ABSTRACT

As a natural product with a long history of medicinal use, parthenolide has aroused great interest of chemists and biologists. Existing studies have shown that it has anti-inflammatory, antitumor and other pharmacological activities, and also revealed its action on NF-κB signaling pathway, DNMT1 enzyme and Wnt/β-catenin signaling pathway. But its biological targets remain to be elucidated systematically. Proteolysis Targeting Chimeras (PROTAC) provides a new strategy for target discovery of natural products, which can be used to explore the panorama of protein changes in cells through proteomic investigation, so as to analyze their potential targets. Based on this idea, current study designed and synthesized 20 parthenolide-derived degraders. After measured their antitumor activity in vitro, selected compounds were carried out the proteomic experiment. Finally, 139 down-regulated differentially expressed proteins were identified and the discovery of parthenolide interacting protein was preliminarily explored.

ABSTRACT

The expression, function and prognostic significance of epoxide hydrolase 2 (EPHX2) in hepatocellular carcinoma (HCC) were comprehensively analyzed through collecting HCC tissues and public database. The GEO and MitoCarta databases were used to identify the mitochondria-related differentially expressed genes (DEGs) in HCC. The Cancer Genome Atlas (TCGA) database was applied to analyze the expression levels of DEGs in HCC, including EPHX2 and its co-expressed genes. The R package was applied to draw the Kaplan-Meier survival curve and gene function enrichment analysis. The STRING database and GSEA software were used to analyze the protein-protein interaction (PPI) network and gene set enrichment analysis. qPCR and GEO database were applied to verify the expression level of EPHX2 in HCC. In the present study, a total of 15 mitochondria-related DEGs were identified in HCC. The expression of EPHX2 in HCC was significantly decreased compared to the normal liver tissues (P < 0. 01). The expression of EPHX2 was related to gender, tumor stage and grade in HCC, but not associated with age, T stage, et al in HCC. Moreover, compared with the patients with lower expression of EPHX2, patients with higher expression of EPHX2 had a better prognosis. EPHX2 was associated with fatty acid degradation. In addition, PPI results indicated that HAO1, AGXT, ACOX1, GSTκ1, SCP-2, CAT, CYP2C8, CYP2C9, CYP2B6, and CYP2J2 were co-expressed with EPXH2 in HCC. Furthermore, GSEA results showed that the group with lower expression of EPHX2 was positively correlated with the gene set of liver cancer cell proliferation and liver cancer recurrence. qPCR and GEO database results verified that the expression of EPHX2 was significantly decreased in HCC. The expression of EPHX2 was decreased in HCC, strongly suggesting that EPHX2 might function as a tumor suppressor gene in HCC. However, the potential mechanism of EPHX2 in HCC needs to be further verified.

ABSTRACT

Objective To explore the morphology and the tissue structure of acromioclavicular joint. Methods Anatomical analysis was performed on 27 fresh adult cadavers and the morphology of the acromioclavicular joint was observed. The relevant bone structure and ligament parameters were measured, and the specimens were randomly divided into group A and group B. Group A reserved the acromioclavicular ligament and coracoclavicular ligament, and group B reserved only the acromioclavicular ligament. The difference in tension between the two groups was compared. Results The distance from the midpoint of the conical ligament to the distal end of the clavicle was (42.68 ± 6.34) mm, the width of the end point was (16.97 ± 4.28) mm, and the thickness of the center point was (5.39 ± 0.34) mm; the distance from the midpoint of the trapezoidal ligament to the clavicle was (20.35 mm ± 4.18) mm, the width of the end point was (10.35± 1.31) mm, the thickness of the center point was (5.19 ± 0.342) mm; the average vertical distance from the base of the coracoid process to the surface of the clavicle was 30.75 mm, and the mean coracoclavicular gap was 12.02 mm; the length of the central axis of the conical ligament was (15.68 ± 3.30) mm and the angle was (117.25 ± 10.80) °, while the length of the central axis of the trapezoidal ligament was (9.67 ± 2.25) mm, and the angle was (75.42± 11.37) °. The distance between the start joint of the trapezoidal ligament and the trapezium was (8.96± 3.00) mm, and the distance between the end points (13.09± 3.50) mm. The average tensile force of group A was higher than that of group B [(610.04 ± 51.24) N vs (560.41 ± 44.63) N, P < 0.05]. Conclusion During distal clavicular resection, the resection of the distal clavicle shall be within 10-30 mm. The depth shall not exceed 42 mm when drilling under the coracoid process. The reconstruction of the coracoclavicular ligament during acromioclavicular joint dislocation has an anatomical and biomechanical basis.

