ABSTRACT
OBJECTIVE@#To investigate the expression of transient receptor potential canonical channels (TRPCs) in the heart and kidney of rat model of obstructive sleep apnea hypopnea syndrome (OSAHS).@*METHODS@#Eighteen male SD rats were randomly assigned to intermittent hypoxia (IH) group (=9 ) and control group (=9). In IH group, rats were placed in a chamber and exposed to intermittent hypoxia for 8h (10AM-6PM) daily. The expression of TRPC-related mRNA and protein in the heart and kidney tissue were detected by qRT-PCR and Western blotting, respectively.@*RESULTS@#The mRNA expressions of TRPC3/TRPC4/TRPC5 in heart tissues of IH group were increased significantly compared with the control group (all >0.05); while there were no significant differences in the mRNA expressions of TRPC1/TRPC3/TRPC4/TRPC5/TRPC6/TRPC7 in kidney tissue between two groups (all 0.05).@*CONCLUSIONS@#The IH rat model shows that TRPC5 channel is likely to be involved in the OSAHS induced pathophysiological changes in the myocardium and may become a target to prevent OSAHS related cardiac damage.

Objective:To establish the quality standard for Xiaotang Zhike pills.Methods:Atractylodis Rhizoma,Puerariae Lobatae Radix and Scrophulariae Radix in the formula were qualitatively identified by TLC. HPLC was used for the determination of puerarin in Xiaotang Zhike pills. The analysis was performed on a Wondasil C18-WR(250mm × 4.6 mm,5 μm)column with methanol-water(25:75) as the mobile phase at a flow rate of 1.0 ml·min-1. The detection wavelength was set at 250 nm, the column temperature was 40℃ and the sample size was 10 μl. Results:TLC showed good specificity without any interference for the identification of atrctylodin,puerarin and harpagoside. The calibration curve of puerarin was in good linearity over the range of 41.20-618.00 ng(r = 0.999 9). The average recovery was 100.16%(RSD = 2.91%). Conclusion:The established methods in the study are simple,feasible and reproducible,which are suitable for the quality control of Xiaotang Zhike pills.

The processing of traditional Chinese medicine(TCM) is a traditional pharmaceutical technology,which meets the development demand of the treatment based on syndrome differentiation of TCM. It is one of the important means to guarantee the quality of Chinese medicine and improve the clinical efficacy of TCM. Medical institutions of traditional Chinese medicine should take the advantages and characteristics of traditional Chinese medicine as the foundation, vigorously carry out processing technology for traditional Chinese medicine, rich clinical drug varieties of traditional Chinese medicine, and develop such characteristics of TCM as treatment based on syndrome differentiation and individualized drug treatment. The processing of temporary prescriptions is one of the important means to reflect the characteristics of TCM, and it is of great significance to improve the curative effect of TCM. Based on working practice, this paper summarized the development current status, significance, technical standards and samples of processing variety and methods etc in order to provide thoughts for the processing technology of temporary prescriptions in medical institutions.

Infectious keratitis is one of the leading causes of irreversible vision loss.With the emergence of multidrug-resistant pathogen,despite appropriate diagnosis and antimicrobial treatment,infectious keratitis can still result in corneal opacity and even melting and perforation.Therefore,it is imperative to develop novel treatment for infectious keratitis.More recently,several articles have demonstrated the effectiveness of collagen cross-linking (CXL) in treating infectious keratitis via direct antimicrobial activity and inhibition of corneal enzymatic degradation.In order to highlight the use of CXL tor infectious keratitis treatment,a new term was proposed at the 9th CXL congress to rename its use in infections as photoactivated chromophore for infectious keratitis-CXL (PACK-CXL).Based on the current literature,PACK-CXL appears to be effective for bacterial keratitis,especially on inhibiting corneal perforation.The effect was limited for fungal and amoebic keratitis,and maybe aggravative on viral keratitis.In this article,we will review the basic profile,technical operation,prevention of complication of CXL,the principle mechanism of PACK-CXL and its clinical application for bacterial,fungal keratitis,amoebic and viral keratitis.

