ABSTRACT

OBJECTIVE@#To characterize the traits of neuropsychological functioning deficits in patients with attention-deficit/ hyperactivity disorder (ADHD) with comorbid disruptive, impulse-control, and conduct disorders (DICCD).@*METHODS@#Twenty out-patients with ADHD, 20 with ADHD with comorbid DICCD, and 20 with DICCD, all aged 6-16 years, were enrolled in this study, with 20 healthy subjects matched for age, gender and IQ serving as the healthy controls. The patients were diagnosed according to the Diagnostic and Statistical Manual of Mental Disorders, Fifth Revision (DSM-5). All the subjects were assessed with Golden Stroop test and emotional Stroop test to evaluate their response inhibition and emotional responding.@*RESULTS@#In Golden Stroop test, the interference scores (IGs) of errors and reaction time both differed significantly among the groups ( < 0.05), and were the highest in patients with ADHD only. In emotional Stroop test, the mean reaction time (MRT) showed significant differences among the groups ( < 0.05); the MRT of positive- congruent trials in ADHD with comorbid DICCD group was shorter than that in ADHD group but longer than that in group DICCD; the MRT in the 3 case groups were all longer than that in the control group. The MRT of both positive-incongruent trials and negative-congruent trials in ADHD with comorbid DICCD group and DICCD group was shorter than that in ADHD group but longer than that in the control group. The MRT of negative- incongruent trials in DICCD group was shorter than that in ADHD group and ADHD with comorbid DICCD group but longer than that in the control group.@*CONCLUSIONS@#The response inhibition deficit and abnormal emotional responding are the core symptoms of ADHD. Bias emotional stimuli may render response inhibitory dysfunction in patients with DICCD with callous-unemotional traits of emotional responding disorder, especially in dealing with negative emotional trials, while the comorbidity of ADHD and DICCD tends to have the emotional response trait of DICCD.

ABSTRACT

The neurocircuitries that constitute the cortico-striato-thalamo-cortical (CSTC) circuit provide a framework for bridging gaps between neuroscience and executive function in attention deficit hyperactivity disorder (ADHD), but it has been difficult to identify the mechanisms for regulating emotional problems from the understanding of ADHD comorbidity with disruptive behavior disorders (DBD). Research based on "cool" and "hot" executive functional theory and the dual pathway models, which are thought of as applied response inhibition and delay aversion, respectively, within the neuropsychological view of ADHD, has shed light on emotional responding before and after decontextualized stimuli, while CSTC circuit-related domains have been suggested to explain the different emotional symptoms of ADHD with or without comorbid DBD. This review discusses the role of abnormal connections in each CSTC circuit, especially in the emotion circuit, which may be responsible for targeted executive dysfunction at the neuroscience level. Thus, the two major domains - abstract thinking (cool) and emotional trait (hot) - trigger the mechanism of onset of ADHD.

ABSTRACT

Objective To explore the predictive value of brain lobe location of stroke lesion to development of long-term post-stroke depression(PSD)in the first-episode ischemic stroke patients for providing evidence for early intervention.Methods In the prospective study,158 patients aged 60 and over with first-episode ischemic stroke were continuously admitted into Department of Neurology of Shangqiu First People's Hospital from January 2013 to July 2013.The 2 to 3 years follow-up after stroke episode was performed in 126 cases for inquiring into correlation between brain lobe location of stroke lesion and development of PSD.The diagnosis of depression was in accordance with Mental Disorders Diagnostic and Statistical Manual 4(DSM-Ⅳ)standard,and divided into groups of stroke with depression(n=52)and stroke without depression(n=74).The degree of depression was evaluated by 17-item Hamilton Rating Scale for Depression(HAMD17).The location,number and volume of stroke lesions were determined by head MRI.The relationship between PSD and pathogenetic loci was analyzed by unconditional Logistic regression analysis.A difference in hemisphere commensalism.Results The morbidity of PSD was 41.3%(52/126),with 21.4%(27/126),12.7%(16/126),7.1%(9/126)in mild,moderate and severe PSD respectively.The frontal lobe(OR=2.824,95%CI=1.189-6.706)and the temporal lobe(OR=3.579,95%CI=1.233-10.393)cerebral infarction were correlated with the occurrence of PSD.The long-term PSD severity was more in frontal lobe than in temporal lobe(χ2=6.399,P<0.05).The average volume of cerebral infarction was larger in PSD group than in non-PSD group(t=3.271,P<0.05),and the average number of cerebral infarction loci was more in PSD group than in non-PSD group(t=3.176,P<0.05).The more severe the degree of depression according to HAMD17,the larger the average volume of cerebral infarction(F=6.280,P<0.05)and the more the average number of lesions(F=6.132,P<0.05).Conclusions The development of long-term PSD in the first-episode elderly patients is affected by the invasion site.The frontal lobe and temporal lobe infarction are independent risk factors for long-term PSD in patients with ischemic stroke,and the PSD was more severe in frontal lobe infarction than in temporal lobe infarction.

