ABSTRACT

Objective@#To investigate the expression of LIM domain binding 2 (LDB2) in lung cancer tissues and its correlation with sphingosine-1 phosphate receptor 1 (S1PR1). @*Methods@#Lung cancer tissues and the corresponding adjacent tissues from 52 patients in Nantong Tumor Hospital during April 2010 and May 2011 were collected as the experimental group and the control group, respectively. The expression levels of LDB2 and S1PR1 were detected by the real-time PCR (qRT-PCR). The expression results of LDB2 gene were further verified by the Oncomine database, and its correlations with clinicopathological parameters were analyzed. The ROC curve was drawn to evaluate the diagnosis value of LDB2 expression in lung cancer. The correlation of LDB2 expression with the prognosis of lung cancer was analyzed by the “Kaplan-Meier Plotter” database. In addition, the relationship between LDB2 and S1PR1 was also analyzed. @*Results@#The expression levels of LDB2 in lung cancer tissues (0.158 [0.062,0.383]) were significantly lower than that in the adjacent tissues (0.403 [0.261,0.711], U=700.0, P< 0.01). A total of 9 eligible studies were retrieved from the Oncomine database, and their expressions of LDB2 were also low (P<0.01). The expressions of LDB2 in lung cancer tissues were not related to gender, age, smoking history, pathological type, tumor size, TNM staging and lymphatic metastasis (P>0.05). The results of ROC curve showed that when the area under the ROC curve (AUC ROC ) was 0.741 (95% CI:0.643-0.839) and the cut-off value was 0.247, the sensitivity and specificity of LDB2 in the diagnosis of lung cancer were 80.8% and 61.5%, respectively. The Kaplan-Meier survival analysis showed that the 5-year overall survival time of the patients with low expression of LDB2 was shorter than that of the patients with high expression of LDB2(P<0.01). In addition, the expression levels of S1PR1 in lung cancer tissues (0.710[0.337,1.523]) were significantly lower than that in the adjacent tissues (1.582[0.913,3.533],U=780.0, P<0.01), and the expression levels of S1PR1 in lung cancer tissues were positively correlated with that of LDB2(r=0.827,P<0.01). @*Conclusion@#The expressions of LDB2 and S1PR1 in lung cancer tissues are down-regulated, and have a positive correlation, and they may play an important role in the occurrence and development of lung cancer.

ABSTRACT

Objective@#To explore the diagnostic value of serum cell-free DNA (cfDNA) concentration and integrity for esophageal carcinoma.@*Methods@#Venous blood samples from 68 patients with esophageal cancer, 36 patients with benign esophageal lesions and 45 healthy subjects were collected. Circulating cfDNA was verified through quantitative real-time PCR (Alu-qPCR) using Alu-115 and Alu-247 primers. DNA integrity index was calculated as the ratio of Alu-qPCR results (Alu247/115). Concentrations of carcino-embryonic antigen (CEA) and squamous cell carcinoma associated antigen (SCC) were detected by chemiluminescence analyzer assay. Statistical analysis was performed using Mann-Whitney U test, Kruskal-Wallis H test and Spearman correlation test. The Receiver operating characteristic (ROC) curve was used to evaluate the diagnostic efficiency of each index to esophageal carcinoma.@*Results@#The median absolute serum Alu115 and the Alu247/115 index (1 162.0 ng/ml, 0.57) in esophageal cancer group were significantly higher than those in benign esophageal disease group (496.7 ng/ml, 0.43) and in healthy control group (432.3 ng/ml, 0.42) (all P<0.01, respectively). The Alu115 and Alu247/115 index of serum DNA in benign esophageal disease group were no statistically different from those in the healthy control group (all P>0.05, respectively). The levels of cfDNA and its integrity were not significantly correlated with age, gender, tumor differentiation, or disease stage according to American Joint Committee on Cancer (AJCC) staging system in the esophageal cancer group (all P>0.05). The serum Alu247/115 index of Stage Ⅲ patients was higher than that of Stage Ⅰ~Ⅱ patients(P<0.05). The serum Alu247/115 index of Stage Ⅳ was higher than that of Stage Ⅲ(P<0.05). In the esophageal cancer group, both of serum Alu115 and Alu247/115 index had no correlation with CEA or SCC (all P>0.05). The area under the ROC curve (AUC) of Alu115 and Alu247/115 index were 0.867 and 0.854, respectively, which were both higher than that of CEA (0.622) and SCC (0.753). The addition of Alu115 or Alu247/115 index to CEA and SCC detection increased the sensitivity of the diagnosis of esophageal cancer by 95.6% and 94.1%, respectively.@*Conclusions@#The detection of serum cfDNA concentration and integrity is helpful to the early diagnosis and monitoring of esophageal cancer. Their diagnostic value of esophageal cancer is better than that of the traditional tumor markers CEA and SCC.

