ABSTRACT

Objective:To investigate the effect of apatinib on radiosensitivity of glioma cells U87MG and its potential mechanism.Methods:U87MG cells were divided into control group, apatinib group, radiation group and combination group treated with apatinib and radiation. The effect of different concentrations of apatinib (5, 10, 20, 40, 80 μmol/L) on cell proliferation was detected by CCK8 assay. The effect of apatinib on cell migration and invasion was detected by wound-healing assay and transwell assay, respectively. The effect of apatinib on cell radiosensitivity was detected by plate cloning assay, the cell apoptosis rate was detected by flow cytometry, and the protein expressions of Bax and Bcl-2 were detected by Western blot.Results:Apatinib significantly inhibited the proliferation of U87MG cells in a manner depended on the drug treatment time and radiation. Compared with the radiation group, the cell proliferation, migration and invasion in the combination group were inhibited much significantly ( t=9.857, 18.704, 4.197, P<0.05), so that the value of D0, Dq and SF2 of the combination group was lower, resulting in a radiosensitivity enhancement ratio (SER D0 ) of 1.3. Moreover, compared with the radiation group, the apoptosis rate of the combination group was increased, the expression of Bcl-2 protein was decreased, and the expression of Bax protein was increased ( t=16.187, 8.890, 5.222, P< 0.05). Conclusions:Apatinib inhibits cell proliferation, invasion and migration, induces apoptosis and increases radiosensitivity of glioma cells.

ABSTRACT

Orbital venous malformation (OVM) is a congenital vascular disease. As a common type of vascular malformation in the orbit, OVM may result in vision deterioration and cosmetic defect. Classification of orbital vascular malformations, especially OVMs, is carried out on the basis of different categories, such as angiogenesis, hemodynamics, and locations. Management of OVM is complicated and challenging. Treatment approaches include sclerotherapy, laser therapy, embolization, surgical resection, and radiotherapy. A satisfactory outcome can be achieved only by selecting the appropriate treatment according to lesion characteristics and following the sequential multi-method treatment strategy. This article summarizes the current classification and treatment advances in OVM.

ABSTRACT

Objective To investigate the association between clinicopathological characteristics and familial history of malignant neoplasms (MN-FH) in patients with colorectal cancer. Methods The clinical data of 652 cancer patients in Zhengzhou University Affiliated Cancer Hospital from December 2014 to December 2015 were analyzed retrospectively. Patients were grouped based on with or without MN-FH. The clinical and pathological features of the patients were analyzed by χ 2test. Results One hundred and thirty cases (19.9 %) of colorectal cancer had MN-FH. Compared with NO MN-FH group, MN-FH group had the features of low differentiation degree, late clinical stage, deep infiltration, and also prone to lymph node metastasis and distant metastasis, associated with cancer nodules, vascular thrombosis, nerve invasion, multiple primary tumor, MSI-H (χ 2values were 30.825, 12.270, 12.122, 8.502, 53.969, 4.502, 12.861, 11.680, 6.272, 17.460, all P < 0.05). Conclusions Colorectal cancer patients with MN-FH has high malignant degree,the early diagnosis and treatment are the key to survival of patients with MN-FH.

ABSTRACT

ObjectiveTo study the synergistic effect of p38MAPK and ERK1/2 in bone marrow mesenchymal stem cells (BMSCs), and to explore their influence on osteogenic differentiation in BMSCs cultures. MethodsMouse BMSCs were cultured in phenol red-free α-MEM containing osteogenic supplements (OS) for inducing osteogenic differentiation. The temporal sequence of osteogenic differentiation in BMSCs cultures was assayed by measuring alkaline phosphatase activity (ALP) and calcium deposition gene expression. The activation of p38MAPK and ERK1/2 was detected by western blotting using phospho-specific MAP kinase antibody. BMSCs were treated with the inhibitor of p38MAPK pathway(SB203580) or ERK1/2 pathway (PD98059), and osteogenic differentiation was measured. BMSCs were treated with SB203580 or sodium arsenite(ARS), a strong activator of p38MAPK, and the phosphorylation of ERK 1/2 was measured. BMSCs were treated with PP2A inhibitor, Okadaic acid(OA), the phosphorylation of ERK1/2 and osteogenic differentiation were measured. lmmunoprecipitation was used to test the binding interaction between PP2A and ERK1/2, and the effect of SB203580 on the interaction. ResultsTreatment of BMSCs with osteogenic supplements resulted in activation of p38MAPK and ERK1/2 that coincided with osteogenic differentiation. Inhibition of p38MAPK activation by SB203580, blocked the osteogenic differentiation, whereas inhibition of ERK1/2 activation by PD98059, enhanced the osteogenic differentiation in a dose-dependent manner.SB203580 treatment resulted in increased ERK1/2 phosphorylation. By contrast, ARS treatment resulted in decreased ERK1/2 phosphorylation. Inhibition of PP2A by OA resulted in increased ERK1/2 phosphorylation. OS-induced osteogenic differentiation was also attenuated by PP2A inhibition. Immunoprecipitation confirmed the association of PP2A with ERK1/2 in BMSCs cultures, which was decreased by SB203580 treatment. ConclusionThe present study demonstrates a synergistic effect between p38MAPK and ERK1/2 signaling pathways via PP2A in BMSCs cultures, which may regulate the osteogenic differentiation of BMSCs.

ABSTRACT

Objective To explore the relationship between trace elements proportion in food and health. Methods 45 mice aged 12 month were randomly divided into 3 groups: control group(n=16) and two experimental groups(n=15).The mice in the experimental groups were given Zn, Li, Fe, Se, Mn, Cu, Co in different proportion by food for 60 days, then all mice were sacrificed, thymus weight, spleen weight, thymus coefficient, spleen coefficient, bone indexes were determined. Results Supplementation of the trace elements in food increased the content of Ca2+ in the bone of the aged mice and proportion variety of trace elements might induce the changes of thymus weight, spleen weight, spleen coefficient and bone indexes. Conclusion The reasonable proportion of trace elements in food could increase the content of bone Ca2+ in aged mice.

ABSTRACT

Objective Artificial mineral water pills were made by loading many kinds of trace elements into the ceramic carrier.Pure water could be mineralized after dipping the pills into the water.To assess the possible hazardous effects on the human health induced by the pills,a series of hygienic and toxicological experiments on the pills were carried out.Methods Acute oral toxicity test,micronucleus test of mice bone marrow cells,sperm abnormality test in mice and Ames test were con-ducted.Contents of trace elements,heavy metals and radioactivity in the mineralized water were determined according to the examination methods in Standard Examination Methods for Drinking Water and Examination Methods for Drinking Natural Min-eral Water.Results The contents of trace elements,heavy metals and radioactivity in mineralized water met the requirements of GB5749-1985Sanitary Standard for Drinking Water and GB8537-1995Sanitary Standard for Drinking Natural Mineral Wa-ter.Mineral pills were classified as actually non-toxic and the results of micronucleus test of mice bone marrow cells,sperm ab-normality test in mice and Ames test were negative.Conclusion Mineralized drinking water treated by artificial mineral pills were safe.