Objective@#To detect the alterations of telomerase activity and the expression for oxidative stress responsive genes related with senescence during cellular replicative senescence and hydrogen peroxide-induced premature senescence in human embryonic lung fibroblasts (HELFs) in vitro.@*Methods@#The HELFs were divided into young cells (22 population doubling levels, 22PDL) , mid-aged cells (35PDL) and replicative senes-cent cells (49PDL) and premature senescent cells induced by H2O2(premature senescence, PS). The telomerase activity was detected by ELISA assay during cellular replicative and premature senescence. The mRNA level of oxidative stress responsive genes related with senescence for Foxo1, Foxo3, Pdx1, apoA-I and MMP1 was per-formed by RT-Q-PCR separately.@*Results@#The mRNA level for Foxo1, Foxo3, apoA-I and Pdx1 was decreased separately during cellular replicative senescence compared to that in the young-stage cells with statistical signifi-cance (P<0.05). The expression of MMP1 was up-regulated 5.1-fold obviously (P<0.05). In premature senes-cence, the mRNA level was only decreased for Foxo1, Foxo3 and apoA-I, but up-regulated 2.3-fold and 6.2-fold for Pdx1 and MMP1 respcetively vs 22PDL significantly (P<0.05). The telomerase activity in young cells was not detected, and it increased in mid-aged cells and replicative senescence stages during cellular replicative se-nescence as compared to 22PDL with statistical significance (P<0.05). The telomerase activity in premature se-nescence was highly active.@*Conclusion@#The expression for genes related with senescence has differences be-tween replicative and premature senescence and hydrogen peroxide modifies their expression levels. The telomer-ase activity has been going up with increased PDLs.

ABSTRACT

Objective · To explore family medical intervention model of senile dementia with behavioral and psychological symptoms. Methods · Four streets of Changning District in Shanghai were randomly selected and subjects were enrolled according to the inclusion criteria, who were randomly divided into the intervention group (n=71) and control group (n=70). The intervention group received door-to-door service from psychiatric doctors, given drug treatment and psychological intervention. Subjects were evaluated by several scales, including Behavioral Pathology in Alzheimer's Disease Rating Scale (BEHAVE-AD), Mini-Mental State of Examination (MMSE), Activity of Daily Living Scale (ADL), Quality of Life-Alzheimer's Disease (QOL-AD), and Generic Quality of Life Inventory-74 (GQOLI-74), at baseline and by the end of 6 months and 12 months. Results · ① There was no significant difference in the total scores and all factor scores of BEHAVE-AD between the two groups before intervention (P>0.05). Repeated measures analysis of variance revealed a significant main effect of time (P0.05). Repeated measures analysis of variance revealed a significant main effect of time (P<0.001). The between-group effect was significant in the scores of MMSE and QOL-AD (P<0.001). The interactive effect of time×group was significant in the scores of MMSE, ADL, QOL-AD, and GQOLI-74 (P<0.05). ④ Inter-group comparison of MMSE, ADL, QOL-AD, and GQOLI-74 scores indicated that by the end of 6 months, scores of MMSE of the intervention group were remarkably better than those of the control group and the differences were statistically significant (P<0.05). By the end of 12 months, scores of MMSE, ADL, QOL-AD, and GQOLI-74 of the intervention group were remarkably better than those of the control group and the differences were statistically significant (P<0.05). Conclusion · The family medical intervention model of door-to-door services from psychiatrists integrating multidisciplinary team is effective to attenuate the mental and behavioral symptoms of senile dementia patients, and can improve the quality of life of patients and caregivers. The effect of persistent implementation will be more remarkable.