ABSTRACT

Objective:To observe the effect of modified Dihuangyin on vascular dementia due to kidney empty phlegm and blood stasis syndrome,and its effect on phosphoinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway [PI3K,microtuble-associated protein 1 light chain 3(LC3)Ⅱ/LC3Ⅰ,Akt,phosphorylatedprotein kinase B(p-Akt)] in peripheral blood. Method:Totally 100 cases were randomly divided into control group (50 cases) and observation group (50 cases),and were given donepezil,modified Dihuangyin combined with donepezil for 30 d,respectively. The main efficacy indicators [mini-mental state examination (MMSE),clinical dementia rating (CDR),activity of daily living scale (ADL),traditional Chinese medcine(TCM) syndrome] were compared in two groups. The cerebral blood flow dynamics [middle cerebral artery (MCA),basilar artery (BA),anterior cerebral artery (ACA),posterior cerebral artery (PCA)] speed,serum inflammatory factors [tumor necrosis factor-<italic>α</italic> (TNF-<italic>α</italic>),interleukin-6 (IL-6),transforming growth factor-<italic>β</italic> (TGF-<italic>β</italic>),C-reactive protein (CRP)],oxidative stress indicators [malondialdehyde (MDA),superoxide dismutase (SOD),glutathione peroxidase (GSH-Px) ,homocysteine (Hcy)],peripheral blood PI3K/Akt signaling pathway (PI3K,LC3Ⅱ/LC3Ⅰ,Akt,p-Akt) were tested. The efficacy and adverse reactions of the two groups were compared. Result:The total effective rate was 95.9% in observation group was higher than that 72.3% in control group (<italic>χ</italic><sup>2</sup>=5.673,<italic>P</italic><0.05). Compared with the control group after treatment,the MMSE,ADL,SOD,GSH-Px,TGF-<italic>β</italic>,PI3K,Akt and p-Akt in the observation group were increased (<italic>P</italic><0.05),CDR,MCA,BA,ACA and PCA were increased (<italic>P</italic><0.05),and the TCM syndromes,MDA,Hcy,TNF-<italic>α</italic>,IL-6,CRP,LC3Ⅱ/LC3Ⅰ were decreased (<italic>P</italic><0.05). There was no significant difference incidence of adverse reactions between two groups. Conclusion:Modified Dihuangyin can significantly improve the clinical symptoms of patients with vascular dementia due to kidney empty phlegm and blood stasis syndrome,and the mechanism of action may be related to PI3K/Akt signaling pathway in peripheral blood.

ABSTRACT

Objective: To study inhibitory effect of total flavonoids from Ampelopsis grossedentata (TF) on transplanted tumors of human hepatocellular carcinoma in nude mice, and predict that its mechanism may be related to relevant factors regulating phosphatidylinositol 3 kinase (PI3K)/protein kinases B(Akt)/p53 pathway in apoptosis. Method: The nude mice transplanted BEL-7404 hepatoma model was established and divided into model group, 5-fluorouracil (5-FU) group (1.0 g·L-1) and TF (30, 15, 7.5 g·L-1) groups. Nude mice were put to death after two weeks of administration. The tumor tissues were excised, and tumor inhibition rate (IR) and relative tumor proliferation rate (T/C) were calculated. Reverse transcription PCR(RT-PCR) was used to detect PI3K, Akt1, p53 gene(p53), Caspase-3, B cell lymphoma/lewkmia-2 (Bcl-2), Bcl-2 associated X protein (Bax) mRNA expressions, immunohistochemical method was used to detect expressions of relevant proteins PI3K, Akt1, p53, Caspase-3, Bcl-2, Bax. Result: The establishment of xenograft tumor in mice showed that TF was administered orally once per day for two consecutive weeks. IRs were 53.26%, 35.94%, and 26.74%, respectively. T/Cs were 59.74%, 69.66%, and 84.82%, respectively. RT-PCR experiments showed that compared with model group, when TF concentration was 30 g ·L-1, mRNA expressions of PI3K, Akt1, and Bcl-2 were significantly down-regulated, and mRNA expressions of tumor suppressor genes p53, Capsase-3, and Bax were significantly up-regulated. Immunohistochemical method results showed that compared with model group, at TF concentrations of 30, 15 g·L-1, all PI3K, Akt1, Bcl-2 protein expressions were significantly down-regulated, while p53, Capsase-3, Bax protein expressions were significantly increased. Conclusion: TF has an obvious anti-liver cancer activity in vivo. Its mechanism may be correlated with up-regulation of expressions of p53, Caspase-3, and activation of apoptosis PI3K/Akt/p53 pathway, thereby inhibiting Bcl-2, increasing expression of Bax, and promoting hepatocellular apoptosis.

