ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effects of the histone deacetylase inhibitor, MS-275, on the immune molecule content and categories in hepatocarcinoma exosomes.</p><p><b>METHODS</b>Exosomes were isolated from the human hepatocarcinoma cell lines, HepG2 and Hep3b, and purified by a combination technique of ultrafiltration centrifugation and sucrose gradient ultracentrifugation. The expressions of heat shock protein (HSP)70, human leukocyte antigen (HLA)-I, HLA-DR, cluster of differentiation (CD) 80 and NY-ESO-1 on exosomes were analyzed with immunoelectron microscopy and Western blotting before and after MS-275 treatment. Intergroup differences were statistically analyzed by the Student's paired t-test.</p><p><b>RESULTS</b>MS-275 treatment of both HepG2 and Hep3b cell types significantly increased the numbers of exosomes, their total protein content, and expression of HSP70, HLA-I and CD80 (per 100 exosomes), as compared to non-treated cells (all, P less than 0.01). MS-275 was also found to induce de novo expression of HLA-DR, but had no significant effect on NY-ESO-1 expression (P more than 0.05). The findings from immunoelectron microscopy confirmed those from Western blotting.</p><p><b>CONCLUSION</b>The histone deacetylase inhibitor, MS-275, can significantly alter the immune molecule content and categories in exosomes of hepatocarcinoma cells. The differential expression profile may reflect an anti-cancer immune response and represent molecular targets for novel anti-hepatoma therapeutic or preventative strategies.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To identify the relationship between angiotensin-converting enzyme (ACE) gene polymorphism and heart rate variability in cerebral stroke patients.</p><p><b>METHODS</b>An insertion/deletion(I/D) polymorphism of the ACE gene was analyzed by polymerase chain reaction in 43 normal subjects, 46 patients with ischemic cerebral stroke, and 40 patients with brain hemorrhage; their heart rate variability(HRV) parameters such as time domain and power spectral component were analyzed.</p><p><b>RESULTS</b>The frequency of DD genotype and the frequency of deletion alleles in cerebral stroke groups were significantly higher than those in control groups (P<0.01), and the measured components of HRV, including total power (TP), low frequency power (LF), high frequency power (HF), LF/HF, and choas, were higher in the patients with the ACE DD genotype when compared with those in the patients with the ACE II, ID genotypes; there was significant difference in effective rate (P<0.05).</p><p><b>CONCLUSION</b>The above related parameters of HRV were correlated with heritability, suggesting that the cerebral stroke patients with the ACE DD genotype are at high risk of cerebrogenic cardiac autonomic nerve function disturbances.</p>

ABSTRACT

Objective To investigate quantitatively the thermal sensation characteristics of the patients with facial palsy and the value of quantitative thermal test (QTT) in prognostication.Methods The QTT threshold of the fore ear and cheek of 30 patients with peripheral facial palsy was tested,their facial nerve conduction velocity was measured,and House-Brackmann facial nerve grading system was used to estimate facial nerve function at 2~3 weeks,a month,two months and half a year post onset.Results It was found that 12 out of 30 patients had abnor- mal QTT threshold value;the majority of them suffered from herpes virus and diabetes.In those with abnormal QTT, 8 were with diabetes mellitus (account for 66.7%),3 with partial shingles (account for 25%),and 1 with positive serum virus infection (account for 8.3%).Those with normal QTT were significantly different from those with abnor- mal QTT,with regard to the House-Brackmann rating scores after 2 and 6 months post onset (P