ABSTRACT

OBJECTIVE: To construct a cell model with simultaneous knockout of RNA methyltransferase-like( METTL) 3 and METTL14 using clustered regularly interspaced palindromic repeats( CRISPR)/CRISPR-associated protein 9( Cas9)system in human bladder cancer T24 cells,in order to provide new tools and means for further studying gene intervention in occupational cancers. METHODS: The small guide RNA( sgRNA) oligonucleotide sequences targeting METTL3 and METTL14 were designed,synthesized and constructed into the Lenti-multi-CRISPR vector. The Lenti-iCas9-neo inducible plasmids were transfected into T24 cells by lentivirus system,and the positive cells were sorted by flow cytometry. Finally,the recombinant plasmid Lenti-multi-METTL3-METTL14 was transfected into positively sorted cells and screened to obtain stable cell lines. T24-Lenti-multi-CRISPR cells and T24-KO-METTL3-METTL14 were used as control group and multigene knockout group. The Western blot was used to examine relative expression of METTL3 and METTL14 protein in the two groups. RESULTS: The targeting sgRNA was successfully inserted into the Lenti-multi-CRISPR plasmid and confirmed by sequencing. The flow cytometry was used to obtain a large amount of the positive cells. The stable cell line inducted of doxycycline with simultaneous knockout of METTL3 and METTL14 gene was obtained. The relative expression of METTL3 and METTL14 protein decreased respectively by 52. 08% and 64. 04% after 96 hours of doxycycline treatment in the multigene knockout group compared with the control group( P < 0. 01). CONCLUSION: The stable T24 bladder cancer cell with simultaneous double gene knockout of METTL3 and METTL14 was obtained for the first time,which will be helpful for providing a foundation for mechanistic study for prevention and treatment of candidate tumor targets and bladder cancer.

ABSTRACT

Anorexia Nervosa(AN),one kind of eating disorder charactered by abnormal eating patterns,low body weight,and weight and shape concerns,is an intractable psychiatric illness which has the highest mortality rates.The etiology of AN is complex,and the neurobiology mechanisms of AN remain unclear.Much of the research provided indications that the pathogenesis of AN is associated with the oxytocin (OT) functions.Summarizing the recent advances on OT in AN,a systematic review of OT research was conducted that it pertains to regulation of feeding,social activity,emotion and bone metabolism in AN.OT probably play an important role in the development and progression of AN.OT system disorder may be an state indicator rather than a trait indicator of AN.Because of the variety physiological effects and the complex effects on AN,OT has the potential applications,especially for the treatment of AN.

ABSTRACT

Objective:To clarify the characteristics and differences between obsessive-compulsive disorder (OCD)and attention-deficit/hyperactivity disorder (ADHD)in male children.Methods:Forty boys with OCD and 40 boys with ADHD were selected from out-patient department,and 40 normal boys matched with age,gender and IQ were recruited as the controls.Patients'diagnosis was made according to the International Statistical Classifica-tion of Diseases and Related Health Problems,Tenth Revision (ICD-10).The choice delay tasks (CDT1 /2)were assessed respectively for the measurements of normal or high ecological validity of delay aversion.In CDT1 /2, waiting time and scores of the tasks were indicators reflecting the delay aversion.Results:The scores of CDTs were higher in children with OCD than in the controls [CDT1,(30.9 ±3.2)vs.(27.6 ±3.8);CDT2,(31.3 ±4.4)vs. (28.5 ±4.9);P <0.01].The scores of CDTs were lower in children with ADHD than in the controls [CDT1, [(25.0 ±4.3)vs.(27.6 ±3.8)];CDT2,[(26.2 ±4.6)vs.(28.5 ±4.9);P <0.001].Conclusion:The results in-dicate that the children with OCD have a trend to choose the task with longer waiting time and higher score,and the children with ADHD trend to choose the task with shorter waiting time and lower score.The results suggest that the pathogenesis of hyperactivity/impulse control may be different between OCD and ADHD in delay aversion.

