ABSTRACT

Patients with glioblastoma (GBM) generally have a bad prognosis and short overall survival after being treated with surgery, chemotherapy or radiotherapy due to the histological heterogeneity, strong invasive ability and rapid postoperative recurrence of GBM. The components of GBM cell-derived exosome (GBM-exo) can regulate the proliferation and migration of GBM cell via cytokines, miRNAs, DNA molecules and proteins, promote the angiogenesis via angiogenic proteins and non-coding RNAs, mediate tumor immune evasion by targeting immune checkpoints with regulatory factors, proteins and drugs, and reduce drug resistance of GBM cells through non-coding RNAs. GBM-exo is expected to be an important target for the personalized treatment of GBM and a marker for diagnosis and prognosis of this kind of disease. This review summarizes the preparation methods, biological characteristics, functions and molecular mechanisms of GBM-exo on cell proliferation, angiogenesis, immune evasion and drug resistance of GBM to facilitate developing new strategies for the diagnosis and treatment of GBM.

ABSTRACT

ObjectiveTo explore the effects of after-school tutoring for school-age children on their emotions and behaviors, and to provide a basis for developing after-school tutoring and psychological support strategies for school-age children. MethodsFour elementary schools in a district of Shanghai were selected by simple random sampling method. Whole group sampling was conducted by class, and parents of students were surveyed by general questionnaire and the scale of strengths and difficulties. Information was collected on the basic conditions of children and families and the results were compared and analyzed. ResultsThis study showed that 88.26% of children attended extracurricular classes, of which 26.16% attended 3 or more classes, 42.5% attended cultural classes, 28.3% attended sports classes, and 59.8% attended art classes. Children who did not attend classes had higher pro-social scores. Children who attended reading class had lower scores for emotional problems that was a protective factor for emotional problems. Children who chose extracurricular programs in sports and reading also had lower overall scores for difficulties. Excessive use of electronic devices by school-age children on weekdays or weekends had a negative effect on emotional and conduct problem scores regardless of whether they chose extracurricular programs. It appeared that the choice of cultural programs, art programs and different levels of exercise intensity had no effect on children's emotions and behaviors. ConclusionFor children with low pro-social level, extracurricular classes should be carefully selected. For extracurricular classes, more reading and sports-related courses can be considered, which have positive effects on school-age children’s emotions and behaviors. Regardless of the choice of extracurricular classes, try to avoid excessive online classes.

ABSTRACT

Objective@#To determine the association between anthropometric parameters and 20 meter shuttle run test (20 m SRT) score among children and adolescents.@*Methods@#The convenient sampling method was conducted to select 3 192 primary and secondary school students in Baoan District, Shenzhen, based on the street school types school from April to May 2019. Height, weight, 20 m SRT score and general demographic indicators were assessed and collected. Individuals were divided into two groups based on the dose response correlation between different anthropometric parameters (the Z score of height, weight and BMI) and 20 m SRT score analyzed with the restricted cubic spline. The association between the Z score of height and 20 m SRT score was further analyzed using the Spearman correlation analysis. Logistic regression analysis was used to analyze separately the relationship different anthropometric parameters and 20 m SRT score.@*Results@#The Z value of weight and 20 m SRT score showed a non linear dose response association ( P <0.01), the significant but weak linear correlation between the Z value of height and 20 m SRT score ( r=0.06, P <0.05). The prevalence rate on the low level of the Z value of 20 m SRT score in 3 192 children and adolescents was 44.7%, and the gender ( χ 2=14.02, P <0.01) and grade difference ( χ 2=93.28, P <0.01) were both statistically significant. There was no significant relationship between the Z value of height and 20 m SRT score grade among total population, different genders and different grades ( P > 0.05). Compared with the reference group on the Z value of weight ≤-0.23, individuals with the Z value of weight >-0.23 had the low level of 20 m SRT score ( OR =0.61, P <0.05). Compared with the reference group on the Z value of BMI ≤ 0.25, individuals with the Z value of weight >0.25 had the low level of 20 m SRT score ( OR =0.45, P <0.05). Stratified for gender and grade, the above significant relationship on the Z value of weight, Z value of BMI and 20 m SRT score were still observed ( P <0.01).@*Conclusions@#The higher height Z value shows on correlations with 20 m SRT score, but the positive association is found between weight and BMI Z value and the 20 m SRT score. The cardiopulmonary fitness improvement may be more effective among children and adolescents when tuking weight and BMI Z scores into consideration.

