ABSTRACT

ObjectiveTo explore the differences in the protective effects of five formulas for promoting blood circulation and removing blood stasis on the aortic endothelial cells of New Zealand rabbits with heart blood stasis syndrome. MethodEighty New Zealand rabbits were randomly divided into a normal group (n=10) and an experimental group (n=70). The heart blood stasis syndrome model was induced by starvation combined with a high-fat diet and adrenaline in the rabbits of the experimental group. Subsequently, the model rabbits were randomly divided into a model group, a Xuefu Zhuyutang group (3.55 g·kg-1·d-1), a Taohong Siwutang group (2.66 g·kg-1·d-1), a Danshenyin group (1.962 g·kg-1·d-1), a Huoluo Xiaolingdan group (2.80 g·kg-1·d-1), a Shixiaosan group (0.56 g·kg-1·d-1), and a c-Jun N-terminal kinase (JNK) inhibitor (SP600125, 5 μg·kg-1)group. The normal group and the model group received the same amount of distilled water. The rabbits in five Chinese medicine groups were treated correspondingly by gavage, and those in the SP600125 group were injected with 0.5 mL of SP600125-dimethyl sulfoxide diluent. After the treatment, the aorta was collected, and terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay was used to detect the apoptosis of aortic endothelial cells. The enzyme-linked immunosorbent assay (ELISA) was used to detect serum levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6). Western blot was used to detect the protein expression of JNK, phosphorylated JNK (p-JNK), B-cell lymphoma-2 (Bcl-2), Bcl-2-associated X protein (Bax), cysteinyl aspartate-specific protease-9 (Caspase-9), and cysteinyl aspartate-specific protease-3 (Caspase-3) in aortic tissues. Real-time fluorescence-based quantitative polymerase chain reaction (Real-time PCR) was used to detect the mRNA levels of JNK, Bcl-2, Bax, Caspase-9, and Caspase-3 in aortic tissues. ResultFive formulas could improve the apoptosis of aortic endothelial cells to varying degrees. To be specific, Xuefu Zhuyutang and Taohong Siwutang were optimal in efficacy, followed by Huoluo Xiaolingdan, Shixiaosan, and Danshenyin, and SP600125 was the worst (P<0.05, P<0.01). Five formulas could reduce the content of TNF-α and IL-6 (P<0.05, P<0.01), down-regulate the protein expression levels of JNK, p-JNK, Bax, Caspase-9, and Caspase-3 (P<0.05, P<0.01), decrease the mRNA expression levels of JNK, Bax, Caspase-9, and Caspase-3 (P<0.05, P<0.01), and up-regulate the protein and mRNA expression levels of Bcl-2 (P<0.05, P<0.01). ConclusionFive formulas can all reduce the apoptosis of aortic endothelial cells in New Zealand rabbits with heart blood stasis syndrome with different efficacies. It may be related to the different effects of five formulas on the JNK signaling pathway.

ABSTRACT

Objective:To establish the high performance liquid chromatography (HPLC) fingerprint of Citri Sarcodactylis Fructus, and to search for makers to characterize the quality difference of Citri Sarcodactylis Fructus from different origins coupled with chemometrics. Method:The analysis was performed on a Thermo Hypersil GOLD C18 column (4.6 mm×250 mm, 5 μm) with mobile phase consisted of acetonitrile-0.05% phosphoric acid solution for gradient elution, and the detection wavelength was set at 254 nm. A total of 31 batches of samples were analyzed to establish the HPLC fingerprint of Citri Sarcodactylis Fructus. Similarity evaluation was performed by Traditional Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System (2012 edition) to confirm the common peaks, which were identified by comparison of reference substances. On the basis, chemometrics methods were used to analyze and evaluate the quality of Citri Sarcodactylis Fructus from different origins. At the same time, 3 batches of 5 species of decoction pieces from the genus Citrus in the family Rutaceae, including Citri Sarcodactylis Fructus, Aurantii Fructus Immaturus, Aurantii Fructus, Citri Reticulatae Pericarpium Viride and Citri Reticulatae Pericarpium, were randomly collected for evaluating the effectiveness and reliability of the established HPLC fingerprint of Citri Sarcodactylis Fructus. Result:HPLC fingerprint of Citri Sarcodactylis Fructus was established and 22 common peaks were identified. And seven common peaks among them were identified as 6,7-dimethoxycoumarin, diosmin, hesperidin, byakangelicin, 5,7-dimethoxycoumarin, bergapten and oxypeucedanin. Except for 2 batches of samples, the similarities of fingerprints between other 29 batches of samples were >0.9. The 31 batches of Citri Sarcodactylis Fructus were basically divided into 3 groups by cluster analysis and principal component analysis, which were consistent with the classification of three different producing areas. Eight differential markers were screened by orthogonal partial least squares discriminant analysis and four of them (5,7-dimethoxycoumarin, bergapten, 6,7-dimethoxycoumarin and diosmin) were identified by reference substances. Similarity evaluation of 5 species of decoction pieces from genus Citrus in the family Rutaceae was carried out by taking the reference fingerprint of Citri Sarcodactylis Fructus as treference chromatogram, similarity of Citri Sarcodactylis Fructus decoction pieces was 0.892-0.977, and the similarities of the other 4 kinds of decoction pieces were 0.215-0.517. Conclusion:The established fingerprint method is reasonable, effective and accurate for quality control of Citri Sarcodactylis Fructus, the characterization information is more comprehensive combined with chemometrics.