Objective To investigate the effect of surgical treatment on survival in colorectal carcinoma patients with synchronous hepatic metastasis.Methods The retrospective case-control study was done on 953 consecutive patients with synchronous colorectal hepatic metastasesl from January 2003 to December 2013.Results Median survival time (46.7 months)and 5-year survival rate (32％) for patients with resected hepatic metastases was significantly superior to that of with nonoperative treatment (17 months,4％).Expanded criteria for hepatic metastases resection raised resection rates (31％ vs.13.6％,P ＜0.05).For patients with resectable hepatic metastases,the inhospital cost for simultaneous resection group was lower than that in the staged resection group (36 698 vs.45 134 RMB,P ＜ 0.05).For patients of asymptomatic primary tumor with unresectable hepatic metastases,resection of the primary tumor was associated with an improved median survival (18.0 vs.15.0 months,P ＜ 0.05) Conclusions Expanding indications of hepatic metastases resection can improve survival in patients with synchronous colorectal hepatic metastases.Simultaneous resection of primary tumor and hepatic metastases were indicated in patients with resectable synchronous colorectal hepatic metastases.Resection of primary tumor was recommended for asymptomatic patients with unresectable hepatic metastases.
ABSTRACT
Objective To evaluate the effects of focal vibration on the physical properties and surface electroyograms of persons with neck-shoulder pain.Methods Thirty-four patients with neck-shoulder pain received 3 to 5 applications of vibratory stimulation of the inner side of the trapezius and scapula for 30 s each time.They reported their pain level using a visual analogue scale.Muscle tone and the elasticity and stiffness of the trapezius muscle were measured bilaterally using a MyotonPRO Muscular Function Evaluation System.Surface electromyography (sEMG) was performed bilaterally during arm extension while standing.The testing was repeated four times:before treatment (baseline),immediately after treatment (IAT),2 hours after treatment (2AT) and 6 hours after treatment (6AT).Results There was a significant reduction in reported pain at IAT and 2AT compared to baseline,but no significant difference was found between the perceived pain at baseline and at 6AT.The values of oscillation frequency,logarithmic decrement and dynamic stiffness had decreased significantly at IAT and 2AT compared to baseline.The average AEMG during upper-arm flexion of the trapezius on the painful side had increased significantly at IAT comparcd to that before the treatment,while the average IEMG and MF were both significantly higher than before the treatment.At 2AT the average AEMG and IEMG were both still significantly higher than at the baseline,but there was no longer a significant difference in the average MF.By 6AT none of the three indicators differed significantly from those before the treatment.Conclusions A single session of focal vibration can relieve pain,increase muscle activation as well as decrease muscle stiffness for people with neck-shoulder pain within 2 hours.

Objective:To establish the quality standard for Qingchang mixture.Methods:TLC was adopted to identify angelica sinensis,paeoniae radix and phellodendri cortex.HPLC was used to determine the content of ferulic acid on a Wondasil C18 (250 mm×4.6 mm,5 μm) analytical column,the mobile phase was composed of acetonitrile-0.1% phosphoric acid solution(17∶83),the flow rate was 1.0 ml·min-1, and the detection wavelength was 316 nm and the column temperature was 30℃.Results:The TLC spots were clear and well-separated without any negative interference.The calibration curve of ferulic acid was in good linearity over the range of 39.6-316.8 μg·ml-1(r=0.999 9),the average recovery was 98.75% and RSD was 1.21%(n=9).Conclusion:The method is fast and accurate with good specificity,which can be used for the quality control of Qingchang mixture.

Methods:TLC was adopted to identify salvia miltiorrhi-za, rhubarb charcoal and panax notoginseng; the contents of tanshinoneⅡA , chrysophanol and physcion in Huayu pills were deter-mined by RP-HPLC with Wondasil C18 (250 mm × 4. 6 mm,5μm) as the analytical column, the mobile phase was composed of metha-nol-0. 1% phosphoric acid solution (85 ∶15), the flow rate was 1. 0 ml·min-1,the detection wavelength was 254 nm, the column temperature was 35℃, and injection volume was 10 μl. Results:The TLC spots were clear and well-separated without any negative in-terference; the calibration curve of tanshinoneⅡA , chrysophanol and physcion was linear over the range of 0. 212-2. 12 μg ( r =0. 999 9),0. 159-1. 59 μg(r=0. 999 9) and 0. 072 6-0. 726 μg(r=0. 999 9), the average recovery was 99. 55 %, 99. 56% and 100. 60%, and RSD was 1. 57%,2. 26% and 2. 28%(n=6), respectively. Conclusion: The method is fast and accurate with good specificity, and can be used for the quality control of Huayu pills.

