ABSTRACT

OBJECTIVE To study the improvement effects of poria acid on insulin resistance in rats with polycystic ovary syndrome (PCOS) and its mechanism. METHODS One hundred and twenty-six female rats were randomly separated into blank group, PCOS group, poria acid low-dose group (8.33 mg/kg), pachymic acid high-dose group (33.32 mg/kg), ethinylestradiol cyproterone group (positive control group, 0.34 mg/kg), recombinant rat high mobility group protein B1 protein (rHMGB1) group (8 μg/kg), and poria acid high dose+rHMGB1 group (33.32 mg/kg poria acid+8 μg/kg rHMGB1), with 18 rats in each group. Except for the blank group, the rats in all other groups were given Letrozole suspension intragastrically to construct the PCOS model. After successful modeling, administration was performed once a day for 4 weeks. After medication, the fasting blood glucose and fasting insulin levels, and insulin resistance index (HOMA-IR) were measured in rats; the levels of follicle-stimulating hormone (FSH), luteinizing hormone (LH) and testosterone (T) in rat serum, and the levels of interleukin-1β (IL-1β) and tumor necrosis factor- α (TNF- α) in ovarian tissue were detected; ovarian coefficients of rats were calculated; the pathological changes of ovarian tissue were observed; the expressions of HMGB1, receptor for advanced glycosylation elaine_ tanghong@sina.com end product (RAGE) and phosphorylated nuclear factor κB p65 (p-NF-κB p65) proteins were determined in ovarian tissue of rats. RESULTS Compared with the blank group, the pathological injury of ovarian tissue of rats in the PCOS group was serious, the levels of fasting blood glucose and fasting insulin, HOMA-IR and ovarian coefficient were increased, the levels of serum LH and T were increased, while the levels of FSH were decreased; the levels of IL-1β and TNF-α, the expressions of HMGB1, RAGE and p-NF-κB p65 protein in ovarian tissue were increased, with statistical significance (P<0.05). Compared with the PCOS group, pathological damage of ovarian tissue was reduced in poria acid low-dose and high-dose groups and ethinylestradiol cyproterone group, and fasting blood glucose, fasting insulin levels, HOMA-IR and ovarian coefficient were decreased; serum LH and T levels were decreased, while FSH levels were increased; the levels of IL-1β and TNF-α and the expressions of HMGB1, RAGE and p-NF-κB p65 protein in ovarian tissue were decreased, with statistical significance (P<0.05). The trend of corresponding indexes in rHMGB1 group was opposite to the above (P<0.05). Compared with poria acid high-dose group, the changes of the above indexes were reversed significantly in poria acid high-dose+rHMGB1 group (P<0.05). CONCLUSIONS Poria acid may improve insulin resistance and inhibit inflammatory reaction in PCOS rats by inhibiting HMGB1/ RAGE pathway.

ABSTRACT

@#To explore the potential anti-inflammatory effects and retinal neuroprotective effects of glycyrrhizin (GL) on diabetic retinopathy (DR),twenty-one male C57BL/6 mice were randomly divided into control group,model group and glycyrrhizin (GL) group. The diabetic mice model was established by intraperitoneal injection of STZ. The GL group was treated with GL (150 mg/kg/d) by gavage for 6 weeks after modeling. Retinal tissue sections were paraffin-embedded and morphological examinated with hematoxylin-eosin (HE). Immunohistochemistry was used to detect the expression of GLUT1,GFAP and GAP43. RT-PCR was adopted to detect retinal inflammation and expression of apoptotic mediators (TNF-α,IL6,iNOS,NF-κB and Tp53). The results showed that the diabetic mice developed retinal disorders and retinal degeneration. The expression of glial cell proliferation marker GFAP was up-regulated,while the expression of neuronal plasticity marker GAP43 was down-regulated;NF-κB,TNF-α,IL6,iNOS and Tp53 transcription in the retina of diabetic mice increased. In the GL group,the degree of down-regulation of GLUT1 expression in the retina of diabetic mice was reduced,retinal inflammation was eliminated and the structure was improved. The above results suggested that GL may be a potential neuroprotective agent for diabetic patients and has anti-inflammatory effects,but its efficacy and safety in DR patients requires further clinical studies.

ABSTRACT

In recent years, with the rapid development of machine learning techniques,the deep learning algorithm has been widely used in one-dimensional physiological signal processing. In this paper we used electroencephalography (EEG) signals based on deep belief network (DBN) model in open source frameworks of deep learning to identify emotional state (positive, negative and neutrals), then the results of DBN were compared with support vector machine (SVM). The EEG signals were collected from the subjects who were under different emotional stimuli, and DBN and SVM were adopted to identify the EEG signals with changes of different characteristics and different frequency bands. We found that the average accuracy of differential entropy (DE) feature by DBN is 89.12%±6.54%, which has a better performance than previous research based on the same data set. At the same time, the classification effects of DBN are better than the results from traditional SVM (the average classification accuracy of 84.2%±9.24%) and its accuracy and stability have a better trend. In three experiments with different time points, single subject can achieve the consistent results of classification by using DBN (the mean standard deviation is1.44%), and the experimental results show that the system has steady performance and good repeatability. According to our research, the characteristic of DE has a better classification result than other characteristics. Furthermore, the Beta band and the Gamma band in the emotional recognition model have higher classification accuracy. To sum up, the performances of classifiers have a promotion by using the deep learning algorithm, which has a reference for establishing a more accurate system of emotional recognition. Meanwhile, we can trace through the results of recognition to find out the brain regions and frequency band that are related to the emotions, which can help us to understand the emotional mechanism better. This study has a high academic value and practical significance, so further investigation still needs to be done.

