ABSTRACT

Aim To investigate the effect of safflower yellow (SY) on learning and memory ability of APP/ PS1 mice at different disease stages, and to explore the mechanism of SY anti- Alzheimer's disease by using 3-,6- and 9-month-old APP/PS 1 transgenic mice as experimental animal models. Methods Behavioral experiments were conducted to observe the effects of SY on learning and memory of APP/PS1 mice of different months. ELISA was used to detect the effect of SY on the expression of inflammatory factors in cortex of mice of different months. Western blot was used to detect the microglia activation marker protein, and its mechanism of action was further analyzed. Results SY could enhance the learning and memory ability of mice aged 3, 6 and 9 months, reduce the content of IL-6 and increase the content of TGF-β1 in brain tissue, up-regulate the expression levels of arginase-1 (arg-1) and triggering receptor expressed on myeloid cells 2 (tREM2) in brain tissue of mice of different months, and down-regulate the expression levels of inducible nitric oxide synthase (iNOS), Toll-like receptors 4 (tlr4) and nuclear factor-kappa B (nf-KB). Conclusions Compared with 3- and 9-month-old mice, SY is the most effective in improving learning memory in 6-month-old APP/PS1 mice. SY inhibits TLR4/NF-KB pathway activation by inducing TREM2 expression in brain tissue of APP/PS 1 transgenic mice, promotes microglia phenotype shift to anti-inflammatory phenotype, reduces chronic neuroinflammatory response, and improves learning memory in APP/PS1 mice at all months of age.

ABSTRACT

AIM: To investigate the changes in anterior chamber depth(ACD), axial length(AL), and corneal curvature(K)after operation in patients with high axial myopia combined with cataract, and the effect on postoperative mean refractive error(MFE)by different surgical approaches.METHODS: A total of 126 patients(126 eyes)performed cataract combined with intraocular lens(IOL)implantation were selected and divided into 3 groups according to different surgical approaches and axial length. Group A included 42 patients(42 eyes)who had cataract combined with high myopia and were performed cataract phacoemulsification combined with IOL implantation; Group B included 42 patients(42 eyes)who had cataract combined with high myopia and underwent small incision cataract extracapsular extraction combined with IOL implantation; Group C included 42 patients(42 eyes)who had cataract with normal axial length and underwent cataract phacoemulsification combined with IOL implantation. Then, the ACD, AL, K value and visual acuity of the three groups at 1d before operation and 3mo after operation were measured, and statistical analysis was performed.RESULTS: The differences in the mean values of preoperative and postoperative changes in ACD(△ACD)and AL(△AL)between groups A and B showed no statistical significance. The differences in the mean values of △ACD and △AL between groups A and C and groups B and C were both statistically significant(both P<0.01). △ACD and △AL in all three groups showed positive correlation(rA=0.855, rB=0.856, rC=0.639, all P<0.05). Furthermore, preoperative AL, △AL, △ACD and MFE in all three groups showed positive correlation(rA=0.874, 0.877, 0.858, rB=0.875, 0.879, 0.858, rC=0.428, 0.766, 0.862, all P<0.05). The standardized regression coefficients of groups A and B were △AL>△ACD(1.32 and 1.31 times), and the standardized regression coefficients of group C were △ACD>△AL(1.66 times).CONCLUSION: Different surgical procedures had no significant effect on the postoperative K value and MFE in patients with high axial myopia combined with cataract, of which main influencing factor is the change in the AL before and after surgery. The postoperative MFE in patients with normal AL was more due to the change of ACD.

OBJECTIVE@#To delineate the origin and content of a mosaicism small supernumerary marker chromosome (sSMC) in a fetus with combined chromosomal karyotyping, chromosomal microarray analysis (CMA) and fluorescence in situ hybridization (FISH).@*METHODS@#The fetus of a 31-year-old pregnant woman who had presented at the Maternal and Child Health Care Hospital of Longhua District of Shenzhen City in 2022 was selected as the study subject. Non-invasive prenatal testing suggested that the fetus has harbored a 8.75 Mb duplication in 4q12q13.1. With informed consent, amniotic fluid and peripheral blood samples were taken from the couple for chromosomal karyotyping analysis. The origin and content of a sSMC was identified by CMA, and its proportion in amniotic fluid was determined with a FISH assay.@*RESULTS@#The karyotypes of the pregnant woman, her husband and the fetus were respectively determined as 46,XX, 46,XY,inv(9)(p12q12), and 47,XY,inv(9)(p12q12)pat,+mar[75]/ 46,XY,inv(9)(p12q12)pat[25]. CMA test of the amniotic fluid sample was arr[hg19]4p11q13.1(48978053_63145931)×3, which revealed no mosaicism. However, FISH analysis showed that 59% of interphase cells from the cultured amniotic fluid sample had contained three signals for the centromere of chromosome 4, whilst 65% of interphase cells from the re-sampled amniotic fluid had three such signals, which confirmed the existence of trisomy 8 mosaicism.@*CONCLUSION@#Chromosomal structural abnormality combined with mosaicism can be delineated with combined chromosomal karyotyping and molecular techniques such as FISH and CMA, which has enabled more accurate counseling for the family.

