ABSTRACT

Objective To explore the monitoring of cerebrospinal fluid (CSF) dynamics in a model of brain herniation induced by acute intracranial hypertension in Guangxi Bama-Mini pigs by phasecontrast cine magnetic resonance imaging (PC cine MRI).Methods Femoral artery blood were extracted from 10 pigs,and injected into the frontal and temporal parietal lobe to make a model of brain herniation induced by acute intracranial hypertension.The mean arterial blood pressure (MAP),intracranial pressure (ICP),and cerebral perfusion pressure (CPP) were monitored.Routine T1WI,T2WI,coronal,sagittal and cerebrospinal fluid flow sequence (fast PC cine slice) which positioned on the cervical 3 (C3) vertebral body as the center and perpendicular to the spinal scans were performed on all experimental animals before and after blood injection with 3.0T Magnetic Resonance Imaging.The ICP,MAP,CPP,the absolute values of CSF peak flow velocity and the absolute value of carotid peak flow velocity before and after blood injection were compared.Results The ICP,MAP,CPP,and the absolute value of CSF peak flow velocity before injection of autologous arterial blood were statistically significant as compared with those after blood injection [(6.80±2.044) mmHg vs (52.20±1.619) mmHg,(76.80±7.068) mmHg vs (142.80±12.399) mmHg,(70.00±6.074) mmHg vs (90.50±12.250) mmHg,and the absolute value of CSF peak flow velocity was (243.20±77.671) mm/s vs (201.40±55.482) mm/s,respectively,P<0.01].The absolute value of the peak velocity of the carotid artery before blood injection was not statistically significant compared with that after blood injection [(876.80±239.908) mm/s vs (799.40±241.829) mm/s,P>0.05].Conclusion After the formation of brain herniation induced by acute intracranial hypertension,the CSF flow in the C3 level spinal canal showed a low dynamic change,and the CSF flow velocity waveform was disordered and malformed.The non-invasive measurement of CSF dynamics by PC cine MRI can provide an important basis for the change of CSF dynamics in the model of brain herniation induced by acute intracranial hypertension,and provide a theoretical basis for further research on damage control neurosurgery in the future.

ABSTRACT

@#Objective To explore the effect of ultrasound therapy on dysphagia caused by radiotherapy for nasopharyngeal carcinoma. Methods From February to December, 2017, 60 patients with radiation-induced dysphagia for nasopharyngeal carcinoma were randomly divided into control group (n = 30) and treatment group (n = 30). All the patients accepted routine rehabilitation and balloon dilatation. The treatment group received ultrasound therapy, and the control group received placebo ultrasound. They were evaluated with Kubota Drinking Water Test (KDWT) and Videofluoroscopic Swallowing Study (VFSS), and the interincisor distance were measured to determine the clinical effect, before and six weeks after treatment. Results The score of KDWT (Z = -2.253, P < 0.05) and the pass time in VFSS (t > 2.4265, P < 0.05) improved more in the treatment group than in the control group after treatment, as well as the incidence of effect (Z = -4.113, P < 0.001). Conclusion Ultrasound therapy can further improve the function of swallowing based on the balloon dilatation.

Methods From September, 2016 to May, 2017, 60 hemiplegic patients after stroke were randomly divided into control group (n=30) and treatment group (n=30). The control group received routine rehabilitation and functional electrical stimulation (FES), while the treatment group received mirror therapy in addition. They were assessed with Fugl-Meyer Assessment-lower limbs (FMA-L), Berg Balance Scale (BBS) and Holden Rating of Walking Function before and eight weeks after treatment. Results The scores of FMA-L and Berg Balance Scale, as well as the walking ability improved in both groups after treatment (t>4.557, Z>4.666, P<0.001), and improved more in the treatment group than in the control group (t>5.832, Z=-2.086, P<0.05).Conclusion Mirror therapy, based on FES, can further improve the walking in hemiplegic patients after stroke.

ABSTRACT

Objective To compare and analyze the advantages and disadvantages of the forearm skin flap and the lateral femoral skin flap in the repair of oral and maxillofacial defects.Methods Of the 34 patients of oral and maxillofacial tumor resection received postopera-tive reconstruction of maxillofacial defects treatment,18 patients repaired by forearm flap( forearm flap group) ,16 patients repaired by antero-lateral thigh flap ( anterolateral thigh flap group) .The clinical effect and the restoration of two groups after surgery were compared.Results The survival rate of disposable surgical flap in forearm flap group was 94.44%,the survival rate of the anterolateral thigh flap group was 87.50%.The difference between two groups was not statistically significant (P>0.05).The difference of swallowing function in 3 months after surgery between two groups was not statistically significant (P>0.05).The difference of survival quality score between the two groups was not statistically significant (P >0.05) in 6 months after surgery.The scar proliferation rate (27.78%), pigment deposition rate (88.89%), temporary sexual dysfunction rate(66.67%),a permanent disability rate (27.78%) of forearm flap group were significantly higher than those of the femoral anterolateral flap group patients (P<0.05).Conclusion The forearm skin flap and the lateral femoral skin flap have a good clincal effect on the repair of oral and maxillofacial defects,and the incidence of complications of the femoral anterior lateral femoral flap is lower.

