ABSTRACT

Objective: To investigate the clinical implications as well as biomechanical and histological changes and in bone tissue induced by ovariectomy in 64 rats. Methods: The rats were divided into two groups: bilateral oophorectomy or placebo, and subdivided into four subgroups, according to time postoperatively: three, six, nine and 12 months. The weight of the animals at the time of sacrifice was taken into consideration. The biomechanical study was performed on the right tibia, to the maximum load and stiffness coefficient. For the histological study we calculated the trabecular bone of the left tibia. Statistical analysis of body weight and mechanical properties was performed by variance analysis, complemented with Tukey's multiple comparison tests; and trabecular area, the non-parametric variance analysis. Results: Ovariectomy-induced menopause caused an increase in body weight, reduction of diaphyseal bone resistance at six months of hormone deprivation, but this effect is equalized over time by aging; bone stiffness was smaller in the ovariectomized group and reduction of bone mass occurred. Conclusion: The removal of the ovaries produced systemic alterations, characterized by metabolic changes that caused weight gain and changes in bone tissue, associated with alteration of the mechanical profile and reduced bone mass. ABSTRACT

Actin-encoding cDNAs of Nile tilapia (Oreochromis niloticus) were isolated by RT-PCR using total RNA samples of different tissues and further characterized by nucleotide sequencing and in silico amino acid (aa) sequence analysis. Comparisons among the actin gene sequences of O. niloticus and those of other species evidenced that the isolated genes present a high similarity to other fish and other vertebrate actin genes. The highest nucleotide resemblance was observed between O. niloticus and O. mossambicus a-actin and b-actin genes. Analysis of the predicted aa sequences revealed two distinct types of cytoplasmic actins, one cardiac muscle actin type and one skeletal muscle actin type that were expressed in different tissues of Nile tilapia. The evolutionary relationships between the Nile tilapia actin genes and diverse other organisms is discussed.

ABSTRACT

OBJETIVOS: O estudo avaliou a influência de dietas ricas em ácidos graxos saturados (AGS) e ácidos graxos insaturados (AGI) sobre a função mecânica, a morfologia e o estresse oxidativo do miocárdio de ratos. MÉTODOS: Ratos Wistar com 60 dias de idade foram alimentados com dieta padrão (n = 8) ou dietas ricas em AGS (n = 8) ou AGI (n = 8) durante 60 dias. A função mecânica foi avaliada em músculo papilar isolado do ventrículo esquerdo (VE) por meio de contrações isométrica e isotônica, em condição basal (1,25 mM de cálcio), após elevação da concentração extracelular de cálcio para 5,2 mM e estimulação beta-adrenérgica com isoproterenol 1,0 µM. Fragmentos do VE foram usados para estudo de estresse oxidativo e microscopias óptica e eletrônica. RESULTADOS: As dietas suplementadas com AGS e AGI não alteraram a função mecânica do músculo cardíaco. Entretanto, ambas provocaram estresse oxidativo, com aumento do hidroperóxido de lipídio e redução da concentração de superóxido dismutase. A dieta AGI diminuiu a expressão da catalase e a AGS reduziu a quantidade de glutationa peroxidase miocárdica. Ambas as dietas promoveram discretas alterações morfológicas visualizadas ultra-estruturalmente, como depósitos lipídicos e lesões das membranas celulares. CONCLUSÃO: Os resultados sugerem que dietas enriquecidas com AGS e AGI não acarretam alteração da função mecânica do músculo cardíaco isolado, mas causam discretas lesões estruturais e estresse oxidativo no miocárdio.


OBJECTIVES: To study the influence of saturated (SFA) and unsaturated fatty acid (UFA) rich diets on mechanical function, morphology and oxidative stress in rat myocardium. METHODS: Male, 60-day-old Wistar rats were fed a control (n=8), a SFA (n=8), or a UFA-rich diet (n=8) for sixty days. Mechanical function was studied in isolated left ventricle papillary muscle under isometric and isotonic contractions, in basal conditions (1.25mM calcium chloride) and after 5.2mM calcium chloride and beta-adrenergic stimuli with 1.0µM isoproterenol. Left ventricle fragments were used to study oxidative stress and morphology under light and electron microscopy. RESULTS: SFA and UFA-rich diets did not change myocardium mechanical function. Both diets caused oxidative stress, with high lipid hydroperoxide and low superoxide-dismutase concentrations. UFA rich diet decreased catalase expression and SFA rich diet decreased the amount of myocardial glutathione-peroxidase. Both diets promoted light ultrastructural injuries such as lipid deposits and cell membrane injuries. CONCLUSION: Results suggest that SFA and UFA rich diets do not alter isolated muscle mechanical function, but promote light myocardial morphological injuries and oxidative stress.

