ABSTRACT

Objective:To investigate the differences between serum lipoprotein-associated phospholipase A2 (Lp-PLA2) and insulin resistance (IR) and the relationship between Lp-PLA2 and IR and homocysteine (Hcy) levels in patients with type H and non-type H hypertension.Methods:A total of 298 patients with essential hypertension (observation group) who visited Lu′an Hospital of Traditional Chinese Medicine in Anhui Province from January 2018 to January 2020 were selected, and 564 healthy subjects were as control group. According to age, gender and body mass index (BMI), propensity score matching was conducted, and the matching relationship was determined according to 1∶1 nearest neighbor matching method. Finally, 166 cases were included in the observation group and the control group. Type H hypertension was defined as essential hypertension with an Hcy level of > 10 mol/L.Results:The smoking proportion, total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), fasting blood glucose (FPG), fasting insulin (FINS), fibrinogen (FIB), hypersensitive C reactive protein (hs-CRP), LP-PLA2, Hcy, and homeostasis model of assessment for insulin resistance index (HOMA-IR) in the observation group were higher than those in control group: 24.7% vs. 13.9%, (5.06 ± 1.65) mmol/L vs. (4.60 ± 1.42) mmol/L, (2.92 ± 0.97) mmol/L vs. (2.62 ± 0.86) mmol/L, (5.80 ± 1.03) mmol/L vs. (5.12 ± 0.94) mmol/L, (13.06 ± 5.14) U/L vs. (9.25 ± 4.67) U/L, (3.28 ± 1.16) g/L vs. (2.17 ± 1.01) g/L, (2.00 ± 1.09) mg/L vs. (0.52 ± 0.43) mg/L, (161.98 ± 86.84) mg/L vs. (126.87 ± 56.84) mg/L, (15.00 ± 5.06) mol/L vs. (11.54 ± 3.63) mol/L and 3.49 ± 1.25 vs. 2.03 ± 1.11 ( P < 0.05), and the high density lipoprotein cholesterol (HDL-C) was lower than that in control group: (1.15 ± 0.56) mmol/L vs. (1.33 ± 0.66) mmol/L ( P < 0.05). The age, BMI, smoking ratio, TC, FPG, FINS, FIB, hs-CRP and Hcy in H-type hypertension group were higher than those in non-H-type hypertension group: (56.41 ± 11.07) years old vs. (49.61 ± 10.58) years old, (27.92 ± 4.02) kg/m 2 vs. (24.23 ± 4.11) kg/m 2, 31.1% vs. 14.3, (5.32 ± 1.54) mmol/L vs. (4.63 ± 1.24) mmol/L, (5.97 ± 1.03) mmol/L vs. (5.51 ± 0.98) mmol/L, (14.29 ± 5.04) U/L vs. (11.06 ± 4.57) U/L, (3.48 ± 1.10) g/L vs. (2.95 ± 0.83) g/L, (2.48 ± 1.21) mg/L vs. (1.22 ± 1.02) mg/L, 16.13 (12.96, 23.20) mol/L vs. 7.63 (6.58, 8.35) mol/L ( P < 0.05), and the HDL-C was lower than that in non-H-type hypertension group: (1.05 ± 0.54) mmol/L vs. (1.31 ± 0.78) mmol/L ( P < 0.05). HOMA-IR was 2.27 (1.60, 3.34) and Lp-PLA2 was 61.64 (53.25, 75.47) in patients with non-H-type hypertension. HOMA-IR was 3.34 (2.63, 443) and Lp-PLA2 was 212.25 (120.35, 278.62) in patients with type H-type hypertension. Lp-PLA2 and Lp-PLA2 were higher in patients with H-type hypertension than those in patients with non-H-type hypertension ( P < 0.05). In patients with essential hypertension, there was a strong positive correlation between Hcy and HOMA-IR and Lp-PLA2 ( r = 0.655 and 0.774, P < 0.05). Age, BMI, TC, FPG, FINS, FIB, hs-CRP, HOMA-IR and LP-PLA2 were independent risk factors for Hcy increase ( P < 0.05), while HDL-C was a protective factor for Hcy increase ( P < 0.05). Conclusions:Patients with type H-hypertension have a higher HOMA-IR and an increased level of Lp-PLA2, which is correlated with an increase in Hcy.

