ABSTRACT

Objective:To investigate the effects of compound fermented milk on intestinal microbiota, short chain fatty acid (SCFA), intestinal motility and mucosal barrier in mice with constipation.Methods:Twenty-seven C57BL/6JNifdc mice were randomly divided into three groups: control group, model group and intervention group. The model group and the intervention group were given loperamide intragastrically for two weeks. Starting from the second week, the intervention group was treated with compound fermented milk for 7 d. The control group was given normal saline. Food intake, water intake, weight change, fecal moisture content, time of first-time black stool and small intestine propulsion rate were detected. Expression of serotonin C receptor (5-HTR2C), zona occludins-1 (ZO-1) and mucin-2 (MUC-2) at mRNA level in colon was analyzed. Western blot was used to measure the expression of Raf/ERK/MAPK-related proteins. SCFA level in intestinal tract was detected by gas chromatography. Intestinal microbiota diversity was analyzed by high-throughput sequencing.Results:Compared with the control group, the first black stool excretion time was significantly prolonged in the model group ( P<0.01). Moreover, fecal moisture content, small intestine propulsion rate and the expression of 5-HTR2C and ZO-1 at mRNA level in colon were significantly decreased ( P<0.01). Compared with the model group, the first black stool excretion time was significantly shortened ( P<0.05); fecal moisture content, small intestine propelling rate ( P<0.05), the expression of colon 5-HTR2C and ZO-1 at mRNA level ( P<0.05), phosphorylation of Raf/ERK/MAPK pathway in the colon, intestinal SCFA-producing bacteria and intestinal SCFA content were increased in the intervention group. Conclusions:Compound fermented milk had a therapeutic effect on constipation in a mouse model by increasing the abundance of SCFA-producing bacteria and SCFA content, enhancing the phosphorylation of the Raf/ERK/MAPK pathway to up-regulate the expression of 5-HTR2C at mRNA level in the colon, and increasing the expression of ZO-1 at mRNA level in the colon. Intestinal peristalsis and intestinal mucosal barrier function were enhanced, thus improving the symptom of constipation.

ABSTRACT

Survivin is a member of inhibitors of apoptosis protein family, compared with other members, survivin has a unique baculovirus apoptosis inhibitory protein—inhibitor of apoptosis proteins (IAP) baculovirus IAP repeat (BIR) and alpha helix molecular structure, determines its important role in the inhibition of cell apoptosis and control in the cell cycle, and is involved in the regulation of cell proliferation and angiogenesis process, and it is closely related to the cell cycle of the IAP family members.In addition, the distribution of survivin in tissues is highly selective.It is highly expressed in malignant tumors and is not expressed in normal adult tissues.Therefore, it is regarded as an important prognostic indicator of malignant tumors.In this paper, the molecular structure, tissue distribution and main functions of survivin gene are reviewed, and the research progress of its relation with ophthalmic diseases, including ocular tumor, ocular surface disease, ocular related immune diseases, lens disease and retinopathy are emphatically expounded.

ABSTRACT

Objective:To evaluate the clinical effect of Huanglian-Wendan Decoction combined with Estazolam tablets in the treatment of post-stroke sleep disorders (PSSD) with syndrome of internal disturbance of phlegm-heat. Methods:A total of 96 patients with PSSD who met the inclusion criteria in the People's Hospital of Shenzhen Luohu District from September 2016 to June 2018 were randomly divided into the control group (47 cases) and the study group (49 cases) by random number table method. The control group took Estazolam tablets based on routine treatment, and the intervention group took Huanglian-Wendan Decoction based on the control group. Both groups were treated for 2 weeks. The TCM syndromes were recorded and scored before and after the treatment. The anxiety symptoms were assessed by Hamilton Anxiety Scale (HAMA) and the depression symptoms were assessed by Hamilton Depression Scale (HAMD) respectively. Sleep quality was assessed by Pittsburgh Sleep Quality Index (PSQI). The level of brain-derived neurotrophic factor (BDNF) was detected by ELISA, the level of 5-hydroxytryp-tamine (5-HT) was detected by high performance liquid-phase electrochemical detector. The adverse reactions were recorded and clinical efficacy was evaluated. Results:The total clinical effective rate of the intervention group was 91.8% (45/49), and 76.6% (36/47) in the control group. There was significant difference between the two groups ( χ2=4.227, P=0.040). After the treatment, the scores of HAMA, HAMD and PSQI in the intervention group were significantly lower than that of the control group ( t value were 4.081, 4.869 and 3.900, respectively, all Ps<0.001). The levels of serum BDNF (42.36 ± 6.78 ng/ml vs. 33.98 ± 6.41 ng/ml, t=6.217), 5-HT (127.21 ± 12.39 ng/ml vs. 101.46 ± 11.70 ng/ml, t=10.460) were significantly higher than that of the control group ( P<0.01). During the treatment, the incidence of adverse reactions was 14.9% (7/47) in the control group and 8.2% (4/49) in the intervention group, and there was no significant difference between the two groups ( χ2=1.071, P=0.301). Conclusions:Huanglian-Wendan Decoction combined with Estazolam tablets can improve the clinical symptoms, relieve anxiety and depression of PSSD patients with syndrome of internal disturbance of phlegm-heat, and improve sleep quality and clinical efficacy.

