ABSTRACT

BACKGROUND@#In China, lung cancer remains the cancer with the highest incidence and mortality rate. Among early-stage lung adenocarcinomas (LUAD), the micropapillary (MPP) component is prevalent and typically exhibits high aggressiveness, significantly correlating with early metastasis, lymphatic infiltration, and reduced five-year survival rates. Therefore, the study is to explore the similarities and differences between MPP and non-micropapillary (non-MPP) components in malignant pulmonary nodules characterized by GGOs in early-stage LUAD, identify unique mutational features of the MPP component and analyze the relationship between the ZNF469 gene, a member of the zinc-finger protein family, and the prognosis of early-stage LUAD, as well as its correlation with immune infiltration.@*METHODS@#A total of 31 malignant pulmonary nodules of LUAD were collected and dissected into paired MPP and non-MPP components using microdissection. Whole-exome sequencing (WES) was performed on the components of early-stage malignant pulmonary nodules. Mutational signatures analysis was conducted using R packages such as maftools, Nonnegative Matrix Factorization (NMF), and Sigminer to unveil the genomic mutational characteristics unique to MPP components in invasive LUAD compared to other tumor tissues. Furthermore, we explored the expression of the ZNF469 gene in LUAD using The Cancer Genome Atlas (TCGA) database to investigate its potential association with the prognosis. We also investigated gene interaction networks and signaling pathways related to ZNF469 in LUAD using the GeneMANIA database and conducted Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis. Lastly, we analyzed the correlation between ZNF469 gene expression and levels of immune cell infiltration in LUAD using the TIMER and TISIDB databases.@*RESULTS@#MPP components exhibited a higher number of genomic variations, particularly the 13th COSMIC (Catalogue of Somatic Mutations in Cancer) mutational signature characterized by the activity of the cytidine deaminase APOBEC family, which was unique to MPP components compared to non-MPP components in tumor tissues. This suggests the potential involvement of APOBEC in the progression of MPP components in early-stage LUAD. Additionally, MPP samples with high similarity to APOBEC signature displayed a higher tumor mutational burden (TMB), indicating that these patients may be more likely to benefit from immunotherapy. The expression of ZNF469 was significantly upregulated in LUAD compared to normal tissue, and was associated with poor prognosis in LUAD patients (P<0.05). Gene interaction network analysis and GO/KEGG enrichment analysis revealed that COL6A1, COL1A1, COL1A2, TGFB2, MMP2, COL8A2 and C2CD4C interacted with ZNF469 and were mainly involved in encoding collagen proteins and participating in the constitution of extracellular matrix. ZNF469 expression was positively correlated with immune cell infiltration in LUAD (P<0.05).@*CONCLUSIONS@#The study has unveiled distinctive mutational signatures in the MPP components of early-stage invasive LUAD in the Asian population. Furthermore, we have identified that the elevated expression of mutated ZNF469 impacts the prognosis and immune infiltration in LUAD, suggesting its potential as a diagnostic and prognostic biomarker in LUAD.

