ABSTRACT

With the development of laparoscopic techniques, the requirement of fine dissection in surgeons is also growing. Laparoscopic technique has the advantages in the choice of anatomical level and nerve vascular protection for surgery. 4K laparoscopy provides more clear field of vision and more detailed anatomic prerequisites. Combined with surgical examples, the authors share experiences in protection of plant nerve during 4K laparoscopic radical resection of colorectal cancer.

ABSTRACT

With the development of laparoscopic technology, surgeon's requirements for precise anatomy are gradually increasing. Laparoscopic system has great advantages in the selection of anatomical level, the protection of nerve and blood vessel in surgery. 4K laparoscopic system can provide high-definition vision of operative field, which is conducive to precise anatomical operation. The authors share the practical team cooperation experience of laparoscope left hemi-colectomy for colon cancer in order to provide references for surgical colleagues.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the effects of different treatments on the prognosis of patients with synchronous colorectal liver metastasis(CLM).</p><p><b>METHODS</b>Clinicopathological data of 121 patients with synchronous CLM in The First Affiliated Hospital of Xinjiang Medical University between January 2010 and December 2014 were retrospectively analyzed. According to the metastatic lesions, patients were divided into simple operation group(22 patients, receiving operation only), simple chemotherapy group(43 patients, receiving chemotherapy only), and combination group(56 patients, receiving chemotherapy based on operation). The prognosis of patients in three groups was investigated. Univariate and multivariate analyses were performed through the long-rank test and Cox model to evaluate the related factors affecting prognosis.</p><p><b>RESULTS</b>The median survival time was 10(3-39) months in simple operation group, 7(3-36) months in simple chemotherapy group, and 18 (4-66) months in combination group. The differences among groups were all statistically significant (all P<0.05). Extent of lymph node metastasis, number of liver metastatic lesion, size of liver metastatic lesion, resection edge extent of liver metastatic lesion, and treatment method were independent factors of synchronous CLM(all P<0.05).</p><p><b>CONCLUSION</b>Combination therapy of a variety of treatment can prolong the survival of patients with synchronous CLM.</p>

ABSTRACT

BACKGROUND:The metastasis ways of colon cancer include blood metastasis, peritoneal metastasis and lymph node metastasis. Among them, the most common metastasis organ is the liver. It is necessary to establish a simple, suitable animal model to study the biological behavior of the colon cancer liver metastasis, the heterogeneity of cancer cells, and the mechanism of tumor metastasis. OBJECTIVE:To establish animal model of colon cancer liver metastasis with splenectomy injection method. METHODS:Human colon cancer celllines HT29 were cultured, and subcutaneously transplanted tumor was produced in BALB/c mice. Models of colon cancer liver metastases were established through spleen injection and excision of spleen method. The tumor growth and metastasis, the pathology of transplanted tumor, DNA content and cellcycle of the transplanted tumor were al determined. RESULTS AND CONCLUSION:In this study, we successful y established the model of colon cancer liver metastases through the spleen injection via the dorsal midline incision, the tumor formation rate reached 8/8 (100%). Observation of tissue slice showed that the transplanted tumor was adenocarcinoma I-II. Flow cytometry analysis results showed that DNA ploid was heteroploid DNA. This model provides a good technical platform for biology research for colon cancer metastasis.

ABSTRACT

BACKGROUND:Adipose stem cells can differentiate into bone cells, cartilage cells, smooth muscle cells, myocardial cells and nerve cells. Adipose-derived stem cells and mesenchymal stem cells have very similar biological properties, and they are also very close in terms of cellsurface marker expression profile. OBJECTIVE:To investigate the expression of human leukocyte antigen II in adipose-derived stem cells and its immunogenicity and immunomodulatory effect, and to explore the specific mechanism. METHODS:After isolation and culture, the adipose-derived stem cells were stimulated with interferon-γ, and the expression of human leukocyte antigen II was detected by flow cytometry, the expression of indoleamine 2,3-dioxygenase was determined by real-time PCR and western blot. Adipose-derived stem cells were used to stimulate lymphocytes, and the cellproliferation was observed. The lymphocytes were stimulated with phytohemagglutinin, and then 1×102-1×105 adipose-derived stem cells were added to inhibit the proliferation;meanwhile, adipose-derived stem cells, a third party, at a dose of 1×102-1×105 were added to the ongoing two-way mixed lymphocyte reaction, and the cellnumber was calculated as count per minute. RESULTS AND CONCLUSION:The expression of human leukocyte antigen II and indoleamine 2,3-dioxygenase was low in adipose-derived stem cells, but up-regulated with stimulation of interferon-γ. Adipose-derived stem cells could not stimulate lymphocyte proliferation, but could inhibit lymphocyte proliferation induced by phytohemagglutinin or al ogeneic lymphocytes, and interferon-γwas highly expressed in the supernatant of co-culture of adipose-derived stem cells and mixed lymphocytes. These results indicate that the adipose-derived stem cells have low immunogenicity and can inhibit cellproliferation of lymphocytes in vitro, and the addition of exogenous interferon-γcan enhance the immunosuppressive effects of adipose-derived stem cells.
