ABSTRACT

The NLRP3 inflammasome's core and most specific protein, NLRP3, has a variety of functions in inflammation-driven diseases. Costunolide (COS) is the major active ingredient of the traditional Chinese medicinal herb Saussurea lappa and has anti-inflammatory activity, but the principal mechanism and molecular target of COS remain unclear. Here, we show that COS covalently binds to cysteine 598 in NACHT domain of NLRP3, altering the ATPase activity and assembly of NLRP3 inflammasome. We declare COS's great anti-inflammasome efficacy in macrophages and disease models of gouty arthritis and ulcerative colitis via inhibiting NLRP3 inflammasome activation. We also reveal that the α-methylene-γ-butyrolactone motif in sesquiterpene lactone is the certain active group in inhibiting NLRP3 activation. Taken together, NLRP3 is identified as a direct target of COS for its anti-inflammasome activity. COS, especially the α-methylene-γ-butyrolactone motif in COS structure, might be used to design and produce novel NLRP3 inhibitors as a lead compound.

ABSTRACT

Objective:To explore the reliability and validity of the 10-item Connor-Davidson resilience scale (CD-RISC-10) in Chinese community-dwelling adults.Methods:Using multistage stratified sampling, totally 2 051 community-dwelling adults were selected and they were investigated by the 10-item Connor-Davidson resilience scale, the center for epidemiological studies depression scale (CES-D) and 10-item Kessler psychological distress scale (K10). The exploratory factor analysis (EFA) and confirmatory factor analysis (CFA) were used to evaluate the CD-RISC-10 by SPSS 22.0 and AMOS 22.0.A parallel analysis and Velicer's minimum average partial (MAP) test were also supplemented to confirm the EFA-derived structure of the scale.Results:The Cronbach α coefficient of the CD-RISC-10 was 0.94, and the Spearman-Brown reliability was 0.89.The total correlation of the questions ranged from 0.74 to 0.81 (all P<0.01). The exploratory factor analysis retained its single dimension, which can explain 63.24% of the total variance.Confirmatory factor analysis revealed the single factor model fit data adequately ( χ2/ df=4.596, CFI=0.967, TLI=0.973, RMSEA=0.065). The resilience(27.02±13.50) was negatively correlated with depression(9.75±5.24) and psychological distress (19.23±6.37)( r=-0.32, -0.35, both P<0.01). Conclusion:The CD-RISC-10 scale has acceptable reliability and validity, with using easily and conveniently.Therefore, it is an effective tool for measuring the resilience of the community-dwelling adults.

ABSTRACT

OBJECTIVE@#To study the changes in mRNA and long non-coding RNA (lncRNA) expression profiles in a mouse model of bleomycin-induced lung fibrosis and identify lung fibrosis-related mRNA for coding-noncoding coexpression (CNC) bioinformatics analysis of the differential lncRNAs.@*METHODS@#Lung fibrosis was induced by intratracheal injection of bleomycin in 10 C57BL/6 mice and another 10 mice with intratracheal injection of saline served as the control group. Lung tissues were harvested from the mice at 14 days after the injections and lung fibrosis was assessed using Masson and HE staining. LncRNA chip technology was used to screen the differentially expressed mRNAs and lncRNAs in mice with lung fibrosis, and GO and KEGG pathway analyses of the differential mRNAs were performed using NCBI database and UCSC database to identify possible fibrosis-related mRNAs, which were validated by qRT-PCR to construct a coding and non-coding co- expression network with the differential lncRNAs.@*RESULTS@#Compared with the control mice, the mice with intratracheal injection of bleomycin showed obvious lung fibrosis. The results of gene chip analysis showed that 127 mRNAs were upregulated and 184 mRNAs were down-regulated in the model group as compared with the control group. GO and pathway analysis suggested that the differentially expressed genes participated mainly in immune response, cell differentiation, and cytoskeletons; the involved signal pathways were associated mainly with cytokine and cytokine receptor interaction and chemokine signal transduction. Bioinformatics analysis identified a significant coexpression network between the fibrosisrelated mRNA and the differentially expressed lncRNA.@*CONCLUSIONS@#In mice with lung fibrosis, the differential expressions of fibrosis-related mRNAs in the lung tissues are closely correlated with the co- expressions of a large number of differential lncRNAs, which points to a new direction for investigation of the pathogenesis of pulmonary fibrosis.