ABSTRACT

Objective:To explore the value of microRNA(miR)-124-3p and its target gene aryl hydrocarbon receptor (AHR) in the diagnosis and prognostic evaluation of gastric cancer, and the related molecular mechanisms in regulating proliferation and invasion of gastric cancer cell.Methods:The clinical and prognostic characteristics of patients with gastric adenocarcinoma expressing miR-124-3p were obtained from The Cancer Genome Atlas database and Genotype-Tissue Expression database. The correlation between miR-124-3p expression level and pathological stage, TNM stage, overall survival (OS), disease-specific survival (DSS) and progression-free interval (PFI) in patients with gastric adenocarcinoma were studied by bioinformatics analysis. The interaction sites between miR-124-3p and AHR mRNA were predicted by Target Scan 7.1 online tool. The target binding sites of miR-124-3p in AHR mRNA were verified by subcutaneous tumorigenesis experiment in mice, immunohistochemistry, dual luciferase assay, quantitative real time-polymerase chain reaction (RT-qPCR) and Western blotting. Nine male Balb/c nude mice, aged 4 to 6 weeks with weight of (18.43±0.29) g were injected with miR-124-3p simulant (miR-124-3p group), negative control simulant (negative control group) and 0.9% sodium chloride solution (sodium chloride control group) through the tail vein. Gastric cancer cell lines (MKN-45, AGS) were transfected with RNA simulants (including miR-124-3p simulant, negative control simulant and 0.9% sodium chloride solution). The expression of AHR and Catenin β 1 gene ( CTNNB1) at mRNA level, the expression of AHR and β-catenin at protein level in 3 mice groups and the effects of miR-124-3p transfection on the proliferation and invasion of transfected gastric cancer cells were analyzed. Pearson correlation analysis and Holm-Sidak corrected multiple t test were used for statistical analysis. Results:Low expression of miR-124-3p was positively correlated with severe pathological stages and TNM stages in patients with gastric adenocarcinoma ( R2=0.83 and 0.86, P=0.031 and 0.023). High expression of miR-124-3p was positively correlated with OS, DSS and PFI ( R2=1.00, 0.99 and 0.99, P=0.029, 0.044 and 0.049). The results of subcutaneous tumorigenesis experiment in mice demonstrated that the number of apoptotic cells in the tumor of miR-124-3p group was more than that of negative control group and sodium chloride control group ((43.33±1.86)/high power field (HPF) vs. (20.00±1.73)/HPF and (18.67±1.76)/HPF), and the differences were statistically significant ( t=8.55 and 8.33, P=0.013 and 0.014). The results of immunohistochemistry showed that the optical density of AHR protein in mice tumor tissue of miR-124-3p group was lower than that of negative control group and sodium chloride control group (0.081±0.008 vs. 0.276±0.019 and 0.273±0.018), and the differences were statistically significant ( t=9.06 and 7.51, P=0.012 and 0.017). The results of dual luciferase assay indicated that the fluorescence intensity in wild-type AHR MKN-45 cells transfected with miR-124-3p simulant was lower than that of negative control group (0.293±0.020 vs. 1.000±0.032), and the difference was statistically significant ( t=18.56, P<0.001). The results of RT-qPCR demonstrated that the mRNA levels of AHR and CTNNB1 in MKN-45 cells transfected with miR-124-3p simulant were both lower than those in untreated MKN-45 cells (0.51±0.09 vs. 1.02±0.02, 0.46±0.03 vs. 1.03±0.01), and the differences were statistically significant ( t=4.51 and 16.60, P=0.046 and 0.004). The results of Western blotting experiments showed that the relative protein expression levels of AHR and β-catenin of MKN-45 cells transfected with miR-124-3p simulant were lower than those of transfected with 0.9% sodium chloride solution and negative control simulant (3 332.94±81.25 vs. 9 041.60±439.79 and 8 276.54±562.52, 2 725.79±167.57 vs. 9 701.94±410.02 and 8 081.66±275.84), and the differences were statistically significant ( t=15.49, 7.91, 17.35 and 19.42, P=0.004, 0.016, 0.003 and 0.003). Conclusions:MiR-124-3p is correlated with diagnosis and prognosis of gastric cancer. MiR-124-3p induces apoptosis of gastric cancer cells in vitro and vivo by negatively regulating AHR expression at mRNA and protein level, thereby down-regulating the expression of CTNNB1 mRNA and β-catenin pathway-related protein. Therefore, miR-124-3p may become a potential diagnostic and prognostic marker of gastric cancer.

