ABSTRACT

Objective:This paper mainly discusses the expression of hsa_circ_002082 in breast cancer and its clinical significance to breast cancer.Methods:The up-regulated hsa_circ_002082 in breast cancer was screened out through the bioinformatics website, and verified by quantitative reverse transcription PCR (qRT-PCR). The differential expression of hsa_circ_002082 in different clinical index groups was analyzed by one-way ANOVA and t-test, and the ROC curve was used to analyze the value of hsa_circ_002082 in diagnosing breast cancer or identifying clinical indexes.Results:Compared with para-cancerous tissue (1.00±0.21), the expression of hsa_circ_002082 in cancer tissue (1.34±0.25) was higher than that in peripheral blood of healthy people (1.00±0.26), and the expression of hsa_circ_002082 in peripheral blood of breast cancer patients (1.39±0.24) increased (all P<0.05). The AUC value of hsa_circ_002082 for breast cancer diagnosis was 0.8520 ( P<0.05). There were differences in the expression of hsa_circ_002082 in tumor grade, estrogen receptor (ER) positive, carcino-embryonic antigen (CEA) positive, Ki-67 proliferation index, tumor metastasis, and TNM grade (all P<0.05). The relative expression level of hsa_circ_002082 was significantly positively correlated with the level of tumor marker CEA ( r=0.368, P=0.009). The AUC values of hsa_circ_002082 for distinguishing tumor grade, ER positive, CEA positive, Ki-67 proliferation index, tumor metastasis, and TNM grade were 0.8744, 0.7005, 0.7890, 0.8075, 0.8317, and 0.8301, respectively (all P<0.05) . Conclusion:hsa_circ_002082 is highly expressed in breast cancer tissue and peripheral blood, and hsa the potential to be a biomarker for the diagnosis of breast cancer.

ABSTRACT

Objective To evaluate the teaching activity on public health courses from clinical medical students in our university in order to provide a scientific basis for improving the curriculum design and teaching reform. Methods The “Questionnaire on Teaching Evaluation in Public Health Courses”, including teaching attitude, teaching content, teaching methods and teaching effectiveness was designed, and a general investigation was conducted among the clinical medical students of five-year program (840 students) and eight-year program (278 students) in these three aspects to under-stand students' evaluation to the course, who had finished the public health courses, including Preven-tive Medicine, Medical Statistics and Epidemiology (hereinafter referred to as: statistics, epidemiology, prevention) in Sun Yat-sen University. Statistical analysis was made using SPSS 13.0 software. Data analysis methods contain descriptive analysis, T-test, ANOVA, LSD, SNK, hierarchical logistic regres-sion analysis, etc. Results The overall score of teaching evaluation is (4.04±0.60). Differences exist between the evaluation in the five-year medical students and the eight-year medical students. The P values were 0.000 (Medical Statistics), 0.269 (Epidemiology), 0.047 (Preventive Medicine). The com-parison of scores among the four dimensions shows: Teaching effectiveness β' effectiveness. Conclusions Clinical Medical students' overall evaluation on the public health courses offered by this university was good. Teaching effectiveness and teaching methods still need improvement. Teaching contents are the most influential factor of overall teaching satisfaction, followed by teaching effectiveness.

ABSTRACT

This study is designed to obtain recombinant human acetylcholinesterase (rhAChE) and apply it in screening acetylcholinesterase inhibitors. The rhAChE was overexpressed in HEK293 cells transfected by plasmid of pCMV-AChE with the cationic liposome and rhAChE was found to be secreted into cell culture medium. AChE activity was assayed according to modified Ellman method to obtain kinetic parameters. IC so50 values for donepezil compounds of rhAChE were calculated to determine their activities of inhibition. The results showed that Km value was 151.9 micromol.L-1 donepezil inhibited rhAChE in a mixed competitive-noncompetitive way (Ki= 16.03 nmol.L-1, Ki = 18.36 nmol.L-1) and that most new compounds tested exhibited high activities of inhibition on rhAChE. The study suggests that rhAChE is available to be applied in screening AChE inhibitors in vitro.

Based on the concept of general education in higher education,Zhongshan school of medicine of Sun Yat-sen University launched general education course-‘ basic medicine introduction' to all non-medical undergraduates.Teaching contents,teaching methods and teaching effects of this course were explored and evaluated.By introducing basic medicine,the overall objective is to guide students to consciously maintain the mental and physical health and to stimulate students' thinking on the meaning of life.

ABSTRACT

Objective To detect the fluctuations of homoeysteine(Hcy),endothelin(ET),mononitrogen monoxide(NO),folic acid(FA) and vitamine Bi2 (VitB_(12)) in the plasma of patients suffered from acute cerebral infarction and to investigate the clinical significance.Methods 140 blood specimens from cerebral infarction patients and 61 specimens from healthy individuals were obtained in this study and a series of items of Hey,ET,NO,FA and VitB_(12) were detected.Results The Hey [(17.15±4.63) μmol/L]and ET [(32.62±18.76) μmol/L]were increased in acute cerebral infarction(ACI) group compared with that in control group[(10.12±2.62) μmol/L,(20.08±13.38) μmol/L respectively]and the difference was significant (P<0.01).The levels of NO [(39.82±7.26) μmol/L],FA [(7.54±4.01) ng/ml]and VitB_(12) [(247.70±119.85) pg/ml]were decreased compared with control group [(60.22±6.28) μmol/L,(10.08±3.86) ng/ml,(411.52±194.58) pg/ml respectively]and the differences significant (P<0.01).Gender and hypertension affected the values of Hey.Conclusions Hcy and ET were increased while NO,FA and VitB_(12) were decreased in acute cerebral infarction.Hcy had positive correlation with ET and negative correlation with FA and VitB_(12).