ABSTRACT

Objective To investigate the global research status and trends ofrehabilitation after arthroplasty.Methods The Wed of Science database was used to search the publications on rehabilitation after arthroplasty from 1994 to 2018.The included publish items were statistically analyzed by bibliometrics.VOSviewer software was used to analyze the visual transformation of literature coupling (including author coupling,mechanism coupling and country coupling) and co-occurrence analysis.The research status and trends of rehabilitation after arthrop]asty in recent years were analyzed and predicted.Results A total of 1 702 studies were included in the present study.The number of literatures increased year by year globally,including 612 in the United States as the top number of studies in the world.The total citation frequency (15 433 times) and H index (61) of the research publications were also the highest in the world.China (79 literatures) ranked 6th in the number of global research publications,with total citation frequency (451 times) and H index (12) ranked 14th.The number of publications published by JOURNAL OF ARTHROPLASTY and ARCHIVES OF PHYSICAL MEDICINE AND REHABILITATION on rehabilitation after arthroplasty was the highest.The University of Pittsburgh and the University of Toronto were the biggest contributors to publications on rehabilitation after arthroplasty.The theme of rehabilitation after arthroplasty can be divided into five categories:pain management,functional exercise,hospital management,complications and clinical trials.Hospital management wasthe main research field recently and the orthopedic specialty hospital would become a hot research topic in the fulure.Conclusion According to the current global trends,rehabilitation study is deepening and the number of publications will increase continuously.The United States is the largest contributor in this area.The current researches focus on the "hospital management" after arthroplasty.The new type of orthopedic specialty hospital may be the next research hotspot for arthroplasty.

ABSTRACT

Objective We aimed to explor the correlation between the serum resistin levels and the collapse process of femoral head necrosis. Methods Eighty-eight patients with osteonecrosis of the femoral head were included in this study (26, 34 and 28 cases at ARCO stage Ⅱ, Ⅲ and Ⅳ, respectively). Fifty healthy controls were enrolled. The serum resistin levels were detected with ELISA method. We compared the serum resistin levels between the patient group and control group. The differences of serum resistin levels between different ARCO stagesand various disease causes were analyzed in the patient group. Results The resistin levels were significantly higher in patients with osteonecrosis of the femoral than healthy control group (P = 0.026). Compared with control group, the resistin levels significantly increased in patients at ARCO stage Ⅲ and ARCO stage Ⅳ respectively (P = 0.001).The resistin levels of procollapse group (ARCO stage Ⅲ and Ⅳ) were significantly higher than that of precollapse group (ARCO stage Ⅱ) (P = 0.000). There was no statistic difference between ARCO stage Ⅲ andⅣ in resistin levels (P> 0.05). No statistical significance was found between different disease causes. ROC curve analysis of resisrin level indicated theertain accuracy (AUC = 0.749) , sensitivity and significant specificity (77.4%, 61.5%, respectively) in the diagnosis of femoral head necrosis. Conclusions Resistin is closely related to the collapse process of femoral head necrosis. The level of resistin was significantly increased after the collapse of femoral head, which could be useful for the clinical diagnosis of the collapse of femoral head necrosis.

