ABSTRACT

Abstract@#Objective To investigate the willingness to receiving multitarget stool DNA (MT-sDNA) testing and factors affecting the payment among individuals receiving colonoscopy screening, so as to provide the evidence for the formulation and health economic evaluation of colorectal cancer screening strategies. @*Methods@# Individuals at ages of 40 to 75 years that received colonoscopy screening in The Affiliated Hospital of Ningbo University Medical School from August 2021 to March 2022 were sampled. Participants' demographics, living behaviors, family history, willingness to receive MT-sDNA testing and willingness to pay for MT-sDNA testing were collected using questionnaire surveys, and factors affecting the willingness to receive and pay for MT-sDNA testing were analyzed using a multivariable logistic regression model.@*Results @#A total of 546 respondents were enrolled, with a mean age of (56.25±8.66) years and including 282 men (51.65%). There were 504 respondents that were willing to receiving MT-sDNA testing (92.31%) and 480 that were willing to pay for the MT-sDNA testing (88.24%). Multivariable logistic regression analysis showed that a family history of colorectal cancer in first-degree relatives (OR=0.246, 95%CI: 0.068-0.888), history of hemorrhoids (OR=0.300, 95%CI: 0.109-0.826) resulted in low willingness to receive MT-sDNA testing, and recognizing the reliability of MT-sDNA testing (OR=5.749, 95%CI: 1.480-22.323), considering no difficulty in sampling for MT-sDNA testing (OR=32.042, 95%CI: 6.666-154.021) and considering a difficulty in sampling for MT-sDNA testing (OR=20.278, 95%CI: 4.405-93.354) resulted in high willingness to receive MT-sDNA testing, while recognizing the reliability of MT-sDNA testing (OR=5.003, 95%CI: 1.761-14.216), concern about the reliability of MT-sDNA testing (OR=4.166, 95%CI: 1.285-13.501), considering no difficulty in sampling for MT-sDNA testing (OR=6.558, 95%CI: 2.105-20.428) and considering a difficulty in sampling for MT-sDNA testing (OR=5.820, 95%CI: 1.810-18.720) resulted in high willingness to pay for the MT-sDNA testing among individuals receiving colonoscopy screening.@*Conclusion @#A family history of colorectal cancer in first-degree relatives, history of hemorrhoids and awareness of MT-sDNA testing are factors affecting the willingness to receive and pay for the MT-sDNA testing among individuals receiving colonoscopy screening.

ABSTRACT

@#Objective To predict the molecular mechanism of Dihuang (Rehmanniae Radix) in the treatment of diabetic nephropathy (DN) complicated with depression based on network pharmacology. Methods The components of Dihuang (Rehmanniae Radix) were identified from the Integrated Pharmacology-based Research Platform of Traditional Chinese Medicine (TCMIP), Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP), and relevant literature. The component targets were detected by combining the SwissTargetPrediction and PubChem databases. Disease targets were collected from the Therapeutic Target Database (TTD), DisGeNET, and Ensembl databases with “diabetic nephropathy” and “depression” as keywords. The disease-component targets were mapped using Venny 2.1.0 to obtain potential targets. A protein-protein interaction (PPI) network was constructed using the Search Tool for the Retrieval of Interacting Genes/Proteins (STRING) database and Cytoscape 3.7.2. The co-expression genes of the key targets were collected based on the COXPRESdb 7.3. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis were performed for potential targets using R language. Target-component docking was verified and evaluated using Discovery Studio 4.5. Results According to the databases and literature reports, Dihuang (Rehmanniae Radix) contained 65 active components, and had 155 related targets for the treatment of DN complicated with depression. PPI screening showed that the key targets included serine/threonine protein kinase 1 (AKT1), signal transducer and activator transcription 3 (STAT3), interleukin 6 (IL-6), mitogen-activated protein kinase 1 (MAPK1), and vascular endothelial growth factor A (VEGFA), etc. GO enrichment analysis mainly involved biological processes, such as lipid metabolism, protein secretion regulation, cell homeostasis, and phosphatidylinositol 3 kinase activity. KEGG pathway enrichment analysis included the role of the AGE-RAGE signaling pathway in diabetic complements, insulin resistance (IR), neurotrophin signal path, Toll-like receptor signaling pathway, relaxin signaling pathway, epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs), etc. Molecular docking showed that the target had high affinity for stachyose, manninotriose, verbascose, nigerose, etc. Conclusion Based on network parmacology, this study preliminarily predict the effects of Dihuang (Rehmanniae Radix) in treating DN complicated with depression by regulating inflammation, glucose metabolism, nution nerve, etc.

