ABSTRACT

Objective To investigate NIMA-related kinase 2 (NEK2) expression in colorectal cancer (CRC) tissues and its effect on the proliferation, migration and invasion of HCT116 cells. Methods Synthesized siRNA targeting NEK2 was transfected into HCT116 cells by lipofectamine method. CCK8 assay, FACS, wound healing assay and Transwell assay were used to detect the effects of NEK2 knockdown on cell proliferation, cell cycle distribution, migration and invasion. Western blot was carried out to detect the expression of E-cadherin, N-cadherin, CDK4 and cyclin D1. Results The expression of NEK2 in CRC tissues was significantly higher than that in adjacent normal tissues (65.0% vs. 35.0%, χ2=14.593, P < 0.01), and its level was closely related to TNM stage, lymph node metastasis and distant metastasis (all P < 0.05). NEK2 protein and mRNA levels in CRC cell lines were also significantly higher than those in normal colorectal mucosal cells (P < 0.01). After NEK2 was knocked down by siRNA, HCT116 cells were arrested in G0/G1 cycle, while cell proliferation, invasion and migration were significantly reduced (P < 0.01). NEK2 interference in HCT-116 cells led to the upregulation of E-cadherin and downregulation of N-cadherin, CDK4 and cyclin D1 at protein levels. Conclusion The high NEK2 expression is correlated with clinicopathological characteristics of CRC patients. NEK2 knockdown by siRNA could effectively inhibit the proliferation, invasion and migration abilities of colorectal cancer cells, suggesting that NEK2 plays an important role in the occurrence and development of colorectal cancer and it can be used as a potential treatment target.

ABSTRACT

AIM: To investigate the mechanism of the involvement of SUMO-ylated Androgen receptor (AR) in tamoxifen resistance and the role of SUMO inhibitor ginkgolic acid in resistance. METHODS: Real-Time PCR was used to detect AR mRNA levels in parental cells MCF-7W and drug-resistant cells MCF-7R, AR protein levels and SUMO levels in MCF-7W and MCF-7R cells was performed by western blot, and CB/IP was applied to detect AR interacts with SUMOE3 ligase PIAS1 and HSP27 in MCF-7R cell chromatin, the transcriptional activity of SUMO AR was also evaluated by the fluorescent reporter gene experiment, the CCK-8 method and the trypan blue exclusion method were used to detect cell viability and cell viability respectively. RESULTS: The mRNA and protein expression levels of AR in MCF-7R cells were significantly higher than those in MCF-7W cells (P<0.05), and there was highly SUMOylated AR in MCF-7R cells. Further research found that there had an obvious interaction between AR and SUMO E3 ligase PIAS1 and HSP27, that was, the SUMOylated AR was modified by E3 ligase. Moreover, androgen R1881 could enhance the transcriptional activity of the SUMOylated AR in a concentration-dependent manner. Compared with ginkgo acid alone, 10 μmol/L of ginkgolic acid combined with 10 μmol/L of enzalutamide treated MCF-7R cells for 3 days, the cell number was significantly reduced, and the number of cell death increased significantly (P<0.05). CONCLUSION: The resistance mechanism of tamoxifen may be due to the enhanced AR transcription and activity increased by the hyperactive SUMOylated AR, SUMO inhibitor ginkgolic acid combined with AR antagonist enzalutamide can be a new strategy for the treatment of tamoxifen resistance.

ABSTRACT

To evaluated the inhibition effect of local injection of odanacatib on orthodontic relapse.Thirty rats were randomly divided into 2 groups(n =15).The maxillary right first molars of all animals were subjected to orthodontic force and moved mesially.Three weeks later,the force was removed,and the teeth relapsed.The rats of experimental group were received local injections of odanacatib.The control group received saline instead.Tooth movement and relapse were measured when appliances removed after 0 day and 2 weeks.The rats were sacrificed after the 2-week relapse.The microcomputed tomography analysis was performed to quantify the alveolar bone,and the values of bone mineral density(BMD) and bone volume fraction(BVF) were measured.The percentage of relapse in the experimental group was significantly lower than that in the control group(P <0.05),and the BMD and BVF of alveolar bone in the experimental group were significantly higher than those in the control group(P <0.05).Local injection of odanacatib effectively inhibit the relapse after orthodontic tooth movement in rats.

ABSTRACT

Objective · To compare the prevalence of tobacco advertisement before and after implementation of the new edition of Advertising Law of the People's Republic of China and evaluate the effects of implementation of the new Advertising Law. Methods · Tobacco sales places, public places, and streets in Shanghai were randomly selected to observe tobacco advertisement and complete the survey. The survey was carried out in 10 districts of Shanghai in June 2015 with 10 tobacco sales places selected in each district, and in 4 districts of Shanghai in June 2016 with 25 tobacco sales places selected in each district. Sales places included convenient stores, tobacco stores, supermarkets, and kiosks. In addition, public transport waiting points, public transport vehicles, hospitals, shopping centers, restaurants, bars, and Internet cafes were selected into survey in 2016. Results · In 2016, the proportion of tobacco sales places having tobacco advertisement was lower than that in 2015, but the proportion of tobacco promotion behaviors was higher. In 2016, 128 public places and streets were investigated and only 1 had outdoor tobacco advertisement. Conclusion · The new edition of Advertising Law has significant effect on controlling tobacco advertisement, but tobacco sales places still have tobacco advertisement with various disguised promotion forms. Tobacco sales places have diversified business styles and minors often enter these places. It is recommended that a clear definition of disguised forms of tobacco advertisement should be made, tobacco advertisement at tobacco sales places needs to be supervised, the release of tobacco advertisement should be strictly reviewed, and existing tobacco advertisement should be severely punished, so as to prevent minors from exposing to tobacco advertisement and information luring smoking.