ABSTRACT

Objective@#To investigate clinicopathologic features and prognosis of adenoid cystic carcinoma (ACC) involving external auditory meatus.@*Methods@#The clinical presentation and follow-up data of 63 patients with ACC of external auditory canal were collected from January 2006 to February 2017 at PLA General Hospital and Hainan Branch of PLA General Hospital. The clinicopathologic features and prognostic factors of external auditory canal ACC were analyzed.@*Results@#(1) There were 28 males and 35 females and the average age of the first diagnosis was 48.9 years (22-81 years). The tumors showed cribriform pattern in 35 cases (15 cases of late stage), tubular pattern in 14 cases (8 cases of late stage), and solid pattern in 14 cases (9 cases of late stage). Cases with solid pattern was relatively more frequent than that of cribriform pattern and tubular pattern, but the difference was not statistically significant (P>0.05). (2) The average follow-up time was 62.4 months (2-228 months) in the 57 available cases. Among the 33 cases with recurrence, 18 cases had local recurrence and 15 cases had distant metastasis. The mean recurrence time was 40.6 months (2-204 months). Nine patients died of ACC: 2 cases in early stage (died at 48 and 102 months after the first treatment), 7 cases in late stage and 57 with (died at 9, 30, 32, 60, 72, 94 and 228 months). (3) Among the 37 patients with perineural invasion, there were 21 cases of cribriform pattern, 4 cases of tubular pattern and 12 cases of solid pattern; the number of cases in early stage and late stage were 15 and 22, respectively; and the differences were statistically significant (P<0.05). In addition, 31 cases had otalgia among the 37 patients with perineural invasion, where differences were not significant (P>0.05). (4) Thirty of 45 cases with tumor resection or partial resection of temporal bone had recurrence, whereas 3 of 12 cases of tumor combined with superficial lobectomy of parotid gland had recurrence. The difference was statistically significant (P<0.05). Postoperative adjuvant radiotherapy was given in 19 cases, including 7 cases of early stage (2 cases of recurrence), and 12 cases of late stage (8 cases of recurrence), among which there was no significant difference (P>0.05).@*Conclusions@#ACC occurring in external auditory canal frequently recurs. Superficial parotid lobectomy at the first operation is necessary to prevent tumor recurrence. Postoperative adjuvant radiotherapy has certain curative effect on patients with early stage tumor, but it does not affect the recurrence rate. Patients at late stage are more prone to perineural invasion than those in early stage. In addition, cribriform and solid patterns are more common that tubular pattern, and there is no significant correlation between perineural invasion and otalgia.

