ABSTRACT

Objective: To observe the intervention effect of auricular point sticking on dry eye in myopia patients after small-incision lenticule extraction (SMILE) surgery.Methods: This was a prospective randomized controlled study conducted among the myopia patients who received SMILE surgery at Eye and ENT Hospital of Fudan University. The post-SMILE operation patients who screened by the inclusion and exclusion criteria were randomized into a control group and a treatment group. Patients in the control group were given 0.1% fluorometholone and 0.5% carboxymethylcellulose sodium eye drops, while the treatment group was given additional unilateral auricular point sticking for 1 month. The patients were estimated using ocular surface disease index (OSDI), Schirmer tear test-1 (STT-1), tear film break-up time (TF-BUT), corneal fluorescein staining (CFS) score, corneal sensitivity (CS) and visual quality (VQ) at 1 d, 1 week and 1 month after surgery; the changes in anxiety and depression were also observed in the patients. Results: Compared with the first day after operation, CS in the nasal region was improved in the treatment group, and the VQ score increased in the control group patients at 1 week after operation (both P<0.05); at 1 month after operation, the TF-BUT increased, CFS score decreased, CS in the central and nasal regions increased (all P<0.05), and VQ score increased (P<0.01) in the treatment group, and the CS in the central, upper, lower and nasal regions were improved (all P<0.05), and VQ score increased (P<0.01) in the control group. The between-group comparison showed that the differences in the change of TF-BUT were statistically significant at 1 week and 1 month after surgery, respectively (both P<0.05). Conclusion: Auricular point sticking therapy can increase the TF-BUT and accelerate the repair of ocular surface function in post-SMILE patients.

ABSTRACT

PURPOSE@#The increasing number of deaths due to road traffic accidents (RTAs) has attracted global attention. However, the influence of road types is rarely considered in the study of RTAs. This study evaluates the influence of different road types in RTAs in northern Guizhou to provide a basis for the formulation of evidence-based policies and measures.@*METHODS@#We obtained the data from the Zunyi Traffic Management Data Platform for the years 2009-2018. The mortality rates of RTAs were calculated. Descriptive methods and Chi-square tests were used to analyze the characteristics of road traffic collisions on different road types. We also examined the associations between the mortality rate per 10,000 vehicles and the growth of per capital gross domestic product (GDP) with Spearman's rank correlation analysis. According to the passing volume and the infrastructure, we defined different types of roads, like administrative road, functional road, general urban road and urban expressway.@*RESULTS@#In 2012, the traffic mortality rate of administrative roads was 8.9 per 100,000 people, and the mortality rate of functional roads was 7.4 per 100,000 people, which decreased in 2018 to 6.1 deaths per 100,000 people and 5.2 deaths per 100,000 people, respectively. The mortality rate per 10,000 vehicles reached the highest level in 2011 (28.8 per 10,000 vehicles and 22.5 per 10,000 vehicles on administrative and functional roads, respectively). The death rate of county roads was the highest among administrative roads (χ@*CONCLUSION@#Although our research shows that RTAs in northern Guizhou have steadily declined in recent years, the range of decline is relatively small. Many measures and sustainable efforts are needed to control road traffic death and accelerate the progress in road traffic safety in northern Guizhou.

