ABSTRACT

OBJECTIVE@#Hypertrophic scars (HS) are a variety of skin tissue fibrosis disease that occurs in human skin, the effective therapeutic method of which is still inaccessible up to now. As a bioactive constituent of a well-known medical plant, Salvia miltiorrhiza (Danshen in Chinese), tanshinone IIA (TSA) is reported to inhibit cell proliferation in HS. Therefore, the aim of this study was to prepare TSA self-soluble microneedles to strengthen its dermal retention and break through the difficulty of significantly thickening epidermal connective tissue and stratum corneum at the HS site. The possible mechanism of action in suppressing HS was studied using human skin fibroblasts (HSF).@*METHODS@#Tanshinone IIA self-dissolving microneedles (TSA-MN) was prepared using a negative mold casting method. The prescription process of microneedle was optimized by Box-Behnken effect surface method. Different media were selected to investigate the ability of transdermal absorption and in vitro release. Furthermore, according to Cell Counting Kit-8 (CCK8) method as well as the Western blot method, the effect of TSA-MN on the biological characteristics of HSF was investigated.@*RESULTS@#With remarkable slow release effect and dermal retention, the release and transdermal properties of TSA-MN in vitro were better than both TSA and ordinary dosage forms. Its effect of HSF confirmed the essential decrease in cell motility during cell proliferation and cell migration in vitro, which plays a significant role in down-regulating the secretion of transforming growth factor-β1 (TGF-β1) in HSF and increasing the expression level of Smad7.@*CONCLUSION@#The prepared TSA self-soluble microneedles is helpful in solving the problem of hypertrophic scars, with a stable dermal retention effect after process optimization.

ABSTRACT

Tetrapyrrole compounds are a class of compounds with important functions. They exist in living organisms and have been widely used in agriculture, food, medicine, and other fields. The cumbersome process and high cost of chemical synthesis, as well as the shortcomings of unstable quality of animal and plant extraction methods, greatly hampered the industrial production and applications of tetrapyrrole compounds. In recent years, the rapid development of synthetic biology has provided new tools for microorganisms to efficiently synthesize tetrapyrrole compounds from renewable biomass resources. This article summarizes various strategies for the biosynthesis of tetrapyrrole compounds, discusses methods to improve its biosynthesis efficiency and future prospects, with the aim to facilitate the research on biosynthesis of tetrapyrrole compounds.

ABSTRACT

Objective:To investigate the correlation of red blood cell distribution width-to-platelet ratio (RPR) with clinical features and prognosis of patients with multiple myeloma (MM).Methods:The clinical data of 137 patients with MM who were admitted to the Affiliated Hospital of Xuzhou Medical University from April 2013 to July 2019 were collected. The receiver operating characteristic (ROC) curve was used to determine the best cut-off value of RPR. According to the best cut-off value of RPR, the patients were divided into high RPR group and low RPR group, and the differences in clinical characteristics and prognosis between the two groups were analyzed.Results:The best cut-off value of RPR was 0.10, and according to the best cut-off value, the patients were divided into high RPR group (RPR ≥ 0.10, 52 cases) and low RPR group (RPR < 0.10, 85 cases). There were statistical differences between the high RPR group and low RPR group in the proportion of patients between different stratification of Durie-Salmon (DS) staging ( χ2 = 17.110, P < 0.01), International Staging System (ISS) staging ( χ2 = 10.817, P = 0.001), red blood cell distribution width standard deviation(RDW-SD) ( χ2 = 26.937, P < 0.01), hemoglobin ( χ2 = 17.140, P < 0.01), lactate dehydrogenase (LDH) ( χ2 = 7.926, P = 0.005), erythrocyte sedimentation rate (ESR) ( χ2 = 9.513, P = 0.002), β 2-microglobulin (β 2-MG) ( χ2 = 7.726, P = 0.005), and bone marrow plasma cell ratio (BMPC) ( χ2 = 6.621, P = 0.010). The overall response rate (ORR) in the low RPR group was higher than that in the high RPR group [82.4% (70/85) vs. 71.2% (37/52)], but the difference was not statistically significant ( χ2 = 2.366, P = 0.124). The deep remission rate in the low RPR group was higher than that in the high RPR group [56.5% (48/85) vs. 19.2% (10/52)], and the difference was statistically significant ( χ2 = 18.327, P < 0.01). The results of multivariate analysis showed that the albumin, RPR and degree of remission were independent influencing factors for the overall survival (OS) of newly treated MM patients (all P < 0.05). Conclusion:MM patients with elevated peripheral blood RPR have shorter OS time, and RPR may be one of the indicators for evaluating the prognosis of MM.

ABSTRACT

The authors, based on barcode analysis and tracking technology, designed a complete supply chain management system for ophthalmic consumables, systematically solving the traceability challenge of high-value medical consumables ( mainly implanted medical instruments) . This innovation can sizably reduce the inbound/outbound error rate at level-1 and level 2 warehouses of the hospital, downsizing inventory cost expenditure and upsizing work efficiency. The management of "one item for one code"constitutes a traceable closed loop for such consumables, improving the informationized, standardized and refined management, and reducing the incidence of adverse events for medical devices. Thus the quality management system of eye hospitals can be elevated.

ABSTRACT

Objective To observe the clinical effect of acupuncture combined with traditional Chinese medicine Tongluo huayu decoction on white matter damage in patients with occlusive cerebral infarction.MethodsA total of 76 patients with cerebral infarction with cerebral hemorrhage were enrolled in this study from April 2013 to January 2015.The random number table was divided into observation group and control group (38 cases).The control group was given routine western medicine treatment, the observation group on the basis of the control group plus traditional Chinese medicine tongluo huayu decoction, and with acupuncture treatment, the two groups were treated for 2 months, compare the effectiveness of treatment, analysis of the two groups before and after treatment platelet aggregation rate MAR (CDCA) and platelet-activated granule membrane glycoprotein 62P (CD62P), and the prognosis was analyzed by NIHSS, Montreal Metabolic Assessment Scale (MoCA) and Barthel Index (BI) Observe adverse reactions.ResultsThe effective rate of the observation group was 94.74% higher than that of the control group (78.95%, P< 0.05).After treatment, the expression of CD62P (21.87±1.55)%, MAR (35.23±1.62)% and NIHSS score (10.27±0.78) (P< 0.05).The incidence of adverse events was 13.16% in the observation group (25.04±1.11) and the BI index (88.15±1.20) were significantly higher than those in the control group (P< 0.05);There was no significant difference in the prevalence of adverse effects between 13.16% and 7.89%.ConclusionAcupuncture combined with traditional Chinese medicine Tongluo huayu decoction can effectively protect the white matter of patients with massive arterial occlusive cerebral infarction, improve its platelet CD62P, MAR and neurological function, cognitive function, improve the quality of life, and mild adverse reactions, it is worth in the clinical promotion application.

ABSTRACT

[Objective] To observe the influence of Red Sage and Propranolol on portal hypertension rats' pressure,liver fibrosis and gastric-intestinal hormone.[Method] At 4w after making models,rats take the said medicines,then measure their portal pressure with biological signal Medlab-Ug4Cs collection and treatment system;after decapitation,measure the indexes with radio-immune method.[Result] In model group(1),the portal pressure is much more increased than normal one(2),that in Red Sage treatment(3) and Propranolol(4) groups is much lowered,so are gastrin and glucagon;compared to group 1,the gastrin,motilin and glucagon in group 3 are much decreased,so are compared to group 4.[Conclusion] Red Sage can effectively reduce portal pressure,liver fibrosis index and regulate gastro-intestinal hormone,with cure effect better than propranolol.