ABSTRACT

Objective To investigate the head and neck CTA findings and cerebral CTPI in patients with transient ischemic attack (TIA),and their relationship with frequency of TIA,duration and CTA detected responsible vessels.Methods 180 TIA patients were selected as the research subjects,the GE Lightspeed 64 spiral CT perfusion CT scan and CTA examination were taken,and the differences in CTPI parameters measurement,head and neck artery disease of patients were analyzed.To observe the correlation between CTPI perfusion abnormalities in patients with TIA seizure frequency,duration and NIHSS score.The CTPI perfusion abnormalities and consistency of CTA display of responsible vessels were analyzed.Results The positive rate of CTPI examination was 75.56%,that of CTA examination was 81.11%.The positive rate of patients(NIHSS ≥4 points) with CTPI was 88.89%,which was higher than 66.67 % in patients (NIHSS < 4 points) with CTPI (x2 =6.87,P < 0.05).The positive rate of patients (seizures time ≥ 10min) with CTPI TIA was 71.43%,which was higher than 45.45 % of the patients (seizures time < 10m in) with CTPI (x2 =7.23,P < 0.05).The CTPI positive rate of patients with the TIA seizure frequency ≥ 2 times/d was 83.33%,which was higher than 61.67% of patients with the seizure frequency < 2 times/d(x2 =5.48,P < 0.05).The CTPI positive and CTA responsibility vascular consistent proportion was 73.33 %,which was significantly higher than 26.67% of both inconsistent and negative (x2 =19.43,P < 0.01).Conclusion The cerebral CTPI and head and neck CTA can response to TIA in patients with intracranial lesions accurately,consistency and clinical manifestations are very good,which have higher value in clinical application.

ABSTRACT

Objective:To explore the differences and similarities of the neuropsychological functioning defi-cits in children between Tourette syndrome (TS)and attention-deficit/hyperactivity disorder (ADHD). Methods:Thirty boys with TS-only,36 with TS-plus-ADHD,36 with ADHD were selected from out-patient department,and 50 normal boys (NC)matched with gender,age and IQ were recruited as the controls. Patients'diagnosis was made according to the International Statistical Classification of Diseases and Related Health Problems,Tenth Revision (ICD-10). They were assessed with the Stroop Color-Word Interference Test (Stroop)and Purdue Pegboard Test (Purdue)to evaluate the response inhibition and fine-motor coordination respectively. Results:The Stroop scores were higher in children with ADHD than in other groups (P0. 05 ). All Purdue scores were higher in children with disease than in normal children (P<0. 05 ). Conclusion:The results indicate that the response inhibition deficit may be found in children with ADHD,but not in those with TS-only and TS-plus-ADHD. The neural compensatory mechanism may be re-sponsible for the response inhibition function in children with TS whilst the fine-motor coordination deficit was as-sociated with the disease groups.

Objective To investigate the clinical efficacy of statins atorvastatin calcium on cerebral carotid atherosclerotic plaques in patients and the effects on C-reactive protein .Methods 76 patients admitted to hospital with cerebral carotid atherosclerotic plaques in patients randomized manner using its divided into the observation group and control group of 38 patients in the control group not using statins , given only hospital conventional therapy , observed in the control group were given atorvastatin group based on calcium statin treatment ,post-treatment comparison groups the size of carotid atherosclerotic plaque and plasma C-reactive protein changes .Results After treatment , plaque size(0.069 ±0.021)cm2,IMT was (1.29 ±0.28)mm,significantly less than before treatment,after treatment difference was statistically significant (t=7.538,7.132,all P<0.05)also significantly less than the control group ,the difference was statistically significant (t=7.131,6.372,all P<0.05);the observation group after treatment C-reactive protein(3.54 ±1.92 ) mg/L, significantly lower than before treatment , before and after treatment difference was statistically significant(t=6.831,P<0.05),and significantly lower than the control group ,the difference was also statistically significant(t=7.125,P<0.05).Conclusion For patients with cerebral carotid atherosclerotic plaque is concerned,the application of atorvastatin calcium treatment can not only effectively reduce plaque area , but also reduce C-reactive protein,and therefore worthy of further clinical application .