ABSTRACT

Objective To propose an airbag-type helmet cushioning lining structure and analyze its protective effect on head injury of two-wheeled bicycle riders. Methods The airbag lining was applied to two typical two-wheeled bicycle helmets for bicycles (half helmets) and motorcycles (full helmets). Then kinematic and biomechanical responses of the human head model were predicted from impact simulations under test conditions of the standard GB 24429-2009 and the regulations ECE R22.05, and conventional expanded polystyrene (EPS) helmets were compared from the perspective of skull fracture and brain injury risk, so as to make comprehensive evaluation on protective performance of the airbag helmet. Results When the airbag pressure was 0.06 MPa, the relevant amount of human skull fracture under protection of airbag helmet (half/full helmet) was smaller than 120 g and 150 g, respectively; the risk of skull fracture was basically lower than 40%; the maximum principal strains of the brain were both smaller than 0.3, which indicated that the risk of mild brain injury was lower than 25%. Generally, the risk of human skull fracture and head injury under protection of airbag helmets was lower than that under protection of EPS helmets. Conclusions The airbag helmet designed in this study has a good protective effect, which can give attention to the protection of both skull fracture and head injury, providing a basic example for the design of novel helmet. Injury risk analysis can also provide the preliminary reference for emergency diagnosis on head injury of cyclists.

ABSTRACT

To analyze the structural specificity of Shenque (CV 8) in terms of vascular biology from the three aspects of structure, tissue and molecular anatomies. On structural anatomy Shenque (CV 8) possessed defined vascular structure and was the only acupoint directly affected vascular intima. The basis of the specific therapeutic effects was owing to its relation with vessel and abundant microcirculation. Endothelial cells and microvascular endothelial cells were the tissue basis of the starting of therapeutic specificity. Molecular anatomy involved in the functions of transient receptor potential vanilloid family (TRPV) pathway and neuronal peptide secretion of endothelial cells in the structural function of the point. The "restoringfor collapse" effect of salt-partition moxibustion at Shenque (CV 8) was analyzed based on the local biological specificity of blood vessels. The mechanisms were concluded as effectively targeting the dysfunction of microvascular endothelial cells and acquiring the maximum quantity of moxibustion by repeated warm-heat stimulation. The vascular biological structural features of Shenque (CV 8) may contribute to direct therapeutic effects on endothelial cells by the point.

ABSTRACT

Objective To explore how the different intervention measures affect the outcome of central venous catheter-related thrombosis (CRT) in children. Methods A retrospective analysis was carried out to collect the clinical data of patients with CRT from the nursing management system of the Children′s Hospital Affiliated to Zhejiang University School of Medicine which reported by each nursing unit from January 1,2015 to December 31,2015. Results Totally 108 cases were included (72 boys and 36 girls), median age of 24.5 months (ranged from 1 month to 14 years old). Nearly 42.59% (46/108) patients suffered from neurological diseases. Nearly 55.56%(60/108) CRT was detected in the first week after catheterization. Once CRT conformed, there were four kinds clinical intervention options applied. Intervention 1: thrombolytic therapy with urokinase combined anticoagulation with nadroparin calcium. Intervention 2: anticoagulant therapy only. Intervention 3: thrombolytic therapy alone. Intervention 4: no medications. The differences of effective between the four kinds of intervention were statistically significant (χ2=13.380, P=0.004). The single-factor regression analysis was done to each relevant factor. Finally the multivariate regression analysis showed four factors had impacts upon the results. The factors were as follows:gender (OR=10.400, 95%CI 1.879-57.563, P=0.007);interval (OR=1.107, 95%CI 1.035-1.184, P = 0.003), size of thrombus(OR = 1.562,95%CI 1.033-2.362,P=0.035; Intervention 2 (OR =11.757, 95% CI 2.254- 61.327, P = 0.003), intervention 4 (OR = 35.397, 95% CI 3.493-358.760, P =0.003). Conclusions The earlier and small size thrombus is more soluble. Thrombolytic therapy or combined anticoagulation is more effective. It is recommended that if no contraindications presents, thrombolytic combined with anticoagulant therapy should start early standardized treatment.