ABSTRACT

We report a case of giant hysteromyoma and complex pelvic adhesion treated by robotic assisted laparoscopic total hysterectomy and bilateral salpingectomy. The patient was diagnosed with uterine fibroids after physical examination in 1998 but did not receive any treatment, and regular examinations reported progressive growth of the fibroids. Ultrasound suggested multiple uterine fibroids, and pelvic MRI indicated large uterine fibroids with bleeding. Robot-assisted laparoscopic total hysterectomy and bilateral salpingectomy were performed after relevant examinations, and the operation was completed smoothly. The patient was discharged 4 days after surgery with good appearance of the abdominal wall and good recovery during the follow-up. With its unique advantages, robot-assisted laparoscopy provides a minimally invasive surgical approach for giant hysterectomy with complex pelvic adhesions.

ABSTRACT

Objective:To screen the active components of sovereign medicinal Achyranthis Bidentatae Radix in Rongjin Niantong formula based on bioinformatics and network pharmacology and observe their effects on therapeutic targets of osteoarthritis (OA) in <italic>in vivo</italic> and <italic>in vitro</italic> animal experiments. Method:The main active components and therapeutic targets of Achyranthis Bidentatae Radix were retrieved from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP), and the differentially expressed genes relevant to OA from the Gene Expression Omnibus (GEO) database for cross analysis. The effects of main active components in Achyranthis Bidentatae Radix on enriched therapeutic targets of rats with OA <italic>in vivo </italic>and <italic>in vitro</italic> were detected by real-time polymerase chain reaction (Real-time PCR) and Western blot. Result:There were 20 active components for Achyranthis Bidentatae Radix against OA, with quercetin being an important one. Among the three target genes, osteopontin (OPN) and plasminogen activator inhibitor-1(PAI-1) were the key ones in the network. Gene Ontology (GO) analysis yielded 227 related terms, involving the regulation of physiological response to trauma (GO: 1903034), negative regulation of trauma response (GO: 1903035), etc. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis revealed 12 related pathways, involving extracellular matrix receptor interaction (hsa04512) and so on. In animal experiments, compared with the normal group, the model group exhibited increased gene and protein expression of OPN and PAI-1. Compared with the model group, the quercetin group displayed decreased gene and protein expression of OPN and PAI-1 (<italic>P</italic><0.05). In cell experiments, the OPN and PAI-1 protein expression levels in the model group were increased as compared with those in the normal group, while the Collagen Ⅱ protein expression was decreased. The OPN and PAI-1 protein expression levels in the quercetin group and the inhibitor group were down-regulated in contrast to those in the model group, whereas the Collagen Ⅱ protein expression levels were up-regulated significantly (<italic>P<</italic>0.05). Conclusion:Achyranthis Bidentatae Radix<italic> </italic>inhibits cartilage degeneration and exerts the preventive and therapeutic effects against OA, which is possibly due to the efficacy of its active component quercetin in down-regulating the expression of OPN and PAI-1 in chondrocytes and up-regulating the Collagen Ⅱ protein expression.

ABSTRACT

Rehabilitation is an essential treatment for knee osteoarthritis. Rehabilitation evaluation is the foundation of rehabilitation treatment. The items of rehabilitation evaluation for knee osteoarthritis include pain, proprioception, range of motion, muscle force, balance, gait, quality of life, and so on. When evaluating the patient's pain, researchers should not only focus on Visual Analogue Score (VAS) outcomes, but also consider the patient's body function and activity level, and whether there is neuropathic pain. Furthermore, isokinetic dynamometer can not only be used for quantitative assessment for muscle strength, but also for the quantitative evaluation for patient's proprioception. Moreover, Community Balance and Mobility Scale has no ceiling and floor effects which also is a reliable and useful tool for assessing the balance function in patients with knee osteoarthritis, compared with Berg Balance Scale. The evaluation for body structure in patients with knee osteoarthritis mainly includes the radiological examination and physical examination. There are a lot of scales to evaluate the quality of life in patients with knee osteoarthritis, and Osteoarthritis of Knee and Hip Quality of Life fits most. Further researches and promotion of evaluation tools for knee osteoarthritis are needed in the future.