ABSTRACT

<p><b>OBJECTIVE</b>To study the radiobiological characteristics of a HepG2 cell line with mitochondrial DNA (mtDNA) deletion.</p><p><b>METHODS</b>HepG2 cells were cultured in a medium containing ethidium bromide, acetylformic acid and uracil. The HepG2 cell line with mtDNA deletion (ρ(0)HepG2 cells) were acquired after 30 subcultures by limited dilution cloning. The cell survival was then observed in the absence of acetylformic acid and uracil, and the total mtDNA deletion in the cells was confirmed by PCR. The radiosensitivity of HepG2 and ρ(0)HepG2 cells was evaluated by exposure to gradient doses of 6 MV X ray irradiation. The cell apoptosis was assessed following a 2 Gy X-ray exposure with Hochest33342 staining, and the invasiveness of ρ(0)HepG2 cells was measured by Transwell assay.</p><p><b>RESULTS</b>HepG2 cells could survive 30 subcultures in the presence of ethidium bromide, and massive cell death occurred after removal of acetylformic acid and uracil from the medium. PCR confirmed total mtDNA deletion from ρ(0)HepG2 cells, whose α/β value was significantly lower than that of HepG2 cells. ρ(0)Hep-G2 cells showed an obviously lowered cell apoptosis rate following X-ray exposure with enhanced cell invasiveness.</p><p><b>CONCLUSION</b>HepG2 cells can be induced by ethidium bromide into ρ(0)HepG2 cells with an increased radiation resistance, anti-apoptosis ability and cell invasiveness.</p>

ABSTRACT

Objective To observe the cardiac function in acute brain injury patients(ABI)and the relationship between ABI and plasma neuropeptideY(NPY),and to inspect the mechanism and find the evidences for preventing cardiac impairment caused by ABI. Methods 89 patients with acute brain injury within 24 hours after the injury were divided into severe group(n =47)and mild group(n = 42)according to Glasgow Coma Scale(GCS),and 35 normal healthy adults were selected as control group.In 24 hours and 72 hours after the brain injury,all patients were examined with echocardiography to observe cardiac structure,Doppler blood flow velocity and cardiac function,and in the same time the plasma NPY were determined by radioimmunoassay.Then the results were compared with controls. Results The parameters of cardiac function such as EF、 SV.AV、CO、CI had statistical change in 24 hours and 72hours after the brain injury between severe ABI group and mild ABI group,and it also had statistical change between severe ABI group and control group(all P <0.05),but no statistical change between mild ABI group and control group(all P <0.05).The level of plasma NPY in ABI patients was significantly higher than that before injury,there was statistically different change between severe ABI group and mild ABI group,and it also had statistical change between severe ABI group and control group(all P<0.05).The parameters of cardiac function was negatively correlated with the rise of plasma NPY by pearson correlation analysis(EF:r =- 0.79,P <0.01; SV:r =- 0.71,P <0.01;AV:r=-0.67,P <0.01 ;E/A:r =-0.63,all P <0.01)and(CO:r =- 0.32,P <0.05;CI:r =-0.35,all P <0.05). Conclusion The parameters of cardiac function were significantly decreased in the patients with acute brain injury,and it was closely related with the level of plasma NPY.

ABSTRACT

Objective To investigate the relationship between the tumor differentiation and the related pathologic factors in Esophago-gastric junction adenocarcinoma. Methods The pathological data from 163 patients of Esophago-gastric junction adenocarcinoma underwent radical surgical resections were randomly divided two groups (group1: well-differentiated,group2: poor-differentiated). The composition of N stages,T stages,morphological type and vessel carcinoma embolus were compared between the two groups ,respectively. The tumor sizes and the number of metastases lymph nodes were compared between the two groups. Results The cases with lower N stages or T stages in group1 were dominant(T1 and T2:55. 74%,N1 and N2:75.41%). Oppositely,those with higher N stages or T stages were dominant(T3 and T4:59.81%,N1 and N2:57.85%)in group 2. The differences were significant Incidence of vessel carcinoma embolus was 44. 26%(27/61)in group 1 and 63. 73%(65/102)in group 2,with a significant difference. There was no significant difference for morphological type in the two groups. The tumor sizes and the number of metastases lymph was 4. 27 ±2. 00 cm and 4. 15 ±5.27 respectively in group 1 ,and 5. 87 ± 3. 26 cm and 8. 80 ± 7.65 respectively in group 2. The differences were significantly different(Ps < 0. 01).Conclusions The tumor differentiation has significant effect on N stages,T stages,vessel carcinoma embolus,tumor size and the number of metastases lymph nodes in Esophago-gastric junction adenocarcinoma.

