ABSTRACT

Identifying data-driven biotypes of major depressive disorder (MDD) has promise for the clarification of diagnostic heterogeneity. However, few studies have focused on white-matter abnormalities for MDD subtyping. This study included 116 patients with MDD and 118 demographically-matched healthy controls assessed by diffusion tensor imaging and neurocognitive evaluation. Hierarchical clustering was applied to the major fiber tracts, in conjunction with tract-based spatial statistics, to reveal white-matter alterations associated with MDD. Clinical and neurocognitive differences were compared between identified subgroups and healthy controls. With fractional anisotropy extracted from 20 fiber tracts, cluster analysis revealed 3 subgroups based on the patterns of abnormalities. Patients in each subgroup versus healthy controls showed a stepwise pattern of white-matter alterations as follows: subgroup 1 (25.9% of patient sample), widespread white-matter disruption; subgroup 2 (43.1% of patient sample), intermediate and more localized abnormalities in aspects of the corpus callosum and left cingulate; and subgroup 3 (31.0% of patient sample), possible mild alterations, but no statistically significant tract disruption after controlling for family-wise error. The neurocognitive impairment in each subgroup accompanied the white-matter alterations: subgroup 1, deficits in sustained attention and delayed memory; subgroup 2, dysfunction in delayed memory; and subgroup 3, no significant deficits. Three subtypes of white-matter abnormality exist in individuals with major depression, those having widespread abnormalities suffering more neurocognitive impairments, which may provide evidence for parsing the heterogeneity of the disorder and help optimize type-specific treatment approaches.

ABSTRACT

Since epileptic seizure is unpredictable and paroxysmal, an automatic system for seizure detecting could be of great significance and assistance to patients and medical staff. In this paper, a novel method is proposed for multichannel patient-specific seizure detection applying the earth mover's distance (EMD) in scalp EEG. Firstly, the wavelet decomposition is executed to the original EEGs with five scales, the scale 3, 4 and 5 are selected and transformed into histograms and afterwards the distances between histograms in pairs are computed applying the earth mover's distance as effective features. Then, the EMD features are sent to the classifier based on the Bayesian linear discriminant analysis (BLDA) for classification, and an efficient postprocessing procedure is applied to improve the detection system precision, finally. To evaluate the performance of the proposed method, the CHB-MIT scalp EEG database with 958 h EEG recordings from 23 epileptic patients is used and a relatively satisfactory detection rate is achieved with the average sensitivity of 95.65% and false detection rate of 0.68/h. The good performance of this algorithm indicates the potential application for seizure monitoring in clinical practice.

ABSTRACT

Neurocognitive deficits are frequently observed in patients with schizophrenia and major depressive disorder (MDD). The relations between cognitive features may be represented by neurocognitive graphs based on cognitive features, modeled as Gaussian Markov random fields. However, it is unclear whether it is possible to differentiate between phenotypic patterns associated with the differential diagnosis of schizophrenia and depression using this neurocognitive graph approach. In this study, we enrolled 215 first-episode patients with schizophrenia (FES), 125 with MDD, and 237 demographically-matched healthy controls (HCs). The cognitive performance of all participants was evaluated using a battery of neurocognitive tests. The graphical LASSO model was trained with a one-vs-one scenario to learn the conditional independent structure of neurocognitive features of each group. Participants in the holdout dataset were classified into different groups with the highest likelihood. A partial correlation matrix was transformed from the graphical model to further explore the neurocognitive graph for each group. The classification approach identified the diagnostic class for individuals with an average accuracy of 73.41% for FES vs HC, 67.07% for MDD vs HC, and 59.48% for FES vs MDD. Both of the neurocognitive graphs for FES and MDD had more connections and higher node centrality than those for HC. The neurocognitive graph for FES was less sparse and had more connections than that for MDD. Thus, neurocognitive graphs based on cognitive features are promising for describing endophenotypes that may discriminate schizophrenia from depression.

ABSTRACT

Objective To investigate the effects of four tyrosine metabolites on the proliferation, cell cycle and chemosensitivity of lung cancer cells.Methods A549 cells were cultured with different concentrations of tyrosine metabolites:tyrosine, 4-hydroxyphenylpyruvate(HPPA), homogentisic acid(HGA) and fumaric acid (FA) in vitro.High content imaging analysis system was used to monitor cell proliferation.Flow cytometry was used to detect cell cycle and apoptosis.Protein and mRNA level of Bax and Bcl-2 were measured by Western blot.Results Tyrosine metabolites had no effect on cell cycle and proliferation of A549 cells.HGA decreased the cell apoptosis of A549 cells induced by Gefitinib with depressed Bax expression and elevated Bcl-2 expression.However, FA increased the apoptosis of A549 cells induced by Gefitinib with up-regulation of Bax and down-regulation of Bcl-2.No obvious effect of tyrosine and HPPA was detected on Gefitinib induced cell apoptosis.Conclusion HGA and FA may play an important role in drug sensitivity of lung cancer cells, indicating a critical role of tyrosine metabolism in lung cancer.

ABSTRACT

<p><b>OBJECTIVE</b>To construct a GFP/Puro double-labeled lentiviral expression vector for CK8 silencing and assess the effects of CK8 silencing on cell apoptosis.</p><p><b>METHODS</b>The siRNA sequences of CK8 were inserted into the lentiviral expression vector GV248 and transfected into 293T cells with the packaging plasmids PMD and SPA. The lentivirus was collected at 24 and 36 h post-transfection. Flow cytometry was used to detect the virus titer and the positive cells were selected with puromycin. The knockdown of CK8 was examined by Western blotting. The effect of CK8 down-regulation on cell apoptosis induced by cisplatin was detected with Annexin V/PI staining.</p><p><b>RESULTS AND CONCLUSION</b>We successfully constructed CK8 interference lentiviral vector and obtained a stable cell line with CK8 knock-down that was sensitive to cisplatin-induced apoptosis.</p>

ABSTRACT

Objective To investigate the effect of early training and touching nursing on intelligent and physical development of premature infants. Methods 80 cases of premature infants in our hospital from January to October 2010 were divided into the observation group and the control group with 40 pa tients in each group,the control group was given routine nursing,the observation group was given early training and touching nursing on the basis of the control group.Intelligent and physical development was compared between two groups. Results the mental development index and psychomotor development index on the 6th and 12th months in the observation group were better than those in the control group,the differences were significant.The height,body weight and head circumference on the 12th month in the observation group were significantly better than those in the control group. Conclusions Early training and touching nursing can promote the intelligent and physical development of premature infants significantly as well as improve the prognosis.

ABSTRACT

Sino 0.5 or 2 mg/kg iv reduced significantly heart rate ( HR), systolic blood pressure ( SBP ), diastolic blood pressure ( DBP ), left ventricular systolic pressure ( LVSP) , dP/dtmax, cardiac index ( CI ) and total peripheral resistance ( TPR ) . VCE- + dP/dtmax was unchanged. T value did't change in 0 .5mg/kg, but it elevated in 2 mg/kg.

The effects of Jiaogulanoside on the cardiovascular system were observed anesthetized chest-opened dogs,JS lmg/kg iv significantly increased BP, LVSP and ?dP/dt_(max); 20mg/kg iv significantly decreased HR, BP, LVSP, +dP/dt_(max),CI, V_(CE)-- +dP/dt_(mxa), These results suggested that the changes of BP and cardiac function was varied with the different doses of JS.