ABSTRACT

OBJECTIVE: To determine and compare the contents of catalpol and aucubin in different parts (root, stem, leaf and flower) of wild Centranthera grandiflora, and to provide reference for the selection of medicinal parts and source development. METHODS: HPLC method was used to determine the contents of catalpol and aucubin in root, stem, leaf and flower of wild C. grandiflora, and the contents of different parts were analyzed comparatively. The determination of catalpol was performed on Agilent TC-C18 column with mobile phase consisted of methanol-0.1% phosphoric acid (1 ∶ 99, V/V) at the flow rate of 1 mL/min; the detection wavelength was set at 210 nm, and sample size was 20 μL. The column temperature was 35 ℃; the determination of aucubin was performed on SPHERI-5RP-C18 column with mobile phase consisted of acetonitrile-water (3 ∶ 97, V/V) at the flow rate of 1 mL/min; the detection wavelength was set at 205 nm, and sample size was 20 μL; the column temperature was 25 ℃. RESULTS: The linear range of catalpol and aucubin were 0.061 5-3.321 and 0.000 36-0.216 mg/mL (all r=0.999 9). The limits of detection were 0.016 and 0.007 μg/mL. The limits of quantitation were 0.052 and 0.023 μg/mL. RSDs of precision, stability (24 h) and reproducibility tests were all lower than 2.00% (n=6). The average recoveries were 99.34% and 99.61%, and RSDs were 1.06% and 1.12%, respectively (n=6). The average content of catalpol in root, stem, leaf and flower wild C. grandiflora were 1.609, 3.030, 11.095 and 1.921 mg/g, respectively. The contents of aucubin in different parts were 0.441, 0.020, 0.005 and 0.006 mg/g,respectively. CONCLUSIONS:The established HPLC method meets the requirements of quantitative analysis. Catalpol is mainly distributed in the leaves of wild C. grandiflora, and aucubin is mainly distributed in the roots of wild C. grandiflora. The experimental conclusion provides a reference for the reasonable selection of different medicinal parts as raw materials to develop medicine with different efficacy.

Teaching design is an important part of the construction of teaching contents,and it plays an important role in improving the quality of education and teaching. At present, teaching design has not received enough attention in the teaching of the physical chemistry for the Chinese pharmacy major. This study discussed the importance of the teaching design in the classroom of physical chemistry for the Chinese pharmacy major and expounded some practices of teaching design in detail, including the design of the affective attitude goals for the purpose of improving students' sense of curriculum identity and stimulating students' interest in learning, the design of "points-lines-planes" for teaching and learning based on the mind map,and the design of the ways of the classroom lead-in such as case-based learning mode.

ABSTRACT

Objective:To investigate the cause, treatment, and prognosis of delayed hemorrhage in patients who underwent radical gastrectomy. Methods:The clinical data of 294 patients who underwent radical gastrectomy in the Second Hospital Affiliated from Nanchang University from January 2015 to October 2015 were retrospectively analyzed. Results:A total of 15 patients suffered from delayed hemorrhage and accounted for 5.1%of the gastric cancer cases in our hospital for the same period of radical gastrectomy. Of the 15 patients, 9 underwent laparoscopic radical gastrectomy and 6 received open radical gastrectomy resection. Large vascular hemorrhage was found in 7 cases. Anastomosis and anastomotic ulcer induced hemorrhage were observed in 3 cases. Duodenal stump rupture induced hemorrhage was detected in 2 cases. Hemorrhage was also observed in some parts in 2 cases. Likewise, hemorrhage occurred in 1 case, but the affected parts were unknown. Of the 11 patients who underwent a second operation, 2 were subjected to digital subtraction angiography (DSA) and transcathete arterial embolization (TAE) to stop hemorrhage. Endoscopic hemostasis was performed to stop hemorrhage in 1 case. Conservative treatment was administered to stop hemorrhage in 1 case. The secondary surgery rate was 73.3%(11/15) with mortality and curative rates of 40%(6/15) and 60%(9/15), respectively. Conclusion:For delayed hemorrhage after D2 of gastric cancer, a second radical surgery and death rates were high. Therefore, patients suffering from hemorrhage should be subjected to comprehensive clinical treatment and positive measures. Major vascular bleeding, anastomotic leakage, anastomotic ulcer, and duodenal stump rupture are relevant risk factors. Anastomotic fistula and celiac artery bleeding complications caused hemorrhage is the leading cause of death. Extensive bleeding and unstable vital signs should be checked. A second operation and abdominal drainage should also be timely conducted to as effective methods. Realistic and conservative treatment can be administered to patients with stable vital signs and low amount of blood loss. Endoscopic hemostasis can be applied to alleviate simple anastomotic ulcer bleeding. DSA can be initially performed to detect unknown bleeding sites. TAE can be subsequently used to treat hemorrhage.