ABSTRACT

Objective To assess the short-term effects and adverse reactions of combination regimen of gemcitabine, nedaplatin and paclitaxel for patients with metastatic cisplatin-resistant nasopharyngeal carcinoma (NPC). Methods A total of 15 patients with metastatic cisplatin-resistant NPC were enrolled. All patients were treated with a combination regimen including gemcitabine with 1000 mg/m2 on day 1st and 8th, nedaplatin with 70 mg/m2 on day 1st and paclitaxel with 135 mg/m2 on day 1st, repeated every 21 days. Response was evaluated according to Response Evaluation Criteria in Solid Tumors (RECIST) criteria 3.0. Results The overall response rate was 40.0 %, with a complete response rate of 6.7 % (1/15) and a partial response rate of 33.3 % (5/15). Six patients (40.0 %) had stable disease and 3 patients (20.0 %) had progressive disease. The median time to progression (TTP) was 4.7 months and the median overall survival was 6.3 months. Hematological toxicities were the adverse reaction with 40.0 % of leucopenia, 6.7 % of anemia and 20.0 % thrombocytopenia. One patient needed for platelet transfusion. Other adverse reactions were mild. Conclusion The combination regimen of gemcitabine, nedaplatin and paclitaxel is feasible as second-line chemotherapy in patients with metastatic cisplatin-resistant NPC.

ABSTRACT

Objective To discuss the role of DNA-dependent protein kinase catalytic subunit (DNA-DPKCS) in human lung adenocarcinoma cell line (A549) by using small interfering RNA (siRNA) to specifically knockdown DNA-DPKCS expression and its effects on cell proliferation, cell cycle and radio-sensitivity. Methods The DNA-DPKCS-siRNA expression vector was constructed and transfected into A549 cell line. The transformed clones were randomly selected and isolated. The cell cycle distribution and apop-tesis were analyzed by flowcytometry analysis. Cell survival was detected by using clonogenic formation as-say. Results With specific inhibition of DNA-DPKCS expression, stable transfected cell line 549pRNA-DNA-DPKCS was constructed by RNA interference technique. The 549pRNA-C and 549pSUPER cell lines were the control cell lines tansfected with control and blank plasmids, respectively. Compared with A549 cells, the expression levels of DNA-DPKCS mRNA (0.110: 1. 000), protein (0. 870: 2.967) and activity of DNA-DPKCS (0.004: 0.266) in 549pRNA-DNA-DPKCS cells were significantly lower (F = 80.55 ,P < 0.01;F=63.96, P<0.01;F=51.62,P<0.01, respectively). The analysis of SF_2(0.25:0.76), D_0 (1.42:1.62) and D_q (0.06: 1. 00) showed significant difference between 549pRNA-DNA-DPKCS and A549 cells (F = 996.86, P < 0.01 ; F = 17.41, P < 0.05 ; F = 68.92, P < 0.01). The number of 549pRNA-DNA-DPKCS cells in S (24.5%: 35.5%) and G_2 (10.7%: 11.0%) phases was significantly decreased (F = 4.83, P<0.05 and F=32.04, P <0.01, respectively). Conclusions In A549 cells, inhibit of DNA-DPKCS gene expression can enhance the radiosensitivity and affect cell cycle distribution.

ABSTRACT

Background and purpose:The local tumor control rate of non-small-cell lung cancer treated with conventionally fractionated radiotherapy is low.Hypofractionated radiotherapy performed by conformal irradiation techniques can improne the local control rate.But further studies for appropriate fraction dose and toxicity for hypofractionation should be done.The purpose of this study was to prospectively evaluate the safety and efficacy of three-dimensional conformal hypofractionated radiotherapy for non-small-cell lung cancer(NSCLC).Methods:According to the dose-volume histogram(DVH) V_(20), patients were divided into three groups:① V_(20)≤20%,②20%30%,grade Ⅲ RP was observes in 2 of 5 patients and grade Ⅳ RP in 1 patient who died of lung function failure.No grade≥Ⅲ radiation esophagitis was observed.25 patients were evaluated with 8 complete responses,13 partial responses,3 stable diseases and 1 progressive disease.Conclusions:For three-dimensional conformal hypofractionated radiotherapy V_(20) level should be controlled below 30%,the treatment plan with V_(20)≥30% should be changed to palliative treatment.More studies are needed to confirm its efficacy.

