ABSTRACT

Smart manufacturing still remains critical challenges for pharmaceutical manufacturing. Here, an original data-driven engineering framework was proposed to tackle the challenges. Firstly, from sporadic indicators to five kinds of systematic quality characteristics, nearly 2,000,000 real-world data points were successively characterized from Ginkgo Folium tablet manufacturing. Then, from simplex to the multivariate system, the digital process capability diagnosis strategy was proposed by multivariate Cpk integrated Bootstrap-t. The Cpk of Ginkgo Folium extracts, granules, and tablets were discovered, which was 0.59, 0.42, and 0.78, respectively, indicating a relatively weak process capability, especially in granulating. Furthermore, the quality traceability was discovered from unit to end-to-end analysis, which decreased from 2.17 to 1.73. This further proved that attention should be paid to granulating to improve the quality characteristic. In conclusion, this paper provided a data-driven engineering strategy empowering industrial innovation to face the challenge of smart pharmaceutical manufacturing.

ABSTRACT

Objective To study the relationship between DNA methylation and pathogenesis of childhood immune thrombocytopenic purpura (ITP) by examining the expression of DNA methyltransferase 1(Dnmt1) and DNA methyltransferase 3a (Dnmt3a) mRNA in peripheral blood lymphocytes of the children with ITP. Methods Expression of Dnmt 1 and Dnmt3a mRNA in the peripheral blood lymphocytes in 36 children with newly diagnosed ITP and 26 healthy children were detected using RT-PCR. Results Dnmt1 mRNA expression in peripheral blood lymphocytes in children diagnosed with ITP was 3.02±0.49, significantly lower than 4.58±0.52 in the control group (t=11.95, P<0.001). Dnmt3a mRNA expression in peripheral blood lymphocytes in children diagnosed with ITP was 1.49±0.44, signiifcantly lower than 2.41±0.32 in the control group (t=9.12, P<0.001). Conclusions Children with newly diagnosed ITP have lower DNA methylation status in peripheral blood lymphocytes as compared to that in healthy children. The DNA methylation may play an important role in the etiology of acute ITP in children.

ABSTRACT

The rat model of dysmenorrhea, pelvic inflammation disease (PID), and uterine myoma were established to observe the effect ofGui-Zhi Fu-Ling Capsule (GZFLC) with the changes of main components. The molecular imprinted technology was used to quantitatively knock out main components in GZFLC (the knocked-out ratios were 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90%). And the observation was made on the writhing times, latency, and effects on ET-1 and PGF2α of uterine tissues in the pitocin-induced dysmenorrhea rat model. Effects on TNF-α and IL-2 were observed in PID rat model induced by mixed bacteria plus mechanical damages. Effects on the weight and index of uterus, levels of E2 and P in serum were detected on the uterine myoma rat model induced by estrogen-burden. The results showed that the GZFLC efficacy was decreased with gradient knockout of 15 main components. When 10%-40% was knocked out, there was still significant difference compared to the model group (P 0.05). Meanwhile, the GZFLC efficacy was decreased significantly compared to the GZFLC group (P <0.05, orP < 0.01). It was concluded that 15 chemical compounds, which included gallic acid, paeoniflorin, paeonol, and etc., may be the main material basis of GZFLC. Meanwhile, it provided a useful reference for the quality control of main components in GZFLC.

ABSTRACT

To study the pathogen distribution, antimicrobial susceptibility and risk factors of postoperative nosocomial infections among children with congenital heart disease. Three hundreds children with congenital heart disease admitted to our hospital to receive surgeries from February 2010 to February 2013 were selected. A total of 120 children were tested as positive by sputum culture, with the infection rate of 40.0%. The top five most common pathogenic microorganisms included Staphylococcus epidermidis, Staphylococcus aureus, Enterococcus, Pseudomonas aeruginosa, and Candida albicans. S. epidermidis, S. aureus and Enterococcus were highly resistant to penicillin, azithromycin and erythromycin, moderately susceptible to levofloxacin and cefazolin, and completely susceptible to vancomycin. Multivariate Logistic regression analysis showed that hospitalization stay length, combined use of antibiotics, systemic use of hormones, mechanical ventilation and catheter indwelling were the independent risk factors of postoperative nosocomial infections [P < 0.05]. Nosocomial infection, which was the most frequent postoperative complication of pediatric congenital heart disease, was predominantly induced by Gram-positive bacteria that were highly susceptible to cephalosporins and vancomycin. Particular attention should be paid to decrease relevant risk factors to improve the prognosis

ABSTRACT

To discuss the treatment effects and costs of surgery and Gamma Knife on hippocampal sclerosis [HS]-induced intractable epilepsy of children below age 10 years. The children below age 10 years who suffered from HS-induced intractable epilepsy from June 2010 to June 2012 were subjected to surgical and Gamma Knife treatments respectively according to their preference. The short-term curative rates of the surgical group and the Gamma Knife group were 93.51% and 54.87%, respectively. The average expenses of the two groups were 10,000 CNY [Chinese Yuan] and 22,000 CNY, respectively. The two groups were treated safely and effectively, but the surgical treatment led to better results at a reduced cost