ABSTRACT

Objective To evaluate the expressions and its clinical significance of Foxj2 in non-small cell lung cancer (NSCLC) patients.Methods Fifty cancer tissues from patients who underwentsurgery were collected.Western blot and immunohisto-chemical methods were used to detect the expressions of Foxj2.Results Results of western blot showed that Foxj2 expres-sion in lung cancer tissue was significantly lower than adjacent tissues.The difference between the two groups was statisti-cally significant(P<0.05).Immunohistoehemical results showed that the number of Foxj2-positive cells in adjacent tissues was significantly higher than that in cancer tissues,and the expression of Foxj2 in non-lymphatic node metastasis group was higher than that in lymphatic node metastasis.The difference between the two groups was statistically significant (P<0.01).Conclusion The expressions and location of Foxj2 was related to the lymphatic node metastasis of lung cancer.This phenomenon indicates that Foxj2 may play an important role in the development of lung cancer.

ABSTRACT

Serum B7-H4 protein levels were measured by enzyme-link immunosorbent assay (ELISA) in 62 patients with epithelial ovarian cancer, 20 patients with benign ovarian diseases, and 30healthy subjects.Serum B7-H4 protein levels in epithelial ovarian cancer, benign ovarian diseases and normal controls were 0.72(0.32,1.88 )μg/L, (0.45±0.21)μg/L,(0.38 ± 0.19 ) μg/L respectively (P<0.01).The serum level of B7-H4 protein in epithelial ovarian carcinoma was correlated with FIGO stages, lymph node metastasis, histological grade and histological types ( all P < 0.01 ), but not with menopause and ascites (P>0.05).The receiver operating characteristics curve analysis revealed that B7-H4protein had a sensitivity and specificity of 69% (43/62)and 86%(43/50) for diagnosis of epithelial ovarian cancer.

ABSTRACT

Objective To investigate the change and clinical significance of the serum level of human epididymis secretory protein 4 (HE4) in elderly patients with epithelial ovarian cancer (EOC).Methods The serum levels of HE4 were determined by ELISA in EOC patients (n= 33), benign ovarian disease patients ( n = 17) and healthy women ( n = 20), all of them aged 60 years or over. And there were other 31 EOC patients and 20 healthy women aged less than 60 years at the same period as controls. Results In healthy people, the serum level of HE4 was higher in the group with age ≥60 years than with age <60 years [(32.25± 13. 15) pmol/L vs. (24.59±8.60) pmol/L]. The serum HE4 level had a positive correlation with age (r=0. 525, P<0.01). The median level of HE4 was significantly higher in elderly EOC patients than in benign ovarian disease patients (81.50 pmol/L vs.45.60 pmol L, U=168.5, P<0.05) and healthy women (81.50 pmol L vs. 33.30 pmol L, U=76.5, P<0. 01). The HE4 had a significantly higher area under ROC curve when compared with CA125 (0. 799 vs. 0. 782), while the cut-off value was set 65.43 pmol L with a sensibility and specificity of 66.67% and 86.49% respectively. The diagnostic efficacy of HE4 was higher than CA125. There was no significantly statistical difference between elderly and non-elderly EOC patients in serum HE4 levels (U= 101.2, P>0. 05). Conclusions Age is an important factor which can affect human serum level of HE4. HE4 determination is helpful in the diagnosis of ovarian epithelial carcinoma in the elderly patients, and the combined detection of HE4 and CA125 is an useful adjuvant diagnostic measure.

Objective To observe the expression levels of CD174, cyclin D1 and cyclin E in gastrointestinal tumor tissues, analyze the correlation among CD174, cyclin D1 and cyclin E, and evaluate the expression level of CD174 for diagnosis and prognosis of gastrointestinal tumors.Methods Flow cytometry was used to determine the expression of CD174, cyclin D1 and cyclin E on surface of the cells which were collected from the tumor tissues and distant part beside the same tumor and suspended.
