ABSTRACT

Cerebral ischemia-reperfusion injury (CIRI) has a very high incidence, disability, and mortality rates, which seriously affects human life and health. In recent years, modern medicine has made some progress in the diagnosis and treatment of CIRI, but there are still problems such as difficulties in postoperative rehabilitation and adverse drug reactions, and new therapeutic drugs for CIRI are urgently needed. As an important class of active ingredients in traditional Chinese medicine, flavonoids can play antioxidant, apoptosis inhibition, anti-inflammatory, and other pharmacological effects to improve brain tissue damage, which is important for improving the quality of life of CIRI patients and slowing down the aging of the social population. Numerous studies have found that flavonoids in traditional Chinese medicine can regulate cell surface receptors Toll-like receptor 4/nuclear factor-kappaB (TLR4/NF-κB), phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt), adenylate-activated protein kinase/mammalian target of rapamycin protein (AMPK/mTOR), Ras homologous gene family member A/Rho-associated coiled-coil protein kinase (RhoA/ROCK), nuclear factor E2-associated factor 2/Kelch-like epoxychloropropane-associated protein-1/haemoglobin oxygenase 1 (Nrf2/Keap1/ HO-1), Notch, and other signaling pathways, so as to regulate the transcription and expression of related proteins after CIRI, alleviate brain tissue injury, and improve CIRI. This paper analyzed the relevant literature in China and abroad in recent years, reviewed the mechanism of action and related pathways of flavonoids in traditional Chinese medicine to improve CIRI, and explored the new therapeutic direction of CIRI at the metabolic level, with a view to providing a basis for the further development and application of flavonoids in traditional Chinese medicine.

ABSTRACT

Objective This study was to detect the expression and level of vascular endothelial growth factor(VEGF)in the acute rejection reaction of orthotopic liver transplantation in rats, which attempted to prove whether VEGF is the key molecule mediating the inter-permeate and inter-enhancement mediated by cells between Angiogenesis and inflammation reaction Methods Expressions of VEGF in plasm in liver and spleen were detected using immunohistochemical staining. The levels of VEGF were measured with enzyme linked immunosorbent assay(ELISA).Results The expression of VEGF in liver, spleen and the level of VEGF in plasma in experiment group were higher than that in the control group(P<0.05),which were the highest in two days after operation. Conclusion VEGF may play a significant role in the acute rejection reaction of orthotopic liver transplantation in rats. There was a close relation between the expression and level of VEGF in liver, spleen and survival time of graft.VEGF was a kind factor which is expressed in early stage.

ABSTRACT

Objective To study the relationship between the expression of vascular endothelial growth factor (VEGF) and invasion, metastasis of primary hepatocellular carcinoma (PHCC). Methods Paraffin-embedded specimens from 30 patients with PHCC undergoing radical resection were studied. Agiogenesis was assessed by the expression of VEGF and microvascular density (MVD) using LSAB immunohistochemical staining. Results (1) The level of VEGF and MVD in PHCC was higher than that in the paratumorous tissue (P<0.05). (2) The level of VEGF expression and MVD in PHCC with tumor metastasis or in nonencapsulated tumors were higher than that in PHCC without metastasis or in well encapsulated tumors (P<0.05). (3) There was no significant differance in the expression of VEGF and MVD between large PHCC (diameter >5cm) and small ones (diameter ≤5cm). (4) The expression of VEGF was positively correlated with MVD (P<0.01). Conclusions The expression of VEGF and MVD are highly correlated with invasion and metastasis in PHCC, may serve as a parameter for determining tumor metastasis and prognosis which may suggest a new idea to supervise and treat PHCC.

ABSTRACT

Objective To observe the manifestations and judgement methods of acute rejection reaction of orthotopic liver transplantation in Wistar-SD rats. Methods The survival conditions after operation in rats was observed, and liver function tests and histopathology examination were used to study the manifestations of acute rejection reaction of orthotopic liver transplantation in Wistar-SD rats. Results Wistar-SD experiment group had moderate to severe acute rejection reaction after OLT.Alanine aminotransferase, aspartate aminotransferase, total bilirubin io clood serum after orthotopic liver transplantation in rats was obviously elevated on 1-3 d and 7 d after OLT,which are significantly different from every corresponding time point of control group(P
