ABSTRACT

Objective To optimize the conditions of extraction process of polysaccharides from seeds of Toona sinensis; To preliminary determinate the antioxidant activity in vitro. Methods The content of polysaccharides from Toona sinensis was chosen as evaluation index; the extraction time, material liquid ratio, and extraction times were chosen as factors; L9(34) orthogonal design was used to optimize extraction process; antioxidant activity of polysaccharides from Toona sinensis was investigated by the total reducing power and 1,1-diphenyl-2-picryhydrazyl (DPPH) method. Results From the statistical data of processing, the optimal extracting conditions were as follows:liquid to material was 1:12, extraction time was 120 min, and extracted for 3 times. Polysaccharides from seeds of Toona sinensis had certain reducing capacity and had better scavenging ability on DPPH? and the scavenging rate significantly increased with the concentration. The effect was simulated by curve equation. Conclusion The process is simple, feasible, stable and reliable and can be used for the small-scale study on extraction of polysaccharides from seeds of Toona sinensis. Polysaccharides from seeds of Toona sinensis have antioxidant activity.

ABSTRACT

OBJECTIVE@#To investigate the bionomics of the olfactory ensheathing cells (OECs) of human olfactory mucosa.@*METHOD@#To separate and cultivate the OECs of human and rat olfactory mucosa. To observe the cell growth, cell grouping and cell migration in vitro of the two types of OECs.@*RESULT@#Successfully separated and cultivated the OECs of human and rat olfactory mucosa. OECs of the human and rat olfactory mucosa had the similar cell growth, cell grouping and cell migration ability in vitro.@*CONCLUSION@#OECs of the human and rat olfactory mucosa have the similar bionomics in vitro, as a result, OECs of the human olfactory mucosa could be a reliable source of cell transplant for nerve injury.

ABSTRACT

OBJECTIVE@#To explore the expression of mycoplasma monoclonal antibody PD4 in different laryngeal carcinoma tissues and their clinical significance.@*METHOD@#Using immunohistochemistry methods to detect 256 cases of laryngeal tissues, including 105 cases of laryngeal carcinoma tissues, 31 cases of neck metastasis lymph node tissues, 18 cases of precarcinoma tissues, 53 cases of vocal cord polyps, 20 cases of normal tissues adjacent to laryngeal carcinoma and 29 cases of normal tissues around laryngeal carcinoma.@*RESULT@#(1) The positive rate of PD4 was 44.76% in laryngeal carcinoma, which was much higher than that in other laryngeal tissues (P < 0.01 or P < 0.05). (2) The PD4 expression in laryngeal carcinoma was higher in the third and fourth stages than in the first and second stages, higher with cervical metastasis than those without cervical metastasis and higher in the third and second histological grade than in the first histological grade (P < 0.01 or P < 0.05). (3) To 3 and 5 years survival, PD4 positive cases had lower chance than PD4 negative cases (P < 0.01).@*CONCLUSION@#These results implicate that the occurrence, development and prognosis of laryngeal carcinoma may be closely related to mycoplasma infection.

ABSTRACT

OBJECTIVE@#To study the expression of PD4, CD44 and PCNA proteins in laryngeal carcinoma and their relationships with the pathogenesis, development and prognosis of laryngeal carcinoma.@*METHOD@#Immunohistochemistry was used to study 140 cases of laryngeal carcinoma tissues, 25 cases of precarcinoma tissues, 36 cases of vocal cord polyps and 13 cases of normal tissues adjacent to laryngeal carcinoma.@*RESULT@#1. The positive rates of PD4, CD44 and PCNA were 45.71% (64/140), 64.29% (90/140) and 77.86% (109/140) in laryngeal carcinoma, which were much higher than in non-carcinoma tissues (P < 0.01). 2. The third and fourth stages laryngeal carcinoma express stronger PD4 and CD44 than those of the first and second stages. Laryngeal carcinoma with cervical metastasis had higher expression than those without cervical metastasis. To 3 and 5 years' survival, PD4, CD44 and PCNA positive cases had lower chance than those negative cases(P < 0.01 or 0.05). 3. The over all positive rate of PD4, CD44 and PCNA was 27.86% (39/140) in laryngeal carcinoma tissues and 5.41% (4/74) in non-carcinoma tissues.@*CONCLUSION@#The high expression of PD4, CD44 and PCNA proteins maybe closely related to the pathogenesis, development and prognosis of laryngeal carcinoma.