ABSTRACT

BACKGROUND@#The clinical trials emerged centromere protein E inhibitor GSK923295 as a promising anticancer drug, but its function in hepatocellular carcinoma (HCC) remain needs to be fully elucidated, especially as chemotherapy after hepatectomy for liver tumors. We aimed to describe anti-HCC activities of GSK923295 and compare its antiproliferative effects on liver regeneration after partial hepatectomy (PH).@*METHODS@#All subjects were randomized to treatment with either vehicle or GSK923295. Antitumor activity of GSK923295 was assessed by xenograft growth assays. The C57BL/6 mice were subjected to 70% PH and the proliferation was calculated by liver coefficient, further confirmed by immunohistochemistry. The proliferation and cell cycle analysis of liver cell AML12 and HCC cells LM3, HUH7, and HepG2 were investigated using the cell counting kit-8 assay and Flow Cytometry. The chromosome misalignment and segregation in AML12 cells were visualized by immunofluorescence.@*RESULTS@#Treatment with GSK923295 induced antiproliferation in HCC cell lines. It also caused delay on HCC tumor growth instead of regression both in a HCC cell line xenograft model and patient-derived tumor xenograft model. With microarray analysis, CENtromere Protein E was gradually increased in mouse liver after PH. Exposure of liver cells to GSK923295 resulted in delay on a cell cycle in mitosis with a phenotype of misaligned chromosomes and chromosomes clustered. In 70% PH mouse model, GSK923295 treatment also remarkably reduced liver regeneration in later stage, in parallel with the mitotic marker phospho-histone H3 elevation.@*CONCLUSION@#The anticancer drug GSK923295 causes a significant delay on HCC tumor growth and liver regeneration after PH in later stage.

ABSTRACT

Aim To investigate the antidepressant effect of imperatorin(IMP) and explore its mechanism. Methods The model of depression was established by prenatal stress (PS) on late pregnant mother, then male offspring rats were randomly divided into five groups: control, PS model, fluoxetine group(5 mg • kg"1), IMP( 15, 30 mg • kg"1). Sucrose preference test, forced swimming test, and open field test were used to evaluate the behavioral changes. 5- hydroxytryptamine ( 5-HT ) concentration, serotonin transporters(5-HTT) and 5-HT,A receptor(5-HT,AR) mRNA expression in the hippocampus and frontal cortex were measured using an enzyme-linked immunosorbent assay and real-time PCR. Results Compared with PS group, the percentage of sucrose preference and the number of total crossings, center crossings, rearing, grooming and 5-IIT concentration significantly increased in IMP group(/> <0. 01). The im-mobility time was significantly reduced in IMP group compared with PS group ( P < 0. 01). The 5-HT concentration and 5-HT1A R mRNA expressions of PS offspring rats significantly increased by the administration ofIMP (P<0.05,P<0.01). And, the increased 5- HTT mRNA markedly decreased in IMP group compared with PS group (P < 0. 05 , P < 0. 01 ). Conclusions IMP can obviously improve rat behavior and show anti-depressant effect, which may relate to the 5- HT concentration, 5-HTT and 5-HT,AR mRNA expression in hippocampus and prefrontal cortex.