ObjectiveTo compare children's resilience and peer relationship in divorced and complete family,and explore the moderating effect of psychological resilience between family structure and peer relationship.Methods208 children from divorced family and 465 children from complete family were surveyed.Results(1)There were significant differences in children's resilience(divorced family (3.58±0.73),complete family(3.83±0.59),t=-2.765,P=0.006) and peer relationship(divorced family (3.47±0.72),complete family(3.73±0.59),t=-2.069,P=0.042) between complete and divorced family.(2) Family structure had remarkable negative correlation with resilience and peer relationship(r1=-0.19,r2=-0.20),and resilience had positive correlation with peer relationship(r=0.43).(3)The moderating effect of resilience between family structure and peer relationship was significant(β=0.641,P<0.05).ConclusionParent divorce has negative influence on children's resilience and peer relationship.Furthermore,the relationship of family structure and peer relationship is mediated by resilience.The negative influence of parent divorce on peer relationship is not significant among children with high resilience,while it is significant on children with low resilience.

ABSTRACT

BACKGROUND:Human umbilical cord mesenchymal stem cells (hUC-MSCs) may be mutated duringin vitro culture based on the spontaneous malignant transformation of adult stem cells and tumor stem cell theory, and there may be a risk of tumorigenesis after in vivo transplantation. Therefore, to establish and perfect the in vitro safety testing procedures will actively promote the clinical application of stem cells. OBJECTIVE:To investigate the tumorigenic mechanism of hUC-MSCs and the expression level of DNA methyltransferase (DNMTs) in hUC-MSCs. METHODS:Primary hUC-MSCs were isolated and expanded by tissue adherent culture. 3-Methycholanthrene was used to cause the malignant transformation in hUC-MSCs (experimental group), followed by morphological observation and tumorigenesis experiment in nude mice. Then, the tumor tissues were obtained and identified by pathological examination and primary cell culture, and the levels of DNMTs mRNA in hUC-MSCs treated with 3-methycholanthrene and dimethyl sulfoxide (control group) were detected by real-time RT-PCR and compared. RESULTS AND CONCLUSION:hUC-MSCs treated with 3-methycholanthrene led to malignant transformation, which showed malignant growth and non-integer ploidy changes in the cell nuclei, and formed a malignant tumor in immune-deficient mice after injection. Compared with the control group, the cells in the experimental group showed higher expression of DNMTs mRNA as detected by real-time RT-PCR. To conclude, hUC-MSCs can trigger malignant transformation in the morphology and the epigenetics under certain conditions. DNMTs can be a candidate for prevention against malignant transformation of transplanted stem cells.

METHODS:Based on specific cases of nephrolo-gy department,the thoughts and ways of clinical pharmacist in nephrology department to review medical orders were investigated from several aspects,including whether the therapeutic drugs had covered all the problems,drug duplication,drug selection,usage and dosage and course of treatments,drug interaction and compatibility. RESULTS & CONCLUSIONS:By establishing rigorous and systematic review thinking mode,based on their professional specialty,clinical pharmacists of nephrology department can sort the reviewing thought,fill in the gaps of medical orders reviewing and enhance the ability of participating in the clinical practice.

ABSTRACT

Objective To investigate the intervention effects of NLP workshop on resilience of children from divorced family.Methods A pre/post test and subsequent follow-up design between experimental group and control group were performed.Thirty-one children from divorced family with low resilience were intervened by NLP workshop,while the other thirty-one children with low resilience were chosen as the control group.Results There was no significant difference between experimental group and control group before the intervention on resilience.But after the intervention,the two groups achieved statistical significant difference on resilience(experimental group (3:68±0.37),control group(2.99±0.18),t=9.60,P=0.00).Furthermore,the results of follow-up showed that the effect of NLP workshop still existed after six months (total average score of resilience before intervention (2.96±0.17),subsequent follow-ups (3.69±0.35),t=10.40,P=0.00).Conclusion NLP workshop can enhance the level of resilience of children from divorced family.Effect of NLP workshop not only has short-term effect,but also has some long-term effect.

ABSTRACT

OBJECTIVE To establish a method of loop-mediated isothermal amplification(LAMP) for detecting Staphylococcus aureus in positive blood culture bottles.METHODS Genomic DNA in 293 positive blood culture bottles was extracted by guanidine hydrochloride and benzenemethanol,then genes ssa and mecA were amplified by LAMP to identify S.aureus.Finally,the results of LAMP were compared with the results of traditional method.RESULTS Twenty-two strains of S.aureus were detected in 293 positive blood culture bottles by LAMP method.Compared with traditional method,the sensitivity and specificity of LAMP method were both 100%,respectively,the detection could be finished in an hour.CONCLUSIONS The LAMP-based assay is simple,rapid,sensitive and specific which can be used to detect S.aureus in positive blood culture bottles rapidly.