ABSTRACT

Objective To investigate the relationship between the interleukin-17 (IL-17),interleukin23 (IL-23) and carotid artery sclerosis in patients with cerebral infarction.Methods One hundred and twentyfive cerebral infarction patients in the first affiliated hospital of Shantou University medical college from June 2010 to June 2012 were recruited.All the subjects were examined with carotid ultrasonography.According to the results of carotid ultrasonography,all patients were divided into carotid atherosclerosis group(86 cases) and non carotid atherosclerosis group(39 cases).Patients with carotid atherosclerosis were divided into soft plaque group (35 cases),hard plaque group (21 cases),mixed plaque group (30 cases) based on plaque feature.Carotid atherosclerosis patients with cerebral infarction plaque determined by semi-quantitative method were grouped into Ⅰ grade(29 cases),Ⅱ grade (43 cases),Ⅲ grade (14 cases) according to the severity atherosclerosis.Serum levels of IL-17 and IL-23 were measured.Results The levels of IL-17 and IL-23 in patients with carotid atherosclerosis group were (31.42 ± 8.73) μg/L and(21.79 ± 9.34) μg/L,higher than that of non-carotid atherosclerosis group((22.81 ±6.52) μg/L,t =5.15,P <0.01 ; (14.33 ±6.21) μg/L,t =4.99,P <0.01).The IL-17 and IL-23 levels in patients with soft plaque,hard plaque and mixed plaque group were significant different (F =10.181,3.835,P <0.05).Serum IL-17 in soft plaque and mixed plaque expression group was higher than that of hard plaque group (P =0.001,P=0.007).IL-23 level in soft plaque group was higher than mixed plaque and hard plaque groups(P =0.017;P =0.045).The IL-17 and IL-23 levels in Ⅰ,Ⅱ,Ⅲ level group were markedly different (F =41.046,3.739 ; P < 0.05),and there was significant difference between Ⅱ,Ⅲ and Ⅰ group(P <0.01,P <0.05).The correlation between IL-17 and IL-23 levels in Ⅰ,Ⅱ,Ⅲ level group were 0.892 and 0.420 (P =0.000 ; P =0.041).Conclusion IL-17 and IL-23 play important roles in the pathologic progress of artery sclerosis and are helpful for early diagnosis of the disease.

ABSTRACT

Objective To investigate the imbalance of T helper 17 (Th17) / CD4 + CD25 + regulatory cell (Treg) in HAM/TSP patients.Methods Enzyme linked immunosorbent assay (ELISA) was performed to measure the levels of interleukin (IL-17) and IL-10 in CSF,and flow cytometry to determine the percentage of Th17 and Treg cells in peripheral blood of patients with HAM/TSP.Results There was a significant increase in the level of IL-17 [(4.58 ± 0.70)pg/ml vs (0.76 ± 0.17)pg/ml,P < 0.01] in CSF and the percentage of Th17 cells [(2.00 ± 0.64) % vs (0.41 ± 0.24) %,P < 0.01],but a decrease in the level of IL-10 in CSF and the percentage of Treg cells in peripheral blood of patients with HAM/TSP compared with the control group.The ratio of Th17/Treg cell (0.55 ±0.10 vs 0.06 ±0.03) in the peripheral blood of patients with HAM/TSP increased significantly compared with control group (t =12.11,P <0.01).Conclusions The imbalance between Thl7 and Treg cells may play an important role in the pathogenesis of the disease.