ABSTRACT

Aim: To investigate the protective effect of Rhodiola rosea extract on alveolar bone injury in diabetic osteoporosis rats. Methods: A diabetic rat model was established by intraperitoneal injection of streptozo-tocin 60 mg · kg-1. After successful establishment of the model, the animals were divided into control group, model group and Rhodiola rosea group. Rhodiola rosea extract was administered orally for eight weeks at a dose of 150 mg · kg-1. The levels of blood glucose, serum ALP, Ca, OPG and RANKL were measured, and the ratio of OPG to RANKL was calculated. At the same time, the alveolar bone histopathology was analyzed, and the number of osteoclasts were counted. Results: After treated with Rhodiola rosea extract for eight weeks, the blood glucose, the levels of serum ALP and RANKL in Rhodiola rosea group were significantly lower than those in model group (P < 0. 01). The serum levels of Ca, OPG and the ratio of OPG/RANKL significantly increased in Rhodiola rosea group compared with those of model group (P < 0. 05). Histological examination showed that alveolar bone trabecule in model group were sparse and fractured, bone resorption was serious, and alveolar bone structure was restored after Rhodiola rosea extract administration. The number of osteoclasts in model group increased significantly, and the number of alveolar osteoclasts decreased after Rhodiola rosea extract administration. Conclusions: Rhodiola rosea extract has protective effect on early alveolar bone injury in diabetic rats, and its mechanism is mainly by regulating ALP and OPG/RANKL/RANK system, and accordingly promoting bone formation and inhibiting bone resorption, and balancing bone turnover.

ABSTRACT

Purpose To investigate the clinical manifesta-tions and morphologic features of placental site nodules (PSNs), and its clinical significance. Methods Twenty patients diag-nosed as PSNs were collected, then a retrospective analysis was conducted, and the characteristics of clinical data and follow-up results were analyzed, including of clinical manifestations, ultra-sonographic evaluation, morphologic and immunohistochemical features. Results The age of patients ranged from 25 to 41 years (32. 48 ± 4. 77 years in average). Three fifths of patients had pregnancy history for at least two times and the interval time to the last pregnancy ranged from 5 to 37 months (15. 33 ± 8. 05 months on average). 15 (75% ) patients went to the hospital because of abnormal vaginal bleeding. In our study, most of the samples showed a membrane-like structure without definite nod-ule. Microscopically, single or multiple, well-circumscribed and oval small nodules were found in endometrial tissue. In most ca- ses, the hyalinization was generally uniform in the center of the nodules, more or less intermediate trophoblasts appeared on the edge of the nodules. Immunohistochemically, the strong diffuse expressed CK (AE1/AE3), CAM5. 2, EMA, GATA-3, Cyclin E and p63 were detected in most of all cases, and PLAP showed strong focal expression, α-inhibin and hPL showed faint focal expression, Ki-67 staining for proliferative index was less than 4% . Conclusion PSN is a benign lesion of the intermediate trophoblast at the chorionic leave. Some diseases including hya-linized decidua, epithelioid trophoblastic tumor, and squamous cell carcinoma with hyalinization need to be identified. Some im-munohistochemical markers may be certain helpful in distinguis-hing as necessary.

