ABSTRACT

· RNA interference (RNAi) is the post-transcriptional gene silencing based on sequence-specific degradation of mRNA,and triggered by double-stranded RNA.RNAi technology is widely used in gene therapy and research of gene function and almost become a standardized technical tool with very broad prospect. The eyes with immune privilege are very suitable for the treatment of RNAi,which has been widely used for a variety of eye diseases, especially cataract. This paper briefly introduces the basic principle of RNAi technology and summarizes the recent research and application of this technology in cataract.

ABSTRACT

AIM: To investigate the alterations of tear film after sutureless large incision manual cataract extraction (SLIMCE). METHODS: Sixty-eight SLIMCE operation eyes were studied with slit-limp microscope, break- up time (BUT), SchirmmerⅠtest (SⅠt),and fluorescence(FL) to observe the alterations of tear film at different time points in postoperation. Impression cytology and microphoto-analyses technique were also applied to observe the goblet cells at different time points postoperation(7,14,30,60,90 days). RESULTS: Subjective complaint of dry eye within 90 days after the operations were significantly increased compare with preoperations(5-27,23,19,16,13; 2-16,14,8,6,3). The schirmmer Ⅰ test were greatly increased in 14 days postoperation(10.1±4.5;15.0±4.7,13.8±5.7),the mean scores of fluorescence increased (0-17,9,5;0-8,3,1) and the mean break-up time decreased in 30 days post-operation(10.3±2.2;5.5±2.3,7.0±2.4,7.9±2.2) (P<0.05). CONCLUSION: SLIMCE operation have effect on the stability of tear film.

ABSTRACT

·AIM: To investigate the effect of vitamin A on the conjunctival goblet cells of rat after corneal transplantation.·METHODS: Rat graft rejection models of corneal transplantation were established. SD rats were receptor and Wistar rats were donors. After corneal allografts were performed, 48 SD rats were randomly divided into three groups, 16 rats in each group. Group A was blank control group; group B was treated by oculotect gel (containing vitamin A); group C was treated by 1g/L dexamethasone eyedrops. Besides, group D was normal unoperated eyes.Slit-lamp microscope was employed to record and compare rejection index (RI) of corneal transplantation. Through HE,PAS staining of conjunctival histological sections and image analysis system, the number and morphology of conjunctival goblet cells were observed and analyzed between operation group and normal group.·RESULTS: The HE, PAS staining detection showed that the number of conjunctival goblet cells in oculotect gel group,1g/L dexamethasone eyedrops group and control group is lower than that in normal group after surgery (P<0.01). The number of conjunctival goblet cells in oculotect gel group and 1g/L dexamethasone eyedrops group is higher than that in control group (P<0.05). The number of conjunctival goblet cells in 1g/L dexamethasone eyedrops group is higher than that in oculotect gel group (P<0.05).·CONCLUSION: The results indicate that vitamin A may inhibit the decrease of conjunctival goblet cells after corneal allograft rejection in rats.

ABSTRACT

·AIM: To investigate a surgical method of modified lensectomy within combined lensectomy and vitrectomy surgery in traumatic eyes.·METHODS: Clinical records were studied retrospectively for a series of consecutive 27 traumatic patients who were performed vitrectomy combined with lensectomy surgery. A modified lensectomy through pars plana incision of sclera was performed to these cases and lens capsule was reserved. It was then combined with vitreous and retinal operations.·RESULTS: All lens were removed completely with lens capsule remained. All cases achieved improved visual acuity.Intraocular pressure (IOP) in all eyes was in normal range after surgery.·CONCLUSION: The modified lensectomy is a safe and effective surgery manner which has few complications and is more suitable for traumatic eyes in the combined lensectomy and vitrectomy surgery.KEYWORDS:ocular trauma; lensectomy; vitrectomy