ABSTRACT

Hirayama disease(HD),also known as juvenile muscular atrophy of the distal upper extremity,is featured by concealed onset clinically and often occurs in male adolescence.In most cases,one or both of the asymmetrical distal muscles of the upper extremities are weak and atrophied without objective sensory abnormalities and this disease naturally stops developing after several years.Early Hirayama disease is similar to motor neuron diseases in clinical symptoms like amyotrophic lateral sclerosis.The limitation of medical diagnostic techniques and the lack of understanding by clinicians lead to a higher rate of misdiagnosis of Hirayama disease,making their treatment plan and prognosis completely different.Therefore,a timely and accurate diagnosis of Hirayama disease is essential.In recent years,domestic and foreign scholars conducted researches in the diagnosis of Hirayama disease mainly through clinical manifestation combined with the magnetic resonance imaging of the cervical flexion position and the application of various neurophysiological detection techniques.As the number of clinical cases gradually increases,Hirayama disease draws more attention of clinicians and there are more and more reports and researches on related cases.With the development of research and application of imaging and neuroelectrophysiological techniques,the importance and effectiveness of early diagnosis and treatment of the disease have been affirmed and emphasized,thus people acquire a better understanding of its pathogenesis,clinical diagnosis and treatment.Especially for those suspected patients with juvenile muscular atrophy of the distal upper extremity,it is more important to be vigilant against the possibility of Hirayama disease.Based on this reality,this paper will discuss the new understanding and research progress of pathogenesis,clinical diagnosis and treatment of Hirayama disease,which is very important for clinicians to better understand and pay more attention to this disease.

ABSTRACT

AIM:To investigate whether ellagic acid (EA) attenuates hypoxic-ischemic encephalopathy (HIE) by down-regulating autophagy.METHODS:In vivo, Sprague-Dawley rats (n=17) were randomly divided into 3groups:5 rats for sham group, 6 rats for HIE group and 6 rats for HIE+EA pretreatment group.The rats in HIE+EA pretreatment group were treated with EA (10 mg/kg, 10 m L/kg, suspended in corn oil, ig).After 24 h of operation, the rats from each group were sacrificed and their brains were collected.TTC staining and HE staining were used to define the infarct areas and brain structure.The autophagy-related proteins beclin-1, P62, LC3-II/-I and Atg5 in the cortex in each group were compared by Western blot.In vitro, PC12 cells were divided into 3 groups:control group, Coand CoEA pretreatment group.Co800μmol/L was added to the PC12 cells to induce an anoxic environment.The PC12 cells were pretreated with EA at 8μmol/L and the cell viability was measured by CCK-8 assay.The production of reactive oxidative species (ROS) in the cells was detected by flow cytometry with DCFH-DA staining.MDC staining and TM-RE staining were applied to reflect the extent of autophagy and the state of apoptosis, respectively.The autophagy-related proteins in PC12 cells were also investigated.RESULTS:In HIE group, 7-day-old rats were given the operations and the their large infarct areas in the hemisphere were observed by TTC staining.HE staining displayed the injured hemispheres which contained few neurons, and exhibited edema status and serious structural damage.EA pretreatment decreased the infarct area and alleviated the damage to hemisphere with more visible neurons, compared with HIE group.Compared with sham group, the levels of autophagy-related proteins Atg5, beclin-1 and LC3-II/-I in the cortex were increased (P<0.01) , and P62 protein expression was decreased (P<0.01) in HIE group.Compared with HIE group, the protein expression of Atg5, beclin-1 and LC3-II/-I was decreased (P<0.01) and P62 protein expression was increased in HIE+EA pretreatment group (P<0.01).In vitro, compared with CoPC12 cells in CoEA pretreatment group showed a lower ROS level.Moreover, the cells in CoEA pretreatment group exhibited higher mitochondrial membrane potential than that in CoMDC staining in Coshowed high value of fluorescence and increased number of autophagosomes.EA pretreatment reduced the number of autophagosomes and the extent of autophagy to protect PC12cells.Furthermore, the protein levels of Atg5, beclin-1 and LC3-II/-I in Cowere higher (P<0.01) , and the protein expression of P62 was lower (P<0.01) than those in control group.In CoEA pretreatment group, the protein levels of Atg5, beclin-1 and LC3-II/-I were decreased (P<0.01) and the protein expression of P62 was increased as compared with Co (P<0.01).CONCLUSION:EA pretreatment attenuates autophagy to protect the neurons against HIE injury.