ABSTRACT

Objective To observe the effects of exogenous interleukin (IL)-6 on survival time and regeneration of liver grafts in an orthotopic liver transplantation model in IL-6 knockout (KO) mice.Methods A model of liver transplantation in C57BL/6 wild type (WT) and IL-6 KO mice with C57BL/6 background was established. Grafts were preserved at 4 ℃ in UW solution before transplantation.Study was divided into 4 groups:IL-6 KO→IL-6 KO control group,IL-6 KO→C57BL/6 WT group,IL-6 KO→IL-6 KO+ rIL-6 group,and IL-6 KO→C57BL/6 WT+ rIL-6 group.Recipient animals were injected subcutaneously with human recombinant IL-6 (1 mg/kg body weight) 1 h before transplantation.Survival of liver grafts after transplantation was followed up.Hepatocyte replication with BrdU uptake after liver transplantation was examined by irnmunohistochemistry.Results A total of 48 liver transplantations were performed.Cold ischemic time was 50 min.The survival time of liver grafts in the control group was 2.2 days,and that was 1.9 days,>17.6 days and >20.4 days in IL-6 KO→C57BL/6 WT group,IL-6 KO+ rIL-6 group and C57BL/6 WT+ rIL-6 group respectively with the the difference being statistically significant among the latter three groups (P<0.01 ).The liver grafts transplanted in control group recipients showed patchy areas of necrosis and hepatocyte ballooning.Slight increase in BrdU uptake was found at 48 h post-transplantation.Minimal injury and no necrosis were observed in IL-6 treated groups.Mild increase of BrdU uptake was demonstrated at 48 h after liver transplantation. Conclusion Exogenous recombinant IL-6 appears to significantly prolong the survival time of IL-6 KO liver grafts after liver transplantation and enhance the regenerative response after liver transplantation in IL-6 KO mice.

ABSTRACT

Objective To observe the effect of electroacupuncture on onset time and effectiveness of Paroxetine for mild or moderate depression patients and on the scores of Hamilton depression rating scale for depression(HAMD) and self-rating depression scale(SDS). Methods Fifty-five patients with mild or moderate depression were randomly divided into A group( Paroxetine group,29 patients) and B group (Electroacupuncture combining with Paroxetine group,26 patients). Therapeutic periods for the two groups were 6 weeks. HAMD and SDS were used to evaluate the efficacy before the research and after treatment on 1 week,2 weeks,4 weeks,6 weeks.Results HAMD scores and SDS scores for A group were significant difference after 2 weeks treatment compared with the beginning of the research (P< 0.05 ) ;HAMD scores and SDS scores for B group were significant difference after 1 week treatment compared with the beginning of the research(P<0. 05 ). Total effective rate of HAMD for A group was 71.4% ,total effective rate of SDS for A group was 75% ,total effective rate of HAMD for B group was 88% ,total effective rate of SDS for B group was 84%. The total effective rate between the two groups ordered by the individual chi-square test was significant difference( X2 = 4. 1790, P= 0. 0408). Conclusion Electroacupuncture can shorten the onset time and increase the effectiveness of paroxetine for mild or moderate depression patients.

ABSTRACT

Objective To investigate the effect of IVIG on pre-existing anti-HLA-A2 Ab levels and graft skin survival.Methods C57BL/6 wild type mice were sensitized to HLA-A2 by intraperitoneal injec-tion (IP) of HLA-A2 TgN mouse spleen cells (C57BL/6-TgN [HLA-2.1]1Enge SC) expressing human HLA-A2 at day 0, week 3 and 4.Sensitized mice were respectively treated with human IVIG , albumin, gly-cine, or PBS for 5 days during week 7.Skin transplantation from TgN mice to C57BL/6 wild type mice was performed at week 10 .Efficacy of IVIG DES was assessed by measuring anti-HLA Ab levels by ELISA pre-/post-Rx and graft skin survival was monitored daily post-Tx.Results Changes of HLA-A2-IgG levels:IgG HLA-A2 class I Ab levels in all groups predictably increased from baseline following spleen cell IP and peaked at week 4 ( P 0.05 ) . Conclusion However, IVIG alone does not inhibit anti-HLA class I Ab production after skin transplanta-tion or prolong SG survival , indicating combination of IVIG with other immunosuppressant or more optimal protocol for desensitization might show more efficacy and should be under explored .

