ABSTRACT

【Objective】 To identify the key factors that regulate the physical vacuum stirring deoxygenation process of bovine hemoglobin and evaluate the stabilities of polyHb solution with different oxygenation degree. 【Methods】 The effects of several factors including gas liquid specific phase boundary area, pH values and polyHb concentrations on deoxygenation process were investigated through monitoring the oxygen saturation of polyHb solution. Furthermore, the MetHb contents of the studied polyHb solutions were measured at regular intervals during the experiments to evaluate the molecular stability of the polyHb through monitoring the oxidation degrees of the experimental Hb solutions under the mimic pasteurevirus inactivation conditions. 【Results】 The results showed that the more gas liquid specific phase boundary area could accelerate deoxygenation rate, the better deoxygenation results could be yielded. On the other hand, lower pH value could promote the deoxygenation process of the polyHb solution due to the decreasing of oxygen affinity of the polyhemoglobin molecules. Furthermore, results of investigation about the Hb concentration presented that Hb concentration had no remarkable effects on the deoxygenation of polyHb. In addition, the results from the stability experiments presented that higher degree of deoxygenation could decrease the oxidation process and thus increase the heat tolerance of the polyhemoglobin. 【Conclusion】 The deoxygenation of polyhemglonbin could improve the stability of the polyhemoglobin and the deoxygenation process could be regulated by changing the gas liquid specific phase boundary area and oxygen affinity. This study could provide reference and preliminary basis for the industrialization and development of HBOCs in the future.

ABSTRACT

ObjectiveTo investigate the effects of lipopolysaccharide (LPS) on mRNA expression of iron metabolism related genes. Methods Ten male mice (2 months) were injected intraperitoneally with lipopolysaccharide(0.5 μg/g). After 6 hours, mice were sacrificed and then sera, liver and spleen were collected. The mice blood routine was measured. The serum iron and total iron binding capacity (TIBC) were determined with reagent kit. The quasi-quantitative reverse transcription polymerase chain reaction (RT-PCR) was performed for mRNA of hepatic hepcidin(HP), ferroportin1(Fpn1), transferrin receptor 1(TfR1) and spleenic HP, Fpn1 and interleukin-6(IL-6). Results The serum iron and TIBC were reduced in mice injected LPS, which exhibited mild anemia(P<0.05) . LPS can increase the expression of hepatic hepcidin and decrease Fpn1 and TfR1 in liver after LPS administration 6 hours(P<0.05). In spleen, IL-6 was upregulated and Fpn1 downregulated(P<0.05). Conclusion LPS can influence serum iron through regulating the mRNA expression of hepatic and spleenic iron metabolism related genes, such as HP, Fpn1 and TfR1.

ABSTRACT

Aquaglyceroporin3(AQP3),a dual function water channel, can transport both water molecule and other small molecular substances such as glycerol.AQP3-mediated glycerol transport in skin is very important for skin function, whose absence can lead to dry skin, decreased elasticity, and impaired barrier function, and whose overexpression is related to higher risk of skin tumorigenesis. The multiple roles of AQP3 in skin function make it a potential target in treatment of skin disease and development of cosmetics. This paper reviews the characteristics, function, application, and research perspective of AQP3.

ABSTRACT

Social rapid progress aggravates air pollution, particulate matter (PM) which obviously affects human health and living quality is the prominent one of pollutants. Particulate air pollutant may increase obviously the incidence of cardiovascular diseases and lung diseases, also has genetic toxicity. Research advance in the field of genetic toxicity of PM has been reviewed in the article.

ABSTRACT

Objective To investigate the changes of iron-related proteins in different sex mice before and after burns. Methods The burn model in mice were prepared with 90 ℃ vapor. Both liver and spleen iron contents were measured by colorimetric method.Hepatic hepcidin and Fpn1 mRNA content were determined with RT-PCR.The Fpn1 protein was determined with Western blot. Results After burns, the content of spleen iron was increased and the expression of hepatic hepcidin was decreased in male mice, and at the same time the Fpn1 of liver and spleen was reduced. The content of iron homeostasis abnormality in female mice was less serious. Conclusion The change of iron-related proteins after burns may be one cause of iron homeostasis abnormality. The differences of iron-related protein expression in different sex after burns suggest that estrogen has a potentially protective function.