ABSTRACT

Cervical spondylotic amyotrophy (CSA) has attracted more and more attention in recent years, according to early studies, it is based on cervical degeneration, and mainly characterized by segmental, asymmetric and stable muscular atrophy of upper extremities, with or without mild sensory abnormalities and spinal cord lesions (manifested as lower extremity symptoms and gait abnormalities), also known as Keegan′s cervical spondylosis. This review will focusing on recent research progress of CSA, and summarize and analyze the definition, pathogenesis, clinical manifestations, diagnosis and treatment of cervical spondylosis muscular atrophy.

ABSTRACT

Objective:To evaluate and compare the identification of several DNA barcoding candidate sequences on Illicium difengpi and its fake I. jiadifengpi. Method:Samples from different origins of I. difengpi and I. jiadifengpi, were collect extraction of total DNA,nuclear gene ITS2 sequence,chloroplast rbcL,matK gene sequence were selected for PCR amplification,product purification and sequencing,and CondonCode Aligner V3.7.1 was used to proofread stitching. Result:PCR amplification and sequencing of rbcL sequences of I. difengpi and I. jiadifengpi were not satisfactory. It is assumed that their rbcL sequences were too long with slow evolution,which was unsuitable for interbreeding. The success rate of matK sequencing of I. difengpi and I. jiadifengpi was 0 and 76.8%,which may be because primer standards were different for matK sequences of different groups. The results of PCR amplification and sequencing of ITS2 on I. difengpi and I. jiadifengpi were successful,with the success rate of sequencing was 89.3% and 91.2%. Analysis sequencing results, the total length of ITS2 sequences was 268 bases,and there were 2 variation sites of I. difengpi. The total length of ITS2 sequences was 430 bases,and there were 4 or 3 variation sites of I. jiadifengpi. It shows that ITS2 sequences of I. difengpi and I. jiadifengpi were short and has obvious variability and can be amplify,that ITS2 sequence was better than rbcL and matK sequence in molecular identification of I. difengpi and I. jiadifengpi. Conclusion:DNA barcoding based on ITS2 sequence was a powerful and efficient tool for identification of I. difengpi and its fake I. jiadifengpi.

[Objective]To investigate the value of deep common iliac region lymphadenectomy in treatment of uterine malignancies.[Methods]88 patients with deep common iliac region lymphadenectomy in Third affiliated hospital of sun yat-sen university from March 2016 to June 2017 were enrolled in this study. The number of removed lymph nodes、metastatic lymph nodes、operative complication and so on were analyzed.[Results]The number of pelvic lymph nodes resection is from 18 to 52,the average is 26;the number of deep common iliac region lymph nodes resection is from 2 to 16,the average is 5. The number of pelvic lymph node metastasis is 11 and the rate is 12.5%;the number of deep common iliac region lymph node metastasis is 3,accounting for 27.3% of the total number of pelvic lymph node metasta-sis,which alone deep common iliac region lymph node positive in 1 case,9.0% of the total number of pelvic lymph node metastasis.The average operation time of single pelvic lymph node resection was 43 min and that of single deep common iliac region lymph nodes was 10 min.There was no severe complication.[Conclusion]Deep common iliac region lymphad-enectomy is feasibility and safety,27%of patients with pelvic lymph node metastasis has positive deep common iliac region lymph nodes,and we believe that the promotion and application of deep common iliac region lymphadenectomy can further improve the therapeutic effect of uterine malignancy.

