ABSTRACT

BACKGROUND:Mesenchymal stem cel transplantation has a certain controversy in the treatment of liver cirrhosis, and its effects on the receptor liver structure and function need further studies. OBJECTIVE:To study the changes in liver ultrastructure, stereology parameters and liver function indexes of rat models with liver cirrhosis treated by bone marrow mesenchymal stem cel transplantation. METHODS:Rat models of liver cirrhosis were made using carbon tetrachloride and treated by bone marrow mesenchymal stem cel transplantation. Liver ultrastructure of al the rats were observed by transmission electron microscope, the stereology parameters of the hepatic sinusoid were analyzed by a stereology analysis software, and the serum liver function indexes were detected by a biochemical analyzer. RESULTS AND CONCLUSION: (1) The hepatic cels in the rat models exhibited acute hypoxia, lots of mitochondria were destroyed, and obvious karyopycnosis and capilarization of the hepatic sinusoid were found. The liver ultrastructure of rats undergoing cel transplantation was improved remarkably, the hepatic cel nucleus was nearly normal, mitochondrial sweling relieved notably and nuclear pore clogging lessened. (2) The number of hepatic sinusoids in the model group was reduced dramaticaly, but the total area and mean diameter of the hepatic sinusoid were enlarged significantly as compared with the cel transplantation and normal groups (P 0.05). These findings indicate that bone marrow mesenchymal stem cel transplantation can improve liver function and structure of rats with liver cirrhosis and lessen pathological changes of hepatic sinusoid, so it is an effective treatment for liver cirrhosis.

ABSTRACT

Objective To study the effects of bone marrow mesenchymal stem cells(BMSCs) transplantation combined with artesunate(Art) on rat model of liver fibrosis.Methods 20 rats from 120 SD rats were selected randomly as normal control group.10 rats from those were executed and extracted marrow to culture BMSCs.The other 90 rats were injected intraperitoneally with CCl4 to make liver fibrosis rat models.10 rats from the 90 rats were selected randomly to ensure the liver fibrosis model was made successfully after 10 weeks.Then,the rest 80 rats were divided randomly into 4 groups:model group,Art treated group,BMSCs transplantation group and Art + BMSCs transplantation group.Fibrous degeneration of the hepatic tissue was detected by Masson stain.The mRNA expression of α-SMA,TGF-β1 in liver were detected by RT-PCR.The hydroxyproline(Hyp) level in liver was detected by enzyme digestion method.Serum TNF-αand TGF-β1 were detected by ELISA.Serum hepatic fibrosis parameters were detected by chemiluminescence and serum liver functional parameters were detected by biochemistry method.Results Liver functional parameters of Art + BMSCs transplantation group were better than those of the other groups.ALB in Art + BMSCs transplantation group was (29.13 ± 3.48) g/L,which was significantly higher than (26.31 ± 2.42) g/L in Art treated group (24.35 ±3.12)g/L in BMSCs transplantation group and (20.14 ±2.86) g/L in model group.There were significant differences among the 4 groups(q =6.27 ~ 19.01,all P < 0.05).Liver fibrosis parameters of Art + BMSCs transplantation group were better than other groups.Hyp in Art + BMSCs transplantation group was (1.03 ±0.16) μg/L,which was significantly lower than (1.56 ± 0.19) μg/L in Art treated group,(1.45 ± 0.24) μg/L in the BMSCs transplantation group and (3.57 ±0.91) μg/L in the model group.There were significant differences among the 4 groups (q =20.47 ~ 27.70,all P < 0.05).The expression of TGF-β1 mRNA of Art + BMSCs transplantation group was lower than other groups.The index of TGF-β1 mRNA expression in Art + BMSCs transplantation group was (20.16 ± 5.21),which was lower than (58.15 ±9.16) in Art treated group,(46.31 ±6.82) in the BMSCs transplantation group and (97.08 ± 14.23) in the model group.There were significant differences among the 4 groups (q =20.35 ~ 44.93,all P < 0.05).Conclusion There're synergistic effects between Art and BMSCs in the treatment of liver fibrosis.