ABSTRACT

Objective:To explore the clinical characteristics, diagnosis, treatment and prognosis of inverted Meckel′s diverticulum in children, and to summarize the clinical experience.Methods:Ten children who received surgical treatment in Beijing Children′s Hospital, Capital Medical University from October 2018 to October 2021 were retrospectively analyzed.The children were diagnosed with Mecker′s diverticulum.Their data were complete and their fo-llow-up information was available.The clinical features, diagnosis, treatment and prognosis were summarized.There were 6 males and 4 females, whose age of onset ranged from 5 months to 12 years and 5 months.The clinical manifestations were abdominal pain in 8 cases, crying in 2 cases and bloody stool in 3 cases.B-ultrasound showed suspected diseases secondary to inverted Meckel′s diverticulum, including small intestinal intussusception in 6 cases and back junction type intussusception in 4 cases.One case of small intestinal intussusception was misdiagnosed after laparoscopic exploration in other hospitals and suffered from recurrent intussusception.All patients underwent segmental bowel resection.Results:Of the 10 children, 8 cases received laparoscopic exploration and 2 cases underwent open surgery for secondary intestinal necrosis.Postoperative pathology revealed Meckel′s diverticulum.Ectopic tissue was accessible in 8 cases, including ectopic gastric tissue in 7 cases, and ectopic pancreas in 1 case.During the postoperative follow-up period (1 month-3 years), the patients were generally in good condition with no complications such as intestinal obstruction.Conclusions:Intussusception secondary to inverted Meckel′s diverticulum is rare and difficult to diagnose before operation.The diagnosis should be confirmed by surgical exploration and postoperative pathology.However, careful surgical exploration is required since inverted Meckel′s diverticulum is prone to be misdiagnosed during exploration.

ABSTRACT

Objective:To explore the effect and mechanism of taxifolin (TAX) on ameliorating cisplatin-induced renal oxidative damage.Methods:(1) Forty male C57BL/6 mice were divided into 4 groups: control group ( n=10), TAX group ( n=10), cisplatin group ( n=10) and cisplatin+TAX group ( n=10). The weight of mice in each group was measured. The level of serum creatinine (Scr) and blood urea nitrogen (BUN) was analyzed. Kidney histopathological change in mice was analyzed by HE staining. The pro-inflammatory cytokines levels of interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-α) were measured by enzyme linked immunosorbent assay. The levels of reactive oxygen species (ROS), malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT) and glutathione (GSH) were measured by multifunctional microplate reader. The expression of inflammatory factors, antioxidant genes, and peroxisome proliferator-activated receptor γ coactivator-1α ( PGC- 1) mRNA were measured by real-time PCR. Evaluation of mitochondrial function by measuring ATP level and mtDNA content. Determination of AMP-activated protein kinase (AMPK) and phosphorylated AMPK protein expression by Western blotting. (2) Evaluate the effect of taxifolin on chemotherapy of cisplatin by establishing Lewis lung cancer transplantation tumor C57BL/6 mice model. Results:Compared with the control group, the weight of the mice in the TAX group was not significantly reduced ( P>0.05), and there was no obvious kidney damage ( P>0.05), indicating that oral TAX had good safety. Compared with the cisplatin group, TAX could significantly delay cisplatin-induced the weight loss of mice, reduce the levels of Scr and BUN, and alleviate the pathological changes of kidney tissue (all P<0.05). TAX could reduce the levels of serum inflammatory factors IL-6 and TNF-α and the expression of renal inflammatory factors IL- 6, TNF- α and IL- 1β mRNA induced by cisplatin in mice (all P<0.05). TAX could significantly reduce the levels of ROS and MDA, and increase the activities of SOD, CAT and GSH in cisplatin-induced acute kidney injury mice (all P<0.01). Meanwhile, TAX could up-regulate the mRNA expression of UCP2, SOD2, CAT antioxidant genes and PGC- 1α in the kidneys of mice with acute kidney injury induced by cisplatin, and increase the levels of ATP and mtDNA in cisplatin-induced acute kidney injury mice (all P<0.01). Western blotting results showed that TAX significantly promoted the expression of phosphorylated AMPK protein in cisplatin-induced acute kidney injury mice ( P<0.01). In addition, through the establishment of Lewis lung cancer transplantation tumor C57BL/6 mice model, it was found that TAX had no significant effect on the anti-tumor efficacy of cisplatin. Conclusions:TAX can ameliorate cisplatin-induced renal oxidative damage, and its mechanism may be related to the activation of AMPK/PGC-1α pathway.