ATP (Adenosine triphosphate) is an important endogenous damage-associated molecular pattern (DAMP). P2X7R is an ATP-gated cation channel. ATP-P2X7R plays a vital role in the pathophysiology of many diseases because P2X7R is distributed on various immune cells. ATP-P2X7R signal transduction pathway has been implicated to participate in the body's im-mune defense against pathogens. This paper reviews the recent progress regarding ATP-P2X7R and its effects on parasitic diseas-es.

ABSTRACT

BACKGROUND: How to establish a stable in vitro culture system, including location of corneal limbal epithelial stem cells, in vitro sample harvest, in vitro culture, vector selection, as well as identification methods, play a key role in corneal limbal epithelial stem cells culture. OBJECTIVE: To culture the isolated rabbit corneal limbal epithelial stem cells and to identify the biological properties of cultured cells. METHODS: The primary rabbit cornel limbal epithelial stem cells were isolated and cultured with tissue inoculation using human amniotic membrane as vector. The growth features of cells were observed under an inverted microscope. The morphology of cells was observed by hematoxylin-eosin staining and a scanning electron microscope. Furthermore, the monoclonal antibody AE5 and P63 two-step immunohistochemical staining were used to identify limbal epithelial stem cell protein expression. RESULTS AND CONCLUSION: The rabbit corneal limbal epithelial stem cells could be successfully cultured and maintained a relatively high value-added potential in vitro. Rabbit corneal limbal epithelial stem cells cultured on the amniotic membrane pull netted cellular layer. The AE5 monoclonal antibody positive rate of primary cultured cells was about 5% and P63 monoclonal antibody positive up to 90%. AE5-positive rate increased and P63-positive rate decreased with the increase in the number of subculture. The rabbit limbal epithelial stem cells can be successful culture and amplified on human amniotic membrane in vitro by limbal tissue culture method. The cultured cells maintain the characteristics of corneal epithelial cells. The rabbit corneal limbal epithelial stem cells can form grafts on the amniotic membrane.

ABSTRACT

Objective To investigate and analyze whether the different types of radial head fracture and internal fixations have influence on the out comeof elbow function after surgery. Methods We reviewed the clinical data of 46 cases of radial head fractures from August 2002 to December 2008, 34 males, and 12 females, aged from 19 to 51years. All these cases underwent open reduction and internal fixation including fixation with Kirschner wire fixation, mini-plate and/or screw fixation and radial head resection. Results Forty-six cases were followed up in this study for a period of 8 months to 4 years (mean 2 yeats and 4 months).According to Mayo assessment of elbow function, the results of 35 cases were excellent or good (35/46 cases, 76.1%). Better function recovery was got in those who were preferred mini-screw internal fixation or combined with mini-plate. The same results were also found in those with Mason type Ⅱ fracture and in those underwent surgery within 2 weeks. Conclusions Good results can be got through surgical treatment as the indication of open reduction is clear, especially in cases with radial head fracture of Mason type Ⅱ and Ⅲ.The type of radial head fracture, the method of internal fixation and the timing of surgery all affect the outcome.

ABSTRACT

<p><b>BACKGROUND</b>To evaluate the diagnostic value of flow cytometric DNA analysis of sputum cells in lung cancer patients.</p><p><b>METHODS</b>The DNA content of sputum cells was determined by flow cytometry in 44 patients (29 patients with lung cancer, and 15 patients with benign pulmonary disease). The results of DNA content of sputum cells were compared with those of cytologic morphology of sputum cells.</p><p><b>RESULTS</b>The last pathologic diagnosis was used as a diagnostic criterion, the sensitivity of DNA heteroploidy analysis was 82.8% , which was significantly higher than that of sputum cytology (82.8% vs 27.6%, P < 0.005); and the positive rate of heteroploidy in sputum samples in stage III and IV disease was significantly higher than that in stage I and II disease (87.5% to 76.9%, P < 0.002 5). In one case, the heteroploidy was found 10 months in advance of cytometry. In the control, heteroploidy was detected in 33.3% (2/6) patients with inflammatory pseudotumor and 50.0% (2/4) patients with tuberculous nodule, who were found with positive staining of Ki67 and PCNA.</p><p><b>CONCLUSIONS</b>Flow cytometry seems to be a valuable adjunctive method for the diagnosis of lung cancer, and probably for the early diagnosis.There are clonal changes in some inflammatory pseudotumor and tuberculous nodule.</p>