ABSTRACT

Objective To investigate the malaria vectors in Nanyang City. Methods Two townships were selected as malaria vector surveillance sites in Nanyang City from 2005 to 2017, including Mazhenfu Township in Tanghe County and Huaiyuan Township in Tongbai County, and the malaria vectors were investigated using the semi-overnight human baiting trapping method and mosquito trapping in nets. Results The mean density of Anopheles sinensis was 0.65 to 22.30 mosquitos/(person · hour) in the two surveillance sites in Nanyang City using the human baiting trapping method from 2005 to 2017, and the peak density was detected in June and July. An. anthropophagus was captured only in Tongbai County during the period between 2008 and 2011, with a mean density of 0.03 to 1.10 mosquitos/(person · hour). The mean density of An. sinensis was 0.08 to 0.24 mosquitos/(net · night) using the mosquito trapping in nets from 2005 to 2011, and the peak density was detected in July and August, while few An. anthropophagus captured only in Tongbai County, with a mean density of 0.01 to 0.02 mosquitos/(net · night). Conclusions There are malaria vectors in Nanyang City, including An. sinensis and An. anthropophagus, and An. sinensis is the dominant mosquito species.

ABSTRACT

Objective The SOX7 gene plays a tumor-suppressive role in a variety of tumors, but there are few reports on whether it plays a role in bladder cancer. This study aims to investigate the expression of SOX7 gene in bladder cancer as well as to investigate the regulation and significance of SOX7 promoter methylation on bladder cancer. Methods GEPIA, Oncomine, MethHC, and cBioPortal databases were used to speculate the SOX7 expression and promoter methylation in bladder cancer tissues. 40 urine samples were collected from January 2017 to October 2017 in the Department of Urology, Tenth People's Hospital of Shanghai City, including 20 samples from bladder cancer patients and the rest 20 from regular patients as a control group. The methylation difference of SOX7 gene was detected by methylation-specific PCR. The bladder cancer cell line was cultured. The medium containing the methylated drug 5-aza-2’ deoxycytidine (5-aza-dc) was added to the Taza cells as the 5-aza-dc group, while T24 cells were added the same volume of DMSO as the control group. The bladder cancer cell line was transfected with the SOX7 plasmid as the plasmid group, and the transfected with the unloaded plasmid was the empty group. Western blot was used to detect the expression of SOX7 in bladder cancer cell lines, and the proliferation, clone formation, and apoptosis of bladder cancer cells after demethylation were detected by CCK-8 experiments, plate cloning experiments, and flow cytometry, respectively. Results The level of methylation in bladder cancer was significantly higher than that in healthy tissues (P<0.005). The higher levels of SOX7 methylation were observed in the urine of 15 bladder cancer patients (75%), compared with only 7 patients (35%) in normal urine, and the proportion was statistically different (P<0.05). The expression of SOX7 protein in the 5-aza-dc group was up-regulated compared to the control group. The expression of SOX7 protein was relatively high when the concentration reached 20 μmol/L. The expression of SOX7 protein in the plasmid group was significantly higher than that in the unloaded group. CCK-8 results showed that the A value of the 5-aza-dc group was statistically lower than that of the control group on the fifth day (P<0.05), and the A value of T24 cells in the plasmid group was significantly lower than that in the unloaded group. The colony formation experiment showed that the number of colony formation per well in the 5-aza-dc group (167.33 ± 13.65) was significantly lower than that in the control group (328.00 ± 20.81) (P<0.05). The number of clone formation per well in the plasmid group (136.00 ± 15.00) was significantly lower than that in the unloaded group (280.67 ± 13.43) (P<0.05). The apoptosis rate of T24 cells in the 5-aza-dc group (27.89%) was significantly higher than that of the control group (3.79%) (P<0.05), and the apoptosis rate of the plasmid group (21.28%) was higher than that of the no-load group (9.90%). Conclusion SOX7 is lowly expressed in bladder cancer, which is regulated by promoter methylation. It is a potential biological marker of bladder cancer and plays a vital role in the occurrence and development of bladder cancer.