ABSTRACT

Objective:To investigate the implementation status of sepsis hour-1 bundle strategy for patients with septic shock in emergency department.Methods:A total of 116 septic shock patients admitted to the emergency department from January 2020 to December 2020 were included in this prospective study, and the implementation of sepsis bundles and the clinical outcomes of patients were recorded.Results:Among 116 patients, 20 cases (17.2%) had lactic acid monitored within 1 h, 20 cases (17.2%) had blood culture before antibiotics, 82 cases (70.1%) received broad-spectrum antibiotics, 16 cases (13.8%) received fluid resuscitation ≥30 ml/kg, and 57 cases (49.1%) received vasoactive drugs during resuscitation. Finally, the sepsis hour-1 bundle strategy was fully implemented only in 13 cases (11.2%). Compared with the group with incomplete implementation of sepsis hour-1 bundle strategy, the volume of fluid recovery in the group with full implementation was significantly increased [33.7 (30.0,37.5) vs. 8.9(7.3,10.8) ml/kg, Z=-4.78, P<0.001], mean artery blood pressure significantly increased [70.0 (70.0,76.7) vs. 67.7 (61.7,76.7)mmHg(1 mmHg=0.133 kPa) , Z=-2.00, P<0.001], and lactic acid significantly decreased [3.0 (2.0,3.2) vs. 4.4 (3.7,7.2) mmol/L, Z=-2.76, P=0.006]. However, there were no significant differences in ICU mortality, in-hospital mortality and 28-day mortality between the two groups ( P>0.05). Conclusions:Septic shock patients in emergency department have poor compliance with the implementation of sepsis hour-1 bundle strategy, and relevant management training should be strengthened.

ABSTRACT

Objective:To investigate the clinical characteristics and prognosis of children with acute pancreatitis(AP)admitted to the pediatric intensive care unit(PICU).Methods:The etiology, clinical data, imaging features, complications, and outcomes of children with AP admitted to PICU at Shanghai Children′s Hospital from July 2016 to June 2021 were retrospectively analyzed.Results:Totally, 47 patients with AP including 24 males and 23 females were enrolled.The mean age was 84(48, 144)months.Four patients with mild AP, 5 patients with moderately severe AP(MSAP)and 38 cases with severe AP(SAP)were diagnosed.The main etiology was drug-related pancreatitis in 23 cases(48.9%). The organ dysfunction mainly included shock in 28 cases(59.6%), acute respiratory failure in 21 cases(44.7%), gastrointestinal disorders in 30 cases(63.8%), and coagulopathy in 29 cases(61.7%). The complications included capillary leakage syndrome in 18 cases(38.3%)and hyperglycemia(>8 mmol/L)in 24 cases(51.1%). Serum amylase and lipase levels were increased in 47 cases(100%). There were significant differences in blood lactate[3.0(2.1, 4.5)mmol/L vs.1.6(1.1, 3.1)mmol/L, P=0.013], and the use of vasoactive drugs[15.0(0, 75.0)vs.0(0, 8.8), P=0.035] between drug-induced pancreatitis and non-drug-induced pancreatitis.All the patients′conditions were improved after treatment and transferred out of PICU. Conclusion:The etiology of acute pancreatitis in children is mainly drug-related, and the main organ dysfunction includes shock, acute respiratory failure, gastrointestinal dysfunction and coagulation dysfunction.The prognosis of childhood pancreatitis is good.

