ABSTRACT

In previous studies, we found interferon-a (IFN-a) could reduce protein levels of p11, 5-hydroxytryptamine receptor 1b(5-HT1b) and 5-hydroxytryptamine receptor 4 (5-HT4), but does not influence their messenger RNA levels in SH-sy5ycells. Thus, we investigated the post-transcriptional modulation of these molecules by IFN-a. SH-sy5y cells were treatedwith IFN-a, NH4Cl or MG132 alone or in combination, and then the protein levels of p11, 5-HT1b and 5-HT4 wereanalyzed by western blots. The regulatory effects of p11 on 5-HT1b and 5-HT4 were also determined in p11 knock-downcells. NH4Cl but not MG132 could reverse the protein level of p11 in IFN-a-treated SH-sy5y cells. MG132 could recoverthe protein levels of 5-HT1b and 5-HT4 in p11 knock-down cells. The down-regulation effects of IFN-a on p11, 5-HT1band 5-HT4 were associated with the lysosome and ubiquitin–proteasome-mediated pathways. p11 was identified as a potentregulator to modulate the ubiquitination of 5-HT1b and 5-HT4. Therefore, it could be potential target therapies in IFN-ainduced depression.

ABSTRACT

Objective:To explore the influence of artificial luminous environment and preventive function of traditional Chinese medicine (TCM) intervention based on “Theory of yin-yang clock” on myopia. Methods:A total of 45 New Zealand young rabbits were randomly divided into 5 groups, 9 for each group. Control group was exposed in natural light. Fluorescent group and full spectrum group were exposed in fluorescent light and full spectrum light, on which basis fluorescent TCM group and full spectrum TCM group were added with “Rizhong Yinyang Formulas”, respectively. Optical parameters were measured and the influence of different lights on the serum and retinal dopamine (DA) levels as well as the retinal histopathological tissues was observed. Results: The spectrum of fluorescent light mainly focused at 420-490 nm with the peak value of wavelength near 450 nm, whereas that of full spectrum was wider (400-800 nm) with the peak value near 600 nm. After 4 and 12 weeks, fluorescent group was evidently lower in serum and retinal DA levels (P0.05). Histopathological observation showed that there was significant difference in pigment epithelium layer, photoreceptor and nerve fiber layer between fluorescent group and control group, but the difference among the test groups was not significant. Conclusions:Fluorescent light has certain influence on retinal histological construction and visual performance. However, TCM intervention may have some degree of protective function on retina.

ABSTRACT

To introduce the application of Stata software to heterogeneity test in meta-analysis.A data set was set up according to the example in the study, and the corresponding commands of the methods in Stata 9 software were applied to test the example. The methods used were Q-test and Ⅰ2statistic attached to the fixed effect model forest plot,H statistic and Galbraith plot. The existence of the heterogeneity among studies could he detected hy Q-test and H statistic and the degree of the heterogeneity could be detected by,Ⅰ2 statistic. The outliers which were the sources of the heterogeneity could be spotted from the Galbraith plot.Heterogeneity test in Meta-analysis can be completed by the four methods in Stata software simply and quickly.H and Ⅰ2 statistics are more robust, and the outliers of the heterogeneity can be clearly seen in the Galbraith plot among the four methods.