ABSTRACT

<p><b>OBJECTIVE</b>Transplantation of adipose-derived stem cells (ADSCs) is associated with potential risks of late complications including tumorigenesis due to the active proliferation of the cells. We aimed to test the effect of transplantation of ADSCs with suppressed proliferation by gamma irradiation in the treatment of thin endometrium in rats.</p><p><b>METHODS</b>ADSCs were isolated from female SD rats and identified by detecting the surface antigens with flow cytometry. After exposure to gamma irradiation at 0, 5 Gy and 10 Gy, the cells were examined for changes in colony-forming ability. Twenty-four female rats with chemically induced thin endometrium were randomized into 4 equal groups and at 6-8 h after modeling, the rats received intrauterine injection of non-irradiated ADSCs (group I), 5 Gy irradiated ADSCs (group II), 10 Gy irradiated ADSCs (group III), or PBS only (group IV). Endometrial pathology was analyzed with HE staining in these rats in the third estrus phase following the cell transplantation.</p><p><b>RESULTS</b>The ADSCs showed a complete loss of proliferative capacity after exposure to 10 Gy irradiation. After the cell transplantation, the endometrium thickness was thicker in group I and II than in group IV (P<0.01), but there was no significant difference between groups III and IV.</p><p><b>CONCLUSIONS</b>Gamma irradiation impairs the proliferative capacity of ADSCs in vitro. Exposure to 10 Gy irradiation causes a total loss of proliferation capacity of the ADSCs, which have no therapeutic potential; 5 Gy irradiation causes partial loss of proliferation capacity of the cells, which still retain the activity to promote endometrial cell regeneration.</p>

ABSTRACT

Twenty-eight compounds were isolated and purified from Clinopodium chinense by Sephedax LH-20, ODS, MCI and preparative HPLC. Their structures were identified as apigenin (1), apigenin-7-O-β-D-glucopyranoside (2), apigenin-7-O-β-D-glucuronopyranoside (3), thellungianol (4), apigenin-7-O-β-D-rutinoside (5), luteolin (6), luteolin-4'-O-β-D-glucopyranoside (7), apigenin-7-O-β-D-pyranglycuronate butyl ester (8), luteolin-7-O-β-D-rutinoside (9), luteolin-7-O-β-D-noehesperidoside (10), acacetin (11), acacetin-7-O-β-D-glucuronopyranoside (12), buddleoside (13), naringenin (14), pruning (15), nairutin (16), isosakuranetin (17), isosakuranin (18), didymin (19), hesperidin (20), kaempferol (21), quercetin (22), kaempferol-3-O-α-L-rahmnoside (23), p-hydroxycinnamic acid (24), caffeic acid (25), cis-3-[2-[1-(3,4-dihydroxy-phenyl)-1 -hydroxymethyl]-1,3-ben-zodioxol-5-yl]-(E)-2-propenoic acid (26), mesaconic acid (27), gentisic acid 5-O-β-D-(6'-salicylyl)-glucopyranoside (28). Among them, compounds 7, 9-10, 12, 23, 26-28 were isolated from the Clinopodium for the first time. The protective effects of compounds 1-6, 8-17 and 19 against H2O2-induced H9c2 cardiomyocyte injury were tested, compounds 15 exhibited significantly protective effects. Compared with the cell viability of (62.12±6.18)% in the model, pruning exhibited viabilities of (84.25±7.36)% at 25.0 mg•L⁻¹, respectively, using quercetin as a positive control [cell viability of (84.55±8.26)%, 20 mg•L⁻¹].

ABSTRACT

Objective To compare the clinical effect of pantoprazole and omeprazole in the treatment of elderly patients with peptic ulcer.Methods 200 patients with peptic ulcer were selected as study subjects,and they were randomly divided into two groups.The control group (100 cases) was orally given clarithromycin,amoxicillin and omeprazole triple therapy.The observation group (100 cases) was given clarithromycin,amoxicillin and pantoprazole triple therapy.The clinical efficacy,clinical symptoms disappeared time and adverse reactions occurred in the two groups were compared.Results The total effective rate of the observation group was 96%,which of the control group was 94%,the difference between two groups was not significant (x2 =0.421,P > 0.05).The disappear time of abdominal pain,belching,burning and bleeding in the observation group after treatment were significantly shorter than those of the control group(t =35.545,56.680,98.896,61.109,all P < 0.05).The incidence rate of adverse reaction of the observation group(5.0%) had no significant difference compared with the control group(7.0%) (x2 =0.354,P > 0.05).Conclusion There is no significant difference in efficacy and safety between pantoprazole and omeprazole in the treatment of elderly patients with peptic ulcer,but the effect of pantoprazole on relieving symptoms is better than omeprazole.