ObjectiveTo investigate the association between catechol-O-methyl transferase (COMT)gene polymorphism and Gilles de la Tourette' s syndrome(GTS).MethodsUsing Amplification Refractory Mutation System(ARMS) PCR genotyping assay method,a polymorphism (val158met) of COMT gene was genotyped in 112 of all GTS patients ( total GTS group) including 54 GTS-alone patients group,48 GTS + ADHD patients group among of them and 71 healthy controls.The correlation between positive association of polymorphism (val158met)of COMT gene in GTS and the age of onset in patients with GTS was also analyzed.ResultsCompared with healthy controls group,genotype of val158met did not differ in total GTS patients group or alone-GTS patients group (χ2 =0.56,P=0.756;χ2 =1.05,P=0.600 respectively).There was also no significant difference (P>0.05)in allele distribution of val158met in total GTS patients group or alone-GTS patients group compared with controls group respectively (χ2 =0.18,P=0.669;χ2 =0.29,P=0.593 respectively).However,genotype distribution of val158met was significantly different between GTS + ADHD patients group and controls group( χ2 =6.35,P =0.041 ).The frequency of the val allele of this locus was significantly higher in GTS + ADHD patients group than those in controls group ( χ2 =5.49,P =0.019 ).The mean age of onset (6.80 ± 1.54 ) in 36 children within GTS + ADHD patients group with the val/val geantype of COMT gene val158met polymorphism was significantly earlier than the mean age of onset (8.04 ± 1.54)in 12 children in val/val genotype (P =0.016 ).ConclusionPolymorphism (val158met) of COMT gene may be associated with GTS children with comorbid ADHD,which may play an important role to make the age of onset in children with GTS become earlier.

ABSTRACT

Objective To study the effects of mosapride and domperidone on the pulmonary infection of acute stroke patients lying in bed and with nasal feeding. Methods Eighty-nine acute stroke patients lying in bed and with nasal feeding were divided randomly into the treatment group (47 cases) and the control group (42 cases). The control group was treated routinely,and the treatment group was treated with mosapride 5 mg and domporidone 20 mg thrice a day for 4 weeks, besides routine therapy. The incidence rate of pulmonary infection, gastric residual volume (GRV) and the number of cases with gastric contents remaining after 3 hours of nasal feeding were studied. All data were analyzed statistically. Results In the treatment group, 13 cases had pulmonary infection,and the incidence rate was 27.66%(13/47). In the control group,25 cases had pulmonary infection,and the incidence rate was 59.52% (25/42). There was significant difference between the two groups (P < 0.01 ). Three hours after nasal feeding,24 cases with gastric contents remaining were discovered in the treatment group,and GRV was (50.80±15.38) ml. Two hundred and thirty-seven cases with gastric contents remaining were discovered in the control group, and GRV was (112.17±32.54) ml. Significance differences were also detected between the two groups (P<0.01). Conclusion As for the acute stroke patients lying in bed and with nasal feeding,mosapride and domperidone can remarkably cut down the pulmonary infection upon common treatment.

ABSTRACT

Objective To investigate the effects of electro-acupuncture on muscular tension and motor function in the flaccid,paralytic limbs of stroke survivors.Methods Seventy-two stroke survivors with flaccid,paralytic limbs were divided randomly into a control group and an electro-acupuncture group.Both grourps received routine therapy and rehabilitation training,but the electro-acupuncture group was also treated with electro-acupuncture.Muscular tension and motor function in the flaccid,paralytic limbs in both groups were evaluated by modified Ashworth scale (MAS)and Fugl-Meyer scores before treatment and at the end of 4 weeks.Results After treatment,in 24 patients of the electro-acupuncture group and 10 patients of the control group muscular tension had increased 1-2 grades.The effectiveness rate(68.57%)in the electro-acupuncture group wag higher significantly than that(27.03%)in the control group(P<0.01).Fugl-Meyer scores in the electro-acupuncture group were significantlv higher than those in the control group(P<0.01).Conclusion Electro-acupuncture supplementing traditional rehabilitation improves muscle tension in flaccid,paralytic limbs and promotes the recovery of motor flunction after stroke.