ABSTRACT

<p><b>OBJECTIVE</b>To explore function and related molecular mechanism of osteopractic total flavone (OTF) on tendon healing in rats.</p><p><b>METHODS</b>Ten male rats aged for 8 weeks were collected and weighted from 180 to 220 g. Tendon stem cells were cultivated, the third tendon stem cells were used for experiment. OTP treated with 0, 0.1, 1, 10 ng/ml were added into tendon stem cells, and expression change of ALP, Runx2, OCN, VEGF, P-S6, P-4E/BP1 were detected after 14 days. Forty male rats aged for 8 weeks (weighted 180 to 220 g) were established extra-articular tendon-bone transplanting healing model, and divided into experimental group and control group. Experimental group were treated with OTF(100 mg·kg⁻¹·d⁻¹), while control group was treated by normal saline with the same volume. Tendon-bone healing degree were detected by biomechanical testing at 3 and 6 weeks after surgery, histological detection were applied to detect tendon-bone healing and number of new vessles.</p><p><b>RESULTS</b>After treated by OTP, ALP staining and active index detection showed there were statistical differences among 0, 0.1, 1, 10 ng/ml group. After 14 days' cultivation, western blotting results showed mTOR downstream marker protein P-S6 protein expression were gradually increased with increase of density of OTP, expression of P-4E/BP1 was reduced, while expression of Runx2, OCN, VEGF were increased. Biological detection results showed that there was no significant difference in mechanical strength between experimental group(0.78±0.05) N/mm and control group (0.51±0.02) N/mm at 3 weeks after surgery, while mechanical strength in experimental group (1.36±0.09) N/mm was higher than control group (1.01±0.08) N/mm at 6 weeks after surgery. Histological results showed maturity of tendon-bone surface cell were higher at 3 and 6 weeks in experimental group, sharpey fiber growth more density, calcification extent of mesenchyme was high, and new bone, vessels were increased.</p><p><b>CONCLUSIONS</b>OTF could promote osteogenic differentiation of tendon stem cells through mTOR signaling in vitro, and stimulate tendon-bone healing in bone tunnel and enhance connection quality between tendon and bone.</p>

ABSTRACT

Objective: To explore the expression and prognostic significance of miR-223 in patients with mantle cell lymphoma (MCL) and to investigate the possible mechanism. Methods: Twenty-one newly diagnosed MCL patients with bone marrow involvement were enrolled in the present study, 20 healthy donors as normal control. The expression level of miR-223 and SOX11 mRNA was determined by RQ-PCR. CCK-8 and flow cytometer assays were used to analyze cell proliferation, cell cycle and apoptosis of the constructed miR-223 overexpressing MCL cell line, Granta519 cells. SOX11 protein expression level was determined by Western blot. The target gene of miR-223 was confirmed by dual luciferase reporter assay. Results: ①Of the 21 newly diagnosed MCL patients, 15 were male and 6 female, the median age was 58 (37-72) years. The expression level of miR-223 was significantly down regulated in MCL patients compared with that of healthy donors (14.7±10.5 vs 1 244.1±1 935.2, P<0.001). The lower expression of miR-223 was inversely correlated with high-risk mantle international prognostic index (P=0.001), elevated LDH (P=0.001), ECOG score ≥2 (P=0.035). ②Using the median relative expression level of miR-223 as the cutoff value, 21 MCL patients were divided into high-expression group (n=10) and low-expression group (n=11) and found that the high-expression group had a significantly superior OS (median OS: 36 vs 12 months, P=0.021). ③In vitro results showed that compared with the control group, the proliferation of miR-223 overexpressed Granta519 cells was inhibited (the most significant reduction on 96h, P<0.001), manifested by lower proportion of cells in G2/M phase (P<0.001) and increased apoptosis (P<0.001), and the expression level of SOX11 protein in Granta519 cells was significantly lower than that of the control group. ④miR-223 could inhibited the 3' untranslated region of SOX11, and the expression level of miR-223 was significantly negatively correlated with mRNA level of SOX11 in MCL patients (r=-0.81, P<0.001). Conclusions: The expression of miR-223 was repressed in MCL and was associated with poor clinical outcomes, which may be probably attributed to its direct targeting SOX11.