Aim To observe the protective effect of epigallocatechin-3-gallate (EGCG) on hypoxia/reoxygenation (H/R) injury of cardiac myocytes and its mechanisms.Methods H9c2 cardiac myocytes were cultured in vitro and randomly divided into five groups:normal group(N group),H/R group,EGCG low dose group (L group),EGCG medium dose group (M group),and EGCG high dose group(H group).The cardiomyocyte H/R injury model was established and EGCG was pretreated.Cell survival rate was tested by CCK-8 method.The cell apoptotic rate was detected using Annexin V-FITC/PI double staining.The contents of total antioxidant capacity(T-AOC) and tumor necrosis factor α(TNF-α) in cell culture medium were tested according to the kit instructions.The protein expression of Akt and p-Akt was observed using Western blot,while the gene expressions of PI3K,Akt,caspase3 were detected by using fluorescence quantitative PCR method.Results Compared with model group,EGCG increased cell survival rate and reduced the apoptosis after H/R injury.Meanwhile,pretreatment EGCG improved the activity of T-AOC,reduced the level of TNF-α,up-regulated the expression of PI3K,Akt and p-Akt,and down-regulated the expression of caspase3.Conclusion EGCG reduces apoptosis and protects cardiac myocytes by influencing PI3K/Akt signal path

ABSTRACT

· AlM: To investigate clinical therapeutic effect and safety of small incision extracapsular cataract extraction combined with intraocular lens implantation in blindness prevention and treatment. · METHODS: Clinical data of 425 patients with cataract (425 eyes) were analyzed retrospectively, who received small incision extracapsular cataract extraction combined with intraocular lens implantation in the No.413 Hospital of Chinese PLA, with the help from “handicapped rehabilitation engineering in Dinghai District of Zhoushan City” from September 2013 to August 2014.Visual acuity before and after operation, average corneal curvature, corneal astigmatism and intraoperative and postoperative complications were compared statistically and analyzed emphatically. ·RESULTS: ln all of the 425 patients with cataract (425 eyes) , the preoperative best corrected visual acuity of 99 patients were less than 0.05, and the other 326 patients were 0.05 to 0.3.Classification of lens nucleus hardness:level Ⅲ, 63 cases; level Ⅳ, 257 cases; and level Ⅴ, 105 cases.The preoperative average corneal curvature and corneal astigmatism of all patients were 44.6 ±1.52D and 1.35±0.96D.All the 425 patients underwent small incision extracapsular cataract extraction combined with intraocular lens implantation.Postoperative follow-up of 3mo results: the best corrected visual acuity: 5 cases were less than 0.05 (blindness-free rate 98.8%);8 cases were 0.05 to · CONCLUSlON: lt is effective and safe to apply small incisionextracapsular cataract extraction combined with intraocular lens implantation in blindness prevention and treatment, and it gives excellent visual rehabilitation to cataract patients.Especially in those areas and hospitals having no condition of phacoemulsification, it’s a safe and effective alternative when carrying out the program of blindness prevention and treatment.