ABSTRACT

Objective To investigate ultrasound guidended nerve block of femoral nerve in emergency operation.Methods 40 patients who planned to accept surgery of feet,lower limb under knee.They were randomly devided into 2 groups:ultrasound guidended group and the control group.Femoral nerve and Sciatic nerve were blocked by ultrasound guidended technique or classic landmark technique.1.5% lidocaine was administered for nerve block.Sensory block was observed every 5 minutes in 30 minutes.Onset time,effect of anesthesia,algesia time and complications were recorded.T-test and varience difference were applied.Results The onset time of local anesthetics was faster in the ultrasound guided group than that of the control group.Effect of analgesia was better in in the ultrasound guided group than that of the control group. Conclusion Nerve block of femoral nerve and the sciatic nerve guided by ultrasound was better than the classic technique.

ABSTRACT

Objective To investigate the coordinate repression of angiostatin(AS)and Fas gene on human colon carcinoma LOVO.Methods Plasmid pcDNA3-AS and pcDNA3-Fas were constructed,and AS,Fas,AS and Fas gene were transfected to human colon carcinoma LOVO cells by liposome method.The expressions of target protein were detected by Western blot.The effects of transfection of AS and Fas gene on the growth of human colon carcinoma cells were detected by MTr methods.AS,Fas,AS and Fas gene were transfected to the human colon carcinoma subcutaneously implanted in nude mice by direct injection into the tumor.The tumor sizes were detected after 14 days of the first gene transfection.Results The effect of gene transfeetion in LOVO cell after twelve hours,24 h,48 h and 72 h were observed and compared with control group,co-transfection of Fas and AS gene group and Fas group significantly inhibited the growth of human colon carcinoma LOVO line cells(P<0.01).Fourteen days after plasmid transfection,the tumor volume in group Fas and group co-transfection were signifieandy smaller than that of control,AS and Fas group(P<0.05).The tumor volume in AS group and Fas group were significantly smaller than that of control(P<0.05).Conclusion Angiostatin and Fas gene have coordinate repression effect on human colon carcinoma in vivo.

ABSTRACT

AIM: To understand the effect of osmotic agents on hyaluronan synthesis in human peritoneal mesothelial cells(HPMCs). METHODS: Cultured HPMCs were stimulated with 90 mmol/L glucose(HG), 30 mmol/L glucose(LG), 5% polyglucose(PG) and 90 mmol/L mannitol. Extracellular cell coats were examined using particle exclusion assay, the expressions of hyaluronan synthase 2 and 3(HAS-2 and HAS-3) mRNA in HPMCs were measured by reverse transcription-polymerase chain reaction(RT-PCR). The concentration of hyaluronan in cultured medium was measured by hyaluronan radio-immunoassay kits. RESULTS: HAS-2 mRNA expressions in the four experimental experimental groups were significantly higher than that of the control group. However, only HG enhanced HAS-3 mRNA expression. The sizes of the extracelluar coat in the four experimental groups were not significantly different from that of the control. In addition, hyaluronan concentrations were significantly higher in the four experimental groups as compared with the control group. The hyaluronan concentration in HG group was much higher than that of the LG, PG and mannitol groups. CONCLUSIONS: Although polyglucose increased HAS-2 mRNA expression, it tended to have no effect on HAS-3 mRNA in HPMCs. This result implies that polyglucose might be more biocompatible to peritoneum in peritoneal dialysis as compared with hypertonic glucose.