ABSTRACT

Objective:To investigate the effects of long non-coding RNA (lncRNA) CDKN2B-AS1 targeting miR-98-5p on proliferation and invasion of lung cancer A549 cells.Methods:A549 cells cultured in vitro were divided into control group, pcDNA group (transfected with pcDNA) , CDKN2B-AS1 group (transfected with pcDNA CDKN2B-AS1) and double transfection group (transfected with pcDNA CDKN2B-AS1 and pcDNA miR-98-5p) . The expression of lncRNA CDKN2B-AS1, miR-98-5p and the protein expression of PCNA, MMP-9 in A549 cells were detected. The activity, clone number, cloning efficiency, and the number of invasive cells of A549 cells were detected.Results:Compared with pcDNA group, the expression level of lncRNA CDKN2B-AS1 [ (2.14±0.14) vs (1.03±0.10) ], OD value in each time points, clone number [ (314.60±18.13) vs (220.08±12.46) ], cloning efficiency [ (85.81±3.06) % vs (60.03±2.85) %], invasive cell number [ (233.30±18.98) vs (140.84±12.30) ], expression levels of PCNA [ (0.78±0.08) vs (0.48±0.07) ] and MMP-9 [ (0.75±0.06) vs (0.38±0.06) ] proteins in A549 cells in CDKN2B-AS1 group were significantly increased ( P<0.05) ; the expression level of miR-98-5p [ (0.23±0.03) vs (0.99±0.09) ] was significantly decreased ( P<0.05) ; compared with CDKN2B-AS1 group, there was no significant difference in the expression level of lncRNA CDKN2B-AS1 in A549 cells in double transfection group ( P>0.05) , while the expression level of miR-98-5p in A549 cells was significantly increased ( P<0.05) . The OD value in each time points, clone number, cloning efficiency, invasive cell number, expression levels of PCNA and MMP-9 proteins were significantly decreased ( P<0.05) . Conclusion:LncRNA CDKN2B-AS1 can promote the proliferation and invasion of lung cancer A549 cells by targetingly inhibiting the expression of miR-98-5p.

ABSTRACT

OBJECTIVE:To establish a method for the simultan eous determination of the contents of 12 flavonoids in Quzhiqiao. METHODS :HPLC method was adopted. The determination was performed on Agilent Extend C 18 column with mobile phase consisted of 0.1% formic acid-acetonitrile (gradient elution )at the flow rate of 1.0 mL/min. The column temperature was set at 35 ℃. The detection wavelength was set at 330 nm,and sample size was 10 μL. The contents of 12 components(such as eriocitrin,narirutin,naringin,naringenin,hesperidin,neohesperidin,hesperide hydrate ,luteolin,hesperide,nobiletin,hesperetin and hesperidolactone )in 10 batches of Quzhiqiao from different collection places were determined. RESULTS :The linear range of eriocitrin,narirutin,naringin,naringenin,hesperidin,neohesperidin,hesperide hydrate ,luteolin,hesperide,nobiletin,hesperetin and hesperidolactone were 1.65-16.51,4.50-45.02,35.41-354.12,4.11-41.12,2.29-22.86,34.96-349.56,1.42-14.15,1.50-15.04, 1.83-18.28,1.51-15.08,1.61-16.12,1.28-12.84 μg/mL,respectively(all r>0.999 7). The detection limits were 0.165 1,0.450 2, 3.541 2,0.411 2,0.228 6,3.495 6,0.141 5,0.150 4,0.182 8,0.150 8,0.161 2,0.128 4 μg/mL,respectively. The limits of quantitation were 0.547 8,1.487 4,11.663 3,1.360 3,0.758 3,11.594 9,0.466 3,0.497 1,0.601 2,0.499 9,0.532 3,0.424 6 μg/mL,respectively. RSDs of precision (n=6),reproducibility(n=6)and stability (24 h,n=7)tests were all lower than 3%. The average recoveries were 99.50%,99.61%,98.18%,98.85%,98.48%,98.50%,98.25%,99.91%,103.13%,98.82%, 98.44% , 100.29% (RSD=1.49% -2.38% , n=6). CONC LUSIONS:The method is accurate ,reliable,simple and efficient,which can be used to simultaneous determination of the contents of 12 flavonoids in Quzhiqiao ,and to provide reference for the establishment of quality control standards of Quzhiqiao.

ABSTRACT

Specialized pro-resolving mediators(SPMs)are lipid mediators derived from essential fatty acids, which have anti-inflammatory/pro-resolution effects.SPMs mainly include lipoxin, resolvin, protectin, and maresin.Different SPMs can bind to different receptors[formyl peptide receptor 2(ALX/FPR2), G protein-coupled receptor 32(DRV1/GPR32), G protein-coupled receptor 18(DRV2/GPR18), or chemokine-like receptor 1(ERV1/CMKLR1)], through nuclear factor-κB, signal transducer and activator of transcription or mitogen-activated protein kinase signal pathways to proresolve inflammation by limiting neutrophil migration and infiltration, regulating inflammatory cytokine expression, and enhancing macrophage phagocytosis.SPMs have potential clinical value in severe disease severity, prognosis prediction and clinical intervention efficacy.This article focused on the role and potential clinical value of SPMs in immune-inflammatory response to severe infection.

