ABSTRACT

To meet the requirement of pathological practice and development, we systematically analyzed the situation of pathological residents training and the importance of initiating the clinical scientific research training. Additionally, we proposed the principle and implementation strategy for clinical scientific research training. According to features of pathological practice, we employed the modularized teaching to divide the training courses into several modules: discussion module for clinical pathology, lecture module for advanced research, and training modules for basic scientific theory, technology and writing skill. With these approaches, the systematic and structured system of standardized residents training is implemented to improve the clinical research ability of pathological residents.

ABSTRACT

Objective To assess the value of brain atrophy and lesion index(BALI),based on magnetic resonance imaging(M RI),in the evaluation of brain aging. Methods 169 healthy older adults were divided into five age groups(40-49,50-59,60-69,70-79,and 80-89 years).MRI scans were evaluated by the 3.0T GE signa and the BALI rating schemes, based on the T1 weighted(T1WI),T2 weighted(T2WI),and T2 weighted fluid attenuated inversion recovery(T2-FLAIR),and T2*weighted gradient-recalled echo(T2 * GRE)images were recorded. Results Based on T1WI,T2WI,T2-FLAIR and T2 * GRE,total scores of BALI increased with age,and showed significant differences between the five age groups(F= 35.35,42.87,46.57,and 54.15,respectively;all P= 0.000).In addition,BALI scores from each sequence were correlated with age(T1WI:r=0.71;T2WI:r=0.73;T2-FLAIR:r=0.73;T2*GRE:r=0.77;all P<0.01).Furthermore,T2*GRE was most sensitive to microbleeds and T2-FLAIR revealed a greater level of deep white matter(χ2 =53.47,P= 0.000)and periventricular lesions(χ2 =29.93,P=0.000)than other sequences. Conclusions The T1WI,T2WI, T2-FLAIR and T2 * GRE,BALI scores can be used to assess whole brain structural changes with aging and provide semi-quantitative indicators for the assessment of brain health.

ABSTRACT

Objective:To compare the proliferative and periodontal specific differentiation abilities of induced pluripotent stem cells (iPSCs) at different passages, and to investigate whether long term culturing would have a negative influence on their proliferation and specific differentiation capacity, thus providing a theoretical basis for further in-depth research on periodontal regeneration and the possible clinical applications of iPSCs.Methods:IPSCs derived from human gingival fibroblasts at passages 5,10,15 and 20 were recovered and cultured in vitro.Their morphology and proliferation rates were observed respectively. We further induced the iPSCs at different passages toward periodontal tissue under the treatment of growth/differentiation factor-5 (GDF-5) for 14 days through the EB routine, then compared the periodontal differentiation propensities between the different passages of iPSCs by detecting their calcified nodules formation by Alizarin red staining and assaying their relative periodontal tissue related marker expressions by qRT-PCR and immunofluorescence staining, including bone related markers:osteocalcin (OCN),bone sialoprotein (BSP);periodontal ligament related markers:periostin, vimentin;and cementum related markers:cementum attachment protein (CAP),cementum protein 1 (CEMP1).The untreated spontaneous differentiation groups were set as negative controls respectively. Results:iPSCs at different passages all showed a high proliferative capacity when cultured in vitro and turned into a spindlelike shape similar to fibroblasts upon periodontal specific differentiation. All iPSCs formed typical calcified nodules upon GDF-5 induction by Alizarin red staining in comparison to their untreated controls. The relative calcium deposition at all passages had been significantly upgraded under the treatment of GDF-5 (P5:t =2.125,P =0.003;P10:t =2.246,P =0.021;P15:t =3.754,P =0.004;P20:t =3.933,P =0.002),but no significant difference in their calcium deposition were detected within passages 5,10,15 and 20 (periodontal differentiation:F =2.365,P =0.109;spontaneously differentiation:F =2.901,P =0.067).Periodontal tissue related marker expressions of iPSCs at all passages had also been significandy upgraded under the treatment of GDF-5 (P < 0.05),but still,no significant difference in their expression levels of periodontal tissue related proteins were detected within passages (BSP:F =0.926 7,P=0.450;vimentin:F=0.917 1,P=0.455;CEMP1:F=2.129,P=0.1367).Conclusion:Our results preliminarily confirmed that long term culturing won't influence the proliferation capacity and periodontal specific differentiation propensity of iPSCs,as they can still proliferate and differentiate toward periodontal cells with high efficiency upon growth factor induction after continuous passaging. Therefore,iPSCs could be recognized as a promising cell source for future possible application in periodontal tissue regeneration.

ABSTRACT

Objective To analyse the changes of cerebral cortical thickness and explore the connectivity of cortical thickness and the clinical syndrome of concomitant strabismus using high-resolution magnetic resonance imaging. Methods Participants were 26 children with primary strabismus and 28 healthy children strictly matched for sex, age, education and socioeconomic level.By using Freesurfer software, the whole-brain-based analysis was perfomrmed to compare the cortical thickness between the two groups. Results Compared with the healthy control group the children with strabismus were found that the cerebral cortex became thinner, including these brain regions related with stereovision, advanced visual-attention,attention and executive control function,as well as brain areas in insula and cingulate circuit. Conclusion Concomitant strabismus in children are not only caused by pathological changes of extraocular muscles, but anatomical changes of the central nervous system, especially the brain areas involved in the advanced visual processing related with visual-motor and visual-attention, and the advanced cognitive brain regions associated with attention and executive control, as well as the anatomical changes in insula and cingulate cortex circuit, which can explain the clinical syndrome of children with concomitant strabismus.