OBJECTIVE@#Study on the role of mycoplasma infection and expression of CD44v6, CD44v9 protein in the pathogenesis, development and prognosis of laryngeal carcinoma (LC).@*METHOD@#Immunohistochemistry was used to study 137 cases of laryngeal carcinoma tissues, 26 cases of precarcinoma tissues, 34 cases of vocal cord polypus and 15 cases of normal tissues adjacent to laryngeal carcinoma.@*RESULT@#The positive rate of PD4 and CD44v6, CD44v9 was 45.3% (62/137), 72.3% (99/137) and 56.2% (77/137) in laryngeal carcinoma, which are much higher than those in non-carcinoma tissues. (2) The positive rate of PD4 and CD44v6. CD44v9 in the third and fourth stages of LC were higher than those in the first and second stages of LC. Laryngeal carcinoma with cervical metastasis had higher expression than those without cervical metastasis (P < 0.01). To 3 and 5 years survival, PD4 and CD44v6, CD44v9 positive cases had lower chance than those negative cases (P < 0.01 [Chinese character: see text] 0.05). (3) The overall positive rate of PD4 and CD44v6, CD44v9 was 27.7% (38/137) in laryngeal carcinoma tissues and 5.3% (4/75) in non-carcinoma tissues.@*CONCLUSION@#These results implicates that the pathogenesis, development and prognosis of laryngeal carcinoma maybe closely related to the high expression of PD4 and CD44v6, CD44v9 protein.

ABSTRACT

OBJECTIVE Study the role of mycoplasma infection and expression of Ki67 protein in the pathogenesis, development and prognosis of laryngeal carcinoma. METHODS Immunohistochemistry method was used to study 145 specimens of laryngeal carcinoma tissues, 25 specimens of precarcinoma tissues, 31 specimens of vocal cord polyps and 15 specimens of normal tissues adjacent to laryngeal carcinoma. RESULTS ①The positive rates of PD4 and Ki67 were 45.52%(66/145) and 82.76 % (120/145) in laryngeal carcinoma tissue, 16.00 % (4/25) and 32.00 % (8/25) in precarcinoma tissue, 12.90 % (4/31) and 22.58 % (7/31) in vocal cords polyps, 6.67 % (1/15) and 0 (0/15) in normal tissues adjacent to laryngeal carcinoma. ②The positive rates of PD4 and Ki67 were higher in the advanced laryngeal carcinoma cases than that in the early laryngeal carcinoma cases. ABSTRACT

OBJECTIVE To construct a prokaryotic expression plasmid encoding HPV16E7-HSP70 fusion gene for further study on the immunity of HPV16E7- HSP70 fusion protein against laryngeal carcinoma. METHODS HPV16E7 was PCR-amplified,digested by NheI and SacI,and ligated into pET28a. HSP70 was cloned into pGEMTeasy,then recut from the vector by SalI and NotI and ligated into pET28a-HPV16E7. PCR amplification, restrict enzyme digestion, DNA sequencing, IPTG induction and Western Blot were used to identify the recombinant plasmid. RESULTS Double digestion and PCR amplification of the recombinant plas- mid have shown that the size of the inserted fragment is as expected. Sequence analysis has demonstrated that the inserted fragment encodes for the HPV16E7- HSP70 fusion gene. IPTG induction and Western Blot have shown that the fusion protein is expressed suc- cessfully in the prokaryotic expression plasmid. CONCLUSION The recombinant prokaryotic expression plasmid pET28a-HPV16E7-HSP70 has been con- structed successfully.