Methods:TLC was adopted to identify Paeoniae Ra-dix Rubra, Cyperi Rhizom and Cinnamomi Cortex; the contents of paeoniflorin and cinnamic aldehyde in Juebi capsules were deter-mined by HPLC with Wondasil C18 (250 mm × 4. 6 mm,5 μm) as the analytical column, the mobile phase was composed of methanol-0. 2％ phosphoric acid solution with gradient elution, the flow rate was 1. 0 ml·min-1 ,the detection wavelengths were 230 nm and 290 nm, the column temperature was 40℃, and the volume injection was 10 μl. Results: The TLC spots were clear and well-separated without any negative interference;the calibration curves of paeoniflorin and Cinnamic aldehyde were in good linearity over the range of 0. 368-3. 680μg(r=0. 9999) and 0. 191-1. 914 μg(r=0. 9999), the average recovery was 101. 9 ％ and 99. 66％, and the RSD was 1. 90％ and 2. 77％ (n=6), respectively. Conclusion:The method is fast and accurate with good specificity, which can be used for the quality control of Juebi capsules.

Objective: To establish the quality standard for Ehong capsules.Methods: A TLC was adopted to identify safflower, pueraria and prepared Radix Polygoni Multiflori.The contents of hydroxyl-safflor yellow A and puerarin in Ehong capsules were determined by a dual wavelength HPLC.A Wondasil C 18-WR (250 mm× 4.6 mm ,5 μm) was used as the analytical column, the mobile phase was composed of methanol acetonitrile-0.7% phosphoric acid aqueous solution (16∶3∶81), the flow rate was 1.0 ml·min-1 ,the detection wavelength was 403 nm for hydroxy safflower yellow A and 250 nm for puerarin.The column temperature was 30℃.Results: The TLC spots were clear and well-separated without negative interference.The calibration curves of hydroxyl-safflor yellow A and puerarin were linear over the ranges, the average recovery was 101.02% and 100.03% , and the RSD was 1.43% and 2.40% (n =6), respectively.Conclusion: The method is fast and accurate with good specificity, which can be used for the quality control of Ehong capsules.

Vascular endothelial growth factor (VEGF),platelet derived growth factor (PDGF) and complements play key roles in the pathogenesis of age-related macular degeneration (AMD).Pegaptanib,the first therapeutic aptamer against VEGF165,has been approved by the Food and Drug Administration (FDA) of US for the treatment of exudative AMD.Another two aptamers El0030 and ARC1905,each target PDGF-B and complement C5 respectively,are undergoing clinical trials.Recent trends to treat AMD are combined therapies targeting multiple key molecules in the pathogenesis of AMD;aptamers against multiple targets may become the preferred drug for AMD.

Objective: To explore the effect of different processing methods on the content changes of ferulic acid and total flavonoids in wheat bran and its processed products by HPLC and UV spectrophotometry.Methods: Ferulic acid was determined by HPLC with SHIMADZU C18 (250×4.6 mm,5 μm) as the analytical column, the mobile phase was composed of acetonitrile-0.1% phosphoric acid solution (16∶84), the flow rate was 1.0 ml·min-1, the detection wavelength was 320 nm and the column temperature was 35℃.Total flavonoids was determined by UV spectrophotometry at the wavelength of 258 nm(±2 nm).Results: The content of ferulic acid in the crude bran, the toasted to brown bran, the toasted to dark brown bran, the over toasted bran and the honey toasted bran was 2.07, 2.33, 1.68, 1.11 and 0.83 μg·g-1, and total flavonoids was 5.96, 7.15, 9.73, 13.61 and 9.42 mg·g-1, respectively.Conclusion: The higher the toasted processing degree is, the lower content of ferulic acid and the higher content of total flavonoids are in wheat bran.