ABSTRACT

Objective: To investigate the effect of acetyltransferase inhibitor Garcinol on estrogen-induced proliferation, cell cycle promotion and apoptosis inhibition of human breast cancer MCF-7 cells and the related mechanism. Methods: The effect of Garcinol on 17β-estradiol (17β-E2)-induced proliferation of MCF-7 cells was investigated using CCK-8 assay, and the optimal concentration of Garcinol was determine according to the inhibition rate. The cell cycle and apoptosis were analyzed by flow cytometry; the nuclear translocation of NF-κB/p65 was examined by immune cell fluorescence. RT-PCR was used to determine the expressions of cyclin D1, Bcl-2 and Bcl-xL mRNA in MCF-7 cells; and the expressions of ac-H3, ac-H4, NF-κB/ac-p65, cyclin D1, Bcl-2, and Bcl-xL protein were determined by Western blotting analysis. Results: Acetyltransferase inhibitor Garcinol inhibited 17β-E2-induced proliferation of MCF-7 cells, arrested MCF-7 cell cycle at G0/G1 phase, and increased the cell apoptosis(P0.05). 17β-E2-induced nuclear translocation of NF-κB/p65 in MCF-7 cells was also significantly inhibited by Garcinol (P<0.01). Garcinol also significantly inhibited 17β-E2-induced transcription and protein expression of cyclin D1, Bcl-2 and Bcl-xL mRNA in MCF-7 cells (P<0.05). Conclusion: 17β-E2-induced proliferation and apoptosis inhibition of MCF-7 cells are associated with elevated acetylation level of histone and nonhistone NF-κB/p65, and acetyltransferase inhibitor Garcinol may inhibit the effect of 17β-E2 by decreasing acetylation, probably through decreasing ac-p65 expression of NF-κB pathway, and subsequently down-regulating cyclin D1,Bcl-2,and Bcl-xL.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the significance of determining ascitic bacterial 16S rRNA by quantitative PCR combined with microarray (PCR-microarray) in the diagnosis of spontaneous bacterial peritonitis (SBP).</p><p><b>METHODS</b>Ascitic bacterial 16SrRNA was determined by real time fluorescent quantitative PCR-microarray in 76 cases of suspected SBP and 6 cases of non-infectious ascites with chronic liver diseases. The results were compared with ascitic bacterial culture simultaneously.</p><p><b>RESULTS</b>Of 76 ascitic samples, 17 were detected bacteria positive by PCR-microarray, including 8 Grams positive(G+) and 9 Grams negative(G-), which was higher than that by bacterial culture which had only 6 ascitic samples detected positive (all G-); the positive rates were 22.4% vs 7.9%, respectively (P < 0.01). The bacterial strains detected by both methods in 6 cases had a consistency with each other. No bacteria were detected in another 6 cases of non-infectious ascites with chronic liver diseases.</p><p><b>CONCLUSIONS</b>Determination of ascitic bacteria 16S rRNA by PCR-microarray has a higher specificity and sensitivity in the diagnosis of SBP as compared with the bacteria culture. Application of this novel method can not only accelerate SBP diagnosis but also stratify the different pathogens.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the expression of galectin-1 in oral squamous cell carcinoma(OSCC) and its clinical significance.</p><p><b>METHODS</b>Detection of the mRNA and protein expression of galectin-1 in the in vitro cellular carcinogenesis model of OSCC, OSCC cell lines and tissue specimens from 30 primary OSCC patients were performed using real-time polymerase chain reaction (PCR), Western blotting and immunohistochemistry, respectively.</p><p><b>RESULTS</b>The value of galectin-1 mRNA and protein level in human immortalized oral epithelia cell (HIOEC) cell was 0.071 ± 0.023, 0.118 ± 0.046, Compared with the HIOEC, galectin-1 mRNA level and protein expression were increased significantly in all the cell lines (0.141 ± 0.049, 0.504 ± 0.33) (P < 0.01). The levels of mRNA and protein expression of galectin-1 were significantly higher in the cancerous tissue (0.059 ± 0.034, 1.5 ± 0.68) than in the normal adjacent tissues (0.029 ± 0.012, 0.4 ± 0.56) (P < 0.01).</p><p><b>CONCLUSIONS</b>The expression of galectin-1 gene up-regulated in carcinogenesis process of OSCC significantly may be related to the tumorigenesis and development of OSCC, which illustrates its potential clinical application as tumor marker for early diagnosis.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the expression of connective tissue growth factor (CTGF) in pancreatic cancer and its influence on the proliferation and migration of cancer cells.</p><p><b>METHODS</b>The expression of CTGF in pancreatic cell line PANC-1 cells was analyzed by real-time PCR and in pancreatic carcinoma (50 cases) tissues by immunohistochemistry. The ability of proliferation and migration in vitro of PANC-1 cells was tested by MTT assay, scratch test and Boyden chamber test after the CTGF gene was overexpressed by Ad5-CTGF or silenced with Ad5-siCTGF transfection.</p><p><b>RESULTS</b>CTGF was overexpressed in both pancreatic cancer cells and tissues. Overxpression of CTGF leads to increased proliferation and migration of PANC-1 cells. The CTGF-transfected PANC-1 cells showed apparent stronger proliferation ability and scratch-repair ability than that of empty vector controls. The results of Boyden chamber test showed that there were 34 cells/field (200× magnificantion) of the CTGF-transfected overexpressing cells, much more than the 11 cells/field of the empty vector control cells; and 6 cells/microscopic field of the Ad5-siCTGF-transfected silenced cells, much less than the 15 cells/field of the control cells.</p><p><b>CONCLUSIONS</b>CTGF is overexpressed in both pancreatic cancer cells in vitro and in vivo, indicating that it may play an important role in the cell proliferation and migration in pancreatic cancer.</p>