Japanese gastric cancer treatment guidelines list options for treatment of each stage of early gastric cancer (EGC).The treatment of EGC is closely related to the lymph node metastasis, and the accurate prediction of lymph node metastasis is related to the choice of the treatment options and the prognosis of the patients.The lymph node metastasis of EGC is evaluated according to the clinicopathological factors, imaging,molecular markers and sentinel lymph node (SLN) tracer biopsy.A two-step method for the management of EGC treated with endoscopic mucosal resection/endoscopic submucosal dissection (EMR/ESD) is recommended.While for those EGC patients not suitable to receive EMR/ESD, imaging or molecular marker and SLN tracer biopsy technology are applied to determine lymph node status.

ABSTRACT

Objective To explore the effect of age on the fracture healing through bioinformatical analysis of gene ex?pression data in GEO, and to screen critical molecular targets and pathways involved in this process. Methods Through R programming language, we identified different expressed genes between 26/52 week old rats and 6 week old rats in every time points of the experiment (No fracture;3 days, 1 week, 2 weeks, 4 weeks and 6 weeks after fracture). By comparison of these different expressed genes, those genes that may contribute to fracture healing were identified. Function annotation was conducted based on DAVID database and PPI network that was constructed via STRING database. Results Compared with 6 week old rat, 52 week old rat show more different genes at 2, 4 and 6 weeks after fracture as well as more than intact rats. At the time point of 6 weeks after fracture, 26 week old rat present 4 different genes while 52 week old rat present 99 differ?ent genes compared with 6 week old rat. We totally found 99 genes that might play important roles in the process of fracture healing. These genes involved in biological process related to bone healing, immune, inflammatory and etc. Also, two screened gene enriched KEGG pathways were identified: ECM-receptor interaction and Arachidonic acid metabolism. Through the analysis of PPI network, Pcna, Fn1, Casp3 and etc, who present high density connectivity in PPI network, were screened out. Conclusion Pcna, Casp3 and Fn1 and etc might play important roles in fracture healing through affecting ECM-receptor interaction and Arachidonic acid metabolism.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the effect of body mass index (BMI) on postoperative short-term outcomes of laparoscopy radical gastrectomy by meta-analysis.</p><p><b>METHODS</b>A literature search was performed in PubMed, EMBASE, Cochrane Library databases, CNKI, and CBM, Wanfang database to screen clinical trials published before October 2014 that compared short-term outcomes between high BMI and low BMI patients undergoing laparoscopy radical gastrectomy. RevMan 5.2 was used to perform the meta-analysis. Begg's and Egger's tests were carried out with Stata 12.0 software to evaluate the publication bias of enrolled literatures.</p><p><b>RESULTS</b>Twelve studies involved a total of 4798 gastric cancer patients after laparoscopy radical gastrectomy. There were 1215 patients in high BMI group(BMI≥25 kg/m2) and 3583 patients in the low BMI group (BMI<25 kg/m2). Compared with the low BMI group, the high BMI group were associated with longer operation time (SMD=0.64, 95%CI:0.35-0.93, P=0.000), more intraoperative blood loss(SMD=0.63, 95%CI:0.24-1.03, P=0.002), less retrieved lymph nodes(SMD=-0.44, 95%CI:-0.72--0.17, P=0.002), and more postoperative complications(OR=1.44, 95%CI:1.19-1.74, P=0.000). There were no significant differences in postoperative hospital stay, the time to first flatus and initial complication(P>0.05).</p><p><b>CONCLUSION</b>The higher BMI may result in a longer operation time, more intraoperative blood loss and a higher rate of postoperative complication after laparoscopy radical gastrectomy.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To compare two methods of bleomycin administration for inducing lung fibrosis in mice.</p><p><b>METHODS</b>Bleomycin (5 mg/kg) was intratracheally instilled or by intratracheal administration of its aerosol (100 µl) in mice. Evans Blue instead of bleomycin was intratracheally administered in the two manners to observe the liquid distribution in the lungs. On Days 14 after bleomycin treatment, the mice were sacrificed and the lungs were removed en bloc to determine the lung coefficient and hydroxyproline concentration, and the histological changes and lung fibrosis were evaluated by HE and Masson's trichrome staining, respectively.</p><p><b>RESULTS</b>Evans Blue intratracheal aerosol distribution in the lung lobes. Intratracheal administration of bleomycin and its aerosol produced no obvious changes in the lung coefficient (P=0.434), and resulted in similar total histopathological scores and fibrosis level in mice (P=0.244 and 0.686, respectively). Bleomycin instillation caused obvious lung injury and lung fibrotic foci with significantly different histological and fibrotic scores between the pulmonary lobes (P=0.016 and 0.038, respectively), while bleomycin aerosol inhalation resulted in similar histological and fibrotic scores in the pulmonary lobes (P=0.466 and 0.29, respectively). A higher level of hydroxyproline was observed in mice with bleomycin aerosol administration (P=0.020).</p><p><b>CONCLUSION</b>Intratracheal bleomycin instillation and bleomycin aerosol inhalation induce similar levels of fibrosis in the lungs of mice, but the latter causes more extensive and more evenly distributed fibrosis in the lungs.</p>

