ABSTRACT

Objective To investigate the effects of heat shock protein Gp96 on alcoholic liver fibrosis in mice. Methods A total of 220 male healthy C57BL/6 J mice were randomly divided into four groups; normal control group (n = 10), saline+alcohol induced liver fibrosis group (n = 70), the injection of CRISPR expression Gp96-sgRNA3 by tail vein+ alcohol induced liver fibrosis group (n = 70), the intraperitoneal injection of nuclear factor kappa B(NF-κB) inhibitors PDTC+alcohol induced liver fibrosis group (n = 70). The blood was got from eyeballs and the mice were killed after 8 weeks of ethanol induction. We detected the activity of serum aspartate aminotransferase (AST) in mice of different groups. The pathological changes were detected by HE staining, sirius red staining and periodic acid-Schiff (PAS) staining in the liver of mice. The expression of Gp96 and transforming growth factor βl ( TGF-βl ) were detected by Western blotting. Results Compared with the normal control group, the AST enzyme activity and liver fibrosis increased significantly, glycogen decreased significantly in other three groups (P<0.01). Compared with the saline+alcohol group, the AST enzyme activity and liver fibrosis increased more significantly, glycogen decreased more significantly, Gp96 expression decreased significantly and TGF-βl expression increased significantly in Gp96-sgRNA3+ alcohol group and NF-κB inhibitors PDTC+ alcohol group (P<0.01 or P<0.05). Conclusion The injection of CRISPR expression plasmid Gp96-sgRNA3 by tail vein significantly inhibited the Gp96 expression, promoted the degree of alcoholic liver fibrosis in mice, and NF-κB signaling pathway played a certain role in regulating the expression of Gp96.

ABSTRACT

The artificial joints containing metal prosthetic components have different degrees of wear particles in the human body,which is the main reason for local osteolysis,inflammatory pseudotumor and prosthesis loosening.The metal wear debris after ar-tificial joint replacement can be derived from the normal wear of the weight-bearing interface volume,or from the edge wear of the non-weight-bearing interface.The particles produced by joint wear are mainly nano-sized particles,and their toxicity is stronger than that of micro-sized particles and ions.The wear particles can cause either local reactions by local accumulation or toxicity symptoms of multiple systems by entering the blood in various forms.Cobalt has the strongest toxicity among various metal particles.Oxidative stress,hyper-sensitivity,endoplasmic reticulum stress,autophagy,and direct destruction are the possible cytotoxic and genotoxic mechanisms,and the corresponding detoxification studies are also related to these mechanisms.There is no definite standard for the increase of the con-centration of metal particles and the clinical symptoms.At present, the main treatment is to remove the prosthesis and symptomatic treatment.Future research will focus on the improvement of prosthetic materials and finding new alternative treatment for joint diseases. Basic research will further explore the mechanism of toxicity of metal particles,especially the discovery of signal pathways and suscepti-bility genes as well as the in vivo and in vitro studies and clinical trials of new antidote.

ABSTRACT

<p><b>OBJECTIVE</b>To measure the axial rotation angles of the carpometacarpal joints during the digital opposition of thumb-index finger, thumb-medial finger, thumb-ring finger, thumb-little finger and the thumb's maximal opposition, then the application of these parameters were studied.</p><p><b>METHODS</b>Twenty neutrality-occupation volunteers (female 10, male 10) with no history of hand injuries or related diseases were involved in the study. First, all the markers' 3-D coordinates were obstained using the 3D motion analysis system (EVaRT4.1) during the digital opposition movements of thumb. Then, the axial rotation angles were calculated.</p><p><b>RESULTS</b>The average rotation angles of carpometacarpal joints during all kinds of digital oppositions were 29.1 degrees +/- 9.4 degrees (male), 24.8 degrees +/- 10.2 degrees (female), while the maximal rotation angles are: 35.3 degrees (male), 28.8 degrees (female).</p><p><b>CONCLUSIONS</b>The combination of video-based 3-D analysis system and mathematics make it possible to measure the axial rotation angles of thumb in vivo, as a result, the rotation angles of thumb carpometacarpal joints are measured precisely for the first time. These results can provide a few parameters for treatment and rehabilitation of carpometacarpal arthrositis.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of recombinant human epithelial growth factor (rhEGF) in promoting the healing of cervical erosion.</p><p><b>METHODS</b>Forty-eight patients with cervical erosion were treated with rhEGF and 30 with 500 kHz high-frequency electromagnetic wave, and the effects of the therapies were compared in terms of healing of the cervical wound, healing time, volume of vaginal discharge and bleeding and the lasting time.</p><p><b>RESULTS</b>In comparison with radiofrequency therapy, the healing of the lesion took significantly shorter time with rhEGF therapy, which also resulted in less vaginal discharge that lasted for shorter time without causing vaginal bleeding.</p><p><b>CONCLUSION</b>rhEGF can obviously accelerate the healing of cervical erosion.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of Qinggan Jiangtang tablet (QJT) in improving the insulin resistance (IR) in patients with multiple metabolic syndrome (MMS).</p><p><b>METHODS</b>Adopting randomized controlled double-blinded method, 60 patients with MMS were divided equally into two groups. The treated group was treated by oral taking of QJT 3 tabs, twice a day and the control group treated by oral taking of Glucophage 3 tabs, twice a day, the therapeutic course for both groups was 1 month, and the observation lasted for 2 successive courses.</p><p><b>RESULTS</b>After treatment, levels of blood glucose, fasting and postprandial blood glucose, C-peptide and IR were significantly reduced with markedly improvement of beta-cell function in both groups, the difference between the two groups showed no significance. Change of plasma level of free fatty acids before and after treatment in both groups was insignificant.</p><p><b>CONCLUSION</b>QJT has the effects on reducing blood glucose, blood lipids, blood pressure and IR and improving function of beta cells.</p>