ABSTRACT

Objective To explore the clinical effects of Ilizarov technology combined with closed minimally invasive osteotomy and slow tissue distraction in the treatment of complex crus malformations.Methods From June 2006 to February 2016,83 cases suffering complex crus malformations in our department were treated by Ilizarov technique combined with closed minimally invasive osteotomy and slow tissue distraction.Of whom,39 cases were traumatic bone defect,36 cases were bone osteomyelitis,8 cases were congenital pseudarthrosis of tibia.Bone defect ranged from 6 to 11 cm,with an average of 8 cm.All cases were conducted by segmental resection of bone lesions combined with closed minimally invasive osteotomy and Ilizarov technique.The functional evaluation was carried out according to the Paley evaluation criterion.Bone healing time,duration of external fixation,postoperative limb lengthening and limb function recovery were recorded.Results Eighty-three patients were followed up for 8 to 36 months,with an average of 16 months.All patients' crus malformations were completely corrected.The external fixation time was from 6 to 18 months,with an average of 10.3 months;the length of the limb lengthening was from 4.5 to 9 cm,with an average of 6.3 cm;and bone healing time was from 6 to 15 months,with an average of 9.8 months.According to the Paley evaluation criterion,53 cases were excellent,24 cases were good,6 cases were general.Conclusion Ilizarov technology combined with closed minimally invasive osteotomy and slow tissue distraction a reliable method to correct the complex deformity of the tibia and fibula.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the safety, effectiveness, and outcomes of holmium laser enucleation of the prostate (HoLEP) for patients with symptomatic enlarged prostate after 11 years of experience.</p><p><b>METHODS</b>The 3162 evaluable patients treated with holmium laser enucleation of the prostate at our institution between August 2001 and August 2011 were retrospectively analyzed. Study variables included International Prostate Symptom Score, quality of life, maximum urinary flow rate, and incidence of complications.</p><p><b>RESULTS</b>HoLEP were performed successfully completed, not patients which occurs as electric cutting syndrome. The operation time was (60.8 ± 18.4) minutes; average resection of prostate quality was (45.4 ± 24.4) g. The hemoglobin reduce though surgery was (1.81 ± 0.93) g/L; percentage of red blood cell change was 1.24% ± 0.43%, and sodium blood drop was (1.14 ± 0.35) mmol/L. Postoperative patients of hospital stay (3.1 ± 1.1) days, average time of indwelling catheter time was (2.3 ± 0.8) days. Patients were followed up for 6-131 months time, an average of 32.4 months. Postoperative patients with international prostate symptom score progressive declined. The quality of life score was 2.2 ± 1.7, and it less than preoperative (5.7 ± 3.3, t = 2.447, P < 0.01). The time of follow-up droped further, and postoperative comparative differences have statistical significance (t = 2.179, 2.228, 2.306 and 2.365, P < 0.05). The maximum urinary flow rate also improved (P < 0.05). Postoperative complications included bladder neck contracture (4 cases), urinary tract infection (107 cases), urethral stricture (11 cases) and urinary incontinence (11 cases). The 11 patients reoperation.</p><p><b>CONCLUSIONS</b>HoLEP treatment of benign prostatic hyperplasia could achieve the advantages of open surgery the same effect. It had fewer damage, faster recovery, fewer complications, and is a good treatment option.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the early efficacy of nedaplatin combined with megestrol in concurrent chemoradiotherapy for advanced cervical cancer.</p><p><b>METHODS</b>Forty-two cases of cervical cancer (FIGO IIb to IVa) were divided randomly into two groups: radiotherapy alone (21 cases) and radiation plus chemotherapy (Nedaplatin) group. The same radiotherapy was given to the two groups. Patients of the RT + C group received nedaplatin 30 mg/m2 in intravenous drip infusion once weekly on day 1, for 4 to 5 weeks, and megestrol 160 mg orally every day during the radiation therapy.</p><p><b>RESULTS</b>The early outcome: the complete remission rate was 81.0% and partial remission rate was 19.0% in the RT + C group, significantly better than the CR (38.1%) and PR (42.9%) in the RT group. The 1-year survival rates in the two groups were 100% (21/21) and 81.0% (17/21), respectively, with a significant difference between the two groups (P<0.05).</p><p><b>CONCLUSIONS</b>The combination of nedaplatin and megestrol with concurrent chemoradiotherapy can improve the early outcome of advanced cervical cancer, with somewhat increased but tolerable adverse effects.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To obtain large amount of differentiated chondrocytes in vitro, examine and compare the biological characterization of rabbits' articular chondrocyte cultured in different density in vitvo.</p><p><b>METHODS</b>From November 2001 to June 2004, articulate tissues were obtained from the joints of the adult rabbits. Chondrocytes were isolated from the cartilage tissue with type II collagenase digestion and cultured in DMEM/F-12 supplemented with 20% fetal bovine serum (FBS). The chondrocytes were cultured with low density of monolayer culture and high density of confluent culture respectively. The differentiated phenotype was evaluated by histochemistry or immunohistochemistry.