ABSTRACT

Objective:To study changes and clinical significance of serum interleukin-33(IL-33) of patients with adult measles complicated acute liver damage. Methods: Totally,88 adult measles patients were selected and divided into acute liver damage group (n=60) and no liver damage group(n=28). At the same time,20 healthy adults were selected as healthy controls. Serum IL-33 levels were measured by enzyme linked immunosorbent assays. Venous blood samples of IL-33 were drawn on the day of admission and were repeated at 7 and 14 days later. The correlation of IL-33 and biochemical indicators were analyzed. 60 patients with liver damage were divided into protecting liver and without protecting liver group in accordance with the method of random numbers. The level of serum IL-33 was compared at 7 and 14 days between the two groups of patients. Results: Patients with adult measles complicated acute liver damage and no liver damage had elevated serum IL-33 levels than that in the controls [( 205. 20 ± 25. 74 ) pg/ml, ( 168. 70 ± 18. 14)pg/ml,vs(132. 17±12. 41)pg/ml,P<0. 05 for all],especially in acute liver damage group. The expression of serum IL-33 in no protecting liver group was significantly higher than protecting liver group at 7th day. IL-33 was no significant difference in patients between protecting liver and without protecting liver groups at 14th day(P>0. 05). The level of IL-33 was a significant decrease in the treatment and there was no significant difference between healthy controls at 14th day(P>0. 05). The level of IL-33 and liver damage were consistent. Serum IL-33 levels in patients with liver damage showed positive correlation with levels of ALT,GGT and IL-6 ( r=0. 392 1,0. 503 9,0. 724 9,P<0. 001). Conclusion: IL-33 may have proinflammatory effects in the phase of adult measles complicated acute liver damage and its serum level reflects the severity of liver inflammation.

ABSTRACT

OBJECTIVE To clone and express toxin C (tcdC) gene pathogenicity locus of Clostridium difficile (CD), and prepare the polyclonal antibody against TcdC protein. METHODS The tcdC gene was amplified from CD (ATCC43255) strain genome DNA and inserted into prokaryotic expression vectors. The correct recombinant plasmids were transformed into E.coli which was induced to express the GST-TcdC/L and IL1-TcdC/L fusion protein. Meanwhile, the fusion proteins were respectively purified through Ni-NTA agarose affinity chromatography and Q-Sepharose Fast Flow. The purified GST-TcdC/L fusion protein was used as an antigen to inoculate rabbits to produce antiserum. Two weeks after the final immunization, the rabbits were sacrificed and serum was collected. The titer of the serum was deter-mined by ELISA and the reactivity of the polyclonal antibody was identified by Western blotting. RESULTS The SDS-PAGE result showed that GST-TcdC/L and IL1-TcdC/L fusion proteins were expressed at the size of 45 ku and 39 ku. The titer of TcdC/L polyclonal antibody was over 1:6.4 × 104. Western blotting detection demonstrated that the TcdC/L polyclonal antibody recognized the TcdC protein in CD (ATCC43255). CONCLUSION The tcdC/L gene is cloned and the polyclonal antibody against TcdC/L is prepared, which will contribute to studing the function and mechanism of tcdC gene in pathogenicity locus.

ABSTRACT

OBJECTIVE@#To determine the relationship between the blood serum brain-derived neurotrophic factor (BDNF) level and cognitive function deterioration in patients with obstructive sleep apnea/hypopnea syndrome (OSAHS), and to explore the possible mechanism of cognitive impairment.@*METHODS@#Twenty-eight male OSAHS patients and 14 normal males (as controls) were enrolled in the study. Polysomnography and the Montreal cognitive assessment (MoCA) were conducted. The blood serum BDNF levels were measured using ELISA.@*RESULTS@#The OSAHS group had significantly decreased blood serum BDNF levels compared with the control group (t = -10.912, P = 0.000). The blood serum BDNF level of the subjects was significantly positively associated with the MoCA score (r = 0.544, P = 0.000), significantly negatively associated with the apnea-hypopnea index (AHI) and shallow sleep (S1+S2) (AHI: r = -0.607, P = 0.000; S1+S2: r = -0.768, P = 0.000), and significantly positively associated with the lowest SaO(2) (LSO), slow wave sleep (S3+S4), and rapid eye movement sleep (REM) (LSO: r = 0.566, P = 0.000; S3+S4: r = 0.778, P = 0.000; REM: r = 0.575, P = 0.000).@*CONCLUSIONS@#OSAHS patients have significantly decreased blood serum BDNF levels compared with the control. Nocturnal hypoxia as well as the deprivation of slow wave sleep and REM may lead to the decreased serum BDNF level of OSAHS patients. This decreased blood serum BDNF level may contribute to the cognitive impairment in OSAHS.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the feasibility of repairing the long defects in rabbits by double-bridging technique and chitosan-collagen chambers.</p><p><b>METHODS</b>Thirty Chinese rabbits were divided into three groups randomly. 15 mm defects in right facial nerve of rabbits were repaired in three groups: double-bridging (group I ), nerve autografts (group II), and normal control (group III). General observation, electrophysiological study, histological study and image analysis were performed 12 weeks postoperatively. All results were used to evaluate the nerve regeneration.</p><p><b>RESULTS</b>Chitosan-collagen chambers were obviously degraded 12 weeks postoperatively and there was no foreign body reaction at this stage. They also restrain the formation of neuroma and provided a good microcircumambience for nerve regeneration. The recovery of nerve regeneration were good in group I. The nerve conduction velocity (NCV) and compound muscle action potential (CMAP) of regenerated nerve were analyzed, which showed that there was no significant difference between the group I and group II (P > 0.05), but they all did not recover to normal level(P < 0.01).</p><p><b>CONCLUSION</b>Double-bridging technique is a simple method for treatment of long nerve defect and its effect is certain. Chitosan-collage chambers is suitable to implant into body in order to repair a nerve defect with excellent biocompatibility.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To determine whether serum homocysteine and folate levels are correlated with the occurrence of neonatal asphyxia and to study the effects of gender and gestational age on serum homocysteine and folate levels.</p><p><b>METHODS</b>Thirty-five neonates with mild asphyxia (19 males and 16 females) and 40 normal neonates (control group,18 males and 22 females) were enrolled in this study. The asphyxia and the control groups consisted of 10 and 11 cases of preterm infants respectively. Serum homocysteine levels were measured using ELASA. Serum folate levels were measured using radioimmunity assay.</p><p><b>RESULTS</b>Serum homocysteine level (14.66+/-2.61 micromol/L vs 7.55+/-0.50 mumol/L; P<0.05) was significantly higher and serum folate level (2.47+/-0.24 ng/mL vs 3.28+/-0.28 ng/mL; P<0.05) was significantly lower in the asphyxia group than that in the control group. There were no significant differences in serum levels of homocysteine and folate between males and females either in the asphyxia group or the control group. The asphyxiated neonates born at premature showed increased serum homocysteine level compared with the full-term neonates with asphyxia (21.25+/-5.01 micromol/L vs 12.34+/-2. 01 micromol/L; P<0.05).</p><p><b>CONCLUSIONS</b>The increased serum homocysteine level and decreased serum folate level are correlated with the occurrence of neonatal asphyxia. Serum homocysteine and folate levels are not associated with the gender. A more significantly increased serum homocysteine level may be found in asphyxiated neonates born at premature.</p>