ABSTRACT

Objective To assess the effect and safety of epalrestat on diabetic peripheral neuropathy ( DPN) . Methods Retrieved from PubMed,Cochrane Library,CNKI,VIP,CBM and Wanfang database ,RCTs about epalrestat in the treatment of DPN were included from establishment to July 2017 and comprehensively evaluated .Two reviewers independently screened literature according to the inclusion and exclusion criteria , extracted data , assessed the risk bias of included studies , and then analysis was performed using the RevMan 5.3 software.Results A total of 14 RCTs involved 1945 patients were included .The results of systematic review and Meta -analysis showed that epalrestat could significantly improve DPN compared with the placebo /blank control group ( OR =8.01,95%CI:4.48-14.32,P<0.01;OR=2.49,95%CI:1.71-3.65,P<0.01).Compared with the placebo group and the blank control group,epalrestat could significantly improve the median sensory nerve conduction velocity ( OR=3.92,95%Cl:1.37-6.47,P<0.01),peroneal nerve conduction velocity (OR=4.87,95%CI:2.35-7.39,P<0.01) and ulnar nerve conduction velocity ( OR =4.58, 95%CI:2.54-6.63, P <0.01 ).The major adverse reactions were gastrointestinal reactions ,nausea and vomiting ,but no serious adverse reactions were observed in the epalrestat group , compared with the placebo/blank control group,the difference was statistically significant (OR =12.87,95%CI:2.35-70.50,P<0.01).Conclusion Epalrestat can effectively treat DPN through improving the body's sensory nerve conduction velocity,and it has no serious adverse reactions .Since the limitation of quality and quantity of included studies,large-scale and high-quality randomized double -blinding controlled trails are needed to verify the above conclusion.

Objective To analysis The clinical efficacy of XiaoErKang granule combined with MAMIAI to 98 children with autumn ped-iatric diarrhea.Methods98 children with diarrhea from January 2015 to December 2015 were divided into 49 cases with treatment group and 49 cases with the control group.The control group received MAMIAI,and the treatment group received XiaoErKang granule combined with MAMIAI,following-up and recording related signs disappearing time,changes of serum factors and the clinical results.Results①The fever subsided, stop vomiting and stool recovery time of treatment group were (18.24±2.13h,1.59±0.51d,3.67±0.51d),shortter than the control group with (27.05±3.09h,2.72±0.64d,5.69±0.86d),and the differences were statistically significant (P<0.05).②The hs-CRP and TNF-ɑ levels of treatment group were (4.58±0.61ng/L,8.14±0.79pg/mL),lowwer than the control group with (8.13±0.75ng/L,11.49±1.87pg/mL),and the differences were statistically significant (P<0.05).③The IL-1 and IL-6 levels of treatment group were(27.49±3.16,9.73±1.25)pg/mL,lowwer than the control group with (40.16±5.72,12.65±1.77)pg/mL,and the differences were statistically significant (P<0.05).④The treatment effect of treatment group were 91.84%,better than the control group with 73.47%,and the differences were statistically significant (P<0.05).ConclusionThe effece of XiaoErKang granule joint MAMIAI to children with diarrhea can reduce levels of inflammation,and it is effective.

ABSTRACT

Objective To investigate CT image features of pleomorphic adenoma and adenolymphoma,and to improve the diagno-sis accuracy.Methods CT data of 1 3 cases of pleomorphic adenomas and 9 cases of adenolymphomas confirmed by pathology were retrospectively analyzed and compared.Results All 1 3 cases of pleomorphic adeonoma were solitary lesions,in which 1 1 cases were located in the non posterior inferior quadrant,and 1 1 cases showed persistent or delayed enhancement,with “slowly forward and slowly quit”enhancement performance.In all of the 9 cases of adenolymphoma,3 cases were multiple lesions.In total of 1 3 lesions, 9 lesions were located in the posterior inferior quadrant,and 9 lesions were significantly enhanced in the arterial phase and enhance-ment was decreased in the delayed phase,which showed “quickly forward and quickly quit”.Conclusion CT image features of pleo-morphic adenoma and adenolymphoma are different.Most of lesions can be diagnosed correctly before surgery by using image fea-tures with clinical information.