ABSTRACT

Bovine skeletal muscle growth characteristics and muscle fiber type frequency are of primary interest because they both play a fundamental role in modeling meat quality and tenderness, although the precise relationship remains undefined. Growth promoters like rbST have been reported to have varying effects on muscle growth performance. The objective of this experiment was to evaluate the histochemistry and growth characteristics of bovine semitendinosus muscle treated with rbST. Animals were divided into two groups: control (saline-injected; n=8) and rbST-injected (15 ug/kg; n=8). Heifers were injected every 14 days from day 210 until day 285 of age. Muscle samples were collected (day 210 and 360) and frozen in liquid nitrogen. Histological sections (10 um) underwent morphological and histochemical analysis (HE, NADH-TR and mATPase), morphometry (fiber area and distribution), and biochemical analysis. Fibers were classified as SO, FOG, and FG. FOG fiber percentage distribution decreased and cross-sectional area increased in rbSTtreated animals. Recombinant bST caused a greater animal body weight gain and FOG fiber hypertrophy, while contributing to a decrease in FOG fiber distribution. We conclude that the phenotypic modulation seen in this muscle fiber suggests a potential role of this muscle in modeling the meat quality.

ABSTRACT

Se describen diversos tipos de fibras misculares de dos músculos flexores y de dos músculos extensores del cerdo (Sus scrofa). Además, en áreas seccionadas transversalmente, se efectuó una estimación relativa del número de mionúcleos por fibras de los músculos examinados. Los músculos fueron seleccionados de acuerdo a la conveniencia de realizar un futuro análisis neurofisiológico como componente del sistema motor segmentario y por su homología con músculos de otros vertebrados. Los músculos seleccionados fueron : sóleo, flexor superficial de los dedos, extensor largo de los dedos y extensor largo del hálux. Las secciones seriadas de estos músculos fueron teñidas con NADH-diaforasa y hematoxilina-eosina. Se discuten las relaciones entre los resultados obtenidos y hallazgos previos en músculos homólogos del miembro posterior de otros mamíferos

ABSTRACT

A rede testicular (RT) da cobaia tem características morfológicas de um tipo intermediário de RT. Esta rede foi caracterizada com cavitária, labiríntica e axial relativamente à disposiçäo de seus canais epiteliais interconectados. Morfologicamente consiste, predominantemente, de câmaras epiteliais interligadas que penetram no parênquima testicular, por uma pequena distância. Os canais e câmaras de todas as partes da rede säo revestidos por epitélio cúbico simples. Reatividades histoenzimáticas fortes foram observadas, predominantemente, na matriz da RT às enzimas fosfatases alcalina e ácida e m-ATPase. Ambas as fosfatases mostraram reatividades médias no epitélio da RT de cobaia, no qual a reatividade da m-ATPase foi fraca. Por outro lado, a NADH-d mostrou reatividade média, porém homogênea, na matriz e no epitélio da RT.

ABSTRACT

Este trabalho teve como objetivos analisar o músculo masseter do coelho (O. cuniculus), após a retirada do gânglio simpático cervical superior direito, quanto aos aspectos histológicos, histoquímicos, imuno-histoquímicos e ultraestruturais. Foram utilizados 2 grupos de animais: 30 com simpatectomia e 10 sem simpatectomia - grupo controle. Os coelhos controles foram sacrificados 6, 12, 22 e 32 meses após o início do experimento, e os com simpatectomia, 3, 6, 9, 12, 15 e 18 meses após a retirada do gânglio simpático. Após o sacrifício, foram retirados os músculos masseteres direito e esquerdo dos animais com simpatectomia e controles; foi estudada a porçäo superficial do músculo, subdividida em 3 regiöes: próxima à inserçäo proximal - "anterior", regiäo mediana e próxima à inserçäo distal - "posterior". Os fragmentos foram congelados em n-hexana, e cortes histológicos foram submetidos às seguintes coloraçöes e reaçöes: HE, Tricrômico de Gomori, PAS, SDH, NADH-TR, ATPase miofibrilar (mATPase), Esterase Inespecífica e Imunohistoquímica, utilizando-se anticorpos específicos para a miosina "fast", (tipo IIa) e "slow", (tipo I). Outros fragmentos musculares foram coletados para a análise ultra-estrutural. O músculo masseter dos grupos controles, apresentou aspectos morfológicos similares, em ambos os lados: as fibras musculares mostraram morfologia geral preservada, à microscopia óptica e eletrônica; distribuiçäo em mosaico; frequência maior de fibras tipo IIa, seguida por fibras do tipo I, e menor frequência de fibras IIb. Nos grupos com simpatectomia, foram observadas lesöes nas fibras musculares, nos animais dos grupos de 6, 12 e em maior grau, no grupo de 18 meses. Em geral, houve predominíio de fibras tipo I, nos grupos com 3, 6, 9, 12 e 15 meses; no grupo de 18 meses, houve predomínio de fibras tipo IIa, sendo observada frequência maior de fibras IIb, em relaçäo aos demais grupos, com ou sem simpatectomia.
