t = 0.117, P > 0.05). (2) The expression of IRP1 mRNA in N group was 0.2616 +/- 0.0785, in ID group was 0.3696 +/- 0.1801, and in IDA group was 0.3971 +/- 0.0902 and was no difference among the three groups (F = 1.845, P = 0.179).</p><p><b>CONCLUSIONS</b>The expression of TfR1 mRNA is increased when maternal iron deficiency progressed while there is no change in the expression of IRP1 mRNA in the placentae of TfR1 mRNA indicated that IRP1 takes part in the regulation of placenta iron transport.</p>

ABSTRACT

Objective To investigate the expressions of transferrin receptor (TfR) mRNA, ferritin(Fn) mRNA and iron regulatory proteinl (IRP-1) mRNA in the placentas complicated with fetal iron deficiency(ID). Methods Depending on the cord SF,the subjects were divided to fetal ID group and fetal iron sufficient( IS) group. TfR mRNA, Fn mRNA and IRP - 1 mRNA in placentas were measured by RT - PCR. Results 1. The expression of TfR mRNA in ID group was 1.10 ? 0. 26, it was significantly higher than that in IS group (t=0.028 P0.05) ;4. TfR mRNA and Fn mRNA correlated with fetal IS respectively. Conclusion Fetal ID induces highly expressed TfR mRNA and lower level of Fn mRNA to furthest satisfy the iron need for fetus.