</p><p><b>RESULTS</b>When chondrocytes cultured in monolayer and in low density, it proliferated rapidly during the three generations, but with the same time, dedifferentiation was also rapid. After the third passage, most of the passage cells lost the phenotype, and the proliferation also stagnated. While chondrocytes cultured in high density, dedifferentiation slowed down. And even the phenotypes of the dedifferentiated chondrocyte which were cultured in low density could reduced partly by followed high density culture.</p><p><b>CONCLUSIONS</b>Culture chondrocytes by high density in vitro can effectively maintain the differentiated phenotype of chondrocyte. It also keeps the proliferation character as monolayer culture. The dedifferentiated chondrocyte caused by many passages could redifferentiate partly. So it is indicated that confluent culture of original or expanded chondrocytes in high density is a better culture methods than culture in low density.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluated the curative effect of arthroscopic Bankart repair with use of bio-absorbable fixation tacks.</p><p><b>METHODS</b>Sixteen patients were with traumatic recurrent anterior instability of the shoulder. Twelve were males, and 4 were females. Nine were in left shoulder, and 7 in right shoulder. The mean age at the time of the operation was 25 years. The average time from injury to operation was six months. Before operation the shoulder image had been done, in which X-ray film in 16, MRA in 7, CTA in 6 and MRI in 5. The arthroscopic technique included of debridement and Bankart repair using 2 to 3 bio-absorbable fixation tacks underwent arthroscopic.</p><p><b>RESULTS</b>All of the patients were followed up, average time 11 months, no redislocation and instability. According to the UCLA (University of California at Los Angeles) score showed all shoulder scores improved after surgery, preoperative (22.5 +/- 5.6) and postoperative (31.4 +/- 5.8), excellent score in 9, good score in 5 and fair score in 2; no poor score.</p><p><b>CONCLUSION</b>Arthroscopic capsulolabral repair with use of bio-absorbable fixation tacks has provided a satisfactory outcomes in function, stability and range of motion. The procedure has many advantages, such as the operation convenient, fixation safety, mini-invasion, rapid recovery and early rehabilitation.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the feasibility of reconstruction of anterior cruciate ligament (ACL) and posterior cruciate ligament (PCL) with hamstring tendons knot implant fixation.</p><p><b>METHODS</b>Fifty-two cases of ACL and PCL old injury were reconstructed under the arthroscopy with double bundles hamstring tendons knot implant fixated in the bottle-necked femoral tunnel. The tibia side were fixated by tendon weave suture cross tied a knot at the bone bridge of tibia. In this group reconstruction of ACL was in 25, PCL and ACL at the same time reconstruction in 15, PCL in 12. The failure test and displacement evaluation were used to study the biomechanics of reconstruction of ACL by hamstring tendon knot implant fixation (n = 13) in porcine knees. The control group were reconstructed with bone-patellar tendon-bone B-PT-B and interference screw (n = 11). The tibia side tendon weave suture immobility by cross tied a knot at the tibia bone bridge (n = 7) and interference screw in the tibia tunnel (n = 8).</p><p><b>RESULTS</b>Forty-nine cases were followed up, average 14.6 months. The results of Lanchman test was negative in 46 cases, positive in 3 cases. Preoperative Lysholm score was 56.7, and postoperative was 92.8. According to the knee joint effective evaluate standard, 46 were excellent and 3 good. The maximal pull-out force of hamstring tendons knot implanted fixation had been greater than B-PT-B fixation by interference screw. There were similar displacement in 100 N and 400 N load between hamstring tendons knot implant fixation and B-PT-B (P > 0.05). Peak force test maximal displacement evaluation and failure energy absorption measurement showed that hamstring tendons knot implant fixation was significantly greater than B-PT-B (P < 0.01). The failure test and displacement evaluation of tendon fixation by weave suture knot at the tibia bone bridge cross tie were greater than B-PT-B fixation by interference screw.</p><p><b>CONCLUSION</b>The methods of reconstruct of ACL and PCL by hamstring tendons knot implant fixation are feasible. The advantages include: it is biological fixation, not machine fixation; it may benefit for tendons with bone tunnel healing and decrease medical cost. Decreasing or eliminating displacement is helpful to prevent relaxation after reconstruction of ACL and PCL.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the feasibility of arthroscopic therapy of Baker's cysts with radiofrequency.</p><p><b>METHODS</b>Since Feb. 2000 to Oct. 2002 a surgical arthroscopic treatment for popliteal cyst in a series of 21 patients. Men 14 and female 7, the age from 45 to 66 year. To do CT scan 11 and MR in 8. To insert a arthroscope to Baker's cyst, and inject salline in the visceral layer of capsule under the arthroscope, so that remove of the capsule easily. The radiofrequency to clear the capsule of cyst and hemostasia.</p><p><b>RESULTS</b>All of the cases studied, a connection between joint space and cyst was found in 13 cases. The popliteal cyst was found to be almost invariably associated with other knee disorders. Follow-up in all of the case. No nerve and blood vessel injure, no infection in this group. The function is well include 3 cases relapsed after re-operation by arthroscope debridement.</p><p><b>CONCLUSIONS</b>Surgical arthroscopic therapy of Baker's cysts is safety and reliable; To removal of capsule integrity and repair of all intraarticular lesions and sealing of the junction between Baker's cyst and the dorsal recessus of the knee joint for preventing recurrent are very important.</p>

