ABSTRACT

Objective To evaluate the relationship between propofol anesthesia and postoperative delirium and inflammatory responses of aged rats.Methods Twenty-four adult male Sprague-Dawley rats,aged 18 months,weighing 550-650 g,were divided into 3 groups (n=8 each) using a random number table method:control group (group C),propofol anesthesia for 2 h group (group P2) and propofol anesthesia for 4 h group (group P4).In P2 and P4 groups,anesthesia was maintained with propofol at a rate of 24 mg· kg-1 · h-1 for 2 and 4 h,respectively,after anesthesia was induced with propofol.Morris water maze test was performed at 1 day before and after anesthesia.Blood and cerebrospinal fluid samples were collected after the end of Morris water maze test for determination of interleukin-1beta (IL-1β),IL-4,IL-6 and tumor necrosis factor-alpha (TNF-α) concentrations in plasma and cerebrospinal fluid by enzyme-linked immunosorbent assay.The hippocampus and cortex were removed for determination of Iba-1 positive cells.Results Compared with group C and the baseline value before anesthesia,no significant change was found in the escape latency or percentage of time of staying at the original platform quadrant in P2 and P4 groups (P>0.05),the concentrations of IL-6 in cerebrospinal fluid and TNF-α in plasma were significantly decreased in group P2,the concentrations of IL-1β in cerebrospinal fluid and IL-4,IL-6 and TNF-α in plasma were significantly increased,and the concentrations of IL-1β in plasma and IL-4 in cerebrospinal fluid were decreased in group P4,and the number of hippocampal Iba-1 positive cells were significantly decreased in P2 and P4 groups (P<0.05 or 0.01).Conclusion Propofol anesthesia dose not induce postoperative delirium and central inflammatory responses within 4 h,and propofol can induce peripheral inflammatory responses when anesthesia time is longer than 2 h.

ABSTRACT

?AIM:To study the changes of ocular surface in cataract patients with Meibomian gland dysfunction ( MGD) who treated with Meibomian gland massage before surgery.?METHODS: Totally 90 patients ( 93 eyes ) with cataract and MGD were randomly divided into two groups. Patients in experimental group were treated with hot compress and Meibomian gland massage every day before surgery, while the patients in the control group were not taken. Two groups of patients received phacoemulsification combined with intraocular lens implantation. Lid margin abnormalities, secretion characteristics, Schimer I test ( SⅠt) and tear film break-up time ( BUT ) were recorded and compared between two groups using slit lamp microscope inspection preoperatively and postoperatively 1wk.?RESULTS:In the control group, the postoperative score of eyelid margin and Meibomian gland secretion was increased significantly than preoperative, while the value of BUT and SⅠt was down significantly. Compared with control group, the postoperative score of eyelid margin shape and Meibomian gland secretion in the experimental group was decreased significantly, while the value of BUT and SIT improved significantly (P<0. 05).?CONCLUSION: Phacoemulsification can aggravate the Meibomian gland dysfunction and have some extent of effects on the ocular surface. Meibomian gland massage before surgery can significantly improve the function of Meibomian gland and the state of ocular surface in patients with MDG.

ABSTRACT

To establish a LC-MS/MS method to determine caffeic acid, chlorogenic acid in rat plasma and study their pharmacokinetics in rats. Six Sprague-Dawley rats were intravenously injected with 4 mL x kg(-1) of Dengzhanxixin injection, respectively. Their drug plasma concentration was determined by LC-MS/MS, with tinidazole as an internal standard. The pharmacokinetic parameters were calculated by DAS 1.0. The linear concentration ranges of caffeic acid, and chlorogenic acid were 2-128 microg x L(-1) (r = 0.998 1) and 3-384 microg x L(-1) (r = 0.998 7), respectively. The methodological test showed conformance to the requirements. The intraday and inter-day variable coefficients were both less than 10.0%, indicating that both of legitimate precise and accuracy were in conformity with the requirements of biological sample analysis. For caffeic acid, the pharmacokinetic parameter t1/2beta AUC0-t, and CL were (130.91 +/- 38.77) min, (4.89 +/- 0.96) mg x min x L(-1) and (0.12 +/- 0.02) L x min(-1) x kg(-1), respectively. For chlorogenic acid, the pharmacokinetic parameter t1/2beta , AUC0-t, and CL were (49.38 +/- 8.85) min, (9.54 +/- 0.95) mg x min x L(-1) and (0.09 +/- 0.003) L x min(-1) x kg(-1), respectively. The LC-MS/MS analysis method established in this study was proved to be so accurate and sensitive that it can be applied to the pharmacokinetic study of caffeic acid and chlorogenic acid.

ABSTRACT

<p><b>OBJECTIVE</b>To study the relations between anthropometric parameters [body mass index (BMI), percent of body fat, waist circumference] and blood lipids of urban children in Beijing and to compare the ability of prediction effect of waist circumference, percent body fat and BMI on lipid profiles.</p><p><b>METHODS</b>852 children aged 9-10 were recruited with multi-stage sampling. Serum lipids of children were measured using automatic biochemical instrument. Fisher's exact test, trend test and median regression were used to analyze the data from 847 subjects with valid information.</p><p><b>RESULTS</b>(1)With the rise of the BMI, percent body fat and waist circumference, total triglycerides (TG), total cholesterol (TC) and low density lipoprotein choleasterol (LDL-C) showed increasing trends while high density lipoprotein choleasterol (HDL-C) had a decreasing trend(P < 0.01). (2)TG was mainly influenced by waist circumference (t = 6.86), sex (t = 3.96) and percent of body fat (t = 1.85); TC was influenced by percent of body fat (t = 5.31); LDL-C was influenced by waist circumference (t = 4.65), height (t = -2.79) and percent age of body fat (t = 2.77); HDL-C was influenced by waist circumference (t = -12.24), sex (1= -5.83).</p><p><b>CONCLUSION</b>With the aggravation of children's fatness (BMI, WC and PBF), serum lipids would increase and WC seemed to be the best predictor for lipids among BMI, WC and PBF.</p>