ABSTRACT

ObjectiveTo investigate the relationship between the level of plasma D-dimer(D-D) and hepatic functional reserve status in patients with hepatitis B (HB).Methods77 cases with HB were divided into two groups, hepatic function compensated group and hepatic function decompensated group.Plasma D-D, hepatic functional indexes and PT of all patients were detected.ResultsThe level of D-D in the hepatic function decompensated group was higher than that in the hepatic function compensated group(299.0 ± 498.5) μg/L vs (27.9 ± 34.0) μg/L(t =3.1, P < 0.01).D-D negatively correlated with ALB and PA(r =-0.6,-0.6, all P < 0.01), and positively correlated with TBIL and PT(r = 0.4,0.6, all P < 0.01).ConclusionThe level of plasma D-D related with hepatic functional reserve status,and detecting the level of D-D could be helpful to estimate hepatic functional reserve and prognosis of patients with HB.

ABSTRACT

Objective To explore the relationship between the expression of Hypoxia-induciable factor-1α (HIF-1α) and changes of hepatitis microcimulation in patients with chronic hepatitis B (CHB).Methods 120 patients with CHB were studied. The expression of HIF-1 α was observed by immunohistochemistry and the morphotogic changes were observed with transmission eletron microscope. Results Stenosis, blockage, dilataltion, capillarization of hepatic sinusoids were the main morphotogic changes of liver. The expression of HIF-1α was positive or stronger positive. There is a positive correlation between the expression of HIF-1α and microcirculation disturbance of liver.Conclusion The disturbance of liver with CHB could exacerbate the dysbolism of nutrient substance and oxygen between hepatic cells and blood circulation. Hypoxia of hepatic cells was an important reason of the liver's pathological changes.

ABSTRACT

Objective To study the effects of inflammation of blood vessels to microcirculation disturbance in liver of patients with chronic hepatitis B(CHB). Methods188 cases of hepatic tissue with CHB were observed by light microscope and electronmicroscope.They were graded according to the inflammation of blood vessels in liver,and to discuss the relationship between the inflammation of blood vessel and microcirculation disturbance. ResultsAccording to the inflammation of blood vessels in liver,188 cases of CHB were divided to 3 grades,67 cases in grade 1,89 cases in grade 2 and 32 cases in grade 3.The rate of hepatic sinusoidal stenosis,hepatic sinusoidal blockage,the formation of microthrombus,decreased sizes and reduced numbers of sinusoidal endothelial cells' penestrate,the formation of collagen in Disse's spaces and the formation of basilar membrane increased along with the aggravation of inflammation of hepatic blood vessels. ConclusionInflammation of hepatic blood vessels was an important factor on microcirculation disturbance in liver of patients with CHB.It should be done something to improve the microcirculation,and decrease inflammation at the same time.

ABSTRACT

Objective To observe the effects of intrabepatic injection with bepatocyte growth factor and dexamethason on stereological quantitative changes of hepatic sinusoidal tissues and expression of IV-collagen on hepatic sinusoidal walls of patients with hepatic cirrhosis.Methods Under the guide of hypersound,98 cases of hepatic cirrhosis were intrahepatic injected with 80mg hepatocyte growth factor and 1mg dexamethason at one time,twice a week,12 times a course,pathological changes of hepatic sinusoidal tissues and the expression of IV-coliagen on hepatic sinusoidal walls were detected by liver biopsy after one course.Results Pathological changes of hepatic sinusoidal tissues were improved obviously(P<0.01 ) and the expression of IV-collagen was alleviated significantly(P<0.01 ) on 98cases of hepatic cirrhosis after one course.Conclusion Pathological changes of hepatic sinusoidal tissues and the expression of IV-collagen on hepatic sinusoidal wails can be improved obviously on patients with hepatic cirrhosis who had been intrahepatic injected with hepatocyte growth factor and dexamethason under the guide of hypersound.

ABSTRACT

Objective To study the effects of melatonin (MT) on hepatic tissue of autoimmune hepatitis (AIH). Methods AIH model rats were divided randomly as model group, MT group and pig hepacyte growth factor (pHGF) group, to observe the pathological changes of hepatic tissue of the three groups. Results Hepatic tissue inflammation and accrementition graded of elastic fibers, reticular fibre, collagenous fibrils of MT group [(0.97±0.16),(1.09±0.17),(1.31±0.17)] were much lighter than model group [(1.55±0.18),(2.00±0.14),(3.00±0.14)](all P<0.01). The expression of VEGF was weaker in MT group than in pHGF group. There was no distinct difference between MT group and pHGF group. Conclusion MT can obviously improve the pathological changes of hepatic tissue of rats with AIH.