ABSTRACT

Objective To evaluate the clinical efficacy and safety of nanoparticle albumin-bound paclitaxel (Nab-P) in the treatment of advanced non-small cell lung cancer (NSCLC). Methods Fifty-six over 65 years old patients with advanced NSCLC treated with Nab-P monotherapy in department of chemotherapy of the People's Hospital of Zhongshan City from January 2014 to January 2017 were analyzed retrospectively. The chemotherapy regimen was Nab-P 260 mg/m2, on d1 or d1 + d8, every 21 day for a cycle, imaging examination was made for efficacy evaluation after every 2 cycles. Results All 56 patients had been evaluated for efficacy, and received a total of 186 cycles of chemotherapy. All patients had been completed 2 cycles or more than 2 cycles of chemotherapy, and the median number of chemotherapy cycles was 3. The treatment response rate (RR) was 25.0 % and the disease control rate (DCR) was 76.8 %. The median progression free survival (PFS) time was 4.7 months. The main adverse reactions were neutropenia, nausea and vomiting, fatigue and peripheral nerve toxicity. However, the vast majorities of adverse reactions were grade 1-2, and can be improved after treatment. Conclusions Nab-P is effective in the treatment of advanced NSCLC in elderly patients. The adverse reactions are mild and tolerant, and it is worthy to be popularized.

ABSTRACT

Wearable devices bring us an innovative human-computer interaction which plays an irreplaceable role in enhancing the users' ability in environmental awareness, acquirements of their own state and "ubiquitous" computing power. Since 2013, wearable devices have quickly appeared around us. In this article we classify most of the wearable devices which have been appeared in the markets or reported in the literature according to their functions and the positions where they are worn. Furthermore, we review the technologies related to wearable devices, such as sensing technology, wireless communication, power manager, display technology and big data. At last, we analyze the challenges which the wearable devices will face in near future, and look forward to development trends of wearable devices.

ABSTRACT

Objective To explore the effect of atorvastatin application on serum ferritin(SF)levels in elderly male patients with acute coronary syndrome(ACS)and its clinical significance. Methods 120 elderly male patients treated in Department of Cardiology in Cangzhou People's Hospital were randomly divided into treatment group and control group(60 cases in each group). All the patients in the two groups took the standard treatment of coronary heart disease,and the patients in treatment group were additionally given atorvastatin 20 mg orally,once-daily for 1 month. After 1 month of treatment,the changes in SF and lipid levels before and after treatment were determined. Results The total cholesterol(TC),low density lipoprotein(LDL)and SF levels of the two groups after treatment were significantly decreased compared with those before treatment. After treatment,triacylglycerol (TG)was decreased,and high-density lipoprotein(HDL)was increased significantly in treatment group,while in the control group,after treatment TG was increased and HDL was decreased. Compared between treatment and control groups after treatment,the differences in SF,TC,TG,HDL and LDL levels were statistically significant〔SF(μg/L):174.33±99.87 vs. 255.66±100.34,TC(mmol/L):4.23±0.60 vs. 5.15±0.56,TG(mmol/L):1.98±0.64 vs. 2.00±1.03,HDL(mmol/L):1.13±0.14 vs. 1.09±0.22,LDL(mmol/L):2.45±0.33 vs. 3.35±0.50,P<0.05 or P<0.01〕. Conclusion In elderly male hospitalized patients with ACS,the level of SF is significantly higher than normal,after atorvastatin specification treatment,the blood fat and SF levels are significantly lower,thus the SF inducing myocardial damage may be reduced and the risk of recurrence of acute coronary events can be decreased.

ABSTRACT

Objective To investigate the relationships between level of serum ferritin(SF)/blood lipid and the degree of coronary artery stenosis in elderly patients with acute coronary syndrome(ACS),and evaluate the value of SF in the diagnosis of coronary heart disease. Methods ①One hundred and twenty-five elderly patients with ACS(65 males,60 females)underwent coronary angiography examination,and the total cholesterol(TC), triglycerides(TG),high-density lipoprotein(HDL-C),low density lipoprotein(LDL-C)and SF were determined;according to the results of coronary angiography,the degree of coronary artery stenosis was obtained by using the Gensini score and the degrees of severity of stenosis were divided into three groups:light(42 cases),medium (45 cases)and severe(38 cases). The differences in levels of SF and blood lipids among the three groups were observed,and the relationships between the level of SF/blood lipids and various degrees of coronary artery stenoses were analyzed.②The elderly patients with ACS were divided into two groups,male and female,to compare the SF numerical value and Gensini score between the two groups. ③The SF numerical value and Gensini score of 65 senile male cases with ACS were compared to those of 60 middle aged male cases with the same disease. Results ①Variance analysis showed that SF,TC,LDL-C distribution had significant differences among light,medium and severe groups〔SF(μg/L):147.287±73.838,162.313±94.882,290.421±194.501;TC(mmol/L):4.423±0.900,4.998±0.740, 5.032±1.009;LDL-C(mmol/L):3.150±0.799,3.439±0.839,3.902±1.053,all P0.05);in elderly patients with ACS,SF was positively correlated with Gensini score(r=0.355,P=0.000),while there was no correlation between Gensini score and any one of the following lipids:TC,TG,LDL-C,HDL-C(r=0.090,-0.170,0.256,-0.060,P=0.532,0.092,0.089,0.537).②The comparisons of SF value and Gensini score between senile male and senile female patients with ACS showed no significant differences(all P>0.05).③The comparisons of SF value and Gensini score between senile and middle aged male patients with ACS also showed no significant differences(all P>0.05). Conclusion The occurrence of ACS has certain correlation with SF and lipids,the degree of severity in coronary artery lesion is associated with SF,and there are no differences in age and sex in relation to the value of SF in the diagnosis of ACS.
