ABSTRACT

Objective: To investigate the relationship between hemoglobin and serum uric acid in adults with various glucose metabolism status. Methods: The demographic data and biochemical indicators of the adult population who had received physical examination in the Second Medical Center of the PLA General Hospital from January 2018 to December 2021 were collected. The subjects were divided into two groups according to the level of serum uric acid: the normal uric acid group and the hyperuricemia group. The relationship between hemoglobin (stratified into four levels of Q1 to Q4 by the quartile) and serum uric acid was quantified by using Pearson correlation and logistic regression analysis. The effects of age and glucose metabolism status on the relationship between hemoglobin and serum uric acid were analyzed. Results: A total of 33 183 adults were enrolled with age (50.6±10.0) years. The level of hemoglobin in the normal uric acid group (142.61±14.24) g/L was significantly lower than that in the hyperuricemia group [(151.79±11.24) g/L, P<0.001]. Univariate Pearson correlation analysis showed that hemoglobin was positively associated with serum uric acid (r=0.444, P<0.001). After adjusting for related confounding factors, multivariate logistic regression analysis showed that hemoglobin was associated with serum uric acid, and the OR values (95%CI) of hemoglobin Q2 to Q4 group were 1.29 (1.13-1.48), 1.42 (1.24-1.62) and 1.51 (1.32-1.72), respectively (Ptrend<0.001) when compared with hemoglobin Q1 group. Subgroup analysis and hierarchical interaction analysis suggested that with the increase of hemoglobin, the serum uric acid in the age<60 years subgroup, normal glucose subgroup and prediabetes subgroup increased gradually (Ptrend<0.05 and Pinteraction<0.001). Conclusion: The association between hemoglobin and serum uric acid in adults is affected by age and glucose metabolism status.

ABSTRACT

OBJECTIVE@#To investigate serum thyroid stimulating hormone (TSH) level and its changes with age in apparently healthy Chinese elderly population and analyze the differences between TSH levels detected using Roche and Snibe electrochemiluminescence immunoassay analyzers.@*METHODS@#General clinical data and frozen fasting serum samples were collected from 5451 apparently healthy Chinese elderly individuals (> 60 years) from 10 centers in different geographic regions in China. Thyroid function indexes including TSH level were detected using Roche and Snibe electrochemiluminescence immunoassay analyzer, and the median (2.5% and 97.5% quantiles) TSH level was calculated. The variations of TSH level among the participants with geographic regions, gender, and age (with an interval of 5 years) were analyzed to determine the influence of these factors on TSH level.@*RESULTS@#The reference ranges of serum TSH level established using Roche and Snibe electrochemiluminescence immunoassay analyzers were 0.42-9.47 mU/L and 0.36-7.98 mU/L, respectively, showing significant differences between the two methods (P < 0.001). The TSH levels measured at two centers in Western China were significantly higher than those at the other centers (P < 0.05). In elderly male population, serum TSH level tended to increase with age, which was not observed in elderly female population. At the age of 60-75 years, women generally had higher serum TSH level than men, but this difference was not observed in the population beyond 75 years.@*CONCLUSION@#In elderly population, serum TSH level can vary with geographic region, gender, and age, but there was no need for establishing specific reference ranges for these factors. The differences between different detection methods should be evaluated when interpreting the detection results of TSH level.

ABSTRACT

Objective To investigate the relationship between promotion of osteogenic differentiation of adipose-derived stem cells (ADSCs) by exosomes secreted by ADSCs and the Wnt/β-catenin signaling pathway. Methods ADSCs were extracted in the Plastic Surgery Department of the First Hospital Affiliated to China Medical University. Exosomes were extracted from ADSCs induced to differentiate into osteoblasts. The ADSCs were separated into four groups and used for measuring activity of alkaline phosphatase (ALP) by using enzyme-labeled assay, formation of calcium nodes by Alizarin red staining, and expression of β-catenin by Western blotting. Results ADSC-derived exosomes promoted differentiation of ADSCs into osteoblasts by upregulating the expression of β-catenin. Addition of Dickkopf-related protein 1 (DKK1), an inhibitor of Wnt/β-catenin pathway, inhibited expression of β-catenin which in turn resulted in reduced potential of ADSCs for osteogenic differentiation. Conclusion The osteogenic differentiation of ADSCs can be promoted by exosomes derived from ADSCs via the Wnt/β-catenin signaling pathway.