ABSTRACT

Objective@#To investigate the response to neoadjuvant chemotherapy (NAC) among different molecular subtypes of breast cancers using molecular classification with Ki-67 (ER+ PR+ HER2+ Ki-67) or without Ki-67 (ER+ PR+ HER2).@*Methods@#One hundred and twenty-seven cases of invasive breast cancer confirmed by core needle biopsy before NAC were collected from January 2007 to December 2009 and diagnosed at West China Hospital, Sichuan University. The cases were classified into different molecular subtypes using molecular classifications with or without Ki-67. Their clinical and pathological response to NAC was evaluated and compared.@*Results@#The different subtypes using both molecular classifications showed significant difference in clinical response(with Ki-67: χ2=22.40, P<0.01; without Ki-67: χ2=9.202, P=0.027)but not pathological(P>0.05) response to NAC. By multivariate analysis, Ki-67 was predictive for a clinical complete response (P=0.041) and clinical overall response (P<0.01); also Ki-67 was the only clinicopathological factor predictive of pathological response(P=0.041).@*Conclusion@#The molecular classification with Ki-67 is better to predict breast cancers responsiveness to NAC than the molecular classification without Ki-67.

ABSTRACT

Objective:To investigate the labial(buccal)bone thickness at alveolar crest zone and alveolar crest height of the maxil-lary anterior teeth and premolars of young adults with normal occlusion.Methods:The alveolar bone of the anterior teeth and premo-lars of 67 eligible Han national young volunteers was scanned by CBCT.Then the facial bone thickness and the distance between the facial alveolar crest and Cemento-enamel Junction(CEJ)of the anterior teeth and premolars were measured and analyzed after recon-struction.Results:The distance between labial (buccal)crest and labial (buccal)CEJ of the maxillary first premolars was the lar-gest(P <0.05);there was a negative correlation between the labial(buccal)crest height and the facial alveolar bone thickness at 2 mm from CEJ toward root derection[(P <0.05),0.6 <|r|<0.8].Conclusion:The labial (buccal)crest of the first premolars was higher than that of other teeth in maxillary aesthetic zone.At alveolar crest zone,when the labial (buccal)bone was thinner,the dis-tance between labial (buccal)crest and labial (buccal)CEJ was larger,and the implant aesthetic risk is higher.

ABSTRACT

Objective To investigate thyroid hormone nuclear receptor expression in peripheral blood mononuclear cells (PBMCs) and its relation with injury severity in patients with severe multiple trauma.Methods Twenty-eight patients with severe muhiple trauma and another 26 healthy subjects as control were enrolled in the study.At 2 days after injury,the levels of free triiodothyronine (FT3),free thyroxine (FT4) and thyroid-stimulating hormone (TSH) in peripheral venous blood were determined by using immunoradiometry,and the expressions of mRNA of the thyroid nuclear receptors (TRα and TRβ) in PBMCs were measured by using real-time RT-PCR.Results FT3 concentration in peripheral venous blood was significantly decreased in patients with severe multiple trauma versus healthy subjects.Concurrently,the expressions of TR mRNA (TRα and TRβ) were significantly decreased in PBMCs of patients with severe multiple trauma compared with healthy subjects (TRα mRNA:3.86 ±0.54 vs.5.24 ± 1.17,P <0.05;TRβ mRNA:9.86±2.27 vs.13.57 ±2.45,P <0.05).Pearson correlation analysis showed that FT3 concentration and the expression of TRβ mRNA correlated negatively with injury severity score (ISS) in patients with severe multiple trauma (r=-0.445,-0.496,P=0.018,0.007).Conclusions These data provide the evidence of a lowered activity of the thyroid signaling pathway in PBMC and a significantly negative correlation between FT3 concentration and the expression of TRβ mRNA in respect of injury severity in patients with severe multiple trauma.Additional investigations are needed to further determine the roles of the thyroid signaling pathway in adverse outcomes in the wake of severe multiple trauma.

Objective:To analyze the drug use in a patient with Crohn’ s disease and provide relevant pharmaceutical service by clinical pharmacists. Methods:Clinical pharmacists participated in the treatment of the patient. The medication was analyzed according to the check results and diagnosis and treatment process, and relevant pharmaceutical service was provided for the patient and nurses. Results: Through the participation in the patient treatment and health education for the patient and nurses performed by clinical pharmacists, the patient was recovered and discharged. Conclusion: Clinical pharmacist participating in pharmaceutical service can promote rational drug use in clinics.
