ABSTRACT

Objective To study the effects of 3,5-diiodotyrosine (DIT) and potassium iodide (KI) on myocardial ATPase activity in hyperthyroidism Wistar rats induced by thyroid tablets.Methods Seventy-two Wistar rats were divided into 8 groups according to body weight by the random number table method (9 rats in each group),respectively,which were control group,hyperthyroidism model group,low,medium and high doses groups (both DIT and KI contents were 25.0,166.7,500.1 μg/kg).Physiological saline was intragastrically administrated to the control group;the hyperthyroidism model group was given thyroid tablet suspension (200.0 mg/kg);DIT and KI groups were given thyroid tablet suspension with corresponding doses of iodine simultaneously.The medicine was given once a day for a mouth,all the rats were sacrificed and heart tissue was collected.The colorimetric method was used to examine the activity of ATPases (Na+-K+-ATPase,Mg2+-ATPase,Ca2+-ATPase).Results The activities of Na+-K+-ATPase,Mg2+-ATPase,Ca2+-ATPase were significantly different statistically between groups (F =2.99,3.03,6.18,all P < 0.01).Compared with the control group [(4.01 ± 0.22),(4.28 ± 0.28),(4.46 ± 0.35) μmol/mg·h],the activities of ATPases (Na+-K+-ATPase,Mg2+-ATPase,Ca2+-ATPase included) were reduced significantly in hyperthyroidism model group [(3.60 ± 0.25),(3.42 ± 0.31),(3.85 ± 0.17)μ mol/mg·h,all P < 0.01];the activities of Mg2+-ATPase in DIT medium dose group [(3.89 ± 0.35)μmol/mg ·h],Ca2+-ATPase in DIT medium and high doses groups [(4.12 ± 0.20),(4.09 ± 0.21)μ mol/mg·h] were reduced significantly (all P < 0.05);the activities of Na+-K+-ATPases,Ca2+-ATPase were decreased significantly in three KI groups [(3.64 ± 0.32),(3.60 ± 0.32),(3.53 ± 0.33),(3.93 ± 0.22),(3.90 ± 0.23),(3.85 ± 0.26)μmol/mg·h],Mg2+-ATPase in KI high dose group [(3.65 ± 0.49)μmol/mg·h] was decreased significantly (P < 0.05or < 0.01).Compared with the hyperthyroidism model group,the activities of ATPase were increased in most of the DIT groups [Mg2+-ATPase in low,medium doses groups:(4.06 ± 0.51),(3.89 ± 0.35)μmol/mg·h;Ca2+-ATPase in low,medium,high doses groups (4.15 ± 0.26),(4.12 ± 0.20),(4.09 ± 0.21)μmol/mg·h,all P < 0.05].Conclusion Supplementation of thyroid tablets in the process of hyperthyroidism formation in Wistar rats will reduce myocardial damage by DTT compared with the same dose of KI.

