ABSTRACT

Objective@#To investigate the dynamic changes of antibodies induced by leptospiral vaccines.@*Methods@#Antigens for antibody detection were screened out. ELISA was used to analyze antibody responses induced at different time points after immunizing guinea pigs with different batches of leptospiral vaccines from different manufacturers. To investigate the relationship between antibody responses induced by leptospiral vaccines and their protective effects in animal model, guinea pigs were challenged with Leptospira after immunization.@*Results@#There was no significant antigen-antibody reaction between the LigA protein or Patoc Ⅰ antigen and the serum samples of guinea pigs immunized with leptospiral vaccines. Notable IgG and IgM antibody reactions were observed in all vaccination groups when using bacterial proteins from seven Leptospira reference strains which were used for the preparation of leptospiral vaccines as envelope antigens. Antigen-specific IgG antibodies peaked at 35 d after the last immunization, and the highest peak of antigen-specific IgM antibodies was reached 11 d after the last immunization. Results of the challenge test showed that non-diluted leptospiral vaccines induced significant IgG and IgM antibody reactions in guinea pigs as compared with those diluted three or nine times, showing good protective effects.@*Conclusions@#Analysis of the dynamic changes of antibodies induced by leptospiral vaccines revealed that there was correlation between the induced serum antibody responses and the protective effects. This study provided reference for further study on alternative methods for evaluating leptospiral vaccine potency.

ABSTRACT

Objective: To study the antipyretic, anti-inflammatory and analgesic effects of Yinhaojiere mixture and the antipyretic effect of Ilex asprella. Methods: The experiments of mice ear edma induced by xylene, the increase of capillary permeability by acetic acid and the sterilized cotton caused rat groin granulation tissue hyperplasia were used to study the anti-inflammatory effect of Yinhao-jiere mixture; the hot-plate tests and the acetic acid writhing test were applied to research the analgesic effect of Yinhaojiere mixture. In addition, the dry yeast induced fever in rats was employed to explore the efficacy of antipyretic agents in Yinhaojiere mixture and Ilex asprella water extract. Results: Yinhaojiere mixture could obviously inhibit the inflammation of the peritoneal cavity of mice ( P <0. 05). The inhibition of Yinhaojiere mixture against ear swelling induced by p-xylene was significant. Yinhaojiere mixture could sig-nificantly increase the pain threshold. Especially, Yinhaojiere mixture had significant antipyretic effect (P<0. 05). In the anti-inflam-matory, analgesic and antipyretic experimetns, there were no significant differences between the middle and high dose groups of Yinha-ojiere mixture and indomethacin group (P>0. 05). Conclusion: Yinhaojiere mixture with the Ilex asprella as the main ingredient has notable anti-inflammatory, analgesic and antipyretic effects, and Ilex asprella exhibits anti-inflammatory, analgesic effects, and its anti-pyretic effect is more significant.

ABSTRACT

Leptospirosis is recognized as an important emerging zoonotic disease caused by pathogenic Leptospira spp.and has a serious impact on people′s health and animal husbandry. Therefore, it has attracted more and more attention. With the development of biotechnology, major breakthroughs have been made in the fields of pathogenicity, molecular epidemic features and evolution mechanism of Leptospira. In this review, we summarize progress in evolution and molecular typing of Leptospira at home and abroad in order to provide a reference for further research on molecular epidemiological surveillance and new vaccine development.

ABSTRACT

Objective To evaluate the quality of immunological diagnosis kits of schistosomiasis. Methods The national reference IgG serum of schistosomiasis was applied for evaluating eight immunological diagnosis kits of schistosomiasis. Results All eight diagnosis kits had no National Production Certificate issued by State Food and Drug Administration. Six kits had good quality with high precision. After the physical examination, two kits did not meet the quality standards. Conclusions The good and bad quality of immunological diagnosis kits of schistosomiasis used in market is intermingled. The quality of these kits should be monitored.

ABSTRACT

Objective To evaluate the stability of dipstick dye immunoassay (DDIA) kit forschisitosomiasis diagnosis. Methods By means of detection of the sera from infected people withSchistosoma japonicum and healthy people, the stability of the DDIA kit, which stored at 37℃,room temperature or 4 ℃ respectively, was evaluated depending on the detective results ofsensitivity, specificity, detectable minimum and coefficient variation ( CV). Results Thesensitivity, specificity, detectable minimum and coefficient variation of the DDIA kit were invariableafter the kits stored at 37 ℃ for 180 days, and at room temperature or 4 ℃ for 360 days. Conclusion The DDIA kit is stable while it stores at 37℃ for 180 days, and at room temperatureor 4℃ for 360 days at least.