ABSTRACT

Objective:To study crown-root morphology of maxillary anterior teeth in ClassⅡ,division 2 malocclusion using cone-beam CT (CBCT)combined with computer aided measurement technology to provide guidance for clinical treatment.Methods:The samples which consisted of 36 cases radiographed with CBCT techniques were selected and divided into two groups (1 8 each )based on the type of maloc-clusion presented:ClassⅡ,division 2 group (groupⅡ2)and ClassⅠgroup (groupⅠ).The measure-ments of crown-root morphology including crown-root angle and surface-shaft angle were got by Multiple Planer Reconstruction of CBCT data uploaded into InvivoDental software 5.0.The data were processed with SPSS 20.0 software package and t test was employed for comparison of angular measurements. Results:In group Ⅰ,crown-root angles of maxillary central incisor,maxillary lateral incisor and maxillary canines were 1 79.08°±3.31 °,1 76.55°±2.77°and 1 84.20°±2.51 °respectively, surface-shaft angles were 21 .00°±2.63°,1 9.63°±2.35°and 1 9.36°±2.30°respectively. While in group Ⅱ 2 ,crown-root angles of maxillary central incisor,maxillary lateral incisor and maxillary canines were 1 76.80°±2.62°,1 74.1 3°±3.28°and 1 81 .79°±2.88°respectively, surface-shaft angles were 23.20°±2.95°,22.29°±2.1 9°and 20.61 °±2.34°respectively. Compared with group Ⅰ,significant statistical differences were observed with the exception of sur-face-shaft angle of maxillary incisor.There was significant difference in crown-root angle between group Ⅱ2 and 1 80 °.Conclusion:The maxillary anterior teeth in ClassⅡ,division 2 malocclusion exhibited significant crown-root morphology which would influence the torque after orthodontic treat-ment.Special attention should be paid to the position of maxillary anterior teeth roots during ortho-dontic treatment for Class Ⅱ,division 2 malocclusion.The ideal position of tooth movement should be decided by the root rather than the location of the crowns.

ABSTRACT

Objective:To compare the accuracyof interactive closet point (ICP)algorithm,Procrustes analysis (PA)algorithm,andalandmark-independent method to construct the mid-sagittal plane (MSP)of the cone beam computed tomography.To provide theoretical basis for establishing coordinate systemof CBCT images and symmetric analysis.Methods:Ten patients were selected and scanned by CBCT before orthodontic treatment.The scan data was imported into Mimics 10.0 to reconstructthree dimensional skulls.And the MSP of each skull was generated by ICP algorithm,PA algorithm and landmark-independ-ent method.MSP extracted by ICP algorithm or PA algorithm involvedthree steps.First,the 3D skull processing was performed by reverse engineering software geomagic studio 2012 to obtain the mirror skull. Then,the original and its mirror skull was registered separately by ICP algorithm in geomagic studio 2012 and PA algorithm in NX Imageware 1 1 .0.Finally,the registered data were united into new data to calcu-late the MSP of the originaldata in geomagic studio 2012.The mid-sagittal plane was determined by SEL-LA (S),nasion (N),basion (Ba)as traditional landmark-dependent methodconducted in software InVivoDental 5.0.The distance from 9 pairs of symmetric anatomical marked points to three sagittal plane were measured and calculated to compare the differences of the absolute value.The one-way ANOVA test was used to analyze the variable differences among the 3 MSPs.The pairwise comparison was performed with LSD method.Results:MSPs calculated by the three methods were available for clinic analysis,which could be concluded from the front view.However,there was significant differences among the distances from the 9 pairs of symmetric anatomical marked points to the MSPs (F=10.932,P=0.001).LSD test showed there was no significant difference between the ICP algorithm and landmark-independent method (P=0.1 1 ),while there was significant difference between the PA algorithm and landmark-independent methods (P =0.01 ).Conclusion:Mid-sagittal plane of 3D skulls could be generated base on ICP algorithm or PA algorithm.There was no significant difference between the ICP algorithm and landmark-independent method.For the subjects with no evident asymmetry,ICP algorithm is feasible in clinical analysis.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the importance of root information in diagnosis set-up by constructing three-dimensional (3D) digital models with individual anatomic roots.</p><p><b>METHODS</b>Pretreatment cone-beam CT (CBCT) and laser scanning data were collected from two patients (extraction and non-extraction each) with skeletal Class I malocclusion. Threshold segmentation of the CBCT was performed to generate a 3D digital model which has individually isolated tooth. This model and the scan model were superimposed to generate an integrated model (Mo) composed of high-resolution surface scan crowns sutured to the CBCT roots. Pretreatment dentition plaster model were made into set-up model. The diagnosis model set-up was performed successively by three orthodontists and one senior orthodontic technician. Set-up model scan of each patient after tooth alignment was obtained. The isolated composite teeth were individually superimposed onto the set-up model surface scan to creat set-up model (Ma, Mb, Mc, Md) containing root position. These isolated composite teeth were also superimposed onto the posttreatment surface scan depicting the posttreatment model (M). In order to observe whether diagnosis model set-up would cause exposure of the root, Ma-Md were compared with Mo, which showed the true positions of alveolus. In order to validate the accuracy of the expected root position setup, Ma-Md were compared with the true root position represented by M. Color displacement maps generated to measure the discrepancies of root positions.</p><p><b>RESULTS</b>Nonparallel and exposure of the root was found in all setup models. Color displacement maps through molar superimpositions showed maximum differences of 8.79 mm for the maxillary teeth and 9.96 mm for the mandibular teeth.</p><p><b>CONCLUSIONS</b>Diagnosis model set-up based only on crown can not absolutely ensure the good alignment of roots without root exposure. It is necessary to construct the integrated model including root for diagnosis during tooth arrangement process.</p>