ABSTRACT

Objective@#To investigate the role of mast cells in chronic prostatitis / chronic pelvic pain syndrome (CP/CPPS).@*METHODS@#Forty-five male SD rats were equally randomized into a control, an experimental autoimmune prostatitis (EAP) model, and an intervention group. The EAP model was made in the latter two groups by subcutaneous injection of mixed suspension of complete Freund's adjuvant and prostate tissue, while the controls were treated subcutaneously with 0.9% sodium chloride. Tactile allodynia was quantified in the pelvic region of the control and EAP animals using Von-Frey filaments at 5, 10, 20, 30 and 40 days. After successful establishment of the EAP model, the rats of the intervention group were injected intraperitonieally with cromolyn sodium for 10 days, and meanwhile tactile allodynia was detected in the rats of the intervention and EAP model groups every other day. Then the prostates of the rats were harvested for HE and toluidine blue staining and measurement of the expression of mast cell tryptase by immunohistochemistry and Western blot.@*RESULTS@#Von-Frey assessment showed a more severe pelvic pain in the EAP model than in the control rats, but milder in the intervention group than in the EAP models. HE staining revealed infiltration of lymphocytes and neutrophils in the prostate and congestion surrounding the gland in the EAP model rats, but none in the controls. However, both the infiltration and congestion were significantly alleviated in the intervention group. Toluidine blue staining shown that. Compared with the control group, the total count of mast cells and the number degranulated mast cells were markedly increased in the EAP models (P <0.01) but decreased in the intervention group (P <0.05). Both immunohistochemistry and Western blot manifested that the expression of tryptase in the mast cells was remarkably upregulated in the EAP (both P <0.01) but down-regulated in the intervention group (P <0.05 and P <0.01).@*CONCLUSIONS@#Both the total count of mast cells and the number of degranulated mast cells are significantly increased in the prostate of EAP rats. Mast cells are one of the most important mediators of type Ⅲ prostatitis-induced chronic pelvic pain, which can be used as a target for the intervention and treatment of type Ⅲ prostatitis.

Cone-beam computed tomography(CBCT) was used to analyze the anatomic morphology of maxillary canine fossa of different sagittal skeletal patterns patients;implants of various dimensions were virtually placed into the maxillary premolars region. The relationships among maxillary canine fossa and implant were analyzed. The depth of the maxillary canine fossa was(4.33±0.73),(3.77±0.58),(5.18±0.93)mm in the 1st premolar region of class Ⅰ,Ⅱ and Ⅲ respectively. The depth of the maxillary canine fossa was(3.20±0.63),(2.81±0.58),(3.90±0.79)mm in the 2nd premolar region of class Ⅰ,Ⅱ and Ⅲ respectively. There were no significant statistical differences in genders, sides, and loss of tooth. And there were significant statistical differences in three classes(P<0.05).The length of the implants was (10.30±1.70)mm(d=3.3 mm) and (8.77±1.58)mm(d=4.1 mm) in the 1st premolar region and (8.09±1.51)mm(d=3.3 mm),(6.69±1.35)mm(d=4.1 mm) in the 2nd premolar region in patients with teeth and buccal perforation;the length of the implants was (8.98±1.54)mm(d=3.3 mm) and (7.67±1.52)mm(d=4.1 mm) in the 1st premolar region and(7.09±1.59)mm(d=3.3 mm) and (5.79±1.34)mm(d=4.1 mm) in the 2nd premolar region in patients with tooth loss, respectively. There were significant statistical differences in loss of tooth(P<0.05).Analyzing of spatial relationships among maxillary canine fossa and implants of different sagittal skeletal patterns patients especially class Ⅲ patients,by using CBCT has important significance in guiding dental implants.

ABSTRACT

Objective To investigate the effect of percutaneous cardiac intervention (PCI) on the clinical prognosis in aged patients with coronary heart disease (CHD) involving multiple coronary arteries who obtained complete revascularization (CR) or incomplete revascularization (ICR) after PCI.Methods A total of consecutive 257 aged patients (>75 years old) with confirmed CHD that involved multiple coronary arteries, who were admitted to authors' hospital during the period from January 2015 to September 2015 to receive PCI,were enrolled in this study.Based on the complete revascularization (CR) or incomplete revascularization (ICR) after PCI,the patients were divided into CR group and ICR group. The basic clinical data,PCI parameters and the occurrence of major adverse cardiovascular and cerebrovascular events (MACCE) in hospitalization days and in the follow-up period were compared between the two groups. Results CR group included 171 patients (66.53%) and ICR group included 86 patients (33.47%).The hypertension history, diabetes history, diagnosis of acute non-ST segment elevation myocardial infarction or myocardial occlusion disease at admission, chest tightness, palpitation and other discomfort symptoms occurring in one and 3 months after PCI,and re-hospitalization rate in ICR group were significantly higher than those in CR group (P<0.05).No statistically significant differences in the incidence of MACCE during hospitalization days and at one,3 and 6 months after PCI existed between the two groups (P>0.05).Conclusion In aged patients with CHD that affects multiple coronary arteries,ICR does not increase the risk of MACCE after PCI,although the re-hospitalization rate and the incidence of postoperative discomfort symptoms will be increased. The long-term prognosis needs to be further studied.