ABSTRACT

Recent scientific achievements in cell and developmental biology have provided unprecedented opportunities for advances in biomedical research. The demonstration that fully differentiated cells can reverse their gene expression profile to that of pluripotent cells, and the successful derivation and culture of human embryonic stem cells (ESCs) have fuelled hopes for applications in regenerative medicine. Ethical issues concerning the use of cloned human embryos for the derivation of stem cells have stimulated the search for alternative methods for reversing differentiated cells into pluripotent cells. The present state of these reprogramming technologies will be reviewed and their relative success will be discussed.

ABSTRACT

OBJECTIVE To establish a method of polymerase chain reaction(PCR) for detecting staphylococci in positive blood culture bottles.METHODS Genomic DNA in 493 positive blood culture bottles was extracted by guanidine hydrochloride and benzenemethanol,then genes 16S rRNA,ssa and mecA were amplified by PCR to identify staphylococci.Finally,the results of PCR were compared with that of traditional method.RESULTS To compare with traditional method,as the golden standard the sensitivity and specificity of PCR method were 98.6% and 100.0%,respectively,the detection could be finished in four hours.Method of PCR was better than traditional method in detecting meticillin-resistant staphylococci.CONCLUSIONS The PCR-based assay is simple,rapid,sensitive and specific,it can be used to detect staphylococci in positive blood culture bottles.

ABSTRACT

Objective:To analyze the effect of interleukin 2(IL-2) on immunoglobulin G(IgG) expression in cervical cancer cell line HeLaS3.Methods:By immunofluorescence-flow cytometry(FCM),we detected membrane and cytoplasmic IgG expression variation of HeLaS3 after IL-2 stimulation.By western blot,we detected IgG expression variation of HeLaS3 after IL-2 stimulation.Meanwhile,by RT-PCR with primers designed to amplify four subclasses of IgG,we detected ? transcript variation in HeLaS3 after IL-2 stimulation at mRNA level.Results:The result of immunofluorescence-FACS showed that after IL-2 stimulation for 48 hours,membrane and cytoplasmic IgG expression elevated in HeLaS3.The result of Western blot showed after IL-2 stimulation for 48 hours,IgG expression elevated in HeLaS3.Meanwhile,the result of RT-PCR showed that ?1 transcript mildly elevated after IL-2 stimulation for 48 hours.Conclusion:IL-2 can promote IgG expression in cervical cancer cell line HeLaS3.

Objective:To investigate the suppression effect and apoptosis of the colon cell line HT-29 treated with adenovirus-expressed human hyperplasia suppressor gene(hHSG)and hyperthermia.Methods:Replication-defective recombinant adenovirus containing hHSG was constructed,and then infected into HT-29 cell line to detect its overexpression by western blot.The synergic effect with hyperthermia was observed,cell survival was assessed using Vi-CELL TMXR Cell Viability Analyzer,and cell apoptosis and cell cycle distribution were determined by flow cytometry.Results:After the cells were treated with 100 MOI(multiplicity of infection)Ad5-hHSG and heated at 42 ℃ for 1 h,the number of viable cells decreased sharply,compared with single treatment with Ad5-hHSG or 42 ℃ for 1 h(P

Background and Purpose:Prostate cancer is one of the most common cancers and the second leading cause of cancer-related death in Europan and North American males.The incidence of prostate cancer has also been increasing during the past few decades in China.It is widely accepted that this heterogeneity,which results from the tumor progression driven largely by genomic instability(genetic and/or epigenetic alterations)of tumor cells in primary tumor,endows specific populations of tumor cells with the unique character needed for invasion,migration,and metastasis colony formation in other organs and only these subpopulations possessing thost character can survive the potentially destructive journey from the primary tumor to the sites of metastases.The purpose of the present study was to explore the genes associated with invasion and metastasis of human prostate cancer cell line PC-3M in nude mice.Methods:After PC-3M cells were inoculated into orthotopic site(prostate) in a male nude mouse for two months,tumor cells were isolated from the primary tumor and lymph node metastasis,separately.Cell invasion and adhesion ability in vitro were first compared between two cells.Then metastasis-related genes differentially expressed between them were analyzed by utilizing cDNA microarray technique.Results:The in vitro cell invasion and adhesion potential of tumor cells from lymph node metastasis was significantly higher than those from primary tumor by 2.5 fold and 1.5 fold,respectively.Metastasis-related genes differentially expressed between those two sublines were identified,all of them were up-regulated in the tumor cells from lymph node metastasis and could be categorilized: 1.genes encoding cellular matrix-degrading proteolytic enzyme including cathepsin and MMP.2.genes encoding transcription factors.3.genes related to heterotypic adhesion of tumor cells.4.genes encoding cell surface receptors.Conclusions:There are significant differences in invasion and adhesion potential between cells from primary tumor and those from lymph node metastasis.Some differentially expressed molecules might be playing pivotal roles in promoting tumor cells to migrate from primary tumors to distant metastases,which may be helpful to elucidate the possible mechanism of metastasis in prostate cancer.
