ABSTRACT

Objective:To investigate the effect of Jianpi Bufei prescription (JPBFP) on airway inflammation, airway hyperresponsiveness (AHR), and cyclic adenosine monophosphate (cAMP) signaling pathway activity in ovalbumin (OVA)-sensitized and challenged juvenile asthma rats. Method:Seventy-five male SD rats were randomly divided into a blank group (n=15) and an experimental group (n=60). The rats in the experimental group were sensitized by aluminum hydroxide gel containing 0.2% OVA and stimulated by aerosol inhalation of normal saline containing 1% OVA to induce an asthma model, followed by assignment into the following groups: a model group (n=15), a JPBFP group (n=15, 8.37 g·kg-1·d-1), an aminophylline group (n=15, 40 mg·kg-1·d-1), and a dexamethasone group (n=15, 0.1 mg·kg-1·d-1). AHR was detected by the pulmonary function analyzer, changes in inflammatory cells by white blood cell (WBC) count and differential blood count in bronchoalveolar lavage fluid (BALF), and pathological changes of lung tissues by hematoxylin-eosin (HE), Masson, and periodic acid-schiff (PAS) staining. The interleukin (IL)-4, IL-5, IL-13, interferon (IFN)-γ, and tumor necrosis factor (TNF)-α levels in serum and the cAMP level in plasma were tested by the enzyme-linked immunosorbent assay (ELISA). Protein kinase A (PKA) expression in lung tissues was detected by immunohistochemistry. The cAMP-response element-binding protein (CREB) mRNA and protein expression in lung tissues was detected by the real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) and Western blot. Result:Compared with the blank group, the model group showed increased lung resistance, decreased pulmonary compliance (P<0.05), elevated WBC count and proportion of eosinophils in BALF (P<0.05), up-regulated levels of IL-4, IL-5, IL-13, and TNF-α in peripheral blood, declining IFN-γ level (P<0.01), severe pathological changes of lung tissues, dwindled cAMP, and down-regulated PKA and CREB expression (P<0.01). Compared with the model group, JPBFP inhibited AHR, reduced WBC count and proportion of eosinophils in BALF and lung resistance (P<0.05), improved pathological changes of lung tissues, increased pulmonary compliance, and up-regulated cAMP in serum and PKA and CREB expression in lung tissues (P<0.01). Conclusion:JPBFP can improve AHR, inhibit airway inflammation, and alleviate lung injury in asthma rats. Its mechanism may be related to the up-regulation of the activity of the cAMP/PKA/CREB signaling pathway.

ABSTRACT

Objective To investigate the effect of cysteine protease inhibitor derived from S chistosoma japonicum(SjCys-tatin)on dextran sodium sulfate(DSS)-induced acute ulcerative colitis in mice. Methods Eighteen C57BL/6 mice were ran-domly divided into three groups:a control group treated with PBS(Group A),a DSS-induced-colitis group treated with PBS(Group B),and a DSS-induced-colitis group treated with SjCystatin(Group C).Colitis was induced in mice by giving 3%DSS orally for 7 days.During this period, the mice were daily injected with 10μg of SjCystatin or PBS only as a control intraperitone-ally.The mice were monitored daily for their clinical manifestations and given scores based on disease activity index(DAI).The severity of colonic inflammation was monitored by the macroscopic score and pathological change. The cytokine profile including TNF-α,IL-4,IL-6 and IL-10 in the supernatants of colon homogenate was detected by ELISA.Results Compared with Group A(0.50 ± 0.28),the DAI score increased significantly in Group B(9.30 ± 1.30)(F=86.86,P<0.01),with remarkable path-ological damages seen in colon tissues.and the levels of TNF-α and IL-6 were(321.33±67.01)and(403.58 ±180.51)pg/mL.The DAI score significantly reduced in Group C(6.67±1.57)as compared to Group B(F=86.86,P<0.01),with improve-ments in the macroscopic and microscopic pathology in mouse colon specimens. As compared to Group B,the levels of TNF-α [(188.14 ± 40.14)pg/mL] and IL-6 [(209.71 ± 48.47)pg/mL] significantly decreased(F=17.46 and 9.89,both P<0.01).Con-clusion SjCystatin has a significantly inhibitory effect for alleviating DSS-induced acute ulcerative colitis in C57BL/6 mice.

ABSTRACT

<p><b>OBJECTIVES</b>To investigate the effect of magnetron sputtered niobium nitride (NbN) on the bonding strength of commercially pure cast titanium (Ti) and low-fusing porcelain (Ti/Vita titankeramik system).</p><p><b>METHODS</b>Sixty Ti specimens were randomly divided into four groups, group T1, T2, T3 and T4. All specimens of group T1 and T2 were first treated with 120 microm blasted Al2O3 particles, and then only specimens of group T2 were treated with magnetron sputtered NbN film. All specimens of group T3 and T4 were first treated with magnetron sputtered NbN film and then only specimens of group T4 were treated with 120 microm blasted Al2O3 particles. The composition of the deposits were analyzed by X-ray diffraction (XRD). A universal testing machine was used to perform the three-point bending test to evaluate the bonding strength of Ti and porcelain. The microstructure of NbN, the interface of Ti-porcelain and the fractured Ti surface were observed with scanning electron microscopy (SEM) and energy depressive spectrum (EDS), and the results were compared.</p><p><b>RESULTS</b>The XRD results showed that the NbN deposits were cubic crystalline phases. The bonding strength of Ti and porcelain in T1 to T4 group were (27.2+/-0.8), (43.1+/-0.6), (31.4+/-1.0) and (44.9+/-0.6) MPa. These results were analyzed by one-way analysis of variance and differences between groups were compared using least significant difference test. Significant inter-group differences were found among all groups (P<0.05). The results of SEM showed that with treatment of Al2O3 or NbN, alone, pre-cracks were found in the interface of Ti-porcelain, while samples treated with both Al2O3 and NbN had better bond. EDS of Ti-porcelain interface showed oxidation occurred in T1, T2 and T3, but was well controlled in T4.</p><p><b>CONCLUSIONS</b>Magnetron sputtered NbN can prevent Ti from being oxidized, and can improve the bonding strength of Ti/Vita titankeramik system. Al2O3 blast can also improve the bonding strength of Ti/Vita titankeramik system.</p>
