ABSTRACT

@#Proanthocyanidin (PA), as a kind of natural plant polyphenol, have a variety of biological functions, such as promoting remineralization, inducing collagen cross-linking, inhibiting protease activity and inhibiting bacteria. Therefore, PA could be broadly used in the clinical application of treatment and repair of deep caries in the future; for example, PA could promote dentin remineralization, improve resin-dentin bonding durability and improve the dentin acid erosion effect. This application potential of PA arises from several features, firstly, PA can not only promote dentin remineralization on its own or with other remineralizers but also exhibits antibacterial effects, which can inhibit acid production while reducing the formation of cariogenic pathogens and their biofilms. Based on the above features, PA can reduce the incidence of caries disease; thus, PA improves deep caries and long-term effects after treatment. In addition, PA added to adhesives or etch agents can improve the etching and bonding effect of dentin by inducing collagen cross-linking and inhibiting protease activity, thus achieving the ultimate goal of improving the bonding performance of deep caries. This paper summarizes recent progress of research on PA for the treatment and repair of deep caries, including the promotion of dentin remineralization and antibacterial activity as well as the improvement in dentin bonding and acid etching effect, to provide a more comprehensive reference for treating and restoring deep caries in clinical practice.

ABSTRACT

Objective@# To investigate the effects of platelet-rich fibrin (PRF) and acellular dermal matrix (ADM) on the repair of oral mucosal defects and to provide the basis for soft tissue growth in oral implant operations.@*Methods@#Thirty-six healthy male Japanese big ear rabbits were randomly divided into the PRF group, ADM group, Autograft group (autologous connective tissue transplantation group) and Control group (blank control group); each group contained nine rabbits. Between the midline and the hard palate maxillary incisors, in an 8-mm location preparation and a 10-mm standard mucosa defect, the ADM group, PRF and Autograft group were implanted with ADM, autologous PRF and autologous cornification mucosa, respectively, whereas the control group had wound gauze compression processing at 7, 14, and 21 days to determine the wound healing rate in the area selected by HE staining. The inflammatory grade and average epithelial thickness were observed, and the results were statistically analyzed.@*Results @#Compared with the control group, the PRF, ADM and Autograft groups had significantly advanced wound healing (P < 0.05). The wound healing degree in the PRF group was similar to that of the ADM group at all time points (P > 0.05). The wound healing degree in the PRF and ADM groups was lower than that of the Autograft group at each time point (P < 0.05). HE staining results showed that compared with the control group, the levels of inflammation in the PRF group, ADM group and Autograft group were reduced, and the difference was statistically significant (P < 0.05). Nevertheless, there was no significant difference between the PRF, ADM and Autograft groups (P > 0.05). The epithelial thickness in the ADM group was similar to that in the Autograft group (P > 0.05). The epithelial thickness in the ADM group was higher than that in the PRF group at 7 d and 14 d (P < 0.05), but there was no significant difference at 21 d (P > 0.05).@*Conclusion @#PRF and ADM have similar healing effects in repairing oral mucosa defects, and they can be used as soft tissue augmentation materials instead of connective tissue transplantation.

ABSTRACT

@#Dental hard tissues lack the ability to self-heal. In dentin and cementum, hydroxyapatite (HA) can exist outside and/or inside collagen fibers. It is difficult to repair or regenerate HA with a highly ordered orientation in the presence of collagen fibers. At present, the biomimetic mineralization of dentin and cementum, mainly carried out by imitating its biological formation process and its physiological structure, can be divided into those originating from the fiber mineralization mechanism and those with HA as the main component. The materials used include natural materials such as demineralized dentin matrix (DDM) and calcined bovine hydroxyapatite (BHA), and synthetic materials such as polymer-induced liquid precursor (PILP) and synthetic HA. In the future, natural materials and synthetic materials should be combined for the restoration and regeneration of dentin and cementum by means of biomimetic mineralization of calcium phosphate released by remineralization solution-HA.

ABSTRACT

Autoimmune is involved in the pathogenesis of ventricular remodeling in acute myocardial infarction (AMI). In the present study, we investigated the effect of anti-cardiac myosin heavy chain antibodies (AMHCA) from patients with AMI on rat cardiomyocyte apoptosis. Cardiomyocyte apoptosis was observed and measured by DNA end labeling and Annexin- V/PI double-staining assay.The expression of apoptosis related p53 and Bcl-2 protein and the second messenger calcium were detected respectively by Western blotting, patch clamp and confocal calcium imaging. The results showed that AMHCA was able to induce cardiomyocyte apoptosis in a dose dependent manner.Apoptosis-accelerating nucleoprotein p53 was up-regulated, while apoptosis-inhibiting cytoplasmic protein Bcl-2 was down-regulated. In parallel, cytoplasmic calcium concentration was elevated.There was no effect on L-type calcium currents. It is concluded that AMHCA in patients with AMI as a novel triggering factor can induce cardiomyocyte apoptosis, which contributes to ventricular remodeling.