ABSTRACT

Objective To understand the epigenetic regulations of P16 during cellular replicative senescence and premature senescence induced by hydrogen peroxide of human embryonic lung fibroblasts(HEFs).Methods The normal HEFs were divided into young cells(22 PDL),mid-aged cells(35 PDL)and replicative senescent cells(49 PDL)during replicative senescence.The synchronously cultured 22 PDL HEFs were exposed for 2 h to 400?mol/L H_2O_2 at half confluence on daily basis.The procedure lasted for 4 consecutive days.And then the treated cells were cultured for another 7 days,called premature senescent cells.The mRNA level of P16 was detected by fluorescence quantitative PCR.The methylation level in the promoter region -846~-639 bp was observed by methylation-specific PCR(MSP).The histone modifications was detected by chromatin immunoprecipitation-QPCR assay,including acetylation for H3,H4 and methylation for H3(Lys4)and H4(Lys20).Results In the process of cellular senescence,the mRNA level of P16 decreased in mid-aged cells,but increased significantly in both replicative and premature senescent cells compared with that of young cells(P

ABSTRACT

<p><b>BACKGROUND</b>Recent researches have found that NiS can cause the malignant transforming activity and carcinogenicity on human bronchial epithelial cells (16HBE). Its molecular mechanism may be involved in mutation of genes and abnormal expression of transcription factors on 16HBE. And so, this study takes advantage of a model of 16HBE transformed by NiS and screens the differentially expressed genes between 16HBE cells and NiS treated 16HBE cells (NiS-16HBE) using cDNA microarray.</p><p><b>METHODS</b>The total RNA was extracted from 16HBE cells and NiS-16HBE cells. The cDNA probes were prepared by labeling with Cy3-dCTP and Cy5-dCTP respectively through reverse transcription. The mixed probes were then hybridized to the cDNA microarray chips containing 4000 human genes. The chips were scanned by ScanArray 4000 laser scanner. The acquired fluorescent signals were analyzed by GenPix Pro 3.0 software. Bioinformation function of those differentially expressed genes was analysed.</p><p><b>RESULTS</b>A total of 151 genes exhibited differential expression between 16HBE cells and NiS-16HBE cells. The expression of 70 genes ( 46.36%) was down-regulated and that of 81 genes (53.64%) was up-regulated.</p><p><b>CONCLUSIONS</b>The regulation of genes including stress response genes, immune related genes, DNA synthesis and repair genes, metabolism genes, pro-oncogenes and tumor suppressor genes may be involved in transforming activity of NiS.</p>

ABSTRACT

Objective: To observe the changes of blood T-cell subpopulations, ASO,IL-6 and IL-8 in children with Tic disorder and explore the pathogenesis. Methods: T-cell subpopulations and concentration of IL-6, IL-8, ASO were measured in 52 children with tic disorder and 30 normal children using SAP and ELISA.Results:The numbers of CD3+、CD4+ and CD8+ cells had no significant difference between 32 children with tic disorder and 30 normal children, and the concentration of ASO in serum of children with tic disorder was markedly higher than those in normal children. The concentrations of IL-6 and IL-8 in serum had no significant difference between the two groups.Conclusion: There may be dysfunction in humoral immunity in tic disorder.

ABSTRACT

Objective:To investigate the relationship between apolipoprotein E (ApoE) genetype and the depressed symptoms in patients with coronary heart disease. Methods:The genotypes and alleles of apolipoprotein E gene in 26 depressed patients with coronary heart disease,30 non-depressed patients with coronary heart disease and 30 healthy control subjects were examined with the polymerase chain reaction (PCR) and restriction fragment length polymorphisms (RFLPs) technique.Meanwhile, the HRSD was completed by all 86 subjects. Results:The frequency of Apolipoprotein E epsilon 4 in depressed patients with coronary heart disease was more than that in normal subjects .Especially the total cholesterol, HRSD and HAD in patients with the apolipoprotein E epsilon 4 were significantly higher than those in patients without the apolipoprotein E epsilon 4. Conclusion:Our results support the polymorphisms of apolipoprotein E gene were associated with the depressed symptoms in coronary heart disease. Apolipoprotein E epsilon 4 may play a major role in the pathogenesis of depressed symptoms in patients with coronary heart disease.