Objective:To establish the quality standard for Tiaojing Zhuyun granules .Methods:A TLC method was used for the qualitative identification of main ingredients of Cornus officinalis and Angelica sinensis, and an HPLC method was used for the determi-nation of loganin in Tiaojing Zhuyun granule with SinoChrom ODS-BP (250 mm ×4.6 mm,5μm) as the analytical column .The mobile phase was composed of acetonitrile-0.1%phosphric acid solution (14∶86) at a flow rate of 1.0 ml· min-1 .The detection wavelength was set at 240 nm and the column temperature was 30℃.Results:The corresponding spots in the granule were clear without any inter -ference from the negative control .The calibration curve of loganin was in good linearity over the range of 50 .40-1008 .00 ng ( r =0.999 9).The average recovery was 98.02%(RSD=2.51%,n=6).Conclusion:The method is simple,feasible and reproducible, which can be used for the quality control of Tiaojing Zhuyun granules .

Objective:To comprehensively evaluate the intrinsic and external quality, and explore the grading of gypsum pieces. Methods:The characteristics, contents of main components and trace elements and X-diffraction exclusive characteristic peaks of gyp-sum pieces from different origins were detected. Combined with the market grading, the grading standard for gypsum pieces was further improved. Results:According to the tested indices, gypsum pieces were graded into two classes:excellent pieces and common decoc-tion pieces. Conclusion:The study shows reference value for guiding the market grading of gypsum pieces.

Objective To investigate the correlation of plasma interleukin ( IL)-17 level and IL-17 receptor (IL-17R) expression in the thymus of patients with myasthenia gravis (MG).Methods The blood samples of 63 patients (38 with glucocorticoid treatment, 25 with thymus removal) who admitted to Henan Provincial People′s Hospital between 2010 and 2014 were collected at three different stages: pre-treatment, 1 week post-treatment and 1 month post-treatment.The blood samples of 42 healthy controls were also collected.Enzyme linked immunosorbent assay was used to evaluate the levels of IL-17 in plasma.Twenty-five thymus tissues from MG patients and another 12 thymus tissues from patients with congenital heart disease who had surgery therapy were also collected.Reverse transcription polymerase chain reaction was used to evaluate the mRNA levels of IL-17R.The possible correlation between the expression of IL-17 and IL-17R with MG was analyzed.Results Before treatment, the levels of IL-17 in the plasma were much higher in all the MG patients ( both ocular and generalized) when compared to the healthy controls ( controls (3.2 ±0.7) pg/ml, MG patients (8.5 ±1.7) pg/ml, t =2.450, P 0.05, compared to the healthy controls).In the surgery therapy cases, the IL-17 levels were also reduced after the thymus removal ( pre-surgery (8.8 ±1.4) pg/ml, 1 week after surgery (5.3 ±0.7) pg/ml, t=1.950, P<0.05;1 month after surgery (3.0 ±0.4) pg/ml, t=2.683, P<0.01).In the thymus tissues of the MG patients, the mRNA levels of IL-17R were much higher than that of the controls ( relative level 2.31 folds, t =2.682, P <0.01).Meanwhile, a positive correlation was found between the plasma IL-17 levels and the relative IL-17R levels in thymus tissues ( r =0.945 4, P <0.01 ).Furthermore, IL-17 was positively correlated with quantitative myasthenia gravis scores (QMGS) either pre-treatment (r =0.798 1, P <0.01) or post-treatment (r=0.906 5, P<0.01).And IL-17R was positively correlated with QMGS pre-treatment (r=0.775 5, P<0.01).Conclusions IL-17 is increased in the plasma of MG patients (both ocular and generalized) , and is decreased upon the glucocorticoid treatment or surgery therapy, suggesting that it can be used as a parameter to determine the therapeutic effects.IL-17R is increased in the thymus tissues of MG patients, suggesting that it can potentially be used as a pathological diagnosis parameter.