Methods With ".net" platform, we use Visual Basic 2005 and database knowledge to compile the software that can analyze the HPLC data of WATERS and AGILENT at the same time but do not depend on the original HPLC system. Results Successfully Compiling the software that can help to execute the research on material basis of effectiveness in Chinese medicine decoction and analyze the experimental data. Condusion With this software, we can clearly execute the methodology line and quickly analyze the data.

ABSTRACT

The study was conducted to investigate the effect of different forms of trivalent chromium on growth hormone (GH) secretion and pituitary mRNA expression in finishing pigs.A total of 96 finishing pigs with an initial average body weight 65 kg were blocked by body weight and randomly assigned to four treatments with three replicates.Pigs were offered one of four diets including a control diet or the control diet supplemented with 200 μg/kg chromium from either chromium chloride (CrCl3),chromium picolinate (CrPic) or Chromium nanocomposite (CrNano) for 40 days.During the trial,all pigs were given free access to feed and water.At the end of the feeding trial,blood samples were taken via auriculares at 15 min intervals for 3 hours.Eight pigs from each treatment were slaughtered for backfat thickness and longissimus muscle area (LMA),and three pituitaries were collected to determine GH mRNA level.The results showed that average daily gain of pigs fed supplemental CrNano was increased by 6.31 (P<0.05),feed gain ratio was decreased by 4.61% (P<0.05),backfat thickness was decreased by 24.32 % (P<0.05),and LMA was increased by 20.22% (P<0.05).The results of GH dynamic secretion showed that supplemental CrNano increased the mean level,lowest value,peak value and peak duration of GH by 42.62% (P<0.05),87.94%(P<0.05),26.60% (P<0.05),and 17.19%(P<0.05),respectively.The mean level and peak value of GH in pigs fed Supplemental CrPic was increased by 36.58% (P<0.05),27.18%(P<0.05),respectively.Pituitary mRNA expression of GH in pigs fed diet containing chromium from CrNano was increased by 27.63%(P<0.05).These results indicated that chromium nanocomposite increased GH mRNA expression and secretion in finishing pigs,through which growth was promoted and carcass characteristic was improved.