ABSTRACT

Objective@#To evaluate the clinical effect of subcutaneous pedicle skin flap for repairing skin and soft tissue defect in the philtrum area.@*Methods@#From November 2015 to July 2016, 6 patients with pigmented nevus and basal cell carcinoma were treated with regional subcutaneous pedicled flaps. The functional and aesthetical outcomes were evaluated.@*Results@#Follow-up was performed at 6-12 months postoperatively. All 6 flaps survived. The wounds healing was good, and the postoperative appearance was satisfactory.@*Conclusions@#The kite subcutaneous pedicled flap is an ideal method for repairing the skin and soft tissue defects in philtrum area, and it can produce satisfactory clinical results.

ABSTRACT

Objective@#To study the protective effect of adipose-derived stem cell conditioned medium against ultraviolet radiation B (UVB)-induced photoaging and replicative senescence in human dermal fibroblasts (HDFs).@*Methods@#HDFs were cultured and passaged in vitro, and treated with ADSC-CM after being irradiated once with UVB. The mRNA expression of CollagenⅠ(Col1), CollagenⅢ(Col3) and Elastin were detected by real-time PCR to define the anti-aging effects of ADSC-CM. The protein expressions of phosphorylated c-Jun N-terminal kinase (p-JNK) and p-p38 were detected by Western blot.@*Results@#①Both successive passage and UVB irradiation reduced the expression of Col1, Col3 and Elastin in HDFs.②ADSC-CM inhibited the reduction of Col1, Col3 and Elastin protein expressions induced by successive passage and UVB. ③ADSC-CM activated p38 and JNK pathway. Downregulation of p38 MAPK by SB203580 decreased the mRNA expression of Col1, Col3, and Elastin. Inhibiting JNK by SP600125 did not induce significant ECM changes.@*Conclusions@#Both intrinsic and extrinsic stimuli can decrease the expression of collagen and elastin, common markers of skin aging in HDFs, and ADSC-CM can attenuate the above decreasing and promoting the expression of ECM by p38 signaling pathway.

ABSTRACT

Objective To explore the temporal and spatial distribution characteristics of C infectious diseases in Changde City from 2010 to 2015,grasp the situation of high priority areas so as to provide theoretical basis for prevention and control.Methods Spatio-temporal scanning was performed by using SaTScan software,and the spatial and temporal clustering regions of C infectious diseases were presented by ArcGIS software.Results The prevalence of C infectious diseases in Changde City was higher than that in other times in 2010 (RR =4.16,P =0.000).(LLY =497.019,RR =1.48,P < 0.001).There was a most likely clustering area in the Wuling area of Changde,and the incidence of C infectious diseases in this area was higher than that in other areas,and the difference was statistically significant.Simple spatial scanning analysis and space-time scan analysis were also found in South Central Deshan Development Zone (LLR =277.131,RR =1.77,P <0.001) and Northern Linli county (LLR =136.683,RR =1.27,P < 0.001) are two secondary gathering area,where the actual number of cases were much more than the theoretical number of cases,the difference was statistically significant.In addition,there are other regions of the existence of sub-aggregation state.Conclusions Spatio-temporal scanning analysis method can be applied to the high incidence of infectious diseases in Changde City.Combined with geographic information system that we can show the incidence of disease and disease in a more intuitive and comprehensive way,which can provide a scientific reference for the future prevention and control of the disease.

ABSTRACT

Objective Comprehensive evaluated the characteristics and differences of prevalence of hypertension in cold and non-cold areas,to provide evidence-based prevention strategies for establishment and perfection hypertension prevention and control suitable for China's national conditions in cold area.Methods Northeast China (Heilongjiang,Jilin,Liaoning),Inner Mongolia,Ningxia,Gansu,Qinghai,Tibet and Xinjiang were classified as cold area,and the rest were non-cold area.Use hypertension as a search term,literatures on the prevalence of hypertension were retrieved in WanFang Data,CqVip,CNKI,SinoMed and Pubmed database from January 1 1995 to May 1 2017,then the relevant data were screened and extracted.Heterogeneity was estimated and corresponding effect model was selected.Publication bias was evaluated by funnel plot,Egger test and Begg test.Comprehensive meta-analysis V2 was used for Meta analysis,stratified analysis of age,gender,province and region was did.Results In China,the pooled prevalence of hypertension was 27.00%.The pooled awareness rate of hypertension was 50.80%,the pooled treatment rate of hypertension was 40.40%,and the pooled control rate was 6.80%.The pooled prevalence in cold area was 29.70% and 26.30% in non-cold area,respectively,the difference of prevalence between cold and non-cold areas was significant (P < 0.05).After the stratified analysis of age and gender,the pooled prevalence in men and women in cold area had a increased trend than that in non-cold area;the pooled prevalence in < 30,40-,50-,and 60-< 70 years old groups in cold area was higher than that in non-cold area.The pooled prevalence was the highest in Tibet (40.70%) and north and northeast China area (30.40%,29.20%),and the lowest in Hainan (16.70%) and southern China area (20.70%).Conclusions In China,the pooled prevalence of hypertension is 27.00%,which is higher in cold area than non-cold area.The pooled prevalence is the highest in Tibet and north and northeast China,and the lowest in Hainan and Southern China.