@#Objective To observe the effect of Tongqiaohuoxue decoction on cognitive impairment in rats with traumatic brain injury and explore the mechanisms. Methods 70 Sprague-Dawley rats were randomized into normal group (n=10), sham operated group (n=20), untreated group (n=20) and treatment group (n=20). Controlled cortical impact device was utilized to establish traumatic brain injury model. The treatment group received Tongqiaohuoxue decoction 5 ml/d, and other groups received distilled water. The cognitive function of rats was evaluated with Morris water maze 1, 2, 3, 4 weeks post operation. And the rats were sampled to test the expression of brain-derived neu-rotrophic factor (BDNF) and synaptophysin (Syn) I in hippocampus through immunohistochemistry. Results The escape latency was longer in the untreated group than in the normal group in all the time points (P<0.05), and was longer in the treatment group than in the normal group 1 and 2 weeks after injury (P<0.05), with no significant difference 3 and 4 weeks after injury (P>0.05). And it was shorter in the treat-ment group than in the untreated group 3 and 4 weeks after injury (P<0.05). The percentage of swimming time was lower in the untreated group than in the normal group in all the time points (P<0.05), and was lower in the treatment group than in the normal group 1, 2, and 3 weeks after injury (P<0.05), with no significant difference 4 weeks after injury (P>0.05). And it was higher in the treatment group than in the untreated group weeks after injury (P<0.05). The BDNF expression was lower in the untreated group than in the normal group 1 and 2 weeks after injury (P<0.05), with no significant difference 3 and 4 weeks after injury (P>0.05). It was higher in the treatment group than in the normal group 4 weeks after injury (P<0.01), and was higher in the treatment group than in the untreated group 2, 3 and 4 weeks after in-jury (P<0.05). The Syn I expression was lower in the untreated group than in the normal group in all the time points (P<0.001), and was low-er in the treatment group than in the normal group 1, 2 and 3 weeks after treatment (P<0.01). And it was higher in the treatment group than in the untreated group 3 and 4 weeks after injury (P<0.01). Conclusion Tongqiaohuoxue decoction could improve the cognitive function in rats with traumatic brain injury. The change in expression of BDNF and Syn I might be associated with the improvements.

Objective:To develop a method for the rapid determination of CaSO4 ·2H2 O in gypsum by Raman spectra. Methods:Totally 40 batches of gypsum from different origins were used as the training set, compared with the results by ENTA titration, and OPUS software was used to establish a quantitative analysis model of CaSO4 ·2H2 O in gypsum by Raman spectra. Results:The estab-lished quantitative analysis model could provide a good prediction result rapidly when the content was between 97. 93% and 99. 81%. Conclusion:The method is accurate, fast and simple, which can be developed as an analysis method for CaSO4 ·2H2 O in gypsum.

Aim To observe the variation of T-cell subsets in spleen in different courses of disease and tis-sue histopathological changes in rats with adjuvant ar-thritis (AA).Methods The model of rat AA was in-duced by complete Freund’s adjuvant into the right hind metatarsal footpad of 20 male Lewis rats.Arthritis was evaluated by total score,arthritis index and paw swelling number. The percentage of total (CD3 +CD4 +), memory (CD4 + CD44 +), no-sensitizated (CD4 + CD62L +) and Th1 7 (CD4 + IL-1 7 +) on CD4 +T cells in spleen were detected by flow cytometry on days 1 4 and 22 after immuniozation.The his-topathological evaluation of heart,liver,spleen,lung, kidney,ankle joints and mesenteric lymph nodes were examined by hematoxylin and eosin (HE)stain.Re-sults The onset of secondary arthritis appeared on a-bout day 1 0 after immunization,with a peak onset on day 1 9.Systematic arthritis symptoms included front and hind paw swelling and redness (swelling was more apparent in the front rather than hind paws),nodule formation and redness on the ears,connective tissue swelling and redness of the nose and evident nodules and redness on the tail.Total score,arthritis index and the paw swelling numbers were enhanced in AA rats. Pathohistology of ankles joints showed hyperplasia, pannus formation,and inflammatory cells infiltration in AA rats.And pathohistology of heart,liver,spleen, kidney and mesenteric lymph nodes showed inflamma-tory changes.Compared with normal group,there was no change in the percentage of total,memory and no-sensitizated CD4 +T cell in the early stage inflammation but they were significantly enhanced in the peak of in-flammation.Th1 7 cell subsets were significantly en-hanced in both of the early and peak of inflammation. Conclusion These findings suggest that abnormal T cell responses and tissue histopathological changes are important features of AA rats.