ABSTRACT

Objective:To study the anti-colon cancer effect and mechanism of magnolol analogue CT2-3, in order to lay a foundation for the application of CT2-3 in anti-colon cancer area. Method:Colon cancer cells SW480 and LoVo were cultured in vitro. Different concentrations (10, 20, 40, 80 μmol·L-1) of CT2-3 and magnolol were used to stimulate colon cancer cells for 24, 48 h. The effect of CT2-3 and magnolol on the cell viability of colon cancer cells was detected by cell counting kit (CCK-8). Colony formation assay was used to detect the colony formation capacity of CT2-3 on colon cancer cells. Flow cytometry and Western blot were used to determine the effect of CT2-3 on the apoptosis of colon cancer cells and the expression of DNA damage marker phosphorylated histone H2AX (γH2AX). Reactive oxygen species (ROS) generation was measured by ROS assay kit. Real time quantitative polymerase chain reaction (Real-time PCR) was used to detect the effect of CT2-3 on expressions of mitochondrial apoptosis-related genes B-cell lymphoma-2 (Bcl-2) and Bcl-2-associated X (Bax) in colon cancer cells. Result:The half maximal inhibitory concentration (IC50) of magnolol in two kinds of colon cancer cells after treatment for 24, 48 h were both higher than 80 μmol·L-1. While the IC50 of CT2-3 in SW480 cells after treatment for 24, 48 h were (54.59±1.73) μmol·L-1 and (29.82±1.13) μmol·L-1, respectively. The IC50 of CT2-3 in LoVo cells after treatment for 24,48 h were (66.68±2.11) μmol·L-1 and (46.70±1.81) μmol·L-1, respectively. Compared with the blank group, the colony formation capacity of colon cancer cells in CT2-3 groups (20, 40 μmol·L-1) was significantly decreased in a dose-dependent manner (P<0.01), apoptotic colon cancer cells were significantly increased (P<0.01), relative expression of DNA damage marker γH2AX was significantly increased (P<0.01), ROS was significantly increased (P<0.01). In addition, relative mRNA expression of Bcl-2 was significantly decreased (P<0.01), while relative mRNA expression of Bax was significantly increased (P<0.01). Conclusion:CT2-3 can remarkably inhibit colon cancer cells, and the underlying mechanism might be that CT2-3 promotes mitochondria dysfunction and ROS generation by regulating expressions of mitochondrial apoptosis-related genes, so as to further induce DNA damage and finally lead to apoptosis.

@#AIM:To study the curative efficacy of aspheric intraocular lens implantation after manual nuclear fragmentation in treatment of cataract and its effects on visual acuity. <p>METHODS: Totally 100 patients of cataract who received therapy from May 2013 to May 2016 in our hospital were selected. According to random number table, those patients were divided into the observation group(50 patients 68 eyes)and the control group(50 patients 67 eyes). The observation group was treated with small incision manual discectomy and aspheric intraocular lens implantation, the control group was treated with phacoemulsification cataract extraction and aspheric intraocular lens implantation, the operation effect of two groups were compared. <p>RESULTS: After operation, the corneal endothelial cell counts of the two groups decreased(<i>P</i><0.05); in the observation group, the results were higher than those of the control group at 1 and 3mo after operation(<i>P</i><0.05). In the observation group, the uncorrected visual acuity at 1mo after operation was better than that of the control group(<i>P</i><0.05); there was no significant difference between the two groups in postoperative visual acuity after operation 3mo(<i>P</i>>0.05). At 1mo after surgery, the corneal astigmatism in the observation group was higher than that of the control group(<i>P</i><0.05); there was no significant difference of corneal astigmatism between the two groups at 3mo after operation(<i>P</i>>0.05). The incidence of intraoperative complications and postoperative complications in the observation group were lower than that of the control group(<i>P</i><0.05). <p>CONCLUSION: Aspheric intraocular lens implantation after manual nuclear fragmentation is well for cataract, which can reduce the damage to corneal endothelial cells, the complications rate is low and the postoperative visual acuity recovered well.

ABSTRACT

<p><b>OBJECTIVE</b>To purify a low-temperature hydroxylamine oxidase (HAO) from a heterotrophic nitrifying bacterium Acinetobacter sp. Y16 and investigate the enzyme property.</p><p><b>METHODS</b>A HAO was purified by an anion-exchange and gel-filtration chromatography from strain Y16. The purity and molecular mass were determined by RP-HPLC and SDS-PAGE. The HAO activity was detected by monitoring the reduction of potassium ferricyanide using hydroxylamine as substrate and ferricyanide as electron acceptor. The partial amino acid sequence was determined by mass spectrometry.</p><p><b>RESULTS</b>The low-temperature HAO with a molecular mass of 61 kDa was purified from strain Y16 by an anion-exchange and gel-filtration chromatography. The enzyme exhibited an ability to oxidize hydroxylamine in wide temperature range (4-40 °C) in vitro using hydroxylamine as substrate and ferricyanide as electron acceptor. It was stable in the temperature range of 4 to 15 °C and pH range of 6.0 to 8.5 with less than 30% change in its activity. The optimal temperature and pH were 15 °C and 7.5, respectively. Three peptides were determined by mass spectrometry which were shown to be not identical to other reported HAOs.</p><p><b>CONCLUSION</b>This is the first study to purify a low-temperature HAO from a heterotrophic nitrifier Acinetobacter sp. It differs from other reported HAOs in molecular mass and enzyme properties. The findings of the present study have suggested that the strain Y16 passes through a hydroxylamine-oxidizing process catalyzed by a low-temperature HAO for ammonium removal.</p>