ABSTRACT

BACKGROUND:Orthodontic tooth movement is based on the periodontal tissue remodeling. In the exogenous factors accelerating orthodontic tooth movement, Chinese herbal medicine has become a research hotspot because of its wide resources, low cost, easy to extract, mild effect, smal toxic, less side effects and drug resistance. OBJECTIVE: To summarize the role of Chinese herbal medicine in the periodontal tissue remodeling during orthodontic tooth movement. METHODS:A computer-based retrieval of CNKI, Wanfang and PubMed databases was performed for articles related to Chinese herbal medicine for improving orthodontic tooth movement published before 2014. The keywords were “Chinese herbal medicine, orthodontic tooth movement, periodontal tissue remodeling” in Chinese and English, respectively. RESULTS AND CONCLUSION:Erigeron breviscapus, Salvia, teasel, Drynaria, baicalin, evening primrose oil as Chinese herbs are most widely used in the promotion of periodontal tissue remodeling, characterized as wide resources, low cost, easy to extract, mild effect, low toxicity, less drug resistance. In the clinical orthodontic treatment, it is hoped to accelerate orthodontic tooth movement and shorten the treatment time. Therefore, under the appropriate corrective force, Chinese herbs can be used properly to improve periodontal tissue repair and remodeling, which can improve the microcirculation of periodontal tissue, increase the local blood flow, promote bone formation and repress bone resorption.

ABSTRACT

Objective To study CT diagnostic value of benign and malignant salivary gland lesions.Methods CT features in 36 cases with pathologically-proved salivary lesions were retrospectively analyzed.There were 19 cases with benign tumors,5 cases with chronic inflammation,1 case with tuberculosis and 11 cases with malignant tumors.Results Benign tumors were regular in 17 and well-defined in 16.All cases with chronic inflammation and 1 case with tuberculosis were regular but ill-defined.Benign lesions showed homogeneous or inhomogeneous density.Of all 11 malignant tumors,7 cases presented ill-defined margin with irregularity in shape,infiltration of adjacent subcutaneous fat tissue and disappearing of epiderm.Salivary glands enlarged and looked like "cast form" in malignant tumors.Most of the malignant tumors and benign tumors presented inhomogeneous density.Conclusion CT scan is of important value in the diagnosis of salivary diseases.