ABSTRACT

Objective To study the synergistic effect of taxol and irradiation on programmed cell death induction in the human nasopharyngeal carcinoma cell line (CNE) in vitro.Methods Cell viability was evaluated by clonogenic assay. Cell cycle perturbation was assessed by flow cytometric analysis and apoptosis was determined with TUNEL.Results The intensity of cytotoxicity depended on the taxol dose used to CNE cells during the first 24 hrs. A G 2+M block was present with exposure to the drug of 100 nmol/L. When CNE cells were irradiated after 24 hrs of taxol exposure, synergistic effect was demonstrated with a taxol SER of 1.23. The same level apoptosis was induced by 10 ?mol/L taxol for 24 hrs or 2 Gy X-ray radiation in CNE cells.By both,the increase of induced apoptotic cells was over their additive results. Conclusion Taxol combined with radiation is able to enhance the radiosensitivity in the CNE cell line.

ABSTRACT

Objective To evaluate 18 F deoxyglucose positron emission tomography (FDG PET) in the detection of suspicious recurrence in head and neck cancers , as compared with CT/MRI imaging. Methods Thirty seven patients with clinically suspicious recurrences in head and neck cancers underwent FDG PET, with 34 checked with CT/MRI imaging. The final diagnosis of recurrence were proved by pathology or clinical following up. Results FDG PET detected recurrence successfully in 32 of 37(86.5%) patients with 3 false positives and 2 false negatives. The FDG PET sensitivity, specificity and accuracy in defining local recurrence were 91.7%, 76.9%, 86.5%, respectively; and those of CT/MRI were 68.2%, 75.0%, 61.8%, respectively. Conclusion In comparison with CT/MRI, FDG PET possesses a higher accuracy in detecting recurrence in head and neck cancers.

ABSTRACT

Objective To investigate the value of imaging diagnosis of endomyocardial fibrosis(EMF).Methods Imaging findings of 28 cases of EMF were restrospectively analyzed.Results There were three types:right ventricle type(n=21),left ventricle type(n=1),and dual ventricle type(n=6).The misdiagnostic rate of EMF by X-ray plain films was high(18/28). On angiography and MRI right ventricle type presented as shortened,deformed inflow-tract,obliteration of apex and dilated outflow-tract,marked tricuspid valve regurgitation(TR) and enlarged right atrium,even dilated superior vena cava,left ventricle type presented as narrowiug and deformed inflow-tract,small and deformed left ventride,even no markedly volume changes in relaxation or contraction,obtuse cardiac apex and mitral regurgitation.Dual ventricle type owned manifestations in the two types mentioned above,but predominated by right ventricle type. Conclusion In diagnosis of EMF X-ray plain films are of less value while MRI and cardioangiography are significant,MRI is a valuable non-invasive method,it can substitutd for cardioangiography as the first choice in diagnosing EMF.

ABSTRACT

The specific anti-rat LH, anti-rat FSH and anti-hCG sera were used in the peroxidase antiperoxidase (PAP) immunohistochemical method to observe the change of pituitary gonadotropic cells in pregnant rats after being injected by a high dosage of LHRH-A. It was observed that 24 hrs after injection the immunohistochemical reaction of gonadotropic cells weakened strikingly; 48 hrs after injection the immunohistochemical reaction began to recover; and 168 hrs after injection, the recovery was still in progress. The number of LH cells of different immunoreactive intensity and their percentage in total LH cells of the control and experimental groups were calculated and tested by statistical method. It was found that the results were the same as the above. Our observations support the view that the high dosage of LHRH-A can stimulate gonadotropic cells in rat to release a great amount of LH rapidly. It inhibits the synthesis of progesterone in the ovary of pregnant rat and results in termination of pregnancy. Moreover it is believed that the effect of high dosage of LHRH-A on the secretory function of gonadotropic cells can recover themseleves.

ABSTRACT

The specific anti-rat LH in the peroxidase anti-peroxidase (PAP) immunohistochemical method was used to demonstrate the pituitary gonadotropic cells in the pregnant rats, and then the morphometric analysis method was used to study the change of the number, size and nucleo-cytoplasmic ratio parameters of the gonadotropic cells in pregnant rats after being injected by a high dose of LHRH-A.It was observed that on 24h after injection the number of gonadotropic cells decreased, their size became smaller and their nucleo-cytoplasmic ratio larger: the cell number per mm~3 pituitary tissue was from (4.25?0.21)?10~4 to (2.49?0.14)?10~4; the cell size from 1295?35?m~3 to 895?31?m~3; the nucleo-cytoplasmic ratio from 0.144?0.005 to 0.229?0.010. On 48h after injection these changes were in the process of recovery. On 168h after injection all parameters were recovered.Our observations suggest that after being injected by a high dose of LHRH-A the change of gonadotropic cells in pregnant rats results from the exogenous high dose of LHRH-A which has stimulated gonadotropic cells to release a great amount of LH. It is believed, however, that the changes of gonadotropic cells caused by a high dose of LHRH-A can still return to normal.