ABSTRACT

Objective To investigate the alterations of interhemispheric resting-state functional connectivity in patients with chro-nic primary trigeminal neuralgia (CPTN).Methods Twenty patients with CPTN and 20 age-,gender-,and education-matched healthy subjects underwent a resting-state functional magnetic resonance imaging (rs-fMRI)scanning.The voxel-mirrored homo-topic connectivity(VMHC)approach and statistic analysis were used to investigate the interhemispheric coordination of the whole brain. Results Compared with healthy controls, decreased VMHC was found in patients with CPTN,including medial and lateral or-bitofrontal cortex, and premotor (P <0.05,GRF corrected). Conclusion Our results suggest that decreased VMHC in brain regions indicate the aberrant perceptual and affective processing of CPTN,and limited complex movements function.

ABSTRACT

Objective To prospectively assess the diagnostic accuracy of MR inflow inversion recovery (IFIR) fast imaging employing steady-state acquisition (FIESTA) for detection of renal artery stenosis (RAS),as compared with renal artery CT angiography (CTA) performed as the reference standard. Methods Thirty patients [18 males,12 females; mean age (65.0±12.1) years] with arterial hypertension and suspected RAS were examined by 3.0-T 3D MR IFIR-FIESTA and renal artery CTA within a week. The IFIR-FIESTA image quality, the IFIR-FIESTA and CTA in display of second-order and third-order branches of renal artery were assessed by two experienced readers. The sensitivity, specificity, accuracy, and negative predictive value (NPV) of IFIR-FIESTA were calculated on artery-by-artery and patient-by-patient bases by the two readers. The inter-observer agreement between the two readers in determining the presence and grade of renal artery stenosis was assessed. Results The scores of qualification of IFIR-FIESTA image quality by reader A and B were (2.16±1.02) and (2.00 ± 0.14),there was no significant difference between them (Z=-0.037,P=0.971).IFIR-FIESTA and CTA in display of second-order and third-order branches of renal artery had no significant difference (x2 =0.298,P=0.585).In the assessment of all 64 main renal arteries by reader A,IFIR FIESTA on artery-by-artery basis had sensitivity, specificity, accuracy, and NPV of 100%,96%,97%,and 100%,respectively; the above parameters assessed by reader B were 100%,90%,93%,and 100%,respectively.There was excellent inter-observer agreement (Kappa=0.803) with the presence or absence of hemodynamically significant RAS.In the assessment of all 30 patients by reader A,IFIR-FIESTA on patient-by-patient basis had sensitivity, specificity, accuracy, and NPV of 100%,95%,97%,and 100%; the above parameters assessed by reader B were 100%,85%,90%,and 100%,respectively; there was good inter observer agreement (Kappa=0.724) with the presence or absence of hemodynamically significant RAS.Conclusions Compared with CTA,MR IFIR-FIESTA can excellently assess RAS without contrast material and radiation exposure, therefore,it can be used for screening of RAS,especially should be widely applied to elderly patients with arterial hypertension.

ABSTRACT

BACKGROUND:The long-term contraction of skeletal muscle can lead to the decrease of resting potential, affect its contraction, and result in the occurrence of fatigue. Whether acupuncture and injury-free magnetic lifting needle are effective on the recovery of resting potential? OBJECTIVE: To observe and compare the influence on the recovery of resting potential of skeletal muscle by taking acupuncture and magnetic needle as the recovery means.DESIGN: A controlled study.SETTINGS: College of Physical Education, Shanxi Normal University;Binzhou College of Shandong Province.MATERIALS: Thirty experimental toads, which were provided by Beijing University of Physical Education, were randomly divided into acupuncture group (n=10), magnetic needle group (n=10), magnetic needle+pressing group (n=10). Left and right semitendinosus muscles were taken as controls.METHODS: The in vitro toad's semitendinosus muscles (experimental side and control side) were treated with electrostimulation for 40 minutes, and then the resting potential of skeletal muscular cells were measured with the intracellular glass microelectrode technique. After the electrostimulation, filiform needle acupuncture, magnetic needle, magnetic needle plus pressing were also given.MAIN OUTCOME MEASURES: The resting potentials were observed before exercise, after electrostimulation and other treatments in the groups.RESULTS: ① The resting potentials of skeletal muscular cells after electrostimulation were decreased as compared with those before electrostimulation [experimental side: (59.48±7.37), (84.54±5.72) mV; control side:(59.43±7.14), (83.62±5.86) mV]. ② After the treatments of acupuncture, magnetic lifting needle, magnetic lifting needle plus pressing, the decreased resting potentials after electrostimulation were obviously recovered (P < 0.01). ③ The recovered percentage of resting potential in the magnetic needle group was obviously smaller than those in the acupuncture group and magnetic needle+pressing group [(42.50±12.41)%, (53.71±10.93)%,(49.10±8.39)%, P < 0. 01].CONCLUSION: Acupuncture, magnetic lifting needle, magnetic lifting needle plus pressing plays a role in promoting the recovery of resting potential of skeletal muscular cells after long-term electrostimulation, and the effect of simple magnetic needle is smaller than that of acupuncture and magnetic needle plus pressing.
