ABSTRACT

Objective To investigate the expression relationship between microRNA-206 and Bcl-2 and its clinical significance in patients with triple-negative breast cancer (TNBC). Methods Thirty- one patients with TNBC from April 2013 to August 2017 were selected. Tumor tissues and matched normal tissue were collected. The Bcl-2 level in tissue samples was detected by immunohistochemistry, and the microRNA-206 expression was detected using real-time fluorescence quantitative polymerase chain reaction (qRT-PCR). The correlation of Bcl-2 and microRNA-206 with the clinical pathological characteristic of patients was analyzed. The TNBC cell line (MDA-MB-231) was cultured and transfected with microRNA-206 mimics in order to up-regulate the microRNA-206 level. The expression changes of Bcl-2 after microRNA-206 up-regulation were estimated by qRT-PCR and Western blot. The proliferation change of MDA-MB-231 was measured by CCK-8 cell viability kit. Results The total Bcl-2 positive rate in TNBC tissues was significantly higher than that in normal tissues:64.52%(20/31) vs. 35.48%(11/31), and there was statistical difference (χ2=5.226, P=0.022). The relative level of microRNA-206 in tissues from TNBC patients with higher expression of Bcl-2 was significantly lower than that in tissues from TNBC patients with lower Bcl-2 level (0.645 ± 0.062 vs. 1.000 ± 0.181), and there was statistical difference (t=6.363, P=0.003). In TNBC patients, the Bcl-2 level in tumor tissues was related to lymphatic metastasis, TNM stage and tumor size (χ2=4.917, 8.791 and 6.091; P = 0.026, 0.003 and 0.013). The microRNA-206 was associated with lymphatic metastasis and tumor size (χ2 = 6.856 and 4.774, P = 0.008 and 0.028). After up-regulation of microRNA-206, the relative mRNA Bcl- 2 decreased to 0.641 ± 0.031, compared with non- transfection control (1.000 ± 0.164), and the difference was statistically significant (t=7.468, P=0.001). Also the protein of Bcl-2 was reduced. Up-regulated microRNA-206 restrained the proliferation of MDA-MB-231 cells. Conclusions MicroRNA-206 may regulate the level of Bcl-2 in TNBC. This regulation relationship is involved with the development of TNBC.

ABSTRACT

Objective To observe the inhibitory effect of the extracts of Nidus Collocaliae on avian influenza A H5N1 virus in vitro. Methods Nidus Collocaliae water extract, artificial gastric juice digestion products of Nidus Collocaliae water extract, and artifitial intestinal juice digestion products of Nidus Collocaliae water extract were prepared for the experimental study. 293T cells transfection in vitro was carried out. The effects of 3 kinds of Nidus Collocaliae extracts on H5N1 pseudovirus and VSV-G pseudovirus were determined by luciferase detection kit. Blood clotting response to erythrocyte hemagglutinin subtypes H5, H7, H9 antigens and the inhibitory effects of 3 kinds of Nidus Collocaliae extracts were observed. The effects of 3 kinds of Nidus Collocaliae extracts on neuraminidase activity were determined by neuraminidase inhibitor screen kit. Results The 3 kinds of Nidus Collocaliae extracts had inhibitory effects on H5N1 avian influenza pseudovirus, the effects being enhanced with the increase of the concentrations of Nidus Collocaliae extracts. Of the 3 extracts, artificial intestinal digestion products had the strongest inhibitory effect, while Nidus Collocaliae water extract had the weakest effect. However, Nidus Collocaliae extracts had no obvious effect on VSV-G pseudovirus. The concentration of H5, H7 antigen for positive blood clotting response was 1 ∶ 128, and that of H9 antigen was 1 ∶ 256. The 3 kinds of Nidus Collocaliae extracts at certain concentrations could inhibit blood clotting response to H5, H7, H9 antigen, but had no obvious effect on neuraminidase. Conclusion The anti-H5N1 virus effect of Nidus Collocaliae extracts has been achieved probably through resisting hemagglutinin.