ABSTRACT

OBJECTIVE To study the expressions of heat shock protein 70 and human papillomavirus16E7 protein in human laryngeal squamous cell carcinoma, and their relationship in the genesis of human laryngeal squamous cell carcinoma. METHODS The expressions of HSP70 and HPV16E7 protein were detected by the immunohistochemical method in 78 specimens with laryngeal squamous cell carcinoma, 24 specimens with vocal cord polyps and 10 specimens of normal laryngeal tissues. RESULTS In human laryngeal squamous cell carcinoma, vocal cord polyps and normal laryngeal tissues, the positive expression rates of HSP70 were 69.2 % , 8.3 % and 0 % respectively, with those of HPV16E7 protein being 43.6 % 4.2% and 0 % respectively. There was a significant difference of the expression rate of HSP70 or HPV16E7 protein between the laryngeal squamous cell carcinoma and the vocal cord polyps(P

ABSTRACT

OBJECTIVE To explore the changes in gene expression in patients with allergic rhinitis(AR). METHODS 14500 gene DNA microarray(Affymetrix) was used to examine the gene expression in 6 AR nasal mucosal samples and 6 normal nasal mucosal samples.The differentially expressed genes were identified and subjected to real time RT-PCR analysis. RESULTS Compared with normal sinus tissues,161 genes were found to be differentially expressed in AR samples.Forty-seven genes were upregulated,114 genes were downregulated.The differentially expressed genes mostly involved in immune transcription regulatory molecules,signal transduction.Real-time RT-PCR results of CCL20,GPK4 were consistent with that of gene chip analysis.CONCLUSION Microarray expression profile of AR samples is differential.GPK4 and CCL20 may play an important role in cell signal transduction in mechanisms of AR.

ABSTRACT

Objective:To detect and analyze the mycoplasma infection in human laryngeal carcinoma and to explore the relationship between the infection with the development,progression and prognosis of laryngeal cancer.Methods: Immunohistochemical methods were employed to detect the mycoplasma infection in 214 specimens of different laryngeal lesions and cervical lymphatic tumors metastasized from laryngeal cancer,including 121 laryngeal cancer specimens,21 laryngeal precancerous specimens,17 vocal cord polypus specimens,14 normal laryngeal tissues adjacent to cancer tissues,9 normal laryngeal tissues opposite to the cancer,and 32 cervical lymphatic tumor specimens metastasized from laryngeal cancer.The infection results were subjected to statistical analysis.Results: The positive rates of mycoplasma 2G10 antibody in laryngeal carcinoma specimens,cervical lymphatic tumor specimens metastasized from laryngeal cancer,laryngeal precancerous specimens,vocal cord polypus specimens,normal laryngeal tissues adjacent to cancer tissues,and normal laryngeal tissues opposite to the cancer were 44.63%(54/121),34.38%(11/32),23.81%(5/21),17.65%(3/17),7.14%(1/14),and 0%(0/9),respectively,with those of laryngeal carcinoma specimens and cervical lymphatic tumor specimens metastasized from laryngeal cancer significantly higher than those of other specimens(P

ABSTRACT

In older to study the interrelationship between laryngeal carcinoma and human papillomavirus (HPV). The distribution and expression of genome types of HPV in laryngeal carcinoma were investigated. HPV DNA of 160 cases of fresh tissue samples with different lesions of the larynx was detected by applying consensus primers and multiple primers of polymerase chain reaction (PCR) . The consensus primers used were able to detect 9 types of HPVs DNAs such as HPV6, 11, 16, 18, 31, 33, 34, 42 and 58. The results turned out to be as follows: The rate of positive cases with HPV infection was 49.3% (35/71) for the group of laryneal carcinoma, 22.7% (5/22) for the group of neck metastatic lymphnode, 11.8% (2/17) for precarcinomous lesions and 6.7% (2/30 ) for the group of vocal cord polypus, 20 case of normal laryngeal tissue adjacent to the carcinoma was HPV DNA negative. The results demonstrated that the occurrence and development of laryngeal carcinoma were closely related to HPV infection.