ABSTRACT

Purpose To evaluate the left ventricular systolic and diastolic function in patients with pulmonary hypertension (PH) using three-dimensional speckle tracking imaging (3D-STI) combined with two-dimensional ultrasound (2D ultrasound),and to investigate the effects of different-degree PH on left ventricular function.Materials and Methods Fifty-two patients who were diagnosed as PHin the First Hospital of Jilin University from September 2011 to November 2015 were retrospectively analyzed,and based on pulmonary artery systolic pressure (PASP),they were assigned into mild group (PASP 36-49 mmHg,n=20),moderate group (PASP 50-70 mmHg,n=18) and severe group (PASP>70 mmHg,n=14).Besides,healthy volunteers were selected as control group (n=30).All the subjects accepted 2D and 3D imaging,their left ventricular diastolic diameter (LVDd),left ventricular ejection fraction (LVEF),left atrial maximum volume index (LAVI),PASP,e',E/e',etc.,were obtained by 2D ultrasound,and their left ventricular global longitudinal strain (GLS),left ventricular global radial strain (GRS),left ventricular global circumferential strain (GCS),left ventricular global area strain (GAS),etc.,were obtained by 3D-STI.The data of different groups were compared,and the correlation between PASP with left ventricular systolic and diastolic function was analyzed.Results E/e'and LAVI in moderate group and severe group were significantly higher than those in control group,and the differences were statistically significant (P<0.05);the absolute values of GLS and GAS in moderate group were lower than those in control group (P<0.05);the absolute values of GLS,GRS,GCS and GAS in severe group were all lower than those in control group,and the differences were statistically significant (P<0.05).The correlation analysis showed that among 2D parameters,the correlation between LAVI and PASP was better (r=0.56,P<0.01);among 3D strain values,the correlation between GLS absolute value and PASP was better (r=0.60,P<0.01).Conclusion 3D-STI combined with 2D ultrasound can effectively assess the left ventricular systolic and diastolic dysfunction in patients with PH,which provides theoretical support for early clinical intervention.

ABSTRACT

Objective: To optimize the ultrahigh pressure extraction ( UPE) process for forsythoside from Forsythia suspensa by Box-Behnken experimental design. Methods:On the basis of single factor screening, a three-factor and three-level Box-Behnken ex-perimental design was employed with liquid/material ratio( X1 ) , extraction pressure ( X2 ) and extraction time ( X3 ) as the independent variables. The response variable was the extraction yield of forsythoside. Results:The optimal UPE conditions for forsythoside were as follows:the ratio of solvent to material (ml·g-1) was 70, the extraction pressure was 151 MPa, and the extraction time was 114 s. With the optimal extraction process, the extraction yield of forsythoside was 13. 15 mg·g-1 . Conclusion:As a novel extraction tech-nology for Chinese herbal medicines, UPE procedure has higher extraction yield, lower extraction temperature, shorter extraction time and less power consumption, which provides a brand-new method for the extraction of forsythoside from Forsythia suspensa.

ABSTRACT

Objective To evaluate the values of integrated backscatterr (IBS) and dobutamine stress echo- cardiography (DSE) in the diagnosis of coronary artery disease(CAD).Methods Thirty-one suspected CAD patients received DSE and IBS analysis before selective coronary angiography(SCA).Using SCA results as the golden standard of the diagnosis,the diagnostic sensitivity(Se),specificity(Sp),and accuracy rate (Ac) of conventional eye-view and DSE-cyclic variation integrated backscatter (CVIB) were compared. Results The CVIB of myocardial segments perfused by stenotic coronary artery was smaller than that of segments perfused by patent coronary artery during high dose of DSE. For conventional DSE eye-view method, the Se,Sp,and Ac were 80%,72% and 77%, while that of DSE-CVIB were 90%,72% and 83%, respectively. Conclusion IBS is a valuable method in assessing the myocardial wall motion in DSE, and the combination of the two methods can improve the diagnostic accuracy rate of CAD.

ABSTRACT

<p><b>OBJECTIVES</b>To study the interaction of beta-2-glycoprotein I (beta 2GPI) with the membrane of hepatocytes and determine whether beta 2GPI participates in HBV infection.</p><p><b>METHODS</b>Ligand blotting, fluorescence microscopy, and fluorescence activated cell sorter (FACS) analysis were used to detect the specific interaction of beta 2GPI with the hepatoma cell line smmc7721, the gastric carcinoma cell line SGC7901, and the lymphoma cell line HL-60.</p><p><b>RESULTS</b>A specific 40 kDa beta 2GPI band was observed by ligand blotting in the case of smmc7721 cells. No such band was observed in SGC7901 or HL-60 cells. Fluorescence microscopy also revealed specific binding of FITC-beta 2GPI to smmc7721 cells, but neither to SGC7901 nor HL-60 cells. FACS analysis demonstrated that the binding rate of FITC-beta 2GPI to smmc7721 cells was significantly higher than these in SGC7901 and HL-60 cells (P < 0.01). The binding rate to smmc7721 cells did not increase with increasing amounts of FITC-beta 2GPI.</p><p><b>CONCLUSIONS</b>There is a specific beta 2GPI-binding protein on the membrane of hepatoma cells in cell line smmc7721 which as the beta 2GPI receptor may participate in HBV infection of hepatocytes.</p>