ABSTRACT

Objective To establish an animal model of high-iodine and low-protein in Wistar rats,and to observe the effect of combined excess-iodine and low-protein diet on growth,metabolism and morphological changes in thyroid.Methods According to body weight[(110 ± 10)g] and sex(half male and half female),one hundred and ninety-two Wistar rats,1 month after weaning,were randomly divided into ① normal iodine control group (NI),② 10-fold excess-iodine group (10HI),③ 50-fold excess-iodine group (50HI),④ 100-fold excess-iodine group (100HI),⑤ low-protein control group (LC),⑥ low-protein and l 0-fold excess-iodine group (L10HI),⑦low-protein and 50-fold excess-iodine group (L50HI),⑧ low-protein and 100-fold excess-iodine group(L100HI).Twenty-four rats were in each group,with the experimental period of 6 months.The iodine content of NI and LC groups was 4.65 μg/d; 10HI,50HI and 100HI groups were 46.50,232.50 and 465.00 μg/d,respectively.The animal's body weight,water and feed consumption were recorded weekly.At the end of 60,120,180 days,urine and blood samples were collected from eight rats in each group.Urinary iodine was tested by arseni cerium catalytic spectrophotometry; serum iodine was tested by the method of chloric acid.Histological change of the thyroid gland was observed by transmission electron microscopy and hematoxylin-eosin (HE) staining at the end of 6 months; apoptosis of thyroid was tested by terminal-deoxynucleoitidyl transferase mediated nick end labeling (TUNEL) method.Results At the end of 4,8,16,18,22 and 24 weeks,the differences of body mass of rats among groups were statistically significant(F =4.26,3.75,4.98,4.09,3.28,3.95,all P < 0.05).At the end of 60,120,180 days,the differences of iodine concentration in urine and blood among groups were statistically significantly (H =5.37,6.03,all P < 0.05).Light microscopy showed that thyroid follicular epithelial cells became flattened,and follicles became distended with colloid following increasing of iodine concentration.Electron microscopy showed increased glial vesicles,condensation of nuclear chromatin,karyopyknosis,and karyolysis with increasing of iodine concentration.The differences of apoptotic indexes among groups were statistically significant (F =4.59,P < 0.01).The apoptotic indexes of L50HI and L100HI groups [(21.50 ± 5.20)‰,(26.70 ± 6.40)‰] were higher than those of 50HI and 100HI groups [(11.20 ± 4.30)‰,(19.40 ± 4.80)‰,P < 0.01 or < 0.05].Conclusion Excessiodine and low-protein can cause growth retardation,abnormal iodine metabolism,and thyroid follicular epithelium damage in Wistar rats.

ABSTRACT

Objective To study the effects of overdose fluoride on blood lipid and hemorheology in rabbits and the protective effect of selenium. Methods Twenty male rabbits were divided into 4 groups: normal group,fluorine group(100 mg/L),selenium group(1 mg/L) and selenium (1 mg/L) plus fluorine (100 mg/L) group. After 6 months of treatment,whole blood rheology,blood serum TC,TG,HDL-C,LDL-C,SOD activity,GSH-Px activity,the content of MDA were determined. Results The whole blood rheology of rabbit in fluorine group was abnormal. Compared with the normal group,significant increase of blood serum TC,HDL-C,LDL-C was seen,decrease of SOD activity and GSH-Px activity and increase of the content of MDA in fluorine group was found. The results of selenium plus fluorine group was more close to the normal group compared with fluorine group. Conclusion Fluorosis can induce hemorheology and lipid metabolism disorder and decrease the capacity of antioxidase and moderate selenium intake may have some protective effect.

ABSTRACT

Objective To study the protective effect of garlicin on radiation injury in mice. Methods Sixty mice were randomly divided into five groups:model control group, solvent control group and garlicin treatment groups[0.17, 0.33, 1.00 g/(kg?d)]. After treated by gavage for 20 consecutive days, the mice were exposed to 60Co-? irradiation for one time and the effect of the irradiation on the amounts of WBC in peripheral blood, the ratio of micronucleus cells in marrow, the content of DNA in marrow cells and the superoxide dismutase (SOD) activity were measured. Results Compared with the exposure group, garlicin increased the amounts of WBC, decreased the micronucleus frequency of marrow cells, increased the content of DNA in marrow cells and increased the SOD activity in mice. Conclusion Garlicin shows a protective effect on irradiation injury in mice.

ABSTRACT

Objective To research the growth of human umbilical vein endothelial cell(HUECV) cultured with different dose of selenium. Methods HUVEC were cultured with selenium in various concentration (50、100、500、1000、2000 nmol/L) for 48 h in vitro. The morphological changes of cells were observed, activity of cell was measured by MTT method, and at the same time the levels of SOD, GSH-Px, MDA, NO, iNOS, eNOS in the culture were determined. Results At the doses of 1 000-2 000 nmol/L, the cells were damaged and revealed related morphological change.