ABSTRACT

Objective To improve the descriptions of health information network resources by analyzing the influen-cing factors of information retrieval.Methods The top 10 papers retrieved on 12 Websites in Chinese and English engines according their titles, keywords and descriptions in the HEAD file of HTML source code with 5 biomedical hot words as the search terms were comparatively analyzed and scored.Results The score of Websites in Chinese and English engines was higher than that of medical and health information network, the score of Chinese network information resources was higher than that of English network information resources .Conclusion The current de-scription of network health information resources is poor .Only by strengthening health information network con-struction and improving the description of health information resources , can more accurate health information be provided for the users.

ABSTRACT

Activation of the innate immune system requires recognition of pathogen-associated molecular patterns, such as NOD-like receptors. The NOD-like receptor protein 3 (NLRP3) inflammasome is involved in induction of the pro-inflammatory cytokine, IL-1beta, and subsequent inflammatory responses. NLRP3 inflammasome plays important roles in the inflammatory and innate immune responses associated with autoimmune/inflammatory syndrome. However, analysis of the tissue distribution and expression profiles in BALB/c mice is still incomplete. In this study, we investigated the tissue distribution and expression pattern of NLRP3 in BALB/c mice to further elucidate its function in innate immunity in this commonly used laboratory animal model. NLRP3 mRNA expression levels and tissue distribution of the protein were investigated by real-time quantitative PCR and immunohistochemical analyses, respectively. NLRP3 mRNA expression was higher in the kidney and inguinal lymph nodes than in other tissues. Cytoplasmic expression of NLRP3 was detected in the epithelial reticular cells of the spleen and thymus, lymphocytes in the inguinal lymph nodes, cardiac muscle cells, cerebral cortex neurons, alveolar macrophages, renal tubule cells and liver sinusoidal endothelial cells. The results of this study will assist investigators in interpreting site-specific functions and roles of NLRP3 in inflammatory responses.

ABSTRACT

Objective To investigate the variation and significance of the expressions of NF-kB, inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in the renal tissue of insulin-resistant rat. Methods Thirty healthy male Wistar rats were bred since 2 months old, and they were randomly divided into normal control (NC) group (n=15) and insulin-resistant (IR) group (n=15). Insulin resistance rat model was reproduced by feeding with high fat and sucrose diet. Hyperinsulinemic-euglycemic clamp test was used to verify the reproduction of the model. The kidneys of the rats were obtained after the successful reproduction of the model. The change in renal histology was observed by HE staining, and the expressions of iNOS and COX-2 in the kidneys were detected by immunohistochemistry staining. The mRNA expressions of NF-κB, iNOS and COX-2 in the kidneys were assessed with RT-PCR. DNA binding activity of NF-κB in the rat's kidney was assessed with electrophoretic mobility shift assay (EMSA). Results HE staining showed that, compared with NC group, the early lesions of the renal tissue, such as glomerular enlargement and mesangial region broadening, could be seen in IR group. Immunohistochemical staining showed that the positive expressions of iNOS and COX-2 were up-regulated significantly in IR group than in NC group (P<0.05). RT-PCR revealed that the expressions of NF-κB mRNA, iNOS mRNA and COX-2 mRNA in renal tissue were significantly higher in IR group than in NC group (P<0.05). EMSA showed that the binding activity of NF-κB in renal tissue increased significantly in IR group than in NC group (P<0.05). Conclusion NF-κB activation is present in the kidney tissue in the insulin resistance rat, which may upregulate the expression of downstream target gene iNOS and COX-2, resulting in damage to kidney tissue. The activation of NF-κB maybe one of the initiative factors that lead to the kidney lesion of the insulin resistance rat.

ABSTRACT

In this paper we study the scaling behavior of nucleotide cluster in 11 chromosomes of Encephalitozoon cuniculi Genome. The statistical distribution of nucleotide clusters for 11 chromosomes is characterized by the scaling behavior of P(S) proportional, variant e(-alphaS), where S represents nucleotide cluster size. The cluster-size distribution P(S(1)+S(2)) with the total size of sequential C-G cluster and A-T cluster S(1)+S(2) were also studied. P(S(1)+S(2)) follows exponential decay. There does not exist the case of large C-G cluster following large A-T cluster or large A-T cluster following large C-G cluster. We also discuss the relatively random walk length function L(n) and the local compositional complexity of nucleotide sequences based on a new model. These investigations may provide some insight into nucleotide cluster of DNA sequence.