ABSTRACT

Objective To study the preparation of nifedipine liposomes (NP) and evaluate their quality. Methods The liposomes were prepared by a pH steps degree-freeze thawing method, with the entrapment and stability as indices. The entrapment was studied by Sephadex G-50 minicolumn centrifugation UV-Vis spectroscopy method. Entrapment, and degree distribution before and after freezing were compared, the effect of freezing times on entrapment was studied. Results The entrapment and the average size of NP liposomes were 96.65% and 2.0 ?m, respectively. After freezing, both the entrapment and stability were increased. Conclusion The selected formulation and preparation of nifedipine liposomes are practicable.

ABSTRACT

Objective To prepare Paracetamol (APP) multiloculated implant loaded with poly-lactide-co-glycolide acid (PLGA) and to study the drug release profile in vitro. Methods APP multiloculated implant was fabricated by micro-electro-mechanical system (MEMS), and high-performance liquid chromato graphy (HPLC) measurement was used to investigate in vitro drug release profile. HPLC analysis was carried out by employing C18 column and a mixture of methanol-water (15∶85) as mobile phase. The detection wavelength was 215nm and flow rate was 0.8mL/min. Results With different multiloculated shape, the rate of the drug release in vitro was varied significantly. Moreover, the releasing of APP multiloculated implant with ecto-tetragonum ento-hexagon in vitro conformed to Higuchi equation. Conclusion The technology of the preparations is feasible, and the structural and morphological characteristics of the multiloculated implant have a significant impact on the release speed of the drug delivery system.

ABSTRACT

Object To preliminarily study the difference of proteins in the seed of spine date and the seed of Indian jujube. Methods Protein contents were measured with Kieldahl method. Amino acids constituting the protein were determined by means of acidolysis. The proteins were analyzed using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and capillary zone electrophoresis (CZE). Results Contents of proteins in the seed of spine date and the seed of Indian jujube were 36.13% and 41.58%, respectively. The content of valine and methionine in the seed of Indian jujube were significantly more than that in the seed of spine date remarkably, and the contents of other amino acids from the seed of spine date and seed of Indian jujube were similar. Their SDS-PAGE spectra showed that the seed of spine date contained a protein with molecular weight 39 800 and the seed of Indian jujube contained a protein with molecular weight 50 100 . In CZE spectra for the seed of spine date, there was a peak at the mobile time of 3.1 min and in CZE spectra from the seed of Indian jujube, there was a peak at the mobile time of 4.8 min. Conclusion Protein content in the seed of Indian jujube is more than that in the seed of spine date; The content of valine and methionine in the seed of Indian jujube is more than that in the seed of spine date remarkably. The spectra of SDS-PAGE and CZE for the seed of spine date are different from that for the seed of Indian jujube clearly.

ABSTRACT

AIM: To apply Differential Scanning Calorimetry(DSC) identifying powder aqueous extracts and alcohol extracts of Radix et Rhizoma Salviae miltiorrhizae,Radix et Rhizoma Glycyrrhizae and Radix Paeonia rubra. METHODS: ?-Al_2O_3 was taken as the reference substance and under static state air condition,with the determination of the measuring range、the heating rate and temperature range of each herb or its extracts,the herbs were(identified) by comparing the characteristics and the peak profiles of their chromatography. RESULTS: There were the tiny differences in DSC chromatography among powder,aqueous extracts and alcohol extracts,DSC chromatography of the same herbs from two different areas were very close to each other. CONCLUSION: With great advantages of DSC method,such as very small amount of drugs,solvent-free.It is easy,quick and convenient to apply chromatography analyses.

ABSTRACT

Objective: To establish the distinguish method of Jianpixiaoshi capsule.Methods: The microscopic distinguish and TLC were used. Results: 12 herbs in Jianpixiaoshi capsule can be distinguished by Microsccopic distinguish, Atractylodes macrocephala, Glycyrrhiza uralensis, Panax ginseng, Citrus reticulate, Ammomum villosum, Crataegus pinnatifida and Codonopsis pilosula can be distinguished respectively by TLC.Conclusion: The method is accurate and simple, and can be used for distinguishing Jianpixiaoshi Capsule.