ABSTRACT

Objective To evaluate the advantages and short term clinical results of arthroscopic release with radiofrequeney vaporization for knee joint stiffness.Methods Arthroscopic release with ra- diofrequency vaporization was done in 32 cases with knee joint stiffness resulted from various causes be- tween January 2002 and June 2005.According to degree of stiffness,32 cases were divided into three groups,ie,Group A(18 cases with suprapatellar bursa adhesions)treated with confectioning at suprapa- tellar bursa and lateral grooves;Group B(eight cases with joint compartment adhesions)managed with debridement at suprapatellar bursa and lateral groove and intereondyloid fossa,bending the knee with pressure and abscising the adhesion part in the joint compartment with radiofrequency vaporization;and Group C(six cases with most part of the joint adhered)treated by making small incision at the lateral or medial upper pole of the patellar as well as a space at the suprapatellar bursa by blunt dissection when it was difficult to insert arthroscope.After that,release combined with massage manipulation and thorough hemostasis by radiofrequency vaporization were carried out.There was no drainage after operation.Pas- sive and active flexion and extension exercises with pressure began two days after operation.Results Pre-operative limitation of flexion was 35?-75?that was increased to 115?-125?three weeks after opera- tion,with mean improvement of 78?in Group A.Pre-operative limitation of flexion was 40?-60?that was increased to 95?-120?three weeks after operation,with mean improvement of 72?in Group B.Pre-opera- tive limitation of flexion was 25?-45?that was increased to 90?-110?three weeks after operation,with mean improvement of 64?in Group C.No haematocele occurred postoperatively,with only slight swelling of the knee.Conclusion Arthroscopic release with radiofrequency vaporization for knee joint stiffness is characterized by less bleeding,minor trauma and reliable release and can be applicable especially for the eases with only adhesion at suprapatellar bursa,lateral groove or joint compartment.

ABSTRACT

Objective To investigate the suppressive effects of collagen from Cyanea nozakii on adjuvant induced arthritis inrat.Methods Rats with adjuvant arthritis received different do- ses of Cyanea nozakii collagen by intragastric administration for two weeks.Incidence and severity of arthritis were assessed by calculation of mean arthritis index,the concentrations of NO,MDA and activity of SOD in the serum were examined.Results Cyanea nozakii colla- gen at different doses could ameliorate the adjuvant induced arthritis,suppress the concen- trations of NO,MDA and increase the activity of SOD in the serum.Conclusion Cyanea noza- kii collagen has therapeutic efficacy in treatment of adjuvant arthritis rats,and the mecha- nism of Cyanea nozakii collagen is probably related to the antioxidation.