ABSTRACT

Objective To study the expression of CDX2 and p21~(WAF1) protein in colorectal cancer and its correlation with the progression of colorectal cancer., and to explore the eorrelativity of CDX2 and p21~(WAF1) genes expression with TCM deficiency Syndrome. Methods Immunohistochemical method was used to detect the expressions of CDX2 and p21~(WAF1) in45 patients of colorectal carcinoma tissues and peri-cancerous tissues, who received the treatment at the People's Hospital of Jiuquan City between April, 2000 and August, 2005. According to TCM syndrome differentiation, the patients were divided into sthenia syndrome group and asthenia syndrome group and sthenia-ashenia syndrome group. Analyze the Correlation between CDX2 and p21~(WAF1) expression and TCM syndrome. Results The positive rate of CDX2 and p21~(WAF1) was 86.7% and 35.6% respectively in colorectal carcinoma tissues. There was no correlation between CDX2 and p21~(WAF1) in colorectal cancer tissues. The positive rate of p21~(WAF1) was different in three groups, which was much lower in the asthenia syndrome group (11.76%) than the sthenia syndrome group (41.67%)and sthenia-asthenia syndrome group (56.25%). Conclusion CDX2 and p21~(WAF1) positive rate was different in cancer tissue and resection margin. The two genes may be associated with colorectal cancer development. CDX2 was not correlated with p21~(WAF1). The lower expression of p21~(WAF1) indicates poor prognosis. The positive rate of p21~(WAF1) was different between deficiency syndrome and excess syndrome, p21~(WAF1) is probably a marker to determine the asthenia and sthenia syndrome. Asthenia syndrome indicates a poor prognosis.

ABSTRACT

BACKGRUND:The high prevalence of hepatitis B virus(HBV) is related with the sexual characteristics in drug abusers.How about the relationship between prevalence of HBV/hepatitis C virus(HCV) co infection and the sexual characteristics in drug abusers? OBJECTIVE:To investigate the relationship between sexual characteristics and HBV/HCV co infection in drug abusers. DESIGN:A cross sectional survey taking the drug abuser as the subjects. SETTING:A department of clinical laboratory of a municipal hospital,an epidemiological department of the college of public health of a university,a professional drug abuser treatment center. PARTICIPANTS:Totally 452 male and 320 female drug abusers were randomly selected from the Changsha Xinkaipu Drug Abusers Treatment Center and Zhuzhou Baimalong Female Drug Abusers Treatment Center as the subjects from 1st to 31st January 2001. INTERVENTIONS:A self designed questionnaire was used to investigate the sexual characteristics of drug abusers.HBV surface antigen and HCV antibody were detected in the serums of the participants with ELISA. MAIN OUTCOME MEASURES:The relationship between HBV/HCV co infection and the sexual behavior, numbers of sexual partners,and the usage of condom in drug abusers. RESULTS:The total prevalence of HBV/HCV co infection in drug abusers was 21.24% ; the prevalence of HBV/HCV co infection in male drug abusers was 29.6% ),which was significantly higher than that in females(9.4% )(χ 2=46.016,P< 0.01).There was no significant difference in the prevalence of HBV/HCV co infection between male and female drug abuser whether they had or had not sexual behaviors(χ 2=3.775,P >0.05).The prevalence of HBV/HCV co infection was in an ascending tendency with the increase of sexual partner(χ 2=9.846,P< 0.05).The using frequency of condom was significantly higher in females than in males(U=14.80,P< 0.01). CONCLUSION:There is no correlation between sexual experience and prevalence of HBV/HCV co infection;male drug abusers have higher prevalence of co infection than females;the number of sexual partners is correlated with the prevalence of HBV/HCV co infection,the more sexual partners,the higher prevalence is.The protective effect of condom is greater in female drug abusers than in males.
